수계환경에서 분변성 오염의 지표로 사용되는 미생물들 (Indicator Microorganisms Used as Fecal Contamination in Aquatic Environments)

  • The direct detection of intestinal pathogens and viruses often requires costly, tedious, and time-consuming procedures. These requirements developed a test to show that the water was contaminated with sewage-borne pathogens by assessing the hygienic quality of water based on indicator microorganisms whose presence indicates that pathogenic microorganisms may also be present. Various groups of microorganisms have been suggested and used as indicator microorganisms. Proposed and commonly used microbial indicators are total coliforms, fecal coliforms, fecal streptococci, Clostridium perfringens, heterotrophic plate count, bacteriophage, and so on. Unfortunately, most, if not all, of these indicators are not ideal because of the sensitivity and resistance to environment stresses and disinfection. However, the development of gene probes and PCR technology may give hope for the discovery of rapid and simple methods toy detecting small number of fecal pathogens in various environments.

  • Coccidiosis, an intestinal infection caused by intracellular protozoan parasites belonging to several different species of Eimeria seriously impairs the growth and feed utilization of livestock and poultry. Due to complex life cycle of organism and intricate host immune responses to Elmeria, coccidia vaccine development has been difficult. Understanding of basic imunobiology of pertinent host-parasite interactions is necessary for the development of novel control strategy. Although chickens infected with Eimeria spp. produce parasite-specific antibodies in both the circulation and mucosal secretions, antibody mediated responses play a minor role in protection gainst coccidiosis. Rather, increasing evidence show that cell-mediated immunity plays a major role in resistance to coccidiosis. T-lymphocytes appear to respond to coccidiosis both through cytokine production and a direct cytotoxic attack on infected cells. The exact mechanisms by which T-cells eliminate the parasites, however, remain to be investigated. Since it is crucial to understand the intestinal immune system in order to develop an immunological control strategy against any intestinal immune system in order to develop an immunological control strategy against any intestinal diseases, this presentation will summarize our current understanding of the avian intestinal immune system and mucosal immune responses to Eimeria, to provide a conceptual overview of the complex molecular and cellular events involved in intestinal immune responses to enteric pathogens.

Protective Effects of a Novel Probiotic Strain of Lactobacillus plantarum JSA22 from Traditional Fermented Soybean Food Against Infection by Salmonella enterica Serovar Typhimurium

  • Lactobacillus species have been shown to enhance intestinal epithelial barrier function, modulate host immune responses, and suppress the growth of pathogenic bacteria, yeasts, molds, and viruses. Thus, lactobacilli have been used as probiotics for treating various diseases, including intestinal disorders, and as biological preservatives in the food and agricultural industries. However, the molecular mechanisms used by lactobacilli to suppress pathogenic bacterial infections have been poorly characterized. We previously isolated Lactobacillus plantarum JSA22 from buckwheat sokseongjang, a traditional Korean fermented soybean food, which possessed high enzymatic, fibrinolytic, and broad-spectrum antimicrobial activity against foodborne pathogens. In this study, we investigated the effects of L. plantarum JSA22 on the growth of S. Typhimurium and S. Typhimurium-induced cytotoxicity by stimulating the host immune response in intestinal epithelial cells. The results showed that coincubation of S. Typhimurium and L. plantarum JSA22 with intestinal epithelial cells suppressed S. Typhimurium infection, S. Typhimurium-induced NF-κB activation, and IL-8 production, and lowered the phosphorylation of both Akt and p38. These data indicated that L. plantarum JSA22 has probiotic properties, and can inhibit S. Typhimurium infection of intestinal epithelial cells. Our findings can be used to develop therapeutic and prophylactic agents against pathogenic bacteria.

