Inhibitory Effect of Corynoline Isolated from the Aerial Parts of Corydalis incisa on the Acetylcholinesterase

  • In the course of screening Korean natural products for acetylcholinesterase (AChE) inhibitory activity, it was found that a methanolic extract of the aerial parts of Corydalis incisa (Papaveraceae) showed significant inhibitory effects on AChE. Corynoline isolated from this plant inhibited AChE activity in a dose-dependent manner, and the $IC_{50}$ value of corynoline was $30.6{\;}{\mu}M$. The AChE inhibitory activity of corynoline was reversible and noncompetitive.

  • The germination of Lactuca sativa seeds was significantly inhibited by the water extract of the fresh leaves of Pulsatilla koreana N. including abundant ranunculin. Germination inhibitory activity increased in a dose-dependantly. Protoanemonin, produced from ranunculin by enzymatic action during maceration process of leaves, was proved to be the active principle with inhibitory activity was above 90% at concentration of 1 mg/ml.

  • Three flavonoids (1 - 3) and three phenolic compounds (4 - 6) were isolated from the whole plant of Geranium thunbergii Sieb. et Zucc (Geraniaceae). Their structures were determined by chemical and spectral analysis. These compounds were examined for the inhibitory activity of IL-6 production in $TNF-{\alpha}$ stimulated MG-63 cell. Among the isolated compounds, gallic acid (4) and gallic acid methyl ester (6) showed potent inhibitory activity.

  • Nineteen hydrolysable tannins isolated from the Euphorbia helioscopia (Euphorbiaceae) were tested for the inhibitory effect on mushroom tyrosinase activity in vitro. Inhibitory effect of gallotannin group exhibited more potent than that of phenolcarboxylic acid and ellagitannin groups against the enzyme activity. The inhibitory activity by pentagalloyl glucose on mushroom tyrosinase was more potent ($IC_{50}$/, 4.9 $\mu$M) than that of kojic acid ($IC_{50}$/, 8.7 $\mu$M).

  • Four derivatives of kojic acid were synthesized and their inhibitory activities against tyrosinase were evaluated. The C-2 hydroxymethyl and C-7 hydroxyl of kojic acid were replaced by carboxylate and amine, respectively. These derivatives were coupled to L-phenylalanine, producing two amide compounds. The carboxylate derivative (3), its amide compound (5), and the amine derivative (7) were weak inhibitors. The amide compound (9) where amine derivative (7) coupled to L-phenylalanine showed strong inhibitory activity ($IC_{50}=24.6{\;}{\mu}M$) comparable to kojic acid.

  • Thirty ellagitannins originated from some Euphorbiaceous plants were tested for the inhibitory activities against AMV reverse transcriptase and replication of HIV-1 using syncytia forming assay. Most of ellagitannins showed strong inhibitory activities against AMV reverse transcriptase. Some ellagitannins including geraniin, mallotusinin, elaeocarpusin, euphorscopin and jolkianin, showed significant inhibitory activities of syncytia formation of supT1 cell line.

The Inhibitory Effects of Cheonsagunja-tang and Leech on the IL-6, IL-16, GM-CSF mRNA Level by Human Epithelial Cells (천사군자탕(喘四君子湯)과 수질(水蛭)(마황초(麻黃炒))가 인간 기관지상피세포의 IL-6, IL-16, GM-CSF 발현에 미치는 영향)

  • Objectives : We aimed to identify the dose-dependent inhibitory effects of Cheonsagunja-tang(喘四君子湯), leech(Hirudo medicinalis/水蛭) roasted with Ephedrae Herba(麻黃) on the mRNA expression of IL-6, IL-16, GM-CSF involved in the asthma model. Methods : In the study BEAS-2B cell lines, human epithelial cells were used. These cells were stimulated with tumor necrosis factor(TNF)-${\alpha}$ for artificial inflammatory expression. ${\beta}$-actin messenger RNA(mRNA) was used by internal standard. After 24 hours of the Cheonsagunja-tang, leech-treatment, total cellular RNAs were collected treating RNA zol directly on the living cells. Then the transcriptional activities of IL-6, 16 and GM-CSF were measured by RT-PCR with electrophoresis. Results: In the Cheonsagunja-tang study, the mRNA expression of IL-6 showed 30% transcriptional inhibitory effect compared to the control group in the $100{\mu}l/ml$ category(p<0.005). In the IL-16, there was 26%, 31% and 31% transcriptional inhibitory effect compared to the control groups in the $4{\mu}l/ml$, $20{\mu}l/ml$ and $100{\mu}l/ml$ categories, respectively(p<0.05). In the GM-CSF, the experimental group had 56% transcriptional inhibitory effect compared to the control group in the $100{\mu}l/ml$ category(p<0.001). In other concentrations, there was no inhibitory effect. In the leech study, the mRNA expression of IL-6 showed 37% transcriptional inhibitory effect compared to the control group in the $100{\mu}l/ml$ category(p<0.001). In the IL-16, there was 63% and 67% transcriptional inhibitory effect compared to the control groups in the $20{\mu}/ml$ and $100{\mu}/ml$ categories, respectively(p<0.001). In the GM-CSF, there was 64% and 68% transcriptional inhibitory effect compared to the control groups in the $20{\mu}l/ml$ and $100{\mu}l/ml$ categories, respectively(p<0.001). In other concentrations, there was no inhibitory effect. Conclusions : This study shows that Cheonsagunja-tang and leech roasted with Ephedrae Herba have dose-dependent inhibitory effects on the mRNA expression of IL-6, IL-16 and GM-CSF in BEAS-2B cell lines, human epithelial cells.

