Mannose-resistant Hemagglutination(MRHA) 및 Colonization Factor Antigen I(CFA I)을 표현하는 대장균의 장관내 존재와 설사증 발현과의 관계 (Gastrointestinal Carriage of Escherichia coli with Hemagglutination Activity and Colonization Factor Antigen I and its Relation to Diarrhea)

  • Colonization factor antigen I(CFA I) has been shown to be one of several virulence factors that promote attachment of enterotoxigenic E. coli(ETEC) to small intestinal epithelial cells of humans. The ability of ETEC to produce mannose-resistant hemagglutination(MRHA) of human blood group A has been used to detect CFA I. To determine gastrointestinal carriage in Korean children of E. coli with MRHA and CFA I, 116 strains of E. coli from diarrheal children admitted to Hanyang University Hospital were examined for MRHA of human erythrocytes and the presence of CFA I. Of 45 ETEC strains, 18(40%) gave a positive MRHA($MRHA^+$) and eight(18%) were positive for CFA I(CFA $I^+$). ETEC with CFA I were all heat-stable enterotoxin(ST) producers and two of these strains were of serogroups $O_{25}$. Of 17 classic enteropathogenic E. coli(EPEC), 7(41%) were $MRHA^+$ but all were negative for CFA I(CFA $I^-$). Of 30 enteroadherent E. coli(EAEC) strains, 11(37%) were $MRHA^+$ and one was CFA $I^+$. Of 24 nonpathogenic E. coli, 4(17%) were $MRHA^+$ but all were CFA $I^-$. It was shown that MRHA was common in all strains of E. coli, CFA I was limited only to ST producing ETEC and EAEC; although MRHA is a useful screening procedure, serologic tests seem to be necessary to comfirm CFA I production. CFA I was associated with a lower proportion of ETEC isolates in Korea than has been reported for other locations.

민들레추출물이 당뇨유발 흰쥐의 장내 미생물 균총에 미치는 영향 (Effect of Dandelion (Taraxacum Officinale) Extracts on the Intestinal Microorganisms of Streptozotocin-Induced Diabetic Rats)

  • 민들레의 추출물을 급여한 흰쥐에게 streptozotocin으로 당뇨를 유발하여 장내 미생물 균총에 미치는 영향을 구명하기 위하여 Wistar계의 이유한 웅성 흰쥐 64마리를 난괴법으로 8군으로 나누어 streptozotocin(55 mg/kg BW)과 식이 kg당 11.45g의 민들레가 함유되도록 부위와 분획별로 나누어 식이에 첨가하여 50일간 사육하였다. 당뇨-대조군의 총혐기성균은 14일간 증가하다가 당뇨로 인하여 감소를 나타내었으며 잎 추출물 급여시 50일에는 당뇨-대조군에 비하여 증가되었다. 뿌리에틸아세테이트 추출물 급여군은 당뇨-대조군에 비하여 총혐 기성균수가 많은 것으로 나타났다. 민들레의 잎과 뿌리의 추출물 급여초기인 14일의 총호기성균은 당뇨-대조군에 비하여 높았으나 35일에는 당뇨-대조군에 비하여 낮은 것으로 나타났으며 50일에 총호기성균수의 변화결과는 총혐기성균의 변화양상과 유사한 것으로 관찰되었다. 장내 미생물의 변화는 당뇨가 유발된 50일에 당뇨-대조군에 비하여 민들레 잎-열수 추출물 군과 일-에틸아세테이트 추출물군에서 Lactobacillus 증가가 현저하였으며, E. cofi 감소는 잎과 뿌리의 열수 추출물군과 에틸아세테이트 추출물군이 당뇨-대조군과 유사한 수준이며 특히 열수 추출물군의 E. coli억제효과가 관찰되었다. 본 실험 결과 민들레추출물이 당뇨유발된 흰쥐의 장내 미생물에 유익한 효과로 숙주의 감염에 대한 저항력이 있는 것으로 나타났다.

