Enhancement of insecticidal efficacy of acetamiprid soluble concentrates using different adjuvants (서로 다른 보조제에 의한 acetamiprid 액제의 살충활성 증진효과)

  • The studies were undertaken to establish optimum formulation recipes of acetamipid 4% SL(soluble concentrate) using different adjuvants and to evaluate enhanced control effect on target insect pests by adjuvants. Adjuvants for the studies used were SS(Sodium alkylcarboxylate) and PE(Polyoxyethylene alkyl ether). After the fundamental recipe for SL being established by using co-solvent for stability at low temperature, four types of SL were formulated. The physical properties and insecticidal efficacies of the tested SL formulations were investigated. Surface tension of SL1 without adjuvant was highly decreased from 44 dyne/cm to 34 dyne/cm by addition of adjuvants. SL4 using mixed adjuvant showed the lowest contact angle. Efficacies of SL1 without adjuvant against the palm thrips (Thrips palmi) and the cotton aphid (Aphis gossypii) on cucumber were increased by adding adjuvants. SL4 using mixed adjuvant showed higher efficacy than SL2 and SL3 using single adjuvant. These results have demonstrated that the selected adjuvants could be used to enhance insecticide efficacy and reduce spray dose of insecticide.

Synthesis and pesticidal activity of ricinine derivatives (Ricinine 유도체(誘導體)의 합성(合成) 및 농약활성(農藥活性))

  • Chemical derivative synthesis of ricinine, an active compound of Ricinus communis which showed high mortality against brown planthopper (Nilaparvata lugens), was performed to improve its pesticidal activity and the toxicity of 12 synthetic derivatives against major insect pests and phytopathogenic fungi were examined. Carbamate derivatives of ricinine could be synthesized from the precursor of ricinine, chloronorricinine and norricinine, whereas the derivatives were not synthesized from chlororicinic acid and ricinic acid having ketone group of pyridine ring. In organophosphates, reaction with oxon type of phosphate gave better yield than thiono type. Among the organophosphate derivatives of ricinine, thiono type of derivative structure gave $96.3%{\sim}100%$ mortality of the brown planthopper and the two-spotted spider mite (Tetranychus urticae) at 500 ${\mu}g/ml$ level. On the other hand, carbamate derivatives did not show insecticidal activity. In the fungicidal activity of ricinine derivatives, the derivative having amino radical at the 2 position of ricinine gave 85 to 100% of mycelium growth inhibition effect against ten major plant pathogens at the 200 ${\mu}g/ml$ level. In particular, the control value of the derivative on the rice blast (Pyricularia grisea) and barley powdery mildew (Erysiphe graminis) at the 250 ${\mu}g/ml$ level in vivo under greenhouse conditions was 92% and 96%, respectively.

Acetylcholinesterase-based Biosensor for Detection of Residual Organophosphates and Carbamates Insecticides (유기인계 및 카바메이트계 농약을 측정할 수 있는 바이오 센서의 개발)

  • Inhibitors of acetylcholinesterase(AchE), such as organophosphates and carbamates, interfere the action of AchE in nerve and may lead to a severe impairment of nerve functions or even death. Therefore, insect AchE is the biological target of predominant insecticides used in agriculture. Biosensors are sensitive and can be used as dispoisable sensors for environmental control. In recent years, the use of AchEs in biosensor technology has gained enormous attention, in particular with respect to insecticide detection. The principle of biosensors using AchE as a biological recognition element is based on the inhibition the catalytic activity by the agents to be detected. We here present a strip-type biosensor based on AchE inhibition. In this study, acetylcholinesterase and PVA-SbQ(polyvinyl alcohol functionalized with methyl pyridinium methyl sulfate) were co-immobilized on immobilone-P membranes. Immobilization of the enzymes showed a stability in 6 months without activity loss in $4^{\circ}C$ storage. Enzymes immobilized on surfaces of membrane responded to organophosphates and carbamate more sensitivitive than enzyme in solution. Organophosphates and carbamates concentrations could be detected by entrapped and surface immobilized enzymes, in 5 min. For chlorpyrifos, carbofuran, cabaryl, and methidathion, the detection limits of AChE-strip were similar to that of HPLC/GC method.

