Drying Characteristics of Large Western Redcedar Timber During Radio Frequency/Vacuum Drying (웨스턴 레드시더 큰 정각재(正角材)의 고주파 진공 건조 특성)

  • Western redcedar timber, 26 by 26cm in cross section and by 200cm long, was dried in a laboratory radio-frequency/vacuum kiln under 65torr of ambient pressure and a fixed frequency of 6.78MHz for the potential rapidly dry large timber. All process data were collected and saved in a computer through a data acquisition system. The temperature in the middle of timber was higher than temperature at the quarter point of timber length and thickness. Temperature gradients developed in the longitudinal and transverse direction of timber. The pressure in the middle of timber was higher than pressure at the quarter point of timber length. The pressure in the middle of timber was lower in the early stage of drying, and higher in the latter stage of drying than pressure at the quarter point of timber thickness. Power density was very highest during heating period and then gradually decreased. The drying curve was approximately linear and the total drying time was 27 hours from an initial moisture content(MC) of 48.6 percent to a final Me of 19.2 percent with only a few mild internal checks in the middle location of timber.

cAMP Response Element-Binding Protein- and Phosphorylation-Dependent Regulation of Tyrosine Hydroxylase by PAK4: Implications for Dopamine Replacement Therapy

  • Parkinson's disease (PD) is characterized by a progressive loss of dopamine-producing neurons in the midbrain, which results in decreased dopamine levels accompanied by movement symptoms. Oral administration of l-3,4-dihydroxyphenylalanine (L-dopa), the precursor of dopamine, provides initial symptomatic relief, but abnormal involuntary movements develop later. A deeper understanding of the regulatory mechanisms underlying dopamine homeostasis is thus critically needed for the development of a successful treatment. Here, we show that p21-activated kinase 4 (PAK4) controls dopamine levels. Constitutively active PAK4 (caPAK4) stimulated transcription of tyrosine hydroxylase (TH) via the cAMP response element-binding protein (CREB) transcription factor. Moreover, caPAK4 increased the catalytic activity of TH through its phosphorylation of S40, which is essential for TH activation. Consistent with this result, in human midbrain tissues, we observed a strong correlation between phosphorylated PAK4S474, which represents PAK4 activity, and phosphorylated THS40, which reflects their enzymatic activity. Our findings suggest that targeting the PAK4 signaling pathways to restore dopamine levels may provide a new therapeutic approach in PD.

Discrete element modelling of geogrids with square and triangular apertures

  • Geogrid application that has proved to be an effective and economic method of reinforcing particles, is widely used in geotechnical engineering. The discrete element method (DEM) has been used to investigate the micro mechanics of the geogrid deformation and also the interlocking mechanism that cannot be easily studies in laboratory tests. Two types of realistically shaped geogrid models with square and triangle apertures were developed using parallel bonds in PFC3D. The calibration test simulations have demonstrated that the precisely shaped triangular geogrid model is also able to reproduce the deformation and strength characteristics of geogrids. Moreover, the square and triangular geogrid models were also used in DEM pull-out test simulations with idealized shape particle models for validation. The simulation results have been shown to provide good predictions of pullout force as a function of displacement especially for the initial 30 mm displacement. For the granular material of size 40 mm, both the experimental and DEM results demonstrate that the triangular geogrid of size 75 mm outperforms the square geogrid of size 65 mm. Besides, the simulations have given valuable insight into the interaction between particle and geogrid and also revealed similar deformation behavior of geogrids during pullout. Therefore, the DEM provides a tool which enable to model other possible prototype geogrid and investigate their performance before manufacture.

Changes of Physico-chemical Characteristic on Swine Manure Using Different Suction Strength in Composting System (돈분 퇴비화 시 공기 흡입 강도에 따른 이화학적 특성변화)

  • The aim of this experiment was to investigate the effect of air suction rate (SR) during the composting process of swine manure mixed with sawdust used as a bulking agent. In the 25 L composting reactors, the suction rate (SR) was at four different treatment levels (100%, 200%, 300%, 400%), and were fixed on the based on constant aeration rate into the composting mixtures. The temperature reached to thermophilic phase within 2 days and it was maintained up to the $5^{th}$ day of the composting process in all reactors and then gradually decreased to room temperature at the end of the composting process. The moisture content (MC, %) of the initial mixtures was 64.27%, and it was reduced to 38.4, 33.08, 14.59 and 11.93 in the different suction rate of 100%, 200%, 300%, 400%, respectively in the end process. During the composting, the level of pH was increased from 6.83 to 8.67 and it gradually decreased to 7.56 in 100% and 200%(SR). At the same time, the pH values were reduced only up to 8.19 at 300%, and 8.08 at 400%(SR), showing that suction strengths of 100% and 200% were the better option for composting than those of 300% and 400%. The total Kjeldahl nitrogen (TKN) of initial composts mixtures was 2.3% and were changed in 3.3, 3.1, 2.5, and 2.3% at the end of the composting period from the 100%-400% (SR) variations respectively. These results also indicated that 100% and 200% (SR) were more affected by the dry mass loss as $CO_2$ and water evaporation. The initial value of C/N ratio was 25.17 and were significantly reduced to 11.88, 11.97, 14.31, and 14.72 at the end of the experiment, respectively from the 100%-400% (SR) variations. These results suggest that the suction rate (SR) of 100% and 200% relative to constant air supply would be the optimal conditions to produce high-quality compost.