  • Prophylactic effects of Bifidobacterium longum HY8001, Korean isolate, against Escherichia coli O157:H7 and Salmonella typhimurium DT104 enteric infection were examined at four groups of specific pathogen free(SPF)-ICR mouse for each pathogen. B. longum HY8001+B. typhimurium DT104+B. longum HY8001(BL+ST+BL) group and B. longum HY8001+E. coli O157:H7+B. longum HY8001(BL+E+BL) group were fed with B. longum HY8001 before and after E. coli O157:H7 or s. typhimurium DT104 challenge, while B. longum HY8001+S. typhimurium DT104(BL+ST) and B. longum HY8001+e. coli O157:H7(BL+E) groups were fed with B. longum HY8001 only before E. coli O157:H7 or S. typhimurium DT104 challenge. E. coli O157:H7(E) and S. typhimurium DT104(ST) groups were challenged with each pathogen without B. longum HY8001 administration and control groups were administered with phosphate buffered solution(PBS). After the oral administration with B. longum HY8001(109cfu), th emice were challenged with E. coli O157:H7(2$\times$1010cfu) or S. typhimurium DT104(108cfu) and the mortality rate and the fecal shedding of challenged pathogen were also examined define the reactivity of the B. longum HY8001. Production of toxin neutralizing substance(s) of B. longum HY8001 was determined by cell cytotoxicity assay using Vero cells. Fecal shedding of th eS. typhimurium DT104 was significantly decreased in BL+ST+BL group fed with B. longum HY8--1 before and after challenge(p<0.05), while the fecal shedding s of S. typhimurium DT104 in BL+ST and St groups remained more than 106cfu. the protective effect of the B. longum HY8001 against E. coli O157:H7 was significantly high only in BL+E+BL group fed with b. longum Hy8001 before and after E. coli O157:H7 challenge from the result of fecal E. coli O157:H7 isolation rate, mortality rate, and intestinal contents culture to detect E. coli O157:H7. the mortality rate of the BL+e and E groups. The cytopathic effect (CPE) of the Vero cytotoxin (Shiga like toxin I & II) in Vero cell was neutralized in B. longum HY8001 culture supernatant added wells which indicate the presence of soluble Vero cytotxin neutralizing substance(s) in B. longum HY8001 culture suprnatant.

Effects of Formic Acid Administration in the Drinking Water on Performance, Intestinal Microflora and Carcass Contamination in Male Broilers under High Ambient Temperature

  • In this study, we examined the effects of formic acid administration to the drinking water on performance, intestinal microflora and carcass contamination in male broilers. A total of 312 day-old male broiler chicks were allocated to two groups with three replicates. The first group (control) received normal drinking water (pH 7.4) during the experiment. The second group consumed acidified drinking water (pH 4.5) after 5 d of age. At 43 d of age, twelve birds were randomly selected from the control group to determine the effect of acidified drinking water on carcass contamination. These birds were only given normal or acidified (pH 3) drinking water for 8 h prior to slaughter. The reduction of water pH from 7.4 to 4.5 significantly decreased body weights of male broilers at 21 and 42 d of age. However, no differences were observed between male broilers given normal and acidified drinking water in terms of feed intake, feed conversion ratio and mortality. The pH value of the gizzard contents was not significantly affected by acid water treatment. There were no significant differences in the intestinal population of E. coli, total organism and Salmonella between the groups. The total organism and E. coli counts of the carcass slightly decreased in the acidified group. No Salmonella was identified in carcass samples of any of the treatment groups. The results showed that drinking water acidification did not provide beneficial effects on performance, intestinal microflora and carcass contamination in male broilers.

  • Porcine epidemic diarrhea (PED), transmissible gastroenteritis (TGE) are an acute viral enteritis. colibacillosis by E coli is a microbial enteritic disease in suckling piglets[1]. These infectious intestinal diarrheal diseases cause severe diarrhea to suckling piglets, so that lead to enormous economical loss in swine-product industries. Ig-Top (AD Biotech, Korea) is a immunomodulator with IgY the specific yolk-antibody for PED, TGE and E. coli and oligosaccharide. The purpose of this study was to investigate protective effects against PED virus, TGE virus E.coli and in suckling piglets by oral administration of the Ig-Top. (omitted)

노화 흰쥐의 장내 균총 조성에 대한 식이섬유질원의 영향 (Effects of Dietary Fiber Sources on the Composition Intestinal Microflora in Aged Rate)

  • This study was performed to investigate the influence of vadrious dietary fiber sources such as mugwort, apple, sea mustard, butterbur on the intestinal in aged rats. Nine groups of aged rats (12months after weanling) were fed each experimental diet containing 5% and 15% dietary fibers for 4 weeks. The contents in large intestine of aged rats used for the measurement of intestinal microflora. Total viable counts and the numbers of Bifidobacterium, Lactobacillus, Clostridium, E. coli and Staphylococcus were determined by nonselective medium and various selective media. The intake of dietary fiber sources employed did not show any differece in the number of total microorganisms, Lactobacillus and Bifidobacterium. It was remarkable that Clostridium perfringens appeared in the colon of aged rats. The prpulations of Peptococcus and Clostridium were significantly larger in the apple 5% group than in other dietary fiber groups while the number of Staphylococcus was significantly smaller in the dietary fiber groups than in cellulose group. And the Streptococcus populations decreased in the mugwort 15% group. Accordingly, the mugwort 15% group is effective in improving intestinal flora of aged rats.

장내 미생물 개선효과가 있는 약용식물소재 탐색 (Search for Medicinal Plants on Improvable Effect of Intestinal Microflora)

  • 장내미생물 개선에 효과가 있는 약용식물소재들을 탐색하여 정장제품 개발하고자 13종의 식물을 물과 에탄올로 각각 추출하여 농도별로 유해균인 C. perfringens, E. coli와 유익균인 B. longum의 생육에 미치는 영향을 in vitro실험으로 시험한 결과는 아래와 같다. 가. C. perfringens은 민들레와 인진쑥 물 추출물에, 돌나물, 오미자, 민들레, 부추, 생강, 쑥, 인진쑥, 질경이 에탄올추출물에 강하게 생육억제 되었다. 나. E. coli은 돌나물, 오미자 물과 에탄올 추출물에서 강하게 생육억제 되었다. 다. B. longum은 추출용매에 관계없이 생육에는 크게 영향을 주지 않았다. 라. 돌나물과 오미자 에탄올추출물이 유익균에 생육억제 없이, 두 유해균을 억제됨으로서 이들 소재가 정장제품으로의 개발 가능성이 있다고 생각되었다.

