Avian adenovirus 관련 심낭수종-봉입체간염 증후군 진단에 관한 연구 (Diagnosis of avian adenovirus-associated hydropericardium hepatitis syndrome)

  • Avian adenoviruses are diverse group of pathogens and recently hydropericardium hepatitis syndrome (HHS) Is an important, emerged disease of poultry. Particularly 2-3 weeks old age broilers increased mortality ranging from 20-30% and brown native chicken 3-7 weeks sudden onset with mortality 20-50%, typically development secondary infection. The infection chicken shows liver enlarged, pale and straw- colored fluid is present in the sac surrounding the heart. Histopathological positive samples have necrotic foci and basophilic intranuclear inclusion bodies in the hepatocytes and polymerase chain reaction (PCR) was useful for detect the fowl adenovirus (FAV) associated with HHS.

Pathological findings of the mixed infection with canine distemper virus and Streptococcus canis on farmed badger

  • Herein, we report a case of badgers showing high morbidity and mortality rate due to the mixed infection of canine distemper virus (CDV) and Streptococcus canis (S. canis) in a farm where wild animal, badger, is being reared for herbal medicine. During the period of about one month, 120 out of 320 badgers showed severe respiratory symptoms and died, and 3 bodies were submitted to the Animal and Plant Quarantine Agency for disease diagnosis. The lung with the most severe necropsy findings failed to collapse and showed dark reddening and had yellowish nodules on the cut surface. The characteristic and common histopathologic findings include multifocal necrosis with hemorrhage of the lung, severe lymphoid depletion of the spleen and intracytoplasmic or intranuclear inclusion bodies in almost all organs. Finally, CDV and S. canis were identified by immunohistochemistry and bacterial isolation, respectively. This is the first mixed infection case of CDV and S. canis in badgers being raised on the farm.

  • Aujeszky's disease virus(ADV) was inoculated intranasally into the Korean native goats to investigate pathological findings and pathogenesis of ADV infection by using of histological and immunohistochemical methods and in situ hybridization(ISH). Clinical signs of salvation, pyrexia, pruritus and staggering were followed by death with five days after inoculation, Pathoanatomical findings were edema of the lung and the urinary bladder with hemorrhage and congestion, petechial hemorrhages on the endo-and epicardium, renal congestion, moderate splenomegaly and cystic edema. Main microsocpic lesions observed in all infected goats were confined to the CNS and charcterized by perivascular cuffing with lymphocytes and macrophages, focal gliosis, neuronal degeneration and necrosis, and intranuclear inclusion bodies in the neurons and glial cells. Positive reactions to ADV were detected more frequently in the nuclei than in the cytoplasms of infected nerve cells in the CNS by immunohistochemistry and ISH. Frequenctly localized sites of ADV in the CNS were olfactory bulb, prietal cortex, callosal sulcus and corpus callosum. Positive reactions were also detected in the tonsillar epithelium, and alveolar macrophage and sloughed epithelium of the lung.

육계에서 아데노바이러스, 전염성빈혈 및 뉴캣슬병 복합감염 증례 (Coinfected cases with adenovirus, chicken infectious anemia virus and Newcastle disease in broiler chickens)

  • There are several immunosuppressive viral diseases in chickens such as avian adenovirus (AAV), chicken anemia virus (CAV), infectious bursal disease (IBD) and Marek's disease (MD). In this study, we have investigated two broiler chicken farms suffered from high mortality in Jeonbuk in July to August 2009. Clinically high fever and growth retardation were observed in the diseased chicken. In necropsy, the hemorrhages in thigh leg and thymus, hemorrhages and enlargement of liver, kidney and proventriculus, and yellowish fluid in heart were seen. Histologically, necrotic foci and basophilic intranuclear inclusion bodies of hepatocytes, hemorrhages and infiltrated lymphocytes in kidney and proventriculus were observed. By using polymerase chain reaction (PCR), the genes of avian adenovirus, CAV and ND virus were detected in specimens. We suggested that these coinfection cases with high mortality were due to primarily infection of immunosuppressive diseases such as avian adenovirus, CAV, followed by secondary infection of Newcastle disease (ND) virus.

