Stigmalactam from Orophea Enterocarpa Induces Human Cancer Cell Apoptosis Via a Mitochondrial Pathway

  • Stigmalactam, an aristolactam-type alkaloid extracted from Orophea enterocarpa, exerts cytotoxicity against several human and murine cancer cell lines, but the molecular mechanisms remain elusive. The aims of this study were to identify the mode and mechanisms of human cancer cell death induced by stigmalactam employing human hepatocellular carcinoma HepG2 and human invasive breast cancer MDA-MB-231 cells as models, compared to normal murine fibroblasts. It was found that stigmalactam was toxic to HepG2 and MDA-MB-231 cells with $IC_{50}$ levels of $23.0{\pm}2.67{\mu}M$ and $33.2{\pm}4.54{\mu}M$, respectively, using MTT assays. At the same time the $IC_{50}$ level towards murine normal fibroblast NIH3T3 cells was $24.4{\pm}6.75{\mu}M$. Reactive oxygen species (ROS) production was reduced in stigmalactam-treated cells dose dependently after 4 h of incubation, indicating antioxidant activity, measured by using 2',7',-dichlorohydrofluorescein diacetate and flow cytometry. Caspase-3 and caspase-9 activities were increased in a dose response manner, while stigmalactam decreased the mitochondrial transmembrane potential dose-dependently in HepG2 cells, using 3,3'-dihexyloxacarbocyanine iodide and flow cytometry, indicating mitochondrial pathway-mediated apoptosis. In conclusion, stigmalactam from O. enterocarpa was toxic to both HepG2 and MDA-MB-231 cells and induced human cancer HepG2 cells to undergo apoptosis via the intrinsic (mitochondrial) pathway.

Regional comparison of dietary intakes and health related behaviors among residents in Asan

  • Inadequate dietary intakes and poor health behaviors are of concern among rural residents in Korea. This study is conducted to compare dietary intakes, dietary diversity score (DDS), mean nutrient adequacy ratio (MAR) and health related behaviors by rural, factory and urban areas in Asan. A total of 930 adults (351 men and 579 women) were interviewed to assess social economic status (SES), health related behaviors and food intakes by a 24-hour recall method. Mean age was 61.5 years with men being older (64.8 years) than women (59.3 years, p<0.001). Men in the factory area were older than rural or urban men while urban women were the youngest. Education and income of urban residents were higher than other area residents. There were more current drinkers in urban area while smoking status was not different by regions. Physical activity was significantly higher in rural or factory areas, whilst urban residents exercised more often (p<0.05). Rural or factory area residents considered themselves less healthy than others while perceived stress was lower than urban residents. Energy intakes were higher in urban residents or in men, however, after SES was controlled, energy intake did not show any differences. Energy-adjusted nutrient intakes were significantly higher in the urban area (p<0.05) for most nutrients except for carbohydrate, niacin, folic acid, vitamin $B_6$, iron and fiber. Sodium intake was higher in factory area than in other areas after SES was controlled. DDS of rural men and MAR of both men and women in the rural area were significantly lower when SES was controlled. In conclusion, dietary intakes, diversity, adequacy and perceived health were poor in the rural area, although other health behaviors such as drinking and perceived stress were better than in the urban area. In order to improve perceived health of rural residents, good nutrition and exercise education programs are recommended.

  • 본 논문은 세계화가 우리나라 제조업 사업체의 성장과 양극화에 미친 영향을 살펴본 것이다. 본 논문은 먼저 이론적으로 세계화가 성장 및 양극화에 미치는 영향 및 그 구체적 메커니즘을 최근 이질적 기업의 무역이론들을 중심으로 논의하고, 나아가 관련 실증적 증거들이 존재하는지를 살펴보았다. 이를 통하여 본 논문은 세계화가 1990년대 이후 우리나라 제조업 사업체의 생산성 향상뿐 아니라 기업 간 생산성 및 숙련도별 임금 양극화 모두를 초래한 공통적 원인으로 작용하였음을 보여주었다. 본 연구는 최근 세계화된 경제환경하에서 왜 극소수의 대기업만이 특출한 성과를 나타내고 있는지를 이해하는 데 도움이 된다. 마지막으로 본 연구는 세계화의 부작용을 줄이고 그 편익을 보다 많은 기업들이 누릴 수 있도록 하기 위한 정책방향을 제시한다.

