Genetic Stock Identification of Common Carp (Cyprinus carpio) by Detection of Intraspecific DNA Sequence Variation in the Mitochondrial 12S rRNA Gene (미토콘드리아 12S rRNA 유전자 변이 조사를 통한 잉어(Cyprinus carpio)의 유전학적 동정)

  • Intraspecific sequence variation was detected by polymerase chain reaction (PCR) and direct sequencing of a 350-nucleotide region of the mitochondrial 12S rRNA gene of two natural populations (Han River and Nakdong River) and one hatchery stock (Jinhae Inland Fisheries Institute) of local strain common carp, one Israeli strain of common carp stock from Pukyong National University (PKU), and one hybrid between Israeli strain of common carp female and local strain common carp male from PKU stock. There is little variation in 350 bases of the mitochondrial 12S rRNA gene sequences among 2 natural and 1 hatchery local strain common carp populatins, representing abut 7 to 20 nucleotide differences (less than 6%). The sequence of specimens from Han River was more similar to that from Nakdong River (identity=98.0%) than to that from Jinhae Inland Fisheries Institute (identity=96.3%). Sequence variation between Israeli strain and wild local strain common carp was higher than the variation within natural stocks. The level of variation was ranged from 15.7 to 17.7%. The hybrid showed very similar nucleotide4 sequence of 12S rRNA gene to the sequence of Israeli strain with the identity of 98.9%.

  • Intraspecific fusants, produced by protoplast fusion of auxotrophic mutants from Trichoderma koningii ATCC 26113, were segregated into various strains including parental types, non-parental auxotrophic hybrids, and prototrophic hybrids on complete plate. Interestingly, some of non-parental prototrophic hybrids revealed to have enhanced cellulolytic activity incomparison with other strains of parents or hybrids derived thereafter. It was also evident that prototrophic hybrids of aneuploid could be constructed after the spontaneous segregation of complementing fusants produced through the protoplast fusion.

  • The nuclear transfer technique was employed to obtain intraspecific hybrids in Aspergillus nidulans. Nuclei isolated from either a wild type or an auxotrophic mutant strain (FGSC 475) were transferred into the protoplasts of a recipient strain (FGSC 514). The frequency of hybrid formation (4.8% and 10.1 %, respectively) by nuclear transfer was higher than the frequency (0.6%) by protoplast fusion. Furthermore, most of the hybrids formed showed increased activity of some components of cellulase system, xylanase system, and mannanase. The hybrids were analyzed to be either diploid or aneuploid. These results suggest that nuclear transfer technique is more efficient the formation of intraspecific hybrids than protoplast fusion method and is useful for the improvement of Aspergillus strains.

  • Antithamnion sparsum Tokida isolated from the southern and western coasts of Korea was investigated in culture, comparing the morphological character with A. defectum Kylin from the Pacific North America. A. sparsum basically showed a Polysiphonia-type life history. However, it sometimes exhibited a monoecious reproducton and the carpospores released from the cystocarp by self-fertilization unexpectedly developed into the plants bearing spermatangia alone. These male plants were not functional up to 60 days culture. The results of intraspecific crosses between populations of A. sparsum were positive and the hybrid carpospores gave rise to normal tetrasporophytes. On the other hand, theinterspecific crosses between A. sparsum and A. defectum were successful partly, evidenced by the gonimoblast development and the release of carpospores in case of A. sparsum (male)$\times$A. defectum (female), but not in case of A. sparsum (female)$\times$A. defectum (male). Thus, the both species were still under the speciation.

Interspecific protoplast fusion of trichoderma koningii and trichoderma reesei (Trichoderma koningii와 trichoderma reesei 원형질체 융합)

  • Intra and interspecfic fusants were produced by the protoplast fusion of auxotrophic mutants from Trichoderma koningii ATCC 26113 and Trichoderma reesei QM 9414. It was found that 0.6M $MgSO_4\;and\;0.6M\;NH_4Cl$ was the best osmotic stabilizer for the preparation of protoplasts from the mycelium of T. koningii and T. reesei respectively. However, $MgSO_4$ was the most suitable one for the regeneration of the protoplasts from both species. The intraspecific protoplast fusion frequencies between the auxotrophic mutants from T. reesei were $1.8{\times}10^{-2}\;to\;5.1{\times}10^{-1}$. Interspecific protoplast fusion frequencies between the auxotrophic mutants from T. koningii and T. reesei were $3.6{\times}10^{-3}$\;to\;8.4{\times}10^{-2}. Interspecific complementing fusants, however, were not alwats produced. Fusants obtained from interspecific potoplast fusion were spontaneously segregated into various strains including parental types, non-parental auxotrophic hybrids, and prototrophic hybrids on complete plate. Interspecific hybrids revealed to have partially enhanced celluloytic activities.

