Synthesis and Nrf2 Activating Ability of Thiourea and Vinyl Sulfoxide Derivatives

  • Thiourea and vinyl sulfoxide derivatives were designed based on the structures of sulforaphane and gallic acid, prepared and tested for HO-1 inducing activity as a measure of Nrf2 activation, and inhibitory effect on NO production as a measure of anti-inflammatory activity. Both series of compounds showed moderate activity on HO-1 induction, and no inhibitory effect on NO production. Interestingly the thiourea compound 6d showed better HO-1 induction (71% SFN) than the corresponding isothiocyanate compound 6a (55% SFN). Overall, it seemed that more efficient electrophile is needed to get more effective Nrf2 activator.

Potential Biological Activities of Magnoflorine: A Compound from Aristolochia debilis Sieb. et Zucc

  • Magnoflorine, an important compound in Aristolochia, was usually used as an anxiolytic chemical. In this study, the magnoflorine was isolated from Aristolochia and the biological activities such as antioxidant, ${\alpha}$-tyrosinase inhibitory, anti-inflammatory, and anticancer activities were investigated. The magnoflorine showed significant antioxidant activity as a 2,2-diphenyl-2-picryl-hydrazyl (DPPH) free radical scavenger, $50{\mu}g/mL$ of the magnoflorine scavenged about 70.8% of all the free radicals. And it was good at ${\alpha}$-tyrosinase inhibiting, $100{\mu}g/mL$ of the magnoflorine inhibited 36.5% of the tyrosinase. High dosage of magnoflorine inhibited the inflammation production nitric oxide (NO), and the magnoflorine protected the murine macrophage cells (RAW 264.7) from LPS-induced apoptosis. The cell viability of human colon cancer calls (HT-29) was around 100% when treated with different dose of magnoflorine, it's suggesting that magnoflorine had no anticancer effect.

Purification and Characterization of Angiotensin I-Converting Enzyme Inhibitors from Sinapis alba L.

  • To separate ACE inhibitors from edible plants, spices, and herbs, 285 extracts of 95 sources were screened for ACE inhibitory activity. The extract of Sinapis alba L. had the most potent ACE inhibitory activity. Mustard seeds were crushed homogeneously and extracted with hexane and water successively. Lyophilized water extract was fractionated with $H_2O$:butanol(1:1). The ACE inhibitor was purified from butanol fraction by methanol precipi-tation, gel filtration, HPLC, and FPLC with Superdex peptide HQ 10/30 column. The active fraction has been purified to homogeneity, which was proven by gel filtration using FPLC system. The yield was 0.02%. The com-pound has a molecular weight of about 640. The compound competitively inhibited ACE activity and the $IC_{50}$ value was 79$\mu\textrm{g}$/ml. The purified compound showed uterus contraction activity in isolated rat uterus.

  • Tyrosinase is an important enzyme involved in the transition steps from tyrosine to melanin. Inhibition of the tyrosinase activity could block melanin formation from tyrosine and thus prevent melanin pigmentation on skin. This may contribute to the development of new whitening agent that would be useful in the prevention of pigmentation. in this study, we isolated tyrosinase inhibitory compound from BuOH fraction of Potentilla bifurca L. var. glabrata Lehm by activity guided fractionation method. Based on spectroscopic data, the active compound was identified as a quercetin 4"-O- glucopyranoside.

Experimental Studies on the Inhibitory Effect of Immediate-Type Allergic Reaction of Tongku-tang (통규탕의 즉각형 알레르기 반응 억제 효과에 관한 실험적 연구)

  • This report describes an inhibitory effect of Tongku-tang(TKT) on mast cell-mediated immediate-type allergic reactions. TKT is an Oriental herbal prescription, which has been successfully applied for the treatment of allergic disorders, mainly skin anaphylactic diseases in eastern medicine. TKT has concentration-dependently inhibited the ear swelling response induced by intradermal injection of non-specific mast cell degranulator compound 48/80 in mice. TKT also inhibited mast cell-dependent passive cutaneous anaphylaxis activated by dinitrophenyl (DNP)-IgE antibody in rats. I studied the effect of TKT on the histamine and β-hexosaminase release from the rat peritoneal mast cells by compound 48/80. TKT did not inhibit significantly the histamine and β-hexosaminase release from the rat peritoneal mast cells by compound 48/80. However, TKT inhibited both TNF-α and IL-1β secretion induced by phorbol 12-myristate 13-acetate and A23187 respectively. These results provide evidence that TKT may be beneficial in the treatment of immediate-type allergic reaction.