  • The arthropods are the most diverse and successful animal group making up more than three quarters of the total animal species. They also live in a great diversity of habitats on the earth. Because of these unavoidable circumstances, the human life has always been affected by them. Therefore, in many countries from the past, governments have supported the studies of the arthropods on the basis of the national policy. However, the present situation of ours is far behind the other advanced contries, Now we urgently need governmental support fro the development of both the basic and the applied fields of arthropod studies in the well organized manner. Facing with this necessity, the present article reviewed the studies of Korean arthopods based on insect and crustacean representing most of the arthopods. Several subjects were reviewed in the section of the past and present studies on Korean arthropods. These are the studies on Korean arthropods in the past and present, their application to the national strategy, and the present condition of curating specimens, etc. From these reviews, several important points were suggested in the section of future plan. These are such subjects as the establishment of national institution for the curation of specimens , computerizing the data, diversification of research techniques, valid strategy concerning the control of harmful contribution to the nature conversation and environmental management , use as a research animal, the enhancement of public welfare, training of specialists, and policies for the national programs.

An Empirical Model for the Prediction of the Onset of Upward-Movement of Overwintered Caccopsylla pyricola (Homoptera: Psyllidae) in Pear Orchards (배과원에서 꼬마배나무이 월동성충의 수상 이동시기 예측 모형)

  • Pear psylla, Caccopsylla pyricola (Homoptera: Psyllidae), is a serious insect pest in pear orchards. C. pyricola overwinters as adults under rough bark scales of pear trees. When the weather warms up in the spring, the overwintered adults become active, climb up to the tree branches, and inhabit on fruit twigs to lay eggs. This study was conducted to develop a forecasting model for the onset of upward-movement of overwintered C. pyricola adults to control them by timely spraying of petroleum oil. The adult population densities were observed under rough barks (B) and on fruit twigs (T) of pear trees. Relative upward-movement rates (R) were calculated as T/(B+T). Low threshold temperatures for the activation of overwintered C. pyricola adults were selected arbitrarily from 5 to $9^{\circ}C$ at a $1^{\circ}C$ interval. Then, the days (D) when daily maximum air temperatures were above each low threshold temperature were counted from 1 February until to the dates with R $\geq$ 0.8. The same methods were applied for the prediction of the first observation of eggs. The variation of coefficients (CV) for the mean Des were lowest with the low threshold temperature of $6^{\circ}C$. At this selected threshold temperature, the upward movement of C. pyricola adults occurred with 12 D and they started laying eggs with 25 D. In the field validation, the model outputs with the $6^{\circ}C$ threshold temperature reasonably well explained the observed data in Suwon and Cheonan in 2002. Practical usages of the model were also discussed.

  • The ecological characteristics of the specialist insect predator, Oligota kashmirica benefica, were investigated for developing artificial rearing method with special interest on the oviposition behavior, the pattern of adult emergence and temperature in storage, and its effect on the density of the citrus red mite, Panonychus citri. Female beetle oviposited eggs mainly, $95\%$ of the eggs, on the bottom of leaves of the yuzu tree, and $91.3\%$ of them were covered with ecdysis skin of the red mites or the feces of themselves. The rate of adult emergence of the beetle rose up to $86.7\%$ when the horticultural media was provided for pupation from $60\%$ when it pupated in upland soil. Most larvae, $88\%$ of them, were found from the surface to the depth of 2 cm in the horticultural media. The optimum temperature for the storage of the adult beetle was found to be $12^{\circ}C$, at this temperature the 96.7, 73.3 and $70.0\%$ of the beetle survived for 10, 20, and 30 days, respectively. The control effect of the citrus red mite was appeared highest by releasing the beetle at the rate of one beetle against 150 mites.

Attractant Effect of Trap Logs Treated with Ethanol to Platypus koryoensis (Coleoptera: Platypodidae) (에탄올 처리에 의한 광릉긴나무좀(Platypus koryoensis) 유인목 효율 증대)

  • The attractant effect to Platypus koryoensis (Murayama), an insect vector of Korean oak wilt disease, was determined using a multi-funnel trap and trap logs treated with ethanol and ethanol extract of the bark of the Mongolian oak Quercus mongolica (Fisher). The number of entrance holes in ethanol and bark ethanol extract treatments was higher than that in the control (blank bottle). However, the number of entrance holes in ethanol and bark ethanol extract treatments was not significantly different. The number of entrance holes in the logs increased with increase in diameter of the logs. The lower part of the trap logs had significantly more number of entrance holes than that in the upper part (p < 0.05). The lower part of trap logs tends to be rarely exposed to direct sunlight resulting in desiccation of the logs, which in turn provides good conditions for microbial growth, a food source for P. koryoensis. The results show, that ethanol could be used effectively in the monitoring and maintenance of P. koryoensis because of the cheaper cost of ethanol than bark ethanol extracts, and thus, having a higher economical advantage. Further research of oak bark ethanol extracts is necessary to improve its efficiency.