Lin28a attenuates TGF-β-induced renal fibrosis

  • Lin28a has diverse functions including regulation of cancer, reprogramming and regeneration, but whether it promotes injury or is a protective reaction to renal injury is unknown. We studied how Lin28a acts in unilateral ureteral obstruction (UUO)-induced renal fibrosis following unilateral ureteral obstruction, in a mouse model. We further defined the role of Lin28a in transforming growth factor (TGF)-signaling pathways in renal fibrosis through in vitro study using human tubular epithelium-like HK-2 cells. In the mouse unilateral ureteral obstruction model, obstruction markedly decreased the expression of Lin28a, increased the expression of renal fibrotic markers such as type I collagen, α-SMA, vimentin and fibronectin. In TGF-β-stimulated HK-2 cells, the expression of Lin28a was reduced and the expression of renal fibrotic markers such as type I collagen, α-SMA, vimentin and fibronectin was increased. Adenovirus-mediated overexpression of Lin28a inhibited the expression of TGF-β-stimulated type I collagen, α-SMA, vimentin and fibronectin. Lin28a inhibited TGF-β-stimulated SMAD3 activity, via inhibition of SMAD3 phosphorylation, but not the MAPK pathway ERK, JNK or p38. Lin28a attenuates renal fibrosis in obstructive nephropathy, making its mechanism a possible therapeutic target for chronic kidney disease.

Drug resistance of bladder cancer cells through activation of ABCG2 by FOXM1

  • Recurrence is a serious problem in patients with bladder cancer. The hypothesis for recurrence was that the proliferation of drug-resistant cells was reported, and this study focused on drug resistance due to drug efflux. Previous studies have identified FOXM1 as the key gene for recurrence. We found that FOXM1 inhibition decreased drug efflux activity and increased sensitivity to Doxorubicin. Therefore, we examined whether the expression of ABC transporter gene related to drug efflux is regulated by FOXM1. As a result, ABCG2, one of the genes involved in drug efflux, has been identified as a new target for FOXM1. We also demonstrated direct transcriptional regulation of ABCG2 by FOXM1 using ChIP assay. Consequently, in the presence of the drug, FOXM1 is proposed to directly activate ABCG2 to increase the drug efflux activation and drug resistance, thereby involving chemoresistance of bladder cancer cells. Therefore, we suggest that FOXM1 and ABCG2 may be useful targets and important parameters in the treatment of bladder cancer.

  • The Interest in disease prevention and rehabilitation is increasing depending on increase of patients with spinal. This is being developed using the spine stabilization device is being studied. So far studies have only evaluated the effect on trunk stabilization exercises but analysis of human movement patterns for active movement and passive movement did not. We assessed the muscle activity of trunk and leg muscle during passive and active tilt mode on eight tilt directions at tilt angle of $30^{\circ}$ using 3-D dynamic postural balance training system. We performed experimental study on the muscular activities of trunk muscle about rectus abdominis, external obliques, latissimus dorsi, erector spinae, and leg muscle about rectus femoris, Biceps femoris, Tibialis Anterior, gastrocnemius. As a result, muscle activation was different depending on the direction of movement and pattern. The results indicate that various patterns of spinal stabilization exercise system could be applied to an effective training of chronic low back pain patients.

  • 각 은행들은 인터넷 뱅킹을 통해 경쟁력 확보하기 위해 서비스 개발에 박차를 가하고 있을 뿐만 아니라, 새로운 뱅킹 비즈니스 모델을 구현하고 있다. 특히 인터넷 뱅킹 경쟁력 평가는 단순히 인터넷 뱅킹 사이트의 서비스 평가와는 다르다. 국내외 인터넷 뱅킹에서 제공하는 서비스 상품들은 성공적인 합병과 다양한 온라인 접점의 구축을 통해 금융서비스를 가장 효율적으로 제공하며, 또한 은행업무뿐만 아니라 증권서비스, 보험, 소비자금융, 리스, 부동산업무 등을 원스톱으로 제공하는 등 다양한 서비스를 제공하며, 사업의 영역을 넓혀가고 있다. 그리고 대부분의 국내외 은행 및 금융기관들이 인터넷 뱅킹 서비스 개발을 통하여 경영효율성을 증대시키고 경쟁력을 제고하고자 노력하고 있다. 특히 국내외 은행들이 일대일 맞춤 서비스, 개인 재무관리 그리고 인터넷뱅킹을 이용한 아파트관리비 납부서비스 상품을 개발하여 제공하고 있을 뿐만 아니라, 통합모바일서비스, 세금 전자납부 징수, 인터넷 무서류 대출 상품 서비스를 개발하여 시장에서 판매하고 있다. 따라서 현재의 고객 및 잠재고객들이 은행의 인터넷 뱅킹 서비스를 적극 이용하도록 유치함으로써 은행 및 금융기관들은 그들의 경쟁적위상을 제고시키고자 노력하고 있다.