Evaluation of Potato Varieties (Solanum tuberosum L.) on Fecal Microflora of Human Volunteers

  • Effects of Dasom Valley and Bora Valley on fecal microflora, fecal moisture, and fecal pH of twelve healthy human volunteers were investigated. Numbers of Bifidobacterium, Clostridium perfringens, Escherichia coli, and Lactobacillus of control group were $9.24{\pm}0.63$, $4.44{\pm}1.21$, $7.75{\pm}0.38$, and $6.98{\pm}0.81$ (Log CFU/g wet feces), respectively. During administration of Dasom Valley, numbers of Bifidobacterium and Lactobacillus were $10.70{\pm}0.44$ and $8.84{\pm}0.77$, whereas those of C. perfringens and E. coli were $2.96{\pm}1.50$ and $6.69{\pm}0.29$, respectively. Administration of Dasom Valley significantly increased growth responses of beneficial bacteria, Bifidobacterium and Lactobacillus, whereas those of harmful bacteria, C. perfringens and E. coli, significantly decreased. Moisture content of feces increased and fecal pH decreased with intake of Dasom Valley. Intake of Bora Valley slightly increased numbers of Bifidbacterium and Lactobacillus and slightly decreased those of C. perfringens and E. coli. Results indicate Dasom Valley has greater intestinal-modulating effect than Bora Valley and Atlantic. Daily intake of Dasom Valley may normalize disturbed physiological functions, resulting in improvement of growth and composition of microbial community within intestinal tract.

Studies on the Intestinal Microflora of Chicken Under Tropical Condition

  • Three media, i. e., MOD-SD, M98-5 and M98-5 supplemented with chicken fecal extract were tested as isolation media for anaerobic bacteria present in the duodenum, jeju-ileum and cecum of chicken. The results showed that the mean colony counts of medium M98-5 were similar with those of MOD-SD medium in all intestinal samples at the incubation periods of 2, 6 and 10 days. Supplementation with chicken fecal extract of M98-5 medium significantly increased (p < 0.05) the colony counts of bacteria from the duodenum, jeju-ileum and cecum. The colony counts at 6-day incubation were similar with those at 10-day incubation, but were much higher than the counts at 2-day incubation. The major types of bacteria found in the duodenum and jeju-ileum of chicken were tentatively identified as Lactobacillus, Streptococcus and E. coli. In the cecum, ten tentatively identified groups of bacteria, namely, Streptococcus, Staphylococcus, Lactobacillus, E. coli, anaerobic coccus, Eubacterium, Propionibacterium, Clostridium, Fusobacterium and Bacteroides were isolated. Anaerobes were found to comprise nearly the entire microbial population of the cecum. Predominating in all sections of the intestine were homofermentative lactobacilli. The main Lactotacillus species in chicken intestine were L. acidophilus, L. fermentum and L. brevis.

Antimicrobial Activity of Quinoline Derivatives Isolated from Ruta chalepensis Toward Human Intestinal Bacteria

  • The growth responses of Ruta chalepensis leaf-derived materials toward human intestinal bacteria were examined. The biologically active constituent of the R. chalepensis extract was characterized as quinoline-4-carboxaldehyde($C_{10}H_{7}NO$). The growth responses varied depending on the bacterial strain, chemicals, and dose tested. At 0.25 and 0.1 mg/disk, quinoline-4-carboxaldehyde strongly inhibited the growth of Clostridium perfringens and weakly inhibited the growth of Escherichia coli without any adverse effects on the growth of three lactic acid bacteria. Furthermore, at 0.05 and 0.025 mg/disk, this isolate showed moderate activity against C. perfringens. In comparison, chloramphenicol at as low as 0.01 mg/disk significantly inhibited the growth of all bacteria tested, and cinnamaldehyde at 0.25 mg/disk did not inhibit Bifidobacterium bifidum, B. longum, E. coli, and Lactobacillus acidophilus, with the exception of C. perfringens. The structure-activity relationship revealed that quinoline-3-carboxaldehyde had strong growth inhibition against C. perfringens, but quinoline, quinoline-3-carboxylic acid, and quinoline-4-carboxylic acid did not inhibit the growth of B. bifidum, B. longum, C. perfringens, E. coli, and L. acidophilus. These results indicate that the carboxyl aldehyde functional group of quinolines seems to be required for growth-inhibiting activity against C. perfringens, thus indicating at least one of the pharmacological actions of R. chalepensis leaf.
