Intergeneric Hybrid Constructed by Nuclear Transfer of Saccharomycopsis into Saccharomyces (핵전이를 이용한 Saccharomycopsis 속과 Saccharomyces 속간의 잡종형성)

  • Intergeneric hybrids between Saccharomyccopsis fiburigera KCTC 7393 and Saccharomyces cerevisiae KCTC 7049 (tyr-, ura-) were obtained by nuclear transfer technique. Nuclei isolated from the wild type S. fiburigera strain were transfered into auxotrophic S. cerevisiae mutants and new strains showing an increased starch degrading capability were selected. Maximum production of protoplasts was obtained from the treatment with 0.1 % Novozym 234 at $30^{\circ}C$ for 90 min, and most effective osmotic stabilizer for the isolation of protoplasts was 0.6 M KCl at pH 5.8. The frequency of protoplast regeneration was 14.64% under the conditions. Genectic stability, conidial size, DNA content, and nuclear stain suggested that the fusants were aneuploidy. The specific activity of ${\alpha}-amylase$ was observed to increase about $1.2{\sim}1.9$ folds.

Fertilization and Embryo Development in Pollination and Culture for Interspecific and Intergeneric Crossing of Forage Crops (기내 수분과 배양에 의한 화본과 사료작물 종속간 수정과 배의 발육)

  • The ovaries or the ovules of grasses were pollinated and cultured in vitro to raise the interspecific or the intergeneric hybrids between tall fescue, meadow fescue, and Italian ryegrass. The isolated and suface-sterili-zed pistils were dusted with compatible pollens on stigma, on stump after removing stigma, or on excised ovule. Furthermore, the fertilized ovaries and ovules were cultured on MS, M6, or White's media and treated with plant growth regulators: IAA, kinetin, BA to promote embryo development and seed maturity. The in vitro fertilization in grass species ranged from 44 to 92% depending on ovary and pollen parents. The stigmatic pollination was resulted in 67.8% fertilization, the stump pollination 89.0%, and the excised ovule pollination 61.0%, repectively. White's medium was the most effective to provide embryo development and seed maturity in grass species. And the combined treatment of IAA 10mg/$\ell$, kinetin 0.2mg/$\ell$, was better than the non-treatment. Only two seedlings, one complete and one abnormal with root formation were obtained from 127 ovaryies cultured. The anatomy of ovules in vitro cultured was revealed the differentiation of vascular system and meristematic tissue, and the formation of sclerenchyma cells inside ovule.

Intergeneric Transfer of Isolated Nuclei from Lentinus edodes into Protoplasts of Pleurotus florida (사철느타리버섯 원형질체내에 표고 핵의 전이)

  • Transfer of the isolated nuclei from Lentinus edodes into protoplasts of Pleurotus florida was induced with polyethlene glycol (PEG) and $CaCl_2$. The intergeneric transfer products were classified into nuclear hybrid, heterokaryon or synkaryon, and reconstituted cell. These progenies except nuclear hybrids formed mature fruiting bodies on sawdust rice bran medium. Formation of fruit bodies was influenced by several factors such as light, temperature, nutrition and physic state of the culture media. Most of fruiting body characters were similar to those of P. florida in synkaryon and L. edodes in reconstituted cell, respectively. All these basidiocarps had clamp connections though initial heterokaryon colonies were lacking. Isozyme patterns of intergeneric progenies were quite different from those of parents. DNA polymorphisms of transfer products were also compared by random amplified polymorphic DNAs (RAPD) analysis based on polymerase chain reaction. The RAPD patterns were different from those of donor and recipient. DNA fingerprints ranged in size from 0.25 to 4.0 Kb. On the basis of RAPD, the transfer products were classified into five groups. Two synkaryon were analysed with distribution of progenies and segregation of genetic markers by random spore analyses. The genetic markers were segregated into wild type and riboflavine requiring auxotrophs.

