Biofunctional Characteristics of the Water Soluble Browning Reaction Products Isolated from Korean Red Ginseng-Study on the Antimutagenic and Nitrite Scavenging Activities (홍삼으로부터 분리한 수용성 갈변물질의 생리기능적 특성- 돌연변이원성 및 아질산염 소거능을 중심으로)

  • The purpose of this study was to investigate the biofunctional characteristics of water soluble browning reaction products(WS-BRPs) isolated from Korean red ginseng. The antimutagenic effect of WS-BRPs(L, S-1 and S-2 fraction) showed 23∼61% inhibition rates against the indirect mutagen, imidazol quinoline(IQ) induced mutagenicity. Especially, L fraction exhibited a little higher mutagenicity than those of other fractions on Salmonella typhimurium TA 98 and TA 100 at a concentration of 400 $\mu\textrm{g}$/m/, respectively, and it showed about 16∼39% inhibition rates on the nitrite scavenging. L fraction exhibited a little higher nitrite scavenging than those of S-1 and S-2 fractions.

Growth Rate and Antimicrobial Activity of Lactobacillus plantarum Reacting with Bacterial Culture Supernatant (세균 배양액과 반응한 Lactobacillus plantarum의 생장속도 및 항균 활성에 대한 연구)

  • In this study, L. plantarum, when reacting with the culture media of potential pathogenic bacteria, exhibited an increase in growth rate and antimicrobial activity. In order to examine the characteristics and the nature of the reaction with the bacteria, this study carried out experiments involving culturing the test bacteria in M9 minimal media. Subsequently, the supernatant was incrassated by the decompression-drying method. Through colony forming unit assay, it was confirmed that L. plantarum had the function of growth inhibition to various bacteria. After culturing L. plantarum with bacterial media, the growth rate of L. plantarum was measured by absorbance (OD600), the results showed that the growth rate (E. coli treatment group: OD600 = 0.848, S. typhimurium treatment group: OD600 = 0.848) increased, as compared with the non-treated control group (OD600 = 0.48). In contrast, the concentrate itself did not induce the growth of L. plantarum. These results were observed as a universal phenomenon of the Lactobacillus species. Moreover, the increase in antimicrobial activity was observed in L. plantarum, which reacted with the culture media of E. coli and S. typhimurium, through a disc diffusion assay, and the result of growth inhibition against various bacteria was induced. Finally, based on the analysis results of the characteristics of bacteria culture media, which increased the growth rate of L. plantarum and antibacterial activity, the bacterial media had a tolerance for catabolic enzymes, pH 2−8 and heat. Therefore, this substance can be said to be a small molecule which is highly stable under various conditions.

  • In order to clarify the functional role of Tyr7 in human glutathione S-transferase P1-1, we extensively investigated the effect of mutation of Tyr7 on the substrate specificity and inhibition characteristics. The mutational replacement of Tyr7 with phenylalanine lowered the specific activities with 1,2-dichloro-4-nitrobenzene and 1,2-epoxy-3-(p-nitrophenoxy) propane for GSH-conjugation reaction to 3~5% of the values for the wild-type enzyme. The pKa of the thiol group of GSH bound in Y7F was about 2.4 pK units higher than that in the wild-type enzyme. The $I_{50}$ of hematin for Y7F was similar to that for the wild-type enzyme and those of benastatin A and S-(2,4-dinitrophenyl)glutathione were only moderately decreased. These results suggest that Tyr7 is considered to be important the catalytic activities not only for GSH-chloronitrobenzene derivatives but also for GSH-epoxide conjugation reaction, rather than to binding of the substrates.

Characterization of culturable yeast species associating with whole crop corn and total mixed ration silage

  • Objective: This study investigated the association of yeast species with improved aerobic stability of total mixed ration (TMR) silages with prolonged ensiling, and clarified the characteristics of yeast species and their role during aerobic deterioration. Methods: Whole crop corn (WCC) silages and TMR silages formulated with WCC were ensiled for 7, 14, 28, and 56 d and used for an aerobic stability test. Predominant yeast species were isolated from different periods and identified by sequencing analyses of the 26S rRNA gene D1/D2 domain. Characteristics (assimilation and tolerance) of the yeast species and their role during aerobic deterioration were investigated. Results: In addition to species of Candida glabrata and Pichia kudriavzevii (P. kudriavzevii) previously isolated in WCC and TMR, Pichia manshurica (P. manshurica), Candida ethanolica (C. ethanolica), and Zygosaccharomyces bailii (Z. bailii) isolated at great frequency during deterioration, were capable of assimilating lactic or acetic acid and tolerant to acetic acid and might function more in deteriorating TMR silages at early fermentation (7 d and 14 d). With ensiling prolonged to 28 d, silages became more (p<0.01) stable when exposed to air, coinciding with the inhibition of yeast to below the detection limit. Species of P. manshurica that were predominant in deteriorating WCC silages were not detectable in TMR silages. In addition, the predominant yeast species of Z. bailii in deteriorating TMR silages at later fermentation (28 d and 56 d) were not observed in both WCC and WCC silages. Conclusion: The inhibition of yeasts, particularly P. kudriavzevii, probably account for the improved aerobic stability of TMR silages at later fermentation. Fewer species seemed to be involved in aerobic deterioration of silages at later fermentation and Z. bailii was most likely to initiate the aerobic deterioration of TMR silages at later fermentation. The use of WCC in TMR might not influence the predominant yeast species during aerobic deterioration of TMR silages.

