In this study, 18 kinds of Korean medicinal herb extracts were examined for anti-microbial activities against pathogenic microorganisms. The methanol (MeOH) extracts from Schizandra chinensis, Rhus javanica and Caesalpinia sappan exhibited antimicrobial activities against most pathogenic microorganisms at concentrations of 5 mg/mL, whereas the other 15 extracts exhibited anti-microbial activities at concentrations of 30 mg/mL. The minimum concentration at which Schizandra chinensis extracts inhibited for S. epidermidis and Bor. bronchiseptica was 0.6 mg/mL. The MeOH extracts from Schizandra chinensis, Caesalpinia sappan, Rhus javanica and Seutellaria baicalensis which had higher anti-microbial activities were subsequently fractionated using 5 different solvents, and further screened for anti-microbial activities. The inhibitory effects of ethyl acetate (EtOAc) extracts on microbial growth were greater compared to any other solvent extracts. In order to investigate the inhibitory effect of Korean medicinal herbs with high anti-microbial activities on microbial proliferation, the MeOH extracts at concentrations of 0, 100, 300 and 500 ppm were added to the media. No addition of extracts caused rapid growth of microbes after 12 hours incubation. As the concentration of extracts from Rhus javanica and Caesalpinia sappan increased, the growth-inhibiting effect on gram-positive bacteria including S. aureus, S. epidermidis, and L. monocytogenes was prominent. Rhus javanica extracts exhibited growth-inhibiting activity for gram-negative bacteria including Sal. Pullorum and Sal. Choleraesuis. The low concentration of extracts from Rhus javanica and Caesalpinia sappan exhibited the growth of Bor. bronchiseptica and E. coli serotype $O_8$. However, the higher concentration of extracts from Rhus javanica and Caesalpinia sappan exhibited a strong inhibitory effect on microbial proliferation.
Angelica acutiloba is one of the most intensively cultivated medicinal plants in Korea. The roots of this plant have been used as an important herbal drug, especially for the treatment of various female disorders, as the traditional therapy in Korea and other Asian countries. Consumption of its fresh leaves as a healthy vegetable has recently increased. In this study, essential oil fractions were extracted from the roots and leaves of this plant by steam distillation. Compositions of the two oils were compared by gas chromatography-mass spectrometry (GC-MS). The antibacterial activities of the essential oil were determined against three strains of Escherichia coli. DPPH radical scavenging and reducing power tests were performed to evaluateits antioxidant activities. The cytotoxic activities of the essential oil against a human breast and a uterine cancer cell line were estimated by MTT tests. Additionally, the morphological changes after treatment of the oil fraction were observed under a microscope. The essential oil fraction and its main components, Z-ligustilide and butylidene phthalide, inhibited the growth of three E. coli strains examined, with minimum inhibiting concentrations (MICs) ranging from 1.0 mg/ml to 8.0 mg/ml. Additionally, the essential oil fraction of A. acutiloba exhibited significant DPPH free radical scavenging activity and reducing power. Significant cytotoxic activities of the A. acutiloba essential oil were observed for human uterine (Hela) and breast (MCF-7) cancer cell lines.
Lentinus edodes contains functional metabolites such as polysaccharopeptides, lectins, and secondary metabolites. Fermented soybean paste is representative fermented materials in Korea, and is gradually increasing due to various biological activities. In the present study, ethanol extracts of fermented products with/without L. edodes were designated as SPL and SP, and prepared to develop safer and therapeutic functional foods with antioxidant and anti-inflammatory activities for treatment of inflammatory disorders. SP and SPL extracts exhibited antioxidant effects via inhibiting radical activities. Inflammatory mediators, nitric oxide (NO), prostaglandin E2 (PGE2), interleukin (IL)-1β, IL-6, IL-8, tumor necrosis factor (TNF)-α, and inducible nitric oxide synthase (iNOS) production and nuclear factor-kappa B (NF-κB) activation were down-regulated by two extracts. SPL extract more strongly enhanced the antioxidant and anti-inflammatory activities than SP extract. Its' activities shown more longer fermentation period and more strong inhibitory effects. Taken together, our results suggested that fermented product with medicinal plant has synergic effect and SPL can be a potential candidate for treatment of inflammatory bowel diseases.
We examined the inhibitory activities of various flavonoids, including the flavanones, flavones/isoflavones and flavonols, on collagenase from Clostridium histolyticum to establish their therapeutic potential against skin inflammation and photoaging. In general, the flavonols were stronger inhibitors than the flavones/isoflavones, and this indicated the importance of the C-3 hydroxyl substitution. Quercetin was the most active flavonoid among those tested, and it showed an $IC_{50}$ of 286 ${\mu}M$. These novel results suggest that certain flavonoids, particularly the flavonols, may prevent collagen breakdown by inhibiting collagenase in inflamed skin as well as photoaged skin.

Protein inhibitors are proteins or peptides capable of inhibiting catalytic activities of proteolytic enzymes. They are grouped primarily as either serine, cysteine, aspartic or metallto-proteinase inhibitors. Pretense inhibitors have been hewn since the end of the last century in nematodes and human blood serum, and their ubiquitous distribution in microorganisms, animals and plants has been widely documented. Effects of dimethipin on the senescence of excised barley first leaves were investigated. Dimethipin markedly inhibited chlorophyll and protein loss and reduced peroxidase activity relevant to senescence phenomena in th excised leaves. Dimethipin decreased hydrogen peroxide content, later malondialdehyde content, and increased the activities of superoxide dismutase and catalase. The antisenescence effects of dimethipin may result from the stabilization of membrane structure through inhibiting the peroxidation of unsaturated lipid and the accumulation of free radicals during senescence.