Changes of Angiotensin I-Converting Enzyme Inhibitory Activity, Fibrinolytic Activity and $\beta$-Secretase Inhibitory Activity of Red Wines During Fermentation and Post-Fermentation (적포도주들의 발효와 후 발효 중 심혈관 관련 Angiotensin I 전환효소 저해활성과 혈전용해활성 및 $\beta$-secretase 저해 활성의 변화)

  • The cardiovascular angiotensin I-converting enzyme inhibitory activity, fibrinolytic activity and bbb-secretase inhibitory activity of four kinds of red wine were investigated during fermentation and post-fermentation. After 10 days of fermentation, the antihypertensive angiotensin I-converting enzyme (ACE) inhibitory activities of all the red wines ranged from 38.6% to 58.8%. However, the ACE inhibitory activities increased with the prolongation of the post-fermentation period; moreover, in the Vitis hybrid red wine, the ACE inhibitory activity reached its highest value, 76.9%, after 120 days of post-fermentation. During the fermentation and post-fermentation of all the red wines, fibrinolytic activity was weak or not detected. After 10 days of fermentation, Vitis labrusca B red wine exhibited the greatest antidementia $\beta$-secretase inhibitory activity of 54.8%, though $\beta$-secretase inhibitory activity decreased significantly to less than 10% during 120 days of post-fermentation. In conclusion, we obtained a highly valuable Vitis hybrid red wine that was fermented for 10 days at $25^{\circ}C$ with Vitis hybrid and S. cerevisiae K-7 and then post-fermentation for 120 days at $4^{\circ}C$.

Optimization of Tyrosinase Inhibitory Activity in the Fermented Milk by Lactobacillus plantarum M23 (Lactobacillus plantarum M23 균주를 이용한 Tyrosinase 저해 활성 발효유 생산의 최적화)

  • The melanin pigment in human skin is a major defense mechanism against ultraviolet light to the skin, but darken skin color. Tyrosinase is mainly responsible for melanin biosynthesis (melanogenesis) in animals and enzymatic browning (melanosis) in plants. The purpose of this study was to optimize the fermented milk process for the melanin formation inhibition by using Lactobacillus plantarum M23 with tyrosinase inhibitory activity. We used 4-factor-3-level central composite design combining with response surface methodology. Yeast extract concentration (%, $X_1$), addition of grape (%, $X_2$), incubation temperature ($^{\circ}C$, $X_3$) and incubation time (h, $X_4$) was used as an independent factor, on the other hand, pH (pH, $Y_1$), overall palatability (score, $Y_2$) and tyrosinase inhibitory activity (%, $Y_3$) was used as a dependant factor. Based on the optimization for the highest tyrosinase inhibitory activity with pH 4.4, the expected data of pH, palatability and tyrosinase inhibitory activity with 14.8 h incubation at $37.1^{\circ}C$ by the addition of 0.127% of yeast extract, 2.95% of grape was 4.42, 7.06 and 86.65%, but the real data was 4.35, 6.86 and 84.05%, respectively. Based on the previous results, fermented milk using Lactobacillus plantarum M23 with the tyrosinase inhibitory activity could contribute for the whitening and antiaging of human skin.

Carbohydrate, Lipid Inhibitory Activity and Antioxidant Activity of Extracts from Several Economic Resource Plants in Vitro

  • The objective of this study was determined to evaluate ${\alpha}$-amylase, ${\alpha}$-glucosidase, pancreatic lipase inhibition in vitro and DPPH radical scavenging activity of the several Korean resources plants. The ${\alpha}$-amylase inhibitory activity of Salicornia herbacea, Erythronium japonicum (flower) and Phragmites communis (root) in water extract showed relatively high 62.8%, 66.5% and 69.3%, respectively. The ${\alpha}$-amylase inhibitory activity of Citrus junos (pericarp) and Cornus officinalis in methanol extract was found to have an effect with 32.8% in Citrus junos (pericarp) and 60.9% in Cornus officinalis. Corylopsis coreana in both water and methanol extract had the highest ${\alpha}$-glucosidase inhibitory activity of 81.7% and 89.5%, while the extract of Portulaca oleracea, Ficus carica and Citrus junos was not measured ${\alpha}$-glucosidase inhibitory activity at given experiment concentration. Depending on the extraction solvent and the plant species, it was observed that there was a significant difference in ${\alpha}$-glucosidase inhibitory activity. The pancreatic lipase inhibitory activity showed relatively higher in the methanol extract than water extract except pericarp of Citrus junos. The DPPH radical scavenging activity of selected plants was much difference between measured plant species, and showed that the increase was proportional to the concentration. These results suggested that selected plants had the potent biological activity on carbohydrate, lipid Inhibitory activity and antioxidant activity, therefore these plant resources could be a good materials to develop medicinal preparations, nutraceuticals or health functional foods for diabetes or obesity.
