Anti-Inflammatory Active Polysaccharide from Postbiotics of Cordyceps militaris Mycelium-Liquid Culture (동충하초(Cordyceps militaris) 균사체 액체발효 포스트바이오틱스로부터 항염증 활성다당 분리)

  • To investigate the anti-inflammatory activity of submerged culture using Cordyceps militaris mycelium, culture-including mycelia was extracted and lyophilized into postbiotics (hot-water extract; CM-HW). HW was fractionated into crude polysaccharide (CM-CP) by ethanol precipitation, and CM-CP was further dialyzed into CM-DCP by dialysis with running water using 12~14 kDa dialysis tube. When the cytotoxicity of subfractions against cells was assessed, no subfraction had a cytotoxic impact that was substantially different from the control groups. In an inflammatory model using LPS-stimulated RAW 264.7 cells, CM-DCP significantly decreased IL-6 and MCP-1 production levels compared to the LPS-control group. CM-DCP also inhibited IL-6 and IL-8 secretion in HaCaT keratinocytes stimulated with TNF-α and IFN-γ. In the meanwhile, the neutral sugar content and mannose ratio of anti-inflammatory CM-DCP were higher than the other fractions, and CM-DCP contained β-1,3/1,6-glucan of 216.1 mg/g. High pressure size exclusion chromatography revealed that CM-DCP contained molecules with a molecular weight range of 5.6 to 144.0 kDa. In conclusion, postbiotics of C. militaris mycelium significantly promoted anti-inflammatory activity, suggesting that neutral polysaccharides including Glc and Man contribute to the anti-inflammation in RAW 264.7 or HaCaT cells.

A STUDY ON THE EFFECTS OF TEXTURE OF FOOD ON CHEWING MOVEMENTS AND EMG OF MASTICATORY MUSCLE (식품의 물성이 제작근의 활성도 및 저작 양태에 미치는 영향)

  • To elucidate the effects of food texture such as hardness, cohesiveness, adhesiveness on mastication procedure, the electromyographic activity (EMG) of the masticatory muscles and the chewing movements were analyzed in eighteen young subjects during ordinary chewing. Seven different foods were selected by sensory texture profiling and mechanical test with texturometer; egg white, sausage, pizza cheese, yang-geng, biscuit, peanut and almond. The indexes of jaw movements used were the chewing number, chewing time, maximum opening, average opening, closing velocity and opening velocity. The EMG indexes analyzed were maximum and average integrated amplitude of the anterior temporal and masseter muscles. The results obtained were as follows. 1. The chewing time and chewing number, mandibular displacement and mandibular movement velocity were not changed by the hardness, cohesiveness and adhesiveness of the food (p>0.05). 2. The harder food materials showed a higher amplitude of the anterior temporal muscle integrated EMGs than the softer ones (p<0.05). 3. The maximum and average integrated EMGs of the masseter muscle increased with the increase of hardness of the food (p<0.05). 4. The integrated EMGs of preferred side and non-preferred side masticatory muscles increased with the increase of hardness of the food (p<0.05). 5. The adhesiveness and cohesiveness of the food were not the determining factor to the integrated EMGs of masticatory muscles (p<0.05).

The Effect of Integrated Instruction for Improving English Ability (Listening & Reading Skills) of Elementary School Students: Using Science Animation (초등학생의 융합수업이 영어 능력(듣기, 읽기) 향상에 미치는 영향: 과학 애니메이션의 활용)

  • The purpose of this study is to examine the effects of English integrated instruction on the 6th grade Korean elementary students' English ability using science animation. Twenty-seven students took English integrated instruction before their regular class for 10 minutes while the other students took review activity instead of it. At first, the students were asked to take the sixth grade diagnostic evaluation sponsored by C office of education in 2018 as the pre-test and ten months later, they were asked to take the first grade diagnostic evaluation of middle school sponsored by C office of education in 2017 as the post-test. Results of the study showed that the English ability and the affective attitude of the students taking English integrated instruction were improved. These results suggest that English integrated instruction can contribute to the improvement of Korean elementary students' English ability.

