Multiplex Polymerase Chain Reaction(PCR)법을 이용한 Staphylococcus aureus, Salmonella enterica subsp., Vibrio parahaemolyticus의 다중동시검출 (Simultaneous Detection of Staphylococcus aureus, Salmonella enterica subsp., Vibrio parahaemolyticus by Multiplex Polymerase Chain Reaction)

  • 본 연구는 국내 주요 식중독 원인균인 Staphylococcus aureus, Salmonella enterica subsp., Vibrio parahaemolyticus를 동시에 검출 및 동정할 수 있는 simultaneous multiplex PCR방법을 개발하고자 하였다. S. aureus의 23s rRNA 유전자(482 bp), V. Parahaemolyticus의 toxR 유전자(368 bp), S. enterica subsp.의 invA 유전자(284 bp)를 특이적으로 검출 및 동정할 수 있는 3개 primer set 즉, STA-5F/STA-5R, ToxR-F/ToxR-R, 139/141을 구축하였으며, 그 결과 정제되어진 각 식중독 원인균의 genomic DNA를 template로 하여 세 균주 모두 10 pg까지 다중동시검출이 가능하였다. 생균수(CFU)와 상응되는 검출한계 결과로써 $10^1\sim10^2$ CFU/reaction의 검출한계를 보였으며 이는 즉, S. aureus $6.0\times10^4$ CFU/mL, S. enterica subsp. $9.5\times10^4$ CFU/mL, V. parahaemolyticus $6.1\times10^5$ CFU/mL의 검출한계를 나타내었다. 균체회수부터 agarose gel 상에서 검출 및 동정까지 3~4 hr의 시간 소요로 single tube 반응으로 세 식중독 원인균의 다중동시검출이 가능하였다. 또한 추가적인 연구를 통하여 세 식중독 원인균주의 검출을 위한 향상된 민감도를 가지는 multiplex PCR법 및 real time PCR을 이용한 다중동시검출법 개발을 위한 기초자료로서 활용 가능할 것이라 사료된다.

A SM2 Elliptic Curve Threshold Signature Scheme without a Trusted Center

  • Threshold signature is very important in identity authentication and some other applications. In December 2010, Chinese Encryption Administration released the SM2 elliptic curve digital signature algorithm as the first standard of the digital signature algorithm in China. At present, the papers on the threshold signature scheme based on this algorithm are few. A SM2 elliptic curve threshold signature scheme without a trusted center is proposed according to the Joint-Shamir-RSS algorithm, the Joint-Shamir-ZSS algorithm, the sum or diff-SS algorithm, the Mul-SS algorithm, the Inv-SS algorithm and the PM-SS algorithm. The proposed scheme is analyzed from correctness, security and efficiency. The correctness analysis shows that the proposed scheme can realize the effective threshold signature. The security analysis shows that the proposed scheme can resist some kinds of common attacks. The efficiency analysis shows that if the same secret sharing algorithms are used to design the threshold signature schemes, the SM2 elliptic curve threshold signature scheme will be more efficient than the threshold signature scheme based on ECDSA.

Establishment and Application of Polymerase Spiral Reaction Amplification for Salmonella Detection in Food

  • Salmonella is a common zoonotic and foodborne pathogen that causes high morbidity and mortality in developing countries. In this study, we established and validated a polymerase spiral reaction (PSR) assay which targeted the conserved invasion gene (invA) of Salmonella by SYBR Green I indicator methods. Subsequently, assays for determination of the optimal conditions for optimal specificity and sensitivity of PSR were performed. We performed comprehensive evaluations using loop-mediated isothermal amplification (LAMP) and real-time PCR. A total number of 532 samples of daily food were analyzed by PSR. Twenty-seven bacterial strains were tested in the specificity assay, from which positive results were obtained only for 14-Salmonella strains. However, none of the 13 non-Salmonella strains was amplified. Similarly with LAMP and real-time PCR, the detection limit of the PSR assay was 50 CFU/ml. The PSR method was also successfully applied to evaluate the contamination with Salmonella in 532 samples of daily food, corroborating traditional culture method data. The novel PSR method is simple, sensitive, and rapid and provides new insights into the prevention and detection of foodborne diseases.