보리새우류(대하)의 전염성 Lymphoid organ 괴사 바이러스 감염에 대한 병리학적 연구 (Pathological study of a infectious lymphoid organ necrosis virus infection in penaeid shrimp (Penaeus orientalis))

  • Since 1993, sudden and massive death has occurred among penaeid shrimp, Penaeus orientalis cultured in Korea. We investigated the pathological characterizations on the spontaneously and experimentally infected penaeid shrimp. The major gross findings of the spontaneous cases were 2-6mm sized-white spots on the inside of the carapace and reddish discolorization. Histopathologically, massive necrosis of lymphoid organ, degeneration and necrosis of epithelia of epidermis and foregut were observed in both spontaneous and experimental cases. Amphophilic to basophilic intranuclear inclusion bodies were readly observed epithelia of epidermis, foregut and gills. Electron microscopy revealed enveloped, non-occuluded ellipsoid to rod shaped virus particles, within the nucleus, in the lymphoid organs and interstitial cells of hepatopancreas of both spontaneously and experimentally infected shrimps. The size of the virion was $375{\times}167nm$, and the nucleocapsid was $290{\times}75nm$. The causative agent causing massive death in penaeid shrimps in Korea resembles baculovirus associated with white spot syndrome (WSBV) occurred in Taiwan in virus morphology and gross and histological changes of the shrimps.

수입산 관상 금붕어(Carassius auratus)의 CyHV-2 감염 증례 (A Case report of Cyprinid Herpesvirus 2(CyHV-2) in Imported Ornamental Goldfish Carassius auratus)

  • In this study, imported ornamental goldfish (Oranda and black moor) with high mortality was investigated with histopathological analysis. Fishes showed clinical symptoms characterized by gill necrosis and cumulative mortality ranging from 60% to 90% within a month after import from Thailand and China in 2021. CyHV-2 was detected in gill, kidney, spleen and liver of goldfishes. Histopathological analysis revealed epithelial necrosis, intranuclear inclusion bodies associated with viral symptoms in epithelial cell, macrophage infiltration and vacuolation in secondary lamella. In addition, disseminated granuloma formation was observed in various organs such as kidney, heart and mesenchymal membranes. These lesions refer to coinfections with CyHV-2 and other pathogens that cause chronic inflammation, which leads to mortality. Therefore, early disease monitoring needs to reduce the mortality when ornamental goldfish is imported.

Aujeszky's disease virus 국내분리주 접종자돈의 병리발생에 관한 연구 I. 병리학적 및 전자현미경적 관찰 (Study on the pathogenesis of the piglets experimentally infected with Korean isolate of Aujeszky′s disease virus I. Histopathologic and electron microscopic observation)

  • This study was conducted to elucidate the pathogenesis of Aujeszky's disease virus(ADV) by histopathologic examination. The first Korean ADV Isolate, which was isolated from piglets with clinical signs of Aujeszky's disease in Yangsan(YS) county, Kyungnam province, was inoculated into 32 days old piglets with a dose of $10^{5.9}$$TCID_{50}/ml$ through intranasal or intramuscular route. These piglets were sacrificed at intervals of every 24hrs for 8 days. The virulence of YS strain was determined by the observation of clinical signs, gross findings, and histopathologic changes in tissues. The virus recovery test was performed from brain, spleen, lung and tonsil in cell culture. The pathogenesis of YS strain was determined by the observation of histopathologlc lesions in CNS and neuronal tracts. The major clinical signs were fever, anorexia, dyspnea, constipation, tremor, ataxia, circling movement, hindleg paralysis and salivation. The clinical signs were more severe in piglets of the group inoculated intranasally than those of the intramuscularly inoculated gorup. Lymphocytopenia was detected on day 5 to day 6 postinoculation (PI). The ADV was recovered from the tissue homogenates of tonsil, lung, spleen and cerebrum in cell culture. The highest virus titer was detected from tonsil between day 6 and day 7 PI. Reddish sublobar consolidation foci were scattered in the apical and cardiac lobes of lung. Although yellowish necrotic foci were detected in tonsil and liver, hemorrhagic lesions were mainly observed in heart, kidney and lymph nodes. Histopathologically, degeneration and necrosis of nerve cells, nonsuppurative meningoe-ncephalitis, nodular gliosis and perivascular cuffings were observed in CNS. Multifocal fibronecrotic foci were observed in lung, liver, lymph nodes and spleen. The major pathologic changes were detected in the midbrain, pons and medulla oblongata. Eosinophilic intranuclear inclusion bodies were mainly observed in epithelia and /or macrophages of tonsil, liver, lung, spleen and submandibular lymph nodes, and neurons of brain, respectively. Observation of viral particles at various stages of replication were possible from the endothelial cells of the alveolar capillaries and tonsillar crypt epithelia by transmission electron microscope.