Intraspecific Variations of Rubus oldhami (덩굴딸기(Rubus oldhami)의 종내변이)

  • Taxonomic studies on morphological, principal component analysis (PCA), palynological, RAPD and PCR-RELP analysis were conducted to intraspecific relationships of Rubus oldhami. Three types of Rubus oldhami based on the flower characters such as petal length and number were used in this study. Among the 14 morphological characters, perianth length, calyx lobe length, apical leaflet shape and leaflet length were used to distinguish for each type. The pollen characters such as shape, aperture number, surface sculpture were showed very similar among three types. Eight primers out of 20 arbitrary primers were screened for three types, and were revealed 33 ($60\%$) polymorphic bands. The phonogram by RAPD data showed incongruent with morphological analysis. Even though ten restriction endonucleases produced 20 restriction sites, polymorphic bands were not observed. Based on the results, three types of Rubus oldhami divided well by morphological characters, but pollen and DNA data were not supported. Therefore, type 1 and 2 which different from type 3 by flower characters considered as a temporary hybrid or ecotype because of their similar habitats.

Total Ammonia Nitrogen Excretion Rates and Feces Production Rates as an Index for Comparing Efficiency of Dietary Protein Utilization of Offsprings from Selected Korean Strain, Cultured Japanese Strain and Their Intraspecific Hybrid Strain of Juvenile Red Sea Bream, Pagrus major (한국산 선발 계통, 일본산 양식 계통 그리고 이들 두 계통간 잡종 계통 참돔 치어의 총 암모니아성 질소 배설률 및 분 배출률을 통한 사료내 단백질 이용 효율 비교)

  • An experiment was conducted to investigate the differences of total ammonia nitrogen (TAN) excretion rates and feces production rates among the offsprings from cultured Japanese strain (JJ, mean BW; $17.1{\pm}0.1g$), intraspecific hybrid strain between cultured Japanese and selected Korean strain (JK, mean BW: $17.1{\pm}0.1g$) and selected Korean strain(KK, mean BW: $21.5{\pm}0.1g$) of red sea bream in order to compare their dietary protein utilization efficiency. Fish were hand-fed with a commercial diet containing 46.7% crude protein for 2 weeks, three times daily 09:00, 13:00 and 17:00. After daily feeding, the TAN excretion rates reached peaks of 49.03, 58.75 and 36.26mg/kg fish/hr for the JJ, JK and KK strain, respectively, during the daytime. The value of the KK strain was significantly lower than that of the JJ and JX shuin (P<0.05), however daily TAN excretion rates of the JJ, JK and KK strain were not different (P>0.05). When fish were fed at satiation after 4 days of starvation, TAN excretion rates reached the maximum values 4 hours after the feeding fur the KK (31.23 mg/kg fish/hr) and 6 hours after the feeding fur the JJ (44.19 mg/tg fish/hr) and JK strain (41.70 mg/kg fish/hr). After 3 days of starvation, the daily endogenous TAU excretion rates (ETE) for the JJ, JK. and KK strain were 286.91, 215.66 and 179.29mg/kg fish/day, respectively. The value of the KK strain was lower than that of the JJ and JK strain (P<0.05). The total feces production rates of the JJ, JK and KK strain were not significantly different, however the proportions of feces production rates by time for the JJ, JK and KK strain were different (P<0.05). As overall results, efficiency of dietary protein utilization of JJ, JK and KK seems to be different and KK strain could offer a desirable option for aquaculture purpose.

Effect of Water Temperature and Photoperiod on the Oxygen Consumption of Four Different Strains of Red Seabream, Pagrus major