Clinical Outcomes of Ultrasound-Guided Transmeniscal Injection in Medial Compartment Knee Osteoarthritis (무릎 내측 구획 골관절염에서 초음파 유도하 경 반월상 연골 주사의 임상적 효과)

  • Purpose: The purpose of this study was to introduce the ultrasound-guided transmeniscal injection in medial compartment knee osteoarthritis and analyze the clinical outcomes. Materials and Methods: The electronic medical records of 36 patients with medial compartment knee osteoarthritis who were treated with an ultrasound-guided transmeniscal injection from March 2019 to July 2019 were accessed for this retrospective review. Using an ultrasound guided spinal needle, the patients received an intra-articular steroid injection at the medial compartment of the knee. A physical examination was conducted at the initial visit (pre-injection), and at one week, four weeks, and eight weeks after the injection. The numeric pain rating scale (NRS), Lequesne index, and Western Ontario and McMaster Universities Osteoarthritis (WOMAC) score were measured at each visit and analyzed over time. The percentage change of the patients who revealed substantial improvement was analyzed. The NRS, Lequesne index, and percentage of patients, who revealed substantial improvement over time classified by osteoarthritis grade, were analyzed. Results: The NRS and Lequesne index decreased at one week, four weeks, and eight weeks after the injection compared to the initial baseline, and the pain-relief effect continued without change until eight weeks. The percentage of patients who showed substantial improvement at one, four, and eight weeks was 50.0%, 47.2%, and 52.8%, respectively. The WOMAC scores decreased at one, four, and eight weeks compared to the initial baseline, and the decrease was continued without any difference until eight weeks. The percentage of patients with osteoarthritis stage 1 or 2 who revealed more than substantial improvement was significantly higher at one, four, and eight weeks than those with osteoarthritis stages 3 or 4 (p<0.05). Conclusion: In patients with medial compartment knee osteoarthritis, the pain reduction and functional improvement persisted for at least eight weeks after the ultrasound-guided transmeniscal injection at the medial compartment. In particular, patients with medial compartment osteoarthritis stage 1 or 2 showed more effective pain reduction.

Study on the Extending Storage Life of Grape by Applying Edible Coating Materials (가식성 코팅물질을 이용한 포도의 저장성 연장 연구)

  • This study was conducted to increase the shelf life of grape by edible coating material such as methyl cellulose (MC) with antimicrobial substances, n-capric acid isopropyl ester (ci) and sodium nitrate (sn), added by spraying method. The quality changes of packaged grapes with wrapping PE film on EPS tray were investigated for 16 days at $30{\circ}C$. The shelf-lives of C and MCci based on the weight reduction ratio of 7% were 6 days and 9 days, respectively. The reduction rate of acidity of C was higher value than those of treatments during 18 days of storage at $30{\circ}C$. The vitamin C reduction ratios of C, MCsn and MCci were 64.8, 51.5 and 49.8%, respectively, after 16 days at $30{\circ}C$. The reduction rates of firmness of C, MCsn and MCci after 16 days at $30{\circ}C$ were 44.2, 26.5, and 23,2%, respectively compared to that of initial storage grapes. The additions of ci and sn had much affected the reductions of bacteria and yeast counts especially early stage of storage. The hedonic sensory evaluation scores of MCci and MCsn had higher values than those of MC.

  • SIR model (Kermack and McKendrik, 1927) is one of the most popular method to explain the spread of disease, In order to construct SIR model, we need to estimate transition rate parameter and recovery rate parameter. If we don't have any information of the two rate parameters, we should estimate using observed whole trajectory of pandemic of disease. Thus, with restricted observed data, we can't estimate rate parameters. In this research, we introduced Reed-Frost model (Andersson and Britton, 2000) to calculate the probability of infection in the early stage of pandemic with the restriction of data. When we have an initial number of susceptible and infected, and a final number of infected, we can apply Reed - Frost model and we can get the probability of infection. We applied the Reed - Frost model to the Vibrio cholerae pandemic data from Republic of the Cameroon and calculated the probability of infection at the early stage. We also construct SIR model using the result of Reed - Frost model.

