The rice belonging to Oryza sativa is not only has significant economic importance, for it is the major source of nutrition for about 3 billion all around the world. But also plays a vital role as a model organism, because it has a number of advantages to be a model plant, such as efficient transformation system and small genome size. Many methods and techniques have been conducted to attempt to distinguish different Oryza sativa species, such as amplified fragment length polymorphism (AFLP), random amplified polymorphic DNA (RAPD), simple sequence repeat (SSR) and so on. However, studies using sequence analysis of internal transcribed spacer (ITS), a region of ribosomal RNA has not been reported until now. This study was undertaken with an aim to understand the phylogenetic relationships among sixteen isolates of Oryza sativa collected from abroad and fifteen isolates collected from Korea, using ribosomal RNA (rRNA) internal transcribed spacer (ITS) sequences to compare the phylogeny relationships among different Oryza sativa species. The size variation obtained among sequenced nuclear ribosomal DNA (nrDNA) ITS region ranged from 515bp to 1000bp. The highest interspecific genetic distance (GD) was found between Sfejare 45 (FR12) and Anapuruna (FR15). Taebong isolate showed the least dissimilarity of the ITS region sequence with other thirty isolates. This consequence will help us further understanding molecular diversification in intra-species population and their phylogenetic analysis.
Branchinotogluma segonzaci (Miura and Desbruyères, 1995) occurs in hydrothermal vent fields of the southwestern Pacific Ocean. We morphologically compared B. segonzaci from the North Fiji Basin with the original description from the Lau Basin and a subsequent study of specimens from the Manus Basin. The main characteristics of all B. segonzaci populations were similar having 21 segments, 10 pairs of elytra, cylindrical-shaped anterior lobes, and ventral papillae on segment 12 and ventral lamellae on segments 13-17 in males. However, the specimens from the North Fiji Basin had rounded to sub-renifrom elytra rather than oval in the original description. Additionally, we newly obtained 11 cytochrome oxidase subunit I (COI) DNA barcodes from the North Fiji Basin and Tonga Arc populations and compared them with known COI DNA barcodes of Branchinotogluma species. Thirteen sequences of B. segonzaci showed 0.0-1.07% intraspecific variation and formed two clades in the COI neighbor-joining tree, whereas the interspecific variation among Branchinotogluma species was 8.19-22.4%. The results of this study contribute to biogeographic studies of B. segonzaci and the evolution of polynoid scale worms in chemosynthesis-based ecosystems.
한국산 미꾸리과 어류의 계통유전학적 관계를 고찰하고자 8종의 mitochondrial cytochrome b의 유전자 서열을 비교한 결과 대부분 이전의 형태학적 연구의 결과와 일치하였다. 그러나 종개속 Orthrias과 쌀미꾸리속 Lefua의 분류학적 위치는 미꾸리과 Cobitidae와 paraphyletic group으로 나타났으며 이 두 속의 sequence divergence는 0.184~0.272으로 나타나 미꾸리과와 잉어과 사이의 divergence와 유사하였다. 한편 참종개속 Iksookimia 2종과 북방종개 Cobitis melanoleuca는 각각 다르게 분화한 결과를 보여 주었으며 또한 중국산 미꾸리와 한국산 영덕 미꾸리의 sequence divergence는 0.099로 종간의 divergence를 보여주어 주목되었다. 미꾸리과 어류 가운데 참종개속의 일부 어류는 분류학적 위치로 보아 이들의 기원이 미꾸리과의 속간 잡종기원으로 생각된다.
This research was conducted in order to understand the hybridization between Quercus aliena Blume and Q. serrata Murray in Korea which show wide range of morphological variations within species and interspecific variations of diverse overlapping characteristics caused by hybridization. Morphological analysis (principal components analysis; PCA) of 116 individuals representing two species and their intermediates were performed. As a result, two species were clearly distinguished in terms of morphology, and intermediate morpho-types assumed to be hybrids between the two species were mostly located in the middle of each parent species in the plot of the principal components analysis. There was a clear distinction between two species in trichome distribution pattern which is an important diagnostic character in taxonomy of genus Quercus, whereas intermediate morpho-types showed intermediate state between two species' trichome distributions. Forty-two individuals representing two species and their intermediates were examined for leaf flavonoid constituents. Twenty-three flavonoid compounds were isolated and identified: They were glycosylated derivatives of flavonols, kaempferol, quercetin, isorhamnetin and myricetin. The flavonoid constituents of Q. aliena were five glycosylated derivatives: kaempferol 3-O-galactoside, kaempferol 3-O-glucoside, quercetin 3-O-galactoside, quercetin 3-O-glucoside, and Isorhamnetin 3-O-glucoside. The flavonoid constituents of Q. serrata had 20 diverse flavonol compounds including five flavonoid compounds found in Q. aliena. It was found that there is a clear difference in flavonoid constituents of Q. aliena and Q. serrata. Flavonoid chemistry is very useful in recognizing each species and putative hybrids. The flavonoid constituents of intermediates were a mixture of the two species' constituents and they generally showed similar characteristics to morpho-types. The hybrids between Q. aliena and Q. serrata showed morphologically and chemically diverse characteristics and it is assumed that there are frequent interspecific hybridization and introgression.