Steroidal Saponins from Paris polyphylla Suppress Adhesion, Migration and Invasion of Human Lung Cancer A549 Cells Via Down-Regulating MMP-2 and MMP-9

  • Background: Tumor metastases are the main reasons for oncotherapy failure. Paris polyphylla (Chinese name: Chonglou) has traditionally been used for its anti-cancer actions. In this article, we focus on the regulation of human lung cancer A549 cell metastases and invasion by Paris polyphylla steroidal saponins (PPSS). Materials and Methods: Cell viability was evaluated in A549 cells by MTT assay. Effects of PPSS on invasion and migration were investigated by wound-healing and matrigel invasion chamber assays. Adhesion to type IV collagen and laminin was evaluated by MTT assay. Expression and protease activity of two matrix metalloproteinases (MMPs), MMP-2 and MMP-9, were analyzed by Western blotting and gelatin zymography, respectively. Results: PPSS exerted growth inhibitory effects on A549 cells, and effectively inhibited A549 cell adhesion, migration and invasion in a concentration-dependent manner. Western blotting and gelatin zymography analysis revealed that PPSS inhibited the expression and secretion of MMP-2 and MMP-9 in A549 cells. Conclusions: PPSS has the potential to suppress the migration, adhesion and invasion of A549 cells. PPSS could be a potential candidate for interventions against lung cancer metastases.

ICT 산업의 여성인력 고용확대 방안 (A Study on the Plan of Creating Jobs for Women in the ICT Industry)

  • 최근 정부는 '창조경제(Creative Economy)'를 통하여 상상력과 창의성, 과학기술에 기반한 경제 운용, 새로운 시장, 새로운 일자리 창출을 목표로 하고 있다. 이는 또한 과학기술과 ICT, 문화, 인문학 등이 융합하여 산업간 경계부분에서 창조경제의 성과를 이룩할 수 있다는 것이 선진국의 창조경제 벤치마킹에서 증명되고 있다. 특히, 여성 과학기술인이 창조형 선진국의 사례를 보면 중요한 역할을 수행하고 있다. 하지만 한국은 여성 과학기술인이 낮은 수준이고 전문적인 여성과학기술자에 대한 적극적인 지원 및 육성정책도 미흡한 것으로 알려져 있다. 또한 경제 활동 중이던 여성들은 육아와 출산 등으로 인한 경력단절, 복귀에서 겪는 어려움 때문에 30대 초 중반 여성이 경제활동에 참여하는 비율이 적다. 창조경제를 지속적으로 발전시키기 위해서는 이를 이끌어갈 여성 과학기술인의 창조적 교육과 육성, 지원에 힘을 기울여야 한다. 따라서 본고에서는 창조경제 시대의 ICT 산업정책과 여성의 역할과 경제활동 참여 현황, 그리고 창조경제시대의 여성 일자리 창출방안과 문제점을 살펴본다.

The Anti-Adipogenic Activity of a New Cultivar, Pleurotus eryngii var. ferulae 'Beesan No. 2', through Down-Regulation of PPAR γ and C/EBP α in 3T3-L1 Cells

  • Adipogenesis is one of the cellular processes and a highly controlled program. Nowadays, inhibition of adipogenesis has received attention as an effective way to regulate obesity. In the current study, we investigated the inhibition effect of a chloroform extract of Pleurotus eryngii var. ferulae 'Beesan No. 2' (CEBT) on adipogenesis in 3T3-L1 murine preadipocytes. Pleurotus eryngii var. ferulae is one of many varieties of King oyster mushroom and has been reported to have various biological activities, including antitumor and anti-inflammation effects. Biological activities of 'Beesan No. 2', a new cultivar of Pleurotus eryngii var. ferulae, have not yet been reported. In this study, we found that CEBT suppressed adipogenesis in 3T3-L1 cells through inhibition of key adipogenic transcription factors, such as peroxisome proliferatoractivated receptor ${\gamma}$ and CCAAT/enhancer binding protein ${\alpha}$. Additionally, CEBT reduced the expression of the IRS/PI3K/Akt signaling pathway and its downstream factors, including mammalian target of rapamycin and p70S6 kinase, which stimulate adipogenesis. Furthermore, ${\beta}-catenin$, a suppressor of adipogenesis, was increased in CEBT-treated cells. These results indicate that Pleurotus eryngii var. ferulae 'Beesan No. 2' effectively inhibited adipogenesis, so this mushroom has potential as an anti-obesity food and drug.