The Effect of Chinemys reevesii Gray Pharmacopuncture for Women with Knee Osteoarthritis (구판(龜板)약침이 여성의 퇴행성 슬관절염에 미치는 영향)

  • Objectives : The aim of this study is to determine whether Chinemys reevesii Gray pharmacopuncture is a clinically effective treatment for women with knee osteoarthritis. Methods : Female patients with knee osteoarthritis were recruited from May $1^{st}$, 2014 to April $15^{th}$, 2015 at the Department of Acupuncture & Moxibustion Medicine of Dong-Eui University Korean Medical Hospital. Patients were randomly assigned to two groups. 4.0 ml of Chinemys reevesii Gray pharmacopuncture was injected at acupuncture points of the experimental group(n = 24) twice a week for three weeks. The control group(n = 24) received 5 % dextrose injections in order to identically stimulate the same acupuncture points of $ST_{35}$, $ST_{34}$, $SP_9$, $SP_{10}$, $GB_{34}$, $LR_8$, $EX-LE_4$, and $EX-LE_2$. The Short-Form McGill Pain Questionnaire(SF-MPQ), the Korean Western Ontario and McMaster Universities Osteoarthritis Index(KWOMAC), and the European Quality of Life-5 Dimensions(EQ-5D) were recorded to evaluate treatment efficacy at first visit, after two weeks and four weeks from the initial visit. Results : 40 patients completed the trial. The experimental group showed more significant improvement than the control group in the scores of SF-MPQ, KWOMAC, and EQ-5D. However, there was no significant difference between groups in a few of the assessment scales. Conclusions : Chinemys reevesii Gray pharmacopuncture can be an effective treatment in control of pain, improvement in function and health-related quality of life for women with knee osteoarthritis.

Cellular and Molecular Roles of $\beta$ Cell Autoantigens, Macrophages and T Cells in the Pathogenesis of Automimmune Diabetes

  • Type I diabetes, also known as insulin-dependent diabetes mellitus (IDDM) results from the destruction of insulin-producing pancreatic $\beta$ cells by a progressive $\beta$ cell-specific autoimmune process. The pathogenesis of autoimmune IDDM has been extensively studied for the past two decades using animal models such as the non-obese diabetic (NOD) mouse and the Bio-Breeding (BB) rat. However, the initial events that trigger the immune responses leading to the selective destruction of the $\beta$ cells are poorly understood. It is thought that $\beta$ cell auto-antigens are involved in the triggering of $\beta$ cell-specific autoimmunity. Among a dozen putative $\beta$ cell autoantigens, glutamic acid decarboxylase (GAD) has bee proposed as perhaps the strongest candidate in both humans and the NOD mouse. In the NOD mouse, GAD, as compared with other $\beta$ cell autoantigens, provokes the earliest T cell proliferative response. The suppression of GAD expression in the $\beta$ cells results in the prevention of autoimmune diabetes in NOD mice. In addition, the major populations of cells infiltrating the iselts during the early stage of insulitis in BB rats and NOD mice are macrophages and dendritic cells. The inactivation of macrophages in NOD mice results in the prevention of T cell mediated autoimmune diabetes. Macrophages are primary contributors to the creation of the immune environment conducive to the development and activation of $\beta$cell-specific Th1-type CD4+ T cells and CD8+ cytotoxic T cells that cause autoimmune diabetes in NOD mice. CD4+ and CD8+ T cells are both believed to be important for the destruction of $\beta$ cells. These cells, as final effectors, can kill the insulin-producing $\beta$ cells by the induction of apoptosis. In addition, CD8+ cytotoxic T cells release granzyme and cytolysin (perforin), which are also toxic to $\beta$ cells. In this way, macrophages, CD4+ T cells and CD8+ T cells act synergistically to kill the $\beta$ cells in conjunction with $\beta$ cell autoantigens and MHC class I and II antigens, resulting in the onset of autoimmune type I diabetes.

Effect of Buffers on Citric Acid Production by Aspergillus niger NRRL 567 in Solid Substrate Fermentation (Aspergillus niger NRRL 567을 이용한 고체배양에서 완충용액이 구연산 생산에 미치는 영향)

  • In the submerged fermentation of fungi, it was known pH had significant effect on the citric acid production. Various growth conditions were applied with different buffer on citric acid production by Aspergillus niger NRRL 567 grown on peat moss to find the optimum pH and most effective buffer solution. The initial pHs of different buffer solutions significantly influenced on the citric acid production and A. niger NRRL 567 produced citric acid more efficiently at high pHs. A phosphate buffer and a carbonate buffer with pH 8.6 and pH 10.0 were identified as suitable buffer solutions for citric acid production. The maximal citric acid production of 564.3 g/kg solid substrate was achieved employing carbonate buffer at pH 10.0.