Device Coupling Effects of Monolithic 3D Inverters

  • The device coupling between the stacked top/bottom field-effect transistors (FETs) in two types of monolithic 3D inverter (M3INV) with/without a metal layer in the bottom tier is investigated, and then the regime of the thickness TILD and dielectric constant εr of the inter-layer distance (ILD), the doping concentration Nd (Na), and length Lg of the channel, and the side-wall length LSW where the stacked FETs are coupled are studied. When Nd (Na) < 1016 cm-3 and LSW < 20 nm, the threshold voltage shift of the top FET varies almost constantly by the gate voltage of the bottom FET, but when Nd (Na) > 1016 cm-3 or LSW > 20 nm, the shift decreases and increases, respectively. M3INVs with TILD ≥ 50 nm and εr ≤ 3.9 can neglect the interaction between the stacked FETs, but when TILD or εr do not meet the above conditions, the interaction must be taken into consideration.

Simultaneous Detection of Listeria monocytogenes, Escherichia coli O157:H7, Bacillus cereus, Salmonella spp., and Staphylococcus aureus in Low-fatted Milk by Multiplex PCR

  • A rapid and specific PCR assay for the simultaneous detection of Listeria monocytogenes, Escherichia coli O157:H7, Bacillus cereus, Salmonella spp., and Staphylococcus aureus in foods was developed to reduce the detection time and to increase sensitivity. Multiplex PCR developed in this study produced only actA, fliC, hbl, invA, ileS amplicons, but did not produce any non-specific amplicon. The primer sets successfully amplified the target genes in the multiplex PCR without any non-specific or additional bands on the other strains. The multiplex PCR assays also amplified some target genes from five pathogens, and multiplex amplification was obtained from as little as 1 pg of DNA. According to the results from the sensitivity evaluation, the multiplex PCR developed in this study detected 10 cells/mL of the pathogens inoculated in milk samples, respectively. The results suggested that multiplex PCR was an effective assay demonstrating high specificity for the simultaneous detection of five target pathogens in food system.

Semi-active control of smart building-MR damper systems using novel TSK-Inv and max-min algorithms

  • Two novel semi-active control methods for a seismically excited nonlinear benchmark building equipped with magnetorheological dampers are presented and evaluated in this paper. While a primary controller is designed to estimate the optimal control force of a magnetorheological (MR) damper, the required voltage input for the damper to produce such desired control force is achieved using two different methods. The first technique uses an optimal compact Takagi-Sugeno-Kang (TSK) fuzzy inverse model of MR damper to predict the required voltage to actuate the MR dampers (TSKFInv). The other voltage regulator introduced here works based on the maximum and minimum capacities of MR damper at each time-step (MaxMin). Both semi-active algorithms developed here, use acceleration feedback only. The results demonstrate that both TSKFInv and MaxMin algorithms are quite effective in seismic response reduction for wide range of motions from moderate to severe seismic events, compared with the passive systems and performs better than original and Modified clipped optimal controller systems, known as COC and MCOC.

수면파형의 독립성분분석 (Independent Component Analysis(ICA) of Sleep Waves)

  • Independent Component Analysis (ICA) is a blind source separation method using unsupervised learning and mutual information theory created in the late eighties and developed in the nineties. It has already succeeded in separating eye movement artifacts from human scalp EEG recording. Several characteristic sleep waves such as sleep spindle, K-complex, and positive occipital sharp transient of sleep (POSTS) can be recorded during sleep EEG recording. They are used as stage determining factors of sleep staging and might be reflections of unknown neural sources during sleep. We applied the ICA method to sleep EEG for sleep waves separation. Eighteen channel scalp longitudinal bipolar montage was used for the EEG recording. With the sampling rate of 256Hz, digital EEG data were converted into 18 by n matrix which was used as a original data matrix X. Independent source matrix U (18 by n) was obtained by independent component analysis method ($U=W{\timex}X$, where W is an 18 by 18 matrix obtained by ICA procedures). ICA was applied to the original EEG containing sleep spindle, K-complex, and POSTS. Among the 18 independent components, those containing characteristic shape of sleep waves could be identified. Each independent component was reconstructed into original montage by the product of inverse matrix of W (inv(W)) and U. The reconstructed EEG might be a separation of sleep waves without other components of original EEG matrix X. This result (might) demonstrates that characteristic sleep waves may be separated from original EEG of unknown mixed neural origins by the Independent Component Analysis (ICA) method.