젖소 송아지와 한우 송아지의 설사병 발생률 비교 (Comparison of enteric pathogen incidence between dairy calves and Korean native calves)

  • From July to December 2022, we investigated the incidence of enteric pathogens causing diarrhea in calves under 100 days old at 22 farms across Korea. A total of 354 calves (including 247 dairy calves and 107 Korean native calves) were included. We used commercially available rapid diagnostic kits to identify five enteric pathogens, including bovine rotavirus (BRV), bovine coronavirus (BCoV), Cryptosporidium. Escherichia coli (E. coli), and Giardia. This study investigated how factors such as age, season, and birth month, which can influence the incidence of intestinal pathogens, affect the incidence of these pathogens in diarrheic dairy calves and Korean native calves. Our study found that the incidence of diarrhea-causing pathogens differed between dairy and Korean native calves. In diarrheic dairy calves, bovine rotavirus (BRV) incidence rates, correlated with season and birth month. Similarly, Cryptosporidium incidence C were associated with age and season, while Giardia incidence rates varied by season and birth month. Korean native calves, on the other hand, exhibited seasonal and birth month-related variations in BRV incidence rates, with Cryptosporidium incidence rates influenced by birth month.

가축의 장관에서 분리한 Lactobacillus reulterii BSA-131의 특성 (Characteristics of Lactobacillus reuterii BSA-131 Isolated from Swine Intestine)

  • The development and industrial application of intestinal anaerbic bacteria is highly important in the munufacture of foodstuffs and medical products. We have conducted studies, screening anaerobic intestinal bacteria for acid and bile tolerance and for pathogens suppression in the intestine of swine. One such study was taxonomic in nature and dealt with the isolate's biochemical responses. The results indicated that the BSA-131 strian ioslated. which suppressed various pathogens (13 typical strains), were identified as Lactobacillus reuterii. The strain, which named L. reuterii BSA-131, was able to tolerate acid condition down to pH2 and could also tolerate bile at 5%. The strain exhibited resistance to a range of antibiotics including, cephalexin, erythromycin, flumequine, furazolidine, gentamycin, penicillin G, norfloxacin, spectinomycin, tetracycline, tiamuline, neomycin, chloramphenicol, kanamycin etc.

  • Intestinal infections are common in growing pigs and can be caused by multiple pathogens, environmental and management factors [1]. Among the most important viruses in swine enteritis are porcine epidemic diarrhea virus (PEDV), transmissible gastroenteritis virus (TGEV), porcine enteric calicivirus (PECV), porcine group A rotavirus (PRV gp A) and bacteria are Escherichia coli and Salmonella spp. and protozoa is Isospora suis [1]. The DNA chip system can serve as a powerful tool that can be utilized for simultaneous detection of specific pathogenic bacteria strains and viruses [2,3]. The combination of PCR and DNA chip technology will provide a novel method for the detection of porcine enteric pathogens thus revolutionize the diagnosis and management of the disease. The aim of this study is to develop DNA chip system for the rapid and reliable detection of five major porcine enteric pathogens based on oligonucleotide DNA chip hybridization. (omitted)

소 장관 유래 Enterococcus faecium의 Enterobacter sakazakii에 대한 생육저해활성 (Growth Inhibitory Activity of Enterococcus faecium Isolated from Bovine Intestinal Tract against Enterobacter sakazakii)

  • 소 장관으로부터 분리한 유산균 중에서 Enterobacter sakazakii에 생육저해활성을 나타내는 균주를 분리한 후 동정한 결과 Enterococcus faecium으로 동정되었고 E. faecium JH95로 명명하였다. 본 균주는 kanamycin과 streptomycin에 대해 $100{\mu}g/mL$가지 매우 높은 내성을 나타내었다. 본 균주의 배양액은 L. monocytogenes, C. perfringens와 E. sakazakii에 대하여 높은 항균 활성을 나타내었으며, S. typhimurium, S. aureus와 E. coli O157:H7에 대해서도 항균활성을 나타내었다. 본 균주의 배양 상등액은 항균활성을 보이지 않았으며, 배양액이 지니고 있는 항균활성은 $100^{\circ}C$에서 5분간 가열하거나 단백질 가수분해효소 처리에 의해 소실되어 식중독 미생물에 대한 생육저해물질이 단백질인 것으로 추정되었다.