Enzymatic Hydrolysis of Hydrophobic Triolein by Lipase in a Mone-phase Reaction System Containing Cyclodextrin; Reaction Characteristics

  • A hydrophobic substrate triolein was hydrolyzed by lipase in a mono-phase reaction system containing cyclodextrin(CD) as emulsifier. The triolein was transformation to an emulsion-like state in the CD containing reaction system in contrast to the oil-droplet like state without CD due to the formation of an inclusion complex between the lipids and CDs. The hydyrolysis reaction increased substantially in the CD containing reaction system, and the optimum reaction conditions including the amount of lipase, ${\beta}$-CD concentration, and mixing ratio of triolein and ${\beta}$-CD, were determined. The performance of the enzyme reaction in a mono-phase reaction system was compared with that of a two-phase reaction system which used water immiscible hexane as the organic solvent. The role of a CD in the mono-phase reaction system was elucidated by comparing the degree of the inclusion complex formation with triolein and oleic acid, Km and Vmax values, and product inhibition by oleic aicd in aqueous and CD containing reaction systems. The resulting enhanced reaction seems to be caused by two phenomena; the increased accessibility of lipase to triolein and reduced product inhibition by oleic acid through the formation of an inclusion complex.

Isolation and Identification of Rice Root Endophytic Antagonistic Serratia marcescens (벼 뿌리 내생 항균성 Serratia marcescens의 분리 및 동정)

  • Twenty-three strains of Serratia sp., isolated from surface-sterilized rice roots collected in Chonbuk and Chungnam province, were identified and characterized. They were Gram-negative, rod shaped and red pigmented typically and their endophytism was confirmed by inoculation and reisolation of the strains in planta. Their antifungal activity against 4 rice pathogenic fungi was compared and ranged from 62.4 to 85.2% against Rhizoctonia solani and 68.0 to 88.5% against Pyricularia grisea. Among the 23 strains tested, strain Rsm220 showed the strongest inhibition activity against 4 pathogenic fungi. The strain was, therefore, selected as a biocontrol candidate for both the pathogens and its bacteriological characteristics and 165 rDNA sequences were analyzed. Phenotypic and biochemical characteristics of the selected Rsm220 were highly related to the type strain of S. marcescens and 165 rDNA sequencing of Rsm220 showed a homology of 98.2% to the type strain of S. marcescens. The strain Rsm220 was identified as S. marcescens and the inhibition result of this endophytic strain indicates that it is a potential biocontrol agent for R. solani and R grisea.

Effect of Filter Aid Treatment on the Quality Characteristics of Salt-Fermented Anchovy Sauce (멸치액젓의 품질특성에 미치는 여과조제 처리의 효과)

  • To obtain the basic data for the improvement of flayer and color in salt-fermented anchovy (Engraulis japonicus) sauce and for the inhibition of non-soluble precipitates formation during storage, experiments were carried out on the changes of chemical properties by treatment with 1, 3, 5 and $7\%$ (w/v) of active carbon and filter aids such as active clay, $Al_2O_3$ diatomaceous earth, and celite. Deceleration rate were $32\~33\%$ in case of $5\%$ (w/v) active carbon or $5\%$ (w/v) active clay treatments, but deodoration effect was not expected in all treatments. The inhibition rate of non-soluble precipitates formation in me of $5\%$ (w/v) active clay, diatomaceous earth and $Al_2O_3$ treatments. was shown as $82\%,\;73\%$, and $70\%$, respectively.