Familial Chromosome No. 9 Pericentic Inversion Producing Scimitar Syndrome

  • Scimitar syndrome is a rare congenital anomaly that is characterized by hypoplasia of the right lung and the right pulmonary artery with anomalous pulmonary venous drainage to the inferior vena cava. The scimitar vein is usually visible on chest radiographs, but may be obscured by the heart. It is essential for surgical correction to establish the point of drainage of the anomalous vein and associated anomalies. There are recent reports of familial total anomalous pulmonary venous return suggesting heritable forms of this anomaly. Although genetic factors are believed to have important roles in congenital heart disease, few genes involved in heart development have been located. We report a case of familial chromosome 9 inversion with Scimitar syndrome in an offspring who presented with dextrocardia. Evaluation with magnetic resonance cineangiograph imaging demonstrated an anomalous pulmonary vein draining into the inferior vena cava above the diaphragm and hypoplasia of the right lung and the right pulmonary artery. Chromsome analysis showed pericentric inversion of chromosome 9, inv 9 (p13, q21), in the patient and his mother as well. A brief review of the related literature is also included.

Investigation into Electrical Characteristics of Logic Circuit Consisting of Modularized Monolithic 3D Inverter Unit Cell

  • Monolithic three-dimensional (M3D) logics such as M3D-NAND, M3D-NOR, M3D-buffer, M3D 2×1 multiplexer, and M3D D flip-flop, consisting of modularized M3D inverters (M3D-INVs), have been proposed. In the previous M3D logic, each M3D logic had to be designed separately for a standard cell library. The proposed M3D logic is designed by placing modularized M3D-INVs and connecting interconnects such as metal lines or monolithic inter-tier-vias between M3D-INVs. The electrical characteristics of the previous and proposed M3D logics were simulated using the technology computer-aided design and Simulation Program with Integrated Circuit Emphasis with the extracted parameters of the previously developed LETI-UTSOI MOSFET model for n- and p-type MOSFETs and the extracted external capacitances. The area, propagation delay, falling/rising times, and dynamic power consumption of the proposed M3D logic are lower than those of previous versions. Despite the larger space and lower performance of the proposed M3D logic in comparison to the previous versions, it can be easily designed with a single modularized M3D-INV and without having to design all layouts of the logic gates separately.

A case of de novo duplication of 15q24-q26.3

  • Distal duplication, or trisomy 15q, is an extremely rare chromosomal disorder characterized by prenatal and postnatal overgrowth, mental retardation, and craniofacial malformations. Additional abnormalities typically include an unusually short neck, malformations of the fingers and toes, scoliosis and skeletal malformations, genital abnormalities, particularly in affected males, and, in some cases, cardiac defects. The range and severity of symptoms and physical findings may vary from case to case, depending upon the length and location of the duplicated portion of chromosome 15q. Most reported cases of duplication of the long arm of chromosome 15 frequently have more than one segmental imbalance resulting from unbalanced translocations involving chromosome 15 and deletions in another chromosome, as well as other structural chromosomal abnormalities. We report a female newborn with a de novo duplication, 15q24- q26.3, showing intrauterine overgrowth, a narrow asymmetric face with down-slanting palpebral fissures, a large, prominent nose, and micrognathia, arachnodactyly, camptodactyly, congenital heart disease, hydronephrosis, and hydroureter. Chromosomal analysis showed a 46,XX,inv(9)(p12q13),dup(15)(q24q26.3). Array comparative genomic hybridization analysis revealed a gain of 42 clones on 15q24-q26.3. This case represents the only reported patient with a de novo 15q24-q26.3 duplication that did not result from an unbalanced translocation and did not have a concomitant monosomic component in Korea.