  • Intergeneric or intraspecific protoplast fusion between Saccharomyces cerevisiae X-2180-1A, Candida pseudotropicalis ATCC 8619 and CBS 607 was attempted to produce ethanol from lactose containing materials. Teh intergeneric fusion frequency between Saccharomyces cerevisiae X-2180-1A (ade rho) and Candida pseudotropicalis CBS 607 (his met) was $1.0\times 10^{-5}$. These values exhibited approximately 2-3.5 fold increase when compared with fusion frequency obtained without the treatment of bovine serum albumin, myoinositol and ergosterol, suggesting that these compounds may improve intergeneric of intraspecific protoplast fusion. Nuclear fusion appears to occur in fusants between intergenera(S. cerevisiae+C. pseudotropicalis) and intraspecies (C. pseudotropicalis strains) as strongly suggested by DNA content, nuclear staining, comparison of survival rate to UV light and isolation of recombinants after mitotic segragation. It was also found that alcohol production from intraspecific hybrids was somewhat increased when compared with that from their parents.

Influence of insect pollinators on gene transfer from GM to non-GM soybeans (GM 콩의 도입유전자 이동에 미치는 화분 매개충의 영향)

  • The cultivation area and use of genetically modified (GM) crops have been increased continuously over the world and concerns about the potential risks of GM crops are also increasing. One of the major concern in risk assessment is the possible development of hybrids through interspecific and intergeneric crosses with related species. This study was conducted to investigate the pollinator have an influence on insect-mediated gene transfer from GM soybeans. Hybrid was induced from GM soybeans by honeybee and western flower thrips, and non-GM soybeans were used as pollen receptor. The analysis for gene-flow was conducted by herbicide selection, immunostrip test, and PCR analysis. In the result of the analysis, three hybrids were detected on the distance 15, 75, 105 cm from pollen source in western flower thrips treatment. In honeybee treatment, one hybrid was detected in the farthest distance (300 cm). These results suggested honeybee and western flower thrips have a possibility they can transfer the introduced gene from GM soybeans to non-GM soybeans.

  • Bagrid catfish hybrid between Korean bullhead (Pseudobagrus fulvidraco) and Ussurian bullhead (Leiocassis ussuriensis) was produced by artificial fertilization. Modal chromosome numbers of Korean bullhead, Ussurian bullhead and their hybrid were the same as 2n=52. The karyotypes were 22 metacentric (M), 22 submetacentacentric (SM), and 8 acrocentric (A) chromosomes for Korean bullhead, 22M+14SM+16A for Ussurian bullhead and 22M+18SM+12A for their hybrid. The hybrid bullhead karyotype consisted of each haploid set of parental species chromosomes. Erythrocytic sizes and DNA contents of hybrids were intermediate between two parental species.

Comparison of Growth Characteristics, Forage Productivity and Quality between Italian Ryegrass and Synthetics Derived from Interspecific and Intergeneric Hybrids and Ecotypes (이탈리안 라이그라스와 종.속간 잡종 및 생태형 유래 합성종간의 생육특성, 수량성 및 사료가치 비교)

  • Forage breeding laboratory of National Livestock Research Institute, R.D.A. has made interspecific hybrids of Lolium multiflorum $\times$ L. pratensis and intergeneric hybrids of Lolium $\times$ Festuca since 1984, and has collected ecotypes of Italian ryegrass since 1991. Growth characteristics of these hybrids and ecotypes were researched, and then these clone lines were named. Among these clone lines, the several clones that have polen fertility, high cold-tolerance, and similar heading time were used for making synthetics, Naehan 6, 7, 8, 9, with polycrossing method in 1997. Field experiments were carried out to compare the mophological and agronomical characteristics and forage productivity and quality of the synthetics with those of Italian ryegrass varieties, Barmultra and Hwasan 101. in Suwon and Yonchun from 1999 to 2000. Heading time of the synthetics were 22th to 24th May that belong to late-mature types to be similar to that of Barmultra and Hwasan 101 in Suwon. The synthetics were 101 to 106 c3n in plant length, medium or thick in thickness of stem, dark peen in leaf color, broad and long in flag leaf, strong in lodging resistance, and excellent in regrowth. Winter survivals of the synthetics were no different from that of Barmultra or Hwasan 101 in Suwon, but better than that of Barmultra or Hwasan 101 in Yonchun where was -10 to -12$^{\circ}C$ of minimum average air temperature in January or February. Dry matter(DM) yields of the synthetics were similar to DM 8,238kg per ha of Barmultra in Suwon, but in Yonchun, were more 7 to 13% than DM 7,291kg per ha of Barmultra. Forage qualities, IVDMD, ADF, NDF and TDN of the synthetics were lower than those of Hwasan 101, but higher than those of Barmultra.