Performance Evaluation of Anaerobic Bioreactors and Effects of Ammonia on Anaerobic Digestion in Treating Swine Wastewaters

  • The operational characteristics of anaerobic bioreactors in treating swine wastewater were evaluated upto hydraulic retention time (HRT) of 1 day and organic loading rate (OLR) of $5.1kg-COD/m^3{\cdot}d$ for 200 days. The bioreactors were effective in treating swine wastewaters with COD removal efficiency of $78.9{\sim}81.5%$ and biogas generation of $0.39{\sim}0.59m^3/kg-COD_r$ at OLR of $1.1{\sim}2.2kg-COD/m^3{\cdot}d$. The two-stage ASBF anaerobic bioreactors was effective in treating different characteristics of swine wastewaters since they showed high and stable COD removal efficiency at high OLR due to effective retention of biomass by media and staging. The effects of ammonia on anaerobic digestion were investigated by operating two-stage ASBF reactors using swine wastewaters as influent without and with ammonia removal at HRT of $1{\sim}2$ days and OLR of $2.2{\sim}9.6kg-COD/m^3{\cdot}d$ for 250 days. The COD removal efficiency and biogas generation of two-stage ASBF reactors was decreased by increasing influent ammonia concentrations to 1,580 mg (T-N)/L with increasing OLR to $6.3kg-COD/m^3{\cdot}d$, while those were increased by maintaining influent ammonia concentrations below 340 mg (T-N)/L by MAP precipitation with increasing OLR to $9.6kg-COD/m^3{\cdot}d$. Initial inhibition of ammonia on anaerobic processes was observed at a concentration of 760 mg (T-N)/L and the COD removal efficiency and biogas generation dropped to 1/2 at ammonia concentration ranges of $1,540{\sim}1,870mg$ (T-N)/L. It is essential to remove ammonia in swine wastewaters to an initial inhibition level before anaerobic processes for the effective removal of COD.

Studies on the Physicochemical Characteristics and NDMA Formation of Orostachys japonicus A. Berger (와송의 이화학적 특성 및 NDMA 생성에 관한 연구)

  • The purpose of this study was to analyze the changes in physicochemical characteristics and NDMA formation of Orostachys japonicus A. Berger according to harvest times. The results showed moisture, carbohydrate, crude fiber and crude protein contents of <$48.5{\pm}1.4%$, $38.2{\pm}1.2%$, $l5.8{\pm}0.4%$ and $13.2{\pm}0.6%$ respectively. The total mineral content was 4,817.1 mg/100 g, where Ca was highest($2,577.0{\pm}3.2$ mg/100 g), followed by K, Mg, Na, P and Fe. The composition amino acid of O. japonicus A. Berger showed high levels of glutamic acid and aspartic acid. And among the five free sugars detected, galactose and glucose were most abundant at $32.2{\pm}0.02$ mg/100 g and $25.3{\pm}0.1O$ mg/100 g, respectively. Upon comparing O. japonicus A. Berger extracts that came from plants harvested during $August{\sim}October$, total phenolic compound($2,703.l{\pm}5.2l{\sim}2,428.0{\pm}3.52$ mg/100 g) and flavonoid($634.2{\pm}2.33{\sim}1,324.6{\pm}1.87$ mg/100 g) contents were higher in the methanol extract than in the water extract. Also, within a reaction system, nitrite scavenging ability and NDMA inhibition were most effective at pH 2.5, and increased in proportion to the extract concentration. Finally, the methanol extract of O. japonicus A Berger harvested during $August{\sim}October$ had the highest nitrite scavenging and NDMA inhibition effects.

Effects of Pentoses on 2-deoxy-D-Glucose Transport of the Endogenous Sugar Transport Systems in Spodoptera frugiperda Clone 9 Cells

  • Insect cells such as Spodoptera frugiperda Clone 9 (Sf9) cells are widely chosen as the host for heterologous expression of a mammalian sugar transport protein using the baculovirus expression system. Characterization of the expressed protein is expected to include assay of its function, including its ability to transport sugars and to bind inhibitory ligands such as cytochalasin B. It is therefore very important first to establish the transport characteristics and other properties of the endogenous sugar transport proteins of the host insect cells. However, very little is known of the transport characteristics of Sf9 cells, although their ability to grow on TC-100 medium strongly suggested the presence of endogenous glucose transport system. In order to investigate the substrate and inhibitor recognition properties of the Sf9 cell transporter, the ability of pentoses to inhibit 2-deoxy-D-glucose (2dGlc) transport was investigated by measuring inhibition constants $(K_i)$. To determine the time period over which of sugar into the Sf cells was linear, the uptake of 2dGlc 0.1mM extracellular concentration was measured over periods ranging from 30 seconds to 30 minutes. The uptake was linear for at least 2 minutes at the concentration, implying that uptake made over a 1 minute time course would reflect initial rates of the sugar uptake. The data have also revealed the existence of a saturable transport system for pentose uptake by the insect cells. The transport was inhibited by D-xylose and D-ribose, although not as effective as hexoses. However, L-xylose had a little effect on 2dGlc transport in the Sf9 cells, indicating that the transport is stereoselective. Unlike the human erythrocyte-type glucose transport system, D-ribose had a somewhat greater apparent affinity for the Sf9 cell transporter than D-xylose. It is therefore concluded that Sf9 cells contain an endogenous sugar transport activity that in some aspects resembled the human erythrocyte-type counterpart, although the Sf9 and human transport systems do differ in their affinity for cytochalasin B.