Implementation of Aerial Application System for Application Uniformity (균일 방제를 위한 항공 살포시스템 구현)

  • The aim of this study was to prevent the decrease in crop output by disease and insect pests and excessive spraying of agricultural pesticides by application uniformity. A 3m height and 15km/h speed is difficult to maintain with an unmanned helicopter for aerial application, which has been affected by the controlling habits and methods or environmental factors, such as changes in the wind. Therefore, in this study, an aerial application system was design to be attached to an unmanned helicopter, which can allow a controlled application width and spray rate automatically and verified experimentally using Rmax of MS-AVIATION. The size of agricultural land was 50 m2 and nine water sensitive cards were arranged at 1.25m intervals in 5 rows with each row having a 10m interval from the position of 5m. The unmanned helicopter was flying at speeds ranging from 7.2km/h to 17.6km/h and heights ranging from 2.32m to 3.47m. The proposed aerial application system allowed application uniformity by making a valid spraying area of 7.5 m2 with 46423 particles distributed on average.

Development of the Hospital Foodservice Facility Evaluation tools based on the General HACCP-based Sanitation Standards and Guidelines (병원급식에 일반위생관리기준과 HACCP 제도 적용을 위한 시설ㆍ설비 위생관리 점검도구 개발)

  • The rapid increase in food borne illness outbreaks in Korea has been one of the major threats to the Nation's Health. Foodservice establishments have been identified as the major place for these outbreaks, mainly due to the lack of sanitary management and sanitary facility management practices. The purposes of the study were to develop hospital foodservice facility evaluation tools, based on the general HACCP-based standards and guidelines, for hospital food service establishments, to ensure the safety of these foodservices and to reduce the risk of food home illness. The scope of this study included: 1) an assessment of the current foodservice sanitation practices and managements for 6 general hospitals, with more than 400 beds, and 3 general hospitals, with less than 400 beds; 2) the development of foodservice establishments sanitation evaluation tools and sanitation standards, based on the HACCP system. The survey data showed varied results between the hospitals surveyed. Most of the hospital foodservice operations had many problems with ventilation and the plumbing. The total dimensional mean scores for the hospitals with more than 400 beds and less than 400 beds were 31.5 and 27.0, respectively. The highest dimension scores were for the water supply facility and lighting, with the lowest for insect and rodent control and toxic materials management. The levels of the mean scores were very low, especially for the general hospitals with less than 400 beds. These low mean scores may have arisen from critical problems within the hospital foodservice operations. The most needed facility management items for improvement were: storage shelf should be spaced 6 inches from the floor and walls, the use of three compartment sinks, utility sinks and cleaning facilities, with a floor drain for cleaning mops or liquid wastes, a ventilation hood designed to prevent dripping onto food, cooking facilities should be disassembled for washing and sanitizing, a separated hand washing sink and a sanitized food board for each area should be provided, all toxic material must have warning labels attached, and be stored in an area away from food preparation under padlock. The evaluation tool consisted of 14 dimensions, with 65 check-off items. The results of this study will provide basic facilities' guidelines to regulators, or foodservice industry personnel, wishing to build, or expend, and establish an efficient flow of food. As a result, food borne illnesses will be effectively prevented, and the Nation's health will be promoted for the development of their own sanitation standards, with a checklist for the safe production of foods.

Isolation and purification of a cecropin-like antimicrobial peptide from the japanese oak silkworm, Antheraea yamamai (천잠 세크로핀 항균펩타이드 분리 및 정제)

  • Cecropin is a well-studied antimicrobial peptide that play important role as key factor in insect humoral immunity. In this study, cecropin-like antimicrobial peptide was isolated and purified from the larval haemolymph of immune-challenged japanese oak silkworm, Antheraea yamamai. To isolate antimicrobial peptide, we separated and compared acidic extracted hemolymph protein bends between control and immune-challenged larvae using SDS-PAGE analysis. In the immune hemolymph extract, but not of non-immune hemolymph, we detected differential expressed peptide band with molecular mass 4,223.01 Da. To understand this peptide better, we successfully purified this peptide using cation exchange chromatography and gel permeation chromatography. Its N-terminal amino acid sequence obtained by Edman degradation evidenced a significant degree of identity with other lepidopteran cecropins. The purified A. yamamai cecropin-like peptide showed a broad spectrum of activity against fungi, Gram-negative and Gram-positive bacteria.