  • Phenylaminopyrimidines represent a large group of new selective anticancer agents, the majority of which exert their action through the inhibition of specific kinases. In this study, a new series of N-substituted-2-aminopyrimidines has been designed and synthesized. A selected group of the synthesized derivatives was screened at a single dose concentration of 10 ${\mu}M$ over a panel of 60 cancer cell-lines. Compound 12e has showed great inhibitory and strong lethal effect over almost all of the 60 cell-lines and accordingly was further tested in a 5-dose testing mode to determine its $IC_{50}$ values, where it showed great efficacies with intermediate potencies over the tested cell-lines. The compound was also tested over a panel of 52 kinases to explore its kinase inhibitory profile, and was found to be a selective but moderate inhibitor over FLT3 kinase.

Clitocybin D, a Novel Human Neutrophil Elastase Inhibitor from the Culture Broth of Clitocybe aurantiaca

  • Clitocybin D, a novel human neutrophil elastase inhibitor, was isolated from the culture broth of Clitocybe aurantiaca. This compound was purified by solvent extraction, silica gel column chromatography, Sephadex LH-20 column chromatography, and preparative HPLC. The compound was determined to be 4-(4,6-dihydroxy-3-methoxy-3H-isoindol-1-yl)-benzoic acid on the basis of 1D and 2D NMRs and MS spectroscopic analysis. Analysis of the human neutrophil elastase (HNE) inhibitory activity of the isolated compound revealed that it showed significant HNE inhibitory activity with an $IC_{50}$ value of $17.8{\mu}M$.

Protective Effects of Chalcone Derivatives for Acute Liver Injury in Mice

  • The hepatoprotective effects of chalcone derivatives were evaluated in D-galactosamine/lipopolysaccharide (D-GaIN/LPS)-induced fulminant hepatic failure in mouse. Thirteen chalcone derivatives were synthesized for study and their hepatoprotective effects were evaluated by assessing aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels in serum. Chalcone preparations were injected into mice at 12 hand 1 h before intraperitoneal injection of D-GaIN/LPS. After abdominal administration, changes in AST and ALT between the control and treated groups were observed. Ten of the synthesized chalcone derivatives exhibited inhibitory effects on D-GaIN/LPS-induced levels of AST and ALT in mice. Compounds 2, 3, 8, 9, and 12 markedly reduced serum AST and ALT at 8 h, inhibited hepatocyte necrosis and showed significant hepatoprotective activities. The activity of compound 3 was compared with the bifendate (DDB) through oral administration. Compound 3 showed much higher inhibitory effects than bifendate for decreasing AST and ALT activity. The results indicate that compound 3 has strong hepatoprotective activity through suppression of tumor necrosis factor­alpha (TNF-alpha) preduction, reduction of the histological change in the liver, and attenuated of hepatocyte apoptosis confirmed by DNA fragmentation assay.

Inhibitory Effect of Mast Cell-dependent Anaphylaxis by Gleditsia sinensis

  • We investigated the effect of aqueous extract of Gleditsia sinensis thorns (Leguminosae) (GSAE) on the mast cell-dependent anaphylaxis. GSAE (0.005 to 1 ${g}/kg$) dose-dependently inhibited systemic anaphylaxis induced by compound 48/80 in rats. GSAE (0.1 and 1 ${g}/kg$) also significantly inhibited local anaphylaxis activated by anti-DNP IgE. When GSAE was pretreated at the same concentrations with systemic anaphylaxis, the plasma histamine levels were reduced in a dose-dependent manner. GSAE (0.001 to 1 ${m}g/ml$) dose-dependently inhibited the histamine release from rat peritoneal mast cells (RPMC) activated by compound 48/80 or anti-DNP IgE. The level of cyclic AMP in RPMC, When GSAE (1 ${m}g/ml$) was added, transiently and significantly increased about fourfold compared with that of basal cells. Moreover, GSAE (0.01 and 0.1 ${m}g/ml$) had a significant inhibitory effect on anti-DNP IgE-induced tumor necrosis factor-$\alpha$ production from RPMC. These results suggest a possible use of GSAE in managing mast cell-dependent anaphylaxis.

Isolation and Structural Analysis of MB4-03, an $\alpha$-Amylase Inhibitor Produced by Streptomyces sp. DMCJ-49 (Streptomyces sp. DMCJ-49 균주가 생산하는 Alpha-Amylase 저해제 MB4-03의 분리와 구조분석)

  • MB4-03, an $\alpha$-amylase inhibitor was isolated from the culture broth of Streptomyces sp. DMCJ-49 and purified through ion-exchange chromatography, adsorption, and gel filtration. The results of various instrumental analyses showed that the inhibitor was one of oligosaccharides that had glucoses as its major component and that its molecular weight was about 2000. And one methyl group which seemed to be related with the inhibitory activity of this compound was identified. From the CMR spectrum, it was elucidated that this compound was composed of $\alpha$ -D-glucopyranoses which were linked together by $\alpha$ (I -, 4) bond configuration. As the inhibitory effect of this compound was reduced after incubation with $\beta$-amylase, the maltose units was seemed to exist at non-reducing terminal side of it.