국내 분리 오제스키병 바이러스 접종자돈의 병리학적 관찰 (Pathological observation on the piglets experimentally infected with Aujeszky′s disease virus isolated in Korea)

  • Pathological studies were performed on the five piglets experimentally infected with Aujeszky's disease virus(pseudorabies), NYJ isolate, isolated from the naturally infected pigs in Korea: two piglets were inoculated intramuscularly, two piglets intranasally, and one piglet subcutaneously at the dose of 1$m\ell$ per animal with the 105.5 $TCID_50$/0.1ml titer. Clinical signs included dyspnea, high fever(>$41^{\circ}C$), anorexia, vomiting, diarrhea or constipation, ataxia, circling movement, posterior paralysis, intermittent convulsion, and coma followed by death although some variations by age and inoculated routes were observed. Gross features included multiple necrotic foci in the liver, congestion and hemorrhage in the lymph nodes and spleen, petechial hemorrhage in the kidney, hemorrhagic pneumonia, marked meningeal congestion, severe sub meningeal hemorrhage in the spinal cord, excessive cerebrospinal fluid retention, and muscular necrosis at the inoculated area. Microscopically, non suppurative meningoencephalitis with gliosis and perivascular cuffing in CNS, ganglioneuritis, necrohemorrhagic splenitis, necrotic hepatitis, tonsillitis and rhinitis, hemorrhagic or interstitial pneumonia, and non-suppurative myositis in the injected area were observed. Eosinophilic intranuclear inclusion bodies were found in a variety of tissues the including the liver, kidney, adrenal gland, spleen, lymph nodes, tonsil, and lung. Ultrastructurally, virus particles were confirmed in nucleus and cytoplasms of pneumocytes around the necrotic areas.

In situ hybridization에 의한 개 디스템퍼의 진단 (Diagnosis of canine distemper by in situ hybridization)

  • We have developed the in situ hybridization(ISH) technique for rapid diagnosis of canine distemper(CD) which is the major infectious disease in dogs. In our experiment, we rapidly detected distribution of the specific canine distemper viral genome without disrupting morphology of tissues or cells. Two oligonucleotide probes for ISH were synthesized chemically and labelled 5' end with nonisotopic biotin by DNA synthesizer. The whole procedures of ISH was completed within 1~2 hours using the Microcapillary action system. On histological study, typical cytoplasmic or intranuclear inclusion bodies were observed in the trachea, bronchiole, brain, and urinary bladder with the presence of prominent red positive signals on ISH, indicating specific CDV genome from the paraffin-embedded tissues of infected 13 cases. The results showed ISH can be used as a rapid and effective diagnostic method for diagnosis of CD.

국내분리 오제스키병 바이러스로 비강접종한 야생집쥐(Rattus norvegicus)의 병리학적 소견 및 in situ hybridization에 의한 바이러스 동정 (Histopathological observations and virus detection by in situ hybridization in wild rats intranasally infected with Aujeszky's disease virus isolated in Korea)

  • The present study was carried out to investigate the pathogenicity and pathogenesis of wild rats(Rattus norvegicus), trapped in nature, intranasally infected with Aujeszky's disease virus(ADV/NYJ-1-87) by histopathology, immunohistochemistry and in situ hybridization(ISH). Fifteen rats inoculated intranasally were roughened haircoat, anorexia, listlessness, and depression second day after inoculation, and three rats died in 66-72 hours. Eight rats showed severe pruritus at the face that was accompanied by frequent face-washing movements of the forelegs, and then became violent and spasmodic for an hour or until they died. Four rats slowly recovered after showing mild clinical signs of the disease. Microscopic lesions in infected rats were characterized by meningitis, perivascular round cell infiltration, focal gliosis, and neuronal degeneration and necrosis. And intranuclear inclusion bodies were frequently detected in the cerebral cortex and medulla. Positive reaction to ADV by immunohistochemistry and ISH were detected in the following areas : trigermimal ganglion, brain, tonsil, nasal mucosa, spleen, lung and liver. The result has suggested that ADV intranasally infected in wild rats is followed by replication in epithelial calls of nasal mucosa and tonsil, then invade local lymph nodes by way of the lymphatics. It is also believed that the virus invades bipolar olfactory cells and trigerminal ganglion; and then spread into central nervous system.