  • We determined the effects of different water temperatures (15, 20, and 25℃) and photoperiod cycles (24L:0D, 12L:12D, and 0L:24D) on the oxygen consumption of the offspring of a cultured Japanese strain (JJ), a selected Korean strain (KK), and intraspecific hybrid strains (JK and KJ) of red seabream, Pagrus major, under starvation conditions. The different fish strains, water temperatures, and photoperiod cycles had effects on the mean oxygen consumption of fish. Oxygen consumption increased with increasing water temperatures for all photoperiod treatments (p<0.001). Fish held in continuous darkness (0L:24D) used consistently less oxygen than fish exposed to continuous light (p<0.05). The oxygen consumption of fish exposed to the light phase in a 12L:12D photoperiod was higher than that of fish in the dark phase of the 12L:12D cycle, and differences were significant in three of the strains: JJ (15℃), KK (15 and 20℃), and KJ (25℃). The oxygen consumption of the inbred (JJ and KK) and intraspecific hybrid (JK and KJ) strains varied with differing water temperatures and photoperiod cycles. The JK strain displayed significantly higher oxygen consumption than the other strains under all experimental conditions except 15℃ with a 0L:24D photoperiod. The JK and KJ strains usually showed the highest and lowest oxygen consumption values, respectively, whereas the inbred strains exhibited intermediate values. Oxygen consumption in the JJ and JK strains was usually higher than that of the KK and KJ strains. We propose that differences in the thermal sensitivity and photosensitization properties of the strains contribute to differences in their ability to adapt to changes in water temperature and photoperiod, thus resulting in differences in the amplitude of their metabolic rates.

Nuclear DNA inheritance of intraspecific somatic hybrids by mono-mono cross in Pleurotus ostreatus based on URP-PCR analysis (URP-PCR 분석에 의한 느타리 단핵 계통간 교잡주의 핵 DNA 유전)

  • The primary objective of the present study is the characterization of the hybrids of monokaryon- monokaryon (mono-mono) crosses in mushroom breeding. We employed this technique for developing superior strains from Pleurotus species strains with 85 mono-mono intraspecific hybrids of 7 combinations between six Pleurotus ostreatus strains and one Pleurotus florida strain. In this study, the results of analysis on hybridization rate, nuclear DNA patterns, and colors and yields of fruit-bodies, are presented as follows. The crossability between mono-mono crossing ranges between 50 and 93.75%. The results of the analysis on the nuclear DNA patterns of 85 hybrid strains of mono-mono crosses share the nuclei of both parents, but their genetic similarities were predominated by either parent. The hybrid strain between P. florida and P. ostreatus showed patterns more similar to P. florida, while the hybrid strain between P. ostreatus and P. ostreatus either had patterns predominated by either parent strain. The fruiting body colors of the mono-mono crosses mostly had combined colors of both parents but showed the tendency of being more similar to that of either parent. 82% of the hybrid strain indicated similar fruiting body yields compared to both parent strains, while 0% was higher and 18% were lower than both parents. The present study was able to find out and suggest superior hybrid trains by identifying the nuclear DNA patterns of hybrids between Pleurotus species as well as the characteristics of their fruiting bodies. This study expects that the advantages of the mono-mono crossing are needs to be fully utilized in mushroom breeding and it is better to develop superior strains of Pleurotus species strains together with the mono-mono crossing.

Forage Yield and Quality of Summer Grain Legumes and Forage Grasses in Cheju Island

  • Soybean [Glycine max (L.) Merr.), mungbean [Vigna radiata (L.) Wilcz.], cowpea [V. unguiculata (L.) Walp.], adzuki bean [V. angularis (Willd.) Ohwi & Ohashi], maize [Zea mays L.], sorghum [Sorghum bicolor (L.) Moench], sorghum $\times$ sudangrass [So bicolor intraspecific hybrid], and Japanese millet [Echinochloa crusgalli var. frumentacea (Link) W.F. Wight] were grown at two planting dates (18 June and 15 July) at Cheju in 1997 to select the best forage legumes adapted to Cheju Island for grass-legume forage rotation. Averaged across planting dates and cultivars, dry matter (DM), crude protein (CP), and total digestible nutrient (TDN) yields were 5,646, 1,056, and 3,637 kg/ha for soybean, 4,458, 676, and 2,661 kg/ha for mungbean, 3,289, 553, and 2,055 kg/ha for cowpea, 3,931, 674, and 2,489 kg/ha for adzuki bean, 12,695, 969, and 7,642 kg/ha for maize, 17,071, 1,260, and 8,857 kg/ha for sorghum, 16,355, 1,163, and 8,543 kg/ha for sorghum $\times$ sudangrass hybrid, and 8,288, 929, and 4,091 kg/ha for Japanese millet. Soybean was higher in CP, ether extract (EE), and TON content but was lower in nitrogen free extract content compared with the three other legumes. The legumes had much higher CP (13.7 to 21.9%), EE (2.42 to 6.23%), and TDN (58.7 to 69.9%) content but lower in crude fiber (CF) content (17.3 to 25.3%) than did the grasses tested except maize which had relatively lower CF content but higher TDN content. These results suggest that soybean could be the best forage legume for grass-legume forage rotation in the Cheju region.