This study deals with the characterization of three populations (two hatchery and one natural) of Indian major carps Catla catla and Labeo rohita from different locations in India. The genetics of Indian major carps has been completely obscure and this is the first report on comparative allozyme variations in natural and hatchery population. The total 10 biochemical genetic markers used to measure interspecific and intraspecific level of diversity. The allele frequency data indicate different level of genetic variability in three populations. The hatchery population exhibited least polymorphism, low level of heterozygosity and genetic diversity.
The green marine algae, Capsosiphon fulvescens has been cultivated in south coast of southern Korea for many years on a commercial scale. This species is very popular in Korean as a food supplement because of its attractive flavor and flexcible taste. It is, therefore, necessary to isolate and utilize qualified germplasms for mass production of this economic seaweed. Several reports have been published on phycological applications of RAPDs including the characterization of interspecific genetic variation, the identification of isolates and hybrids, and the study of phylogenetic relationships. However few authors have used RAPDs to assess the genetic variability among populations of a seaweed species(van Oppen et al., 1994; Alberto et al., 1997). The present study was undertaken for characterizing the identities of Capsosiphon fulvescens populations cultivating in Korea through the analysis of PCR based random amplified polymorphic DNAs (Welsh and MacClelland, 1990; Willams et al., 1990) and chemical composition aimed to isolate the useful strains for aquaculture. Species of the genus Pseudodiaptomus are reported as the major constituents in the fresh or brackish waters in the northeastern Asia (Suh et al., 1991; Oka et al., 1991). Of these species, P. inopinus (Burckhardt) and P. poplesia (Shen) are particularly numerous in spring and autumn (Suh et al., 1991; Shen and Song, 1979). Recently. we found a presence of two types of Pseudodiaptomus inopinus during the course of investigation on zooplankton from Korean estuaries. (omitted)
Recently many studies show that genetic characters can be used to provide unambiguous taxonomic discrimination when morphological characters are not practical for routine identification to species level (Bucklin et al., 1996, 1998; Lindeque et al., 1999). The internal transcribed spacer (ITS) regions of the nuclear ribosomal DNA are also one of appropriate markers for species-level studies because it contains sufficient diversity to address intra- and interspecific phylogenetic relationships in invertebrates (Odorico and Miller,1997; Schizas et al., 1999). The pulmonate gastropod Ellobium chinense (Pfeiffer, 1864) is an endangered marine species along the South Korean coasts due to habitat destruction and population declines. We sequenced the cytochrome c oxidase subunit 1 (COI) of 25 E. chinense specimens collected from five coastal sites in South Korea, and identified 16 unique haplotypes. The maximum intraspecific variation among individuals was 1.6%, while interspecific differences from another ellobiid species, Auriculastra duplicata (Pfeiffer, 1854), ranged from 21.9 to 23.0%. Our barcoding data will be useful to elucidate the phylogenetic relationships among pulmonate gastropods and infer the population genetic structure of E. chinense.
A taxonomic examination of hydroids collected from the subtidal zones of Ulleungdo and Dokdo located in the East Sea of Korea from 2017 to 2023 was conducted. Through this study, Antennella aurantia n. sp., a new species belonging to the family Halopterididae, was newly added to the hydrozoan fauna of Korea. This new species is clearly distinguished from similar species by the different morphology and size of the hydrothecae, lateral nematothecae, and gonothecae. By comparing the 16S rRNA sequence with 7 recorded species within the Antennella, the interspecific genetic distance to the new species was 8.2-28.9%, which is very high compared to the intraspecific variation of 0.0%, showing that it was clearly distinguished from other species. As a result of this study, a total of 4 Antennella species are reported in the hydrozoan fauna of Korea.