Development of Cellobiose-utilizing Recombinant Yeast for Ethanol Production from Cellulose Hydrolyzate

  • A cellobiose-utilizing recombinant yeast having $\beta$-glucosidase activity was developed for ethanol production from a mixture of glucose and cellobiose. Using $\delta$-sequences of Tyl transposon of yeast as target sites for homologous recombination, a heterologous gene of $\beta$-glucosidase was integrated into the chromosome of Saccharomyces cerevisiae. The $\delta$-integrated recombinant yeast, Saccharomyces cerevisiae L2612 (Pb-BGL), showed perfect mitotic stability even in nonselective media and showed ca. 1.5 fold higher $\beta$-glucosidase activity than the recombinant yeast harboring the $2\mu$-based plasmid vector system. A mathematical model was developed to describe the $\beta$-glucosidase formation and ethanol production from the Saccharomyces cerevisiae L2612 ($p\delta-BGL$). The model newly described that the heterologous $\beta$-glucosidase production mediated by ADH1 promoter is regulated by glucose and repressed by ethanol.

  • This paper purposes to suggest subjects integration method for effective cut-out animation production education activity. So we suggest concrete method to integrate subjects focusing on animation production. We gave cut-out animation production lessons to a group of students. As a result, their degrees of study accomplishment were higher than those of students under a general teaching style. And we conducted a questionnaire survey of participants, they gave an affirmative answer in understanding, satisfaction, participation, interest. With this, we confirmed that the animation production class activity with integrated subjects raised the creative learning ability.

Application of Time-Driven Activity-Based Costing(TDABC) for Total Productive Maintenance(TPM) and Cost of Quality(COQ) Processes (TPM과 COQ 프로세스에서 시간동인 ABC시스템의 활용)

  • This study introduces the methods to apply and develop the integrated Cost of Quality (COQ) and Time-Driven Activity-Based Costing (TDABC) model for seeking not only quality improvement but also reduction of overhead cost. Inefficient and uneconomical COQ activities can be identified by using time driver which also maximizes the quality improvement for Prevention-Appraisal- Failure (PAF) quality costs. In contrast, reduction of the indirect cost of unused capacity resource using Quality Cost Capacity Ratio (QCCR) of TDABC minimizes overhead cost for COQ activities. In addition, linkage between Overall Equipment Effective (OEE) and Time Driver develops the integrated system of Total Productive Maintenance (TPM) and TDABC model. Lean OEE maximizes when an Unused Time (UT) of TDABC that are TPM losses and lean wastes reduces whereas the TPM Cost Capacity Ratio (TCCR) of TDABC minimizes indirect cost for non-value added TPM activities. Numerical examples are derived to better understand the proposed COQ/TDABC model and TPM/TDABC model from this paper. From the proposed model, process mapping and time driver of TDABC are known to lessen indirect cost from general ledger of comprehensive income statement with a better quality innovation and improvement of equipment.

Anti-inflammatory Action of Herbal Medicine Comprised of Scutellaria baicalensis and Chrysanthemum morifolium

  • Various mixtures were prepared depending on the mixing ratio of Scutellaria baicalensis hot water extract (SB-HW) and Chrysanthemum morifolium ethanol extract (CM-E) and their anti-inflammatory activity were compared. Among them, SB-HW (80 ㎍/mL)/CM-E (120 ㎍/mL) or SB-HW (40 ㎍/mL)/CM-E (160 ㎍/mL) significantly inhibited LPS-stimulated NO and IL-6 levels in RAW 264.7 cells. The SB-HW (80 ㎍/mL)/CM-E (120 ㎍/mL) mixture, which was determined as active mixture, significantly reduced MUC5AC secretion in PMA and LPS-induced NCI-H292 cells. The active mixture also reduced the production of PGE2 and IL-8 in PMA-induced A549 cells. LC-MS/MS analysis showed that the active mixture was composed of high contents of flavone glycosides, such as baicalin and cynaroside. Western blot analysis indicated that the active mixture suppressed phosphorylation of ERK, JNK, and p38, associating with the inhibition of MAPK signaling. Taken together, our results suggest that the active mixture could be applied as a new anti-inflammatory herbal medicine