Objectives : Oxidative stress seems to play a major role in mechanisms by which ethanol causes liver injury. Previous studies have shown that treatment with Chungganhaeju-tang (Qingganjiejiu-tang, CGHJT) has protective effects on alcoholic liver disease. The aim of this study was to investigate the effects of Chungganhaeju-tang on oxidative stress. Materials and Methods : In vitro, we evaluated the inhibitory activities of CGHJT on DPPH (1,1-diphenyl-2-picryl-hydrazyl), xanthine oxidase, trypsin, and hyaluronidase, and measured cell viability, and proliferation. In the cell culture model, we measured the activities of superoxide dismutase (SOD), and catalase (CAT) after CGHJT treatment in C34 and E47 cell lines, HepG2 cells transfected with/without the cytochrome P450 2E1 (CYP2E1) gene. In vivo, we measured malondialdehyde levels in the liver tissue and alcohol concentration in the blood. Results : CGHJT showed significant free radical scavenging activity against DPPH and xanthine oxidase in the in vitro study, and increased cell viability, proliferation, and activities of superoxide dismutase, catalase in C34 and in E47 cell lines. CGHJT reduced malondialdehyde levels and blood alcohol concentration in vivo, as well. Conclusions : This study suggests that CGHJT has antioxidant effects on oxidative stress by reducing lipid peroxidation and inhibiting the ethanol induced suppression of antioxidant enzyme activities.

This study was Performed to investigate the effect of kimchi intake on antiaging characteristics in the brain of senescence-accelerated mouse (SAM) in terms of free radical production and anti-oxidative enzymes. Two hundreds twenty SAM (20 mice) were divided into four groups and fed kimchi diet for 12 months. Experimental groups were kimchi free AIN-76 diet (control) group, Korean cabbage kimchi diet (KCK)group, 30% mustard leaf added Korean cabbage kimchi diet (MKCK) group, and mustard leaf kimchi diet (MLK) group. Concentrations of total free radical, OH radical and $H_2O_2$ of control group increased up to 123%, 262% and 174% of initial value (p<0.05) after one year. Increase in free radical production in kimchi groups due to aging was decreased by kimchi feeding. Among kimchi groups. MKCK and MLK groups showed greater inhibiting effect against free radical production than KCK. The concentration of TBARS in the bruin of control group also significantly increased up to 362% of initial value as aged (p<0.05) and production of TBARS in kimchi groups were decreased. When the activities of Cu, Zn-SOD, Mn-SOD, GSH-px and catalase of kimchi groups were compared to those of control at the same experimental period, anti-oxidative enzyme activities of kimchi groups were lower than those of control (p<0.05). But GSH/GSSG in kimchi groups were higher compared to control. In conclusion, decrease in free radical production and increase in anti-oxidative enzyme activities were observed from kimchi groups suggesting that kimchi might have important role on retarding aging. Among kimchi variety tested in this experiment, MKCK and MLK seem to have greater effect on inhibiting free radical production and increasing anti-oxidative enzyme activities than KCK.
With 32 fungicides, it was examined the inhibitory effects on the mycelial growth of Alternaria dauci KACC42997 causing Alternaria leaf blight of carrot. Showing the results of the agar dilution method, the fungicides belonging to C2, C5, G1, E2, and E3 group were excellent in inhibiting mycelial growth. Protective fungicides belonging to M group, except for iminoctadine tris-albesilate, and pyraclostrobin belonging to C3 group were effective in inhibiting spore germination of pathogens. The fungicides included into C2 group inhibiting succinate dehydrogenase activity and the G1 group inhibiting demethylase activity showed the excellent inhibitory effect on mycelial growth but the inhibitory effect of spore germination was very low. However, fluazinam belonging to C5 group was excellent in inhibiting spore germination as well as mycelial growth. Especially, when 100 ㎍/ml of fluxapyroxad belonging to the C2 group was treated, 47.1% of spore formation was inhibited on the medium. In comparison of the resistance factors of 3 fungicide groups, as G, C, and E group, in populations of A. dauci isolates collected from Gumi, Pyeongchang, and Jeju, resistance factor in the population of Jeju was the lowest. However, two isolates resistant to fludioxonil belonging to E2 group were found in the isolate group of Pyeongchang, and both showed cross-resistance to iprodione and procymidone.
The antioxidant effect of methanol extract (ME) and water extract (WE) from Clematis trichotoma was evaluated as primary study to scavenge stable 1,1-diphenyl-2-picrylhydrazyl radicals (DPPH), inhibited iron-induce lipid peroxidation in linoleic acid emulsion, peroxidation of liposome induced by $Fe^{3+}/H_2O_2/ascorbie$ acid, and on $Fe^{2+}/H_2O_2$ induced the mitochondrial lipid peroxidation. In secondary study, five flavonoids as luteolin (1), quercetin (2), apigenin (3), hirsutrin (4), kaempferol-3-O-glucoside were isolated (5). Among them, compounds 1 and 2 showed good activities in all the model systems. Compound 3 exhibited moderate antioxidant activities in both radical scavenging and these lipid peroxidation systems tested. Compound 4 showed significant inhibitions in liposome peroxidation and compound 5 displayed weak inhibition in all four tested systems. All the results presented herein indicate that products of C. trichotoma maybe useful in inhibiting membrane lipid peroxidation and preventing free radical-linked diseases.
