  • The purpose of this study was to examine the skin regeneration ability of Lespedeza cuneata extract (LC) as well as verify its wound healing effects on an open wound in rats. Rats were divided into six groups (NO, saline treated group; CO, 1% carboxy methyl cellulose (CMC) treated group; PC, fucidin treated group; LCL, 1% LC treated group; LCM, 3% LC treated group; LCH, 5% LC treated group), and the experimental material was applied for 5 weeks. Elastase inhibition rate of the LCM group was 2.7% lower than that of butylated hydroxy anisole (BHA), which is an antioxidant. Futher, the collagenase inhibition rate of the LCM showed 7% higher activity than that of BHA. The left wound areas in the LCL group, LCM group, and LCH group after the 21st day were noticeably reduced in wound area by 54.2%, 53.5%, and 48.7%, respectively, compared to the CO group. This suggests that Lespedeza cuneata extract has healing effects on surgical wounds by promoting regeneration of skin epithelial tissue and synthesis of collagen.

  • The insecticide p-nitropheny diethyl thiophospate is alse known by the symbol E.605 and a legion of trade names including “parathion”. The insecticide is widely used in agriculture, but it is highly toxic and now clear that parathion behaves like a cholinergic drug by inhibiting the enzyme cholinesterase. In order to know acute toxicity and the changes of glucose concentrations and activity according to time lapsed in female mice given orally single with the half dose to $LD_{50}$ of parathion, glucose contents and cholinesterase activities in serum as well as cholinesterase activities in whole brain and spinal cord were investigated, otherwise median lethal dose ($LD_{50}$) of parathion given orally against female mice was determined. The results obtained were summerized as follows ; 1. $LD_{50}$ value of parathion given orally to female mice was 7.1mg/kg(95% confidence limits, 3.8-13.1mg/kg) 2. The inhibition rate of cholinesterase activities in serum of parathion-administrated mice according to time lapsed were peakly decreased to 61% after 30 minutes in comparison to control group, but activities were completely recovered after 48 hours. 3. The inhibition rate of cholinesterase activities in whole brain of parathion-administrated mice according to time lapsed were peakly decreased to 49% after 2 hours in completely recovered after 24 hours. 4. The inhibition rate of cholinesterase activities in spinal cord of parathion-administrated mice according to time lapsed were peakly decreased to 57% after 2 hours in comparison to control group, but activities were completely recovered after 48 hours. 5. The changes of glucose contents in serum of parathion-administrated mice according to time lapsed and in directly after death due to parathion poisoning were no significantly difference.

  • The isolated ${\kappa}-Casein$ on gel permeation chromatography was hydrolyzed by chymosin, trypsin, and pepsin. The 3% TCA soluble portion of the hydrolysates were dialyzed on the angiotensin-I converting enzyme (ACE) inhibition rate (%,) and inhibitory activity $(IC_{50})$ were determined. The trypsin hydrolysate exhibited the highest ACE inhibition rate while the chymosin hydrolysation showed the lowest activity. The hydrolysate was dialyzed using dialysis membrane with various molecular cut-offs, and $IC_{50}$ was determined. As the pore size of the dialysis tubing increased, the ACE inhibitory activity decreased.

  • It was proved that the cell growth followed a photo-inhibition model in light-limited turbidostat cultivation, having 1.06 (1/h) of maximum specific growth rate and 0.00094(kcal/$cm^2$/h) and 0.063 (kcal/$cm^2$/h) as half saturation and light inhibition constants, repectively. ${\beta}$-carotene production showed a growth related porcess. And the activation energy of Dunaliella salina was roughly estimated as 12.36 (kcal/mole) in employing Arrhenius relationship. It should also point out that relatively much porduction of ${\beta}$-carotene was observed at hight light intensity with yieding 1.04 (mg-carotene/g-dry cell/day) of specific product production rate while the cell growth was decreased. The optimal conditions of producing ${\beta}$-carotene in turbiodostat cultivation were as follows: $7.5{\times}10^{-3}$(kcal/$cm^2$/h)of light intensity, 2 (mM) and 50(mM) of nitrate and sodium bicarbonate concentrations and 100(ml/h) of $CO_2$ flow rate.