  • Plant regeneration has been optimized increasingly by organogenesis and somatic embryogenesis using a range of explants with tissue culture improvements focusing on factors, such as the age of the explant, genotype, media supplements and $Agrobacterium$ co-cultivation. The production of haploids and doubled haploids using microspores has accelerated the production of homozygous lines in Brassicaceae crop plants. Somatic cell fusion has facilitated the development of interspecific and intergeneric hybrids in sexually incompatible species of $Brassica$. Crop improvement using somaclonal variation has also been achieved. Transformation technologies are being exploited routinely to elucidate the gene function and contribute to the development of novel enhanced crops. The $Agrobacterium$-mediated transformation is the most widely used approach for the introduction of transgenes into Brassicaceae, and $in$ $vitro$ regeneration is a key factor in developing an efficient transformation method in plants. Although many other Brassicaceae are used as model species for improving plant regeneration and transformation systems, this paper focuses on the recent technologies used to regenerate the most important Brassicaceae crop plants.

Plant Regeneration via in Vitro Culture of Ovule Obtain by Intergeneric Crossing Between Citrus junos Sieb. et Tanaka and Poncirus trifoliata Raf. (유자와 탱자의 속간교잡후 배주배양에 의한 식물체 유기)

  • As a basic research for breeding new varieties, reciprocal -intergeneric crosses between Citrus junos and P.trifoliata were made. F$_1$ hybrid production using in vitro ovule culture, gametogenesis, and fertilization phenomena were investigated. Frequency of fruit set resulting from crossing of Citrus junos and Poncirus Trifoliata was 16.6% while that of Poncirus Trifoliata and Citrus junos was 11.7%. Callus formation occurred well when ovules at the 6th week after pollination were cultured on MT (Murashige and Tucker) medium supplemented with zeatin 0.5 mg/L and NAA 1.0 or 3.0 mg/L. Immature ovules developed into mature embryos of the MT medium supplemented with 2,4-D 0.1 or 3.0 mg/L. Immature ovules developed into mature embryos of the MT medium supplemented with 2,4 D 0.1 or 0.5 mg/L. The invitro germination rates of 20-week-old ovules set C. junos $\times$ P. Trifoliata and P. Trifoliata $\times$ C. junos were 54.5% and 48.6%, respectively. The emergence ratios of trifoliate hybrids obtained by C. junos $\times$ P. Trifoliata and P. Trifoliata $\times$ C. junos were 56.7% and 100%, respectively. The chromosome number of C. junos and P. Trifoliata was n = 9 or 2n = 18, and the sizes of their pollen grain were 33.75 $\mu$ and 25.0 $\mu$. The length and width of embryo sac in C. junos and P. Trifoliata were 69.38~79.23 $\mu$ and 27.50~38.56 $\mu$, and those of egg cells were 17.50~41.50 $\mu$ and 6.25~8.12$\mu$. Fertilization of C. junos and P. trifoliata terminated 72 h after pollination.

  • Intraspecific fusion frequency between Filobasidium capsuligenum CBS6122-2 ade and met trp or met strains was $2.9\times 10^{-3}$ and $8.5\times 10^{-3}$, respectively. And intergeneric fusion frequency between Folobadidium capsuligenum CBS 4318 (cys his) and Saccharomyces cerevisiae 262-12-1 (hom3 thr1) or X2180-1A (ade thr) was $8.8\times 10^{-6}$ and $9.5\times 10^{-4}$, respectively. Nuclear fusion appears to occru in fusants between intraspecies and intergenera as strongly suggested by DNA content, nuclear staining, comparison of survival rate to UV light and mitotic segregation analysis. It was also found that $\alpha$-amylase and glucoamylase activity from intraspecific hybrids was 1.6-2.1 fold increased when compared with that from thier parents.