Immunogenomics approaches to study host innate immunity against intestinal parasites

  • Poultry products including meat and eggs constitute a major protein source in the American diet and disease - causing pathogens represent major challenges to the poultry industry. More than 95 % of pathogens enter the host through the mucosal surfaces of the respiratory, digestive and reproductive tracts and over the past few decades, the two main mechanisms used to control diseases have been the use of vaccines and antibiotics. However, in the poultry industry, there are mounting concerns over the ability of current vaccines to adequately protect against emerging hyper - virulent strains of pathogens and a lack of suitable, cost effective adjuvants. Thorough investigation of the immunogenetic responses involved in host-pathogen interactions will lead to the development of new and effective strategies for improving poultry health, food safety and the economic viability of the US poultry industry. In this paper, I describe the development of immunogenomic and proteomic tools to fundamentally determine and characterize the immunological mechanisms of the avian host to economically significant mucosal pathogens such as Eimeria. Recent completion of poultry genome sequencing and the development of several tissue-specific cDNA libraries in chickens are facilitating the rapid application of functional immunogenomics in the poultry disease research. Furthermore, research involving functional genomics, immunology and bioinformatics is providing novel insights into the processes of disease and immunity to microbial pathogens at mucosal surfaces. In this presentation, a new strategy of global gene expression using avian macrophage (AMM) to characterize the multiple pathways related to the variable immune responses of the host to Eimeria is described. This functional immunogenomics approach will increase current understanding of how mucosal immunity to infectious agents operates, and how it may be enhanced to enable the rational development of new and effective strategies against coccidiosis and other mucosal pathogens.

Ribosomally Synthesiszed Antimicrobial Peptides (Bacteriocins) in Lactic Acid Bacteria: A Review

  • Bacteriocins in Gram-positive bacteria have attracted much attention because many have a strong antimicrobial activity also against bacteria outside the genera of the producers. Lantibiotics and the pediocin-like bactericins have attracted most attention since they kill a broad spectrum of Gram-positive bacteria including important pathogens. But many other promising Gram-positive bacteriocins have been thoroughly characterized. Recent studies have shown that bacteriocins may playa role in the intestinal flora to protect us against the food-borne pathogens. Bacterial genome sequencing has demonstrated that there may be an arsenal of such compounds and we are only seeing the top of the iceberg. The present review gives a short outlook of the field of bacteriocins with focus on lactic acid bacteria and includes recent findings.

Adhesion Properties of Indigenous Dadih Lactic Acid Bacteria on Human Intestinal Mucosal Surface

  • Dadih is Indonesian traditional fermented buffalo milk believed by the natives to have beneficial effects on human health. This may be due to the probiotic properties possessed by the lactic acid bacteria (LAB) involved in its fermentation process. It was discovered that ten strains of dadih lactic isolates possessed some probiotic properties in vitro. In this study, the adhesion properties of dadih LAB, in comparison with documented probiotic strains, were investigated in vitro by using mucin extracted from human faeces and Caco-2 cells as the models for human intestinal mucosal surface and intestinal cells respectively. The adhesion results showed the distinction of Lactobacillus reuteri IS-27560 in adhering to both mucus layer and Caco-2 cells. The competition assay for adhesion to the mucus layer between dadih LAB and selected pathogens indicated the competence of Lactococcus lactis IS-16183 and Lactobacillus rhamnosus IS-7257 in significantly inhibiting the adhesion of Escherichia coli O157:H7. Accordingly, these two strains may be potential candidates for use as probiotic strains. Overall, the adhesion properties of all dadih LAB strains were relatively comparable to that of Lactobacillus casei Shirota and Lactobacillus rhamnosus GG, the documented probiotic strains.