  • Denitrification rate was determined for various freshwater wetland types in the Mississippi River Coastal delta plain. Site 1 and 4 were collected from forested-tupelo dominated wetland, and site 2 and 3 were from floating emergent marsh. The maximum $N_2O$ emission was $7.47mg\;N\;m^{-2}$ for site 1 at day 6 after the addition of nitrate, $6.96mg\;N\;m^{-2}$ for site 2 at day 4, $6.63mg\;N\;m^{-2}$ for site 3 at day 3, and $9.64mg\;N\;m^{-2}$ for site 4 at day 4. The denitrification rate was determined using the acetylene inhibition method $1.24mg\;N\;m^{-2}d^{-1}$ for site 1, $1.93mg\;N\;m^{-2}d^{-1}$ for site 2, $2.24mg\;N\;m^{-2}d^{-1}$ for site 3, and $2.78mg\;N\;m^{-2}d^{-1}$ for site 4. The maximum denitrification rate was in the order of site 4 > site 3 > site 2 > site 1.

  • Huperzine A-loaded microspheres composed of poly(D,L-lactide-co-glycolide) were prepared by an O/w emulsion solvent evaporation method. The characterization of the microspheres such as drug loading, size, shape and release profile was described. The in vitro release in the initial 7 days was nearly linear with $10\%$ released per day. Thereafter drug release rate became slow gradually and about $90\%$ drug released at day 21. The in vitro release rate determined by dialysis bag method had a good correlation with the in vivo release rate. Huperzine A aqueous solution was intramuscularly injected (i.m.) at 0.4mg/kg and microspheres were intramuscularly injected at 8.4 mg eq huperzine A/kg in rats. The maxium plasma concentration $(C_{max})$ after i.m. microspheres was only $32\%$ of that after i.m. solution. Drug in plasma could be detectd until day 14 and about $5\%$ of administered dose was residued at the injection site at day 14. The relative bioavailability of huperzine A microspheres over a period of 14 days was $94.7\%$. Inhibition of acyecholinesterase activity (AchE) in rat's cortex, hippocampus and striatum could sustain for about 14 days. In conclusion, huperzine A-loaded microspheres possessed a prolonged and complete drug release with significant inhibition of AchE for 2 weeks in rats.

  • Spin-filters are used as cell separation devices for achieving high cell density and high productivity in animal cell culture. We have proposed a model for the cell growth in a spin-filter perfusion culture and examined the effects on cell growth by several parameters including ammonia inhibition, specific growth rate, specific feeding rate, and cell retention. Results from computer simulation and sensitivity analysis indicate that the cell retention affects the cell growth mostly while there is a significant inhibition on cell growth by the ammonia accumulated during the culture. The specific feeding rate has minimal effects on cell growth, which is consistant with the fact that the cell growth with a step feeding is quite similar to that with a continuous feeding.

  • Using Pseudomonas sp. B3, identified and isolated from nature, wastewater containing phenol was treated in a continuous stirred tank reactor and its reaction characteristics were studied. Average concentrations of phenol and COD in effluents were 1.5mg/L and 124mg/L at 0.059h-1 dilution rate, respectively. At the dilution rate higher than 0.063h-1, phenol and COD increased abruptly to 19mg/L and 318mg/L. At the dilution rate higher than 0.059h-1, biomass concentration suddenly decreased and was "washed out". Biomass concentration was 150mg/L at a dilution rate of 0.067h-1. Maximum biomass production rate was 15.98mg/L$.$h at a dilution rate of 0.067h-1. When dilution rate increased above 0.059h-1, effluent phenol concentration abruptly increased and biomass production rate decreased. Maximum cell growth rate(${\mu}$max) and Michaelis-Mentens kinetic constant(Ks) were 0.074h-1 and 0.424mg/L, respectively. From the above result low phenol concentration can be expected at a maximum dilution rate, but reactor becomes unstable due to phenol inhibition.

  • Powdery mildew is one of the most devastating diseases in cucurbits. Crop yield can decline as the disease severity increases. In this study, we evaluated the effect of silver nanoparticles against powdery mildew under different cultivation conditions in vitro and in vivo. Silver nanoparticles (WA-CV-WA13B) at various concentrations were applied before and after disease outbreak in plants to determine antifungal activities. In the field tests, the application of 100 ppm silver nanoparticles showed the highest inhibition rate for both before and after the outbreak of disease on cucumbers and pumpkins. Also, the application of 100 ppm silver nanoparticles showed maximum inhibition for the growth of fungal hyphae and conidial germination in in vivo tests. Scanning electron microscope results indicated that the silver nanoparticles caused detrimental effects on both mycelial growth and conidial germination.