Implementing a 1-Night, 2-Day Mental Health Healing Center Program for Dementia Patients and Their Caregivers

  • Objectives: This study aimed to assess the effects of a 1-night, 2-day mental health healing center program on the physical stress, autonomic nervous system health, brain activity levels, brain stress, concentration levels, and Patient Health Questionnaire-9 (PHQ-9) scores of dementia patients and their caregivers. Methods: Forty-eight dementia patients (average age 80 years, 14 males and 34 females) and 48 caregivers (average age 65.23 years, 14 males and 34 females) participated in the program. Pre- and post-assessments were conducted to measure the variables. Results: Dementia patients experienced reduced physical stress, increased brain activity levels, decreased brain stress, improved concentration levels, and a significant decrease in PHQ-9 scores (p<0.05). However, there was a tendency for a decline in autonomic nervous system health among dementia patients. A significant decrease in physical stress was seen in caregivers, but no other significant differences were observed. Conclusions: While the 1-night, 2-day mental health healing center program did not produce significant changes in the caregivers of dementia patients, it exhibited overall positive effects in dementia patients. Consequently, mental healing programs should be utilized effectively for dementia patients. Furthermore, investigating the significance of ongoing programs for the mental well-being of dementia caregivers is imperative, mirroring the attention given to dementia patient care.

Perceptions of Elementary Teachers on Teaching Science-Arts Integrated Activities (과학-예술통합 활동에 대한 초등학교 교사들의 인식)

  • This study investigated the perceptions and the characteristics of elementary teachers on teaching science-arts integrated activities. Twelve elementary teachers who have taught in the third to sixth grade science lessons participated in this study. Data was collected by individual interviews lasting 60 to 90 minutes. Interview questions were composed of three aspects: 1) how elementary teachers perceive science-arts integrated activities, 2) in what ways elementary teachers address science-arts integrated activities, and 3) what difficulties elementary teachers experienced in addressing science-arts integrated activities. The results showed that, first, elementary teachers agreed that science-arts integrated activities have educational value in science learning. Second, most teachers focused on improving understanding of science knowledge through teaching science-arts integrated activities, while several teachers put emphasis on having experience of arts as well as understanding of science knowledge. And third, teachers experienced difficulties due to insufficient teaching materials and time to prepare for teaching science-arts integrated activities. Based on the results, we suggested educational implications for utilization of science-arts integrated activities in elementary education.

A design method of activity cost pool for activity based costing in logistics systems (물류체계에서의 활동기준원가의 활동원가군 설계방법)

  • When logistics system is integrated with production system and marketing system, it takes a very important role of the business management. In general, measurement of logistics cost in logistics system uses the conventional cost assignment method. However the conventional method may result in the incorrect cost because the overhead cost may be incorrectly assigned to the products. Activity-Based Costing(ABC) was proposed as an alternative method which will distribute the overhead cost to each cost obeject more accurately. ABC assigns cost to activities based on the amounts of resources used by resource driver, and assigns cost to cost objects based on the amount of activities driver. This study proposes two heuristic algorithms. The first algorithm selects the best activity driver for each cost object by using correlation analysis. The best activity driver is the one that minimizes the sum of loss cost and measurement cost of activity driver. The second algorithm selects the best number of activities by using correlation analysis. The pair of activities with the highest correlation are combined into one if the saving of measurement cost is no less than the loss due to inaccurate distribution of overhead cost. In order to demonstrate the procedure and validity of the algorithms, Real data of one year from a paper manufacturer are used.
