Antibacterial Activities of Persimmon Roots-derived Materials and 1,4-Naphthoquinone's Derivatives against Intestinal Bacteria

  • The growth-inhibiting activities of persimmon roots-derived materials against intestinal bacteria were evaluated and compared with that of 1,4-naphthoquinone as a positive control. The active constituent isolated from persimmon roots was characterized as 5-hydroxy-2-methyl-1,4-naphthoquinone using various spectroscopic analyses. Treatment with 1,4-naphthoquinone at a dose of 1.0 mg/disc strongly inhibited the growth of 6 intestinal bacteria. Furthermore, when the structure-activity relationships of 1,4-naphthoquinone's derivatives were evaluated, 5-hydroxy-2-methyl-1,4-naphthoquinone and 2-methyl-1,4-naphthoquinone were found to strongly inhibit the growth of Clostridium difficile, Clostridium perfringens, and Escherichia coli without adversely affecting the growth of Bifidobacterium adolescentis, Bifidobacterium longum, and Lactobacillus acidophilus. Additionally, 2-hydroxy-1,4-naphthoquinone and 5-hydroxy-1,4-naphthoquinone strongly inhibited the growth of C. difficile and C. perfringens, but did not inhibit the growth of E. coli. Taken together, these results indicate that persimmon roots-derived materials and some of 1,4-naphthoquinone's derivatives could be useful preventive agents against diseases caused by harmful intestinal bacteria.

사궁산(莎芎散)이 실험동물(實驗動物)의 지혈(止血), 뇌압(腦壓), 혈압(血壓) 및 심혈관계(心血管系)에 미치는 영향(影響) (Effects of Sagungsan on the Hemostasis, Intracranial Pressure, Blood Pressure and Cardiovascular System in Expreimental Animals)

  • According to the original documents, Sagungsan is considered as an effective drug for controlling the hypertensive epistaxis induced by tension of autonomic nerve and it's hyperfunction. The present experiment was designed to understand the effect of Sagungsan extract on the hemostatic action, intracranial pressure, blood pressure and cardiovascular system in experimental animals. And thus the bleeding time, prothrombin time, capillary dilation, blood pressure, Intracranial pressure, and enzymatic analysis of the ATPase activities were studied. The result obtained here were as followings: 1. Sagungsan water extract reduced the bleeding time in mouse, and prolonged the prothrombin time in rabbits. 2. The drug extract increased the tail volume by capillary dilation in rats. 3. The drug extract inhibited the increase of intracranial pressure and arterial blood pressure in rabbits. 4. At the early time, the increase of arterial blood pressure by the drug extract significantly inhibited by pretreated atropin and regitine in rabbits. 5. The drug extract relaxed the smooth muscle by stimulating the Mg2+-Ca2+-ATPase activities of gastric sarcoplasmic reticulum isolated from rabbit stomach. 6. The drug extract stimulated the heart contraction by inhibiting the $Mg^{2+}-Ca^{2+}-ATPase$ activities of cardiac sarcoplasmic reticulum isolated from rabbit heart. The inhibitory mechanism was reversible and noncompatitive. 7. The drug extract increased the hepatic blood volume by stimulating the hepatic total ATPase activities and hepatic metabolism. 8. The drug extract acted as a tranquilizer by inhibiting the neural Na+-K+-ATPase activity. According to the results, Sagungsan water extract dilated the capillaries, stimulated the heart beat, and thus increased the blood flow with decreasing the intracranial pressure and blood pressure. These effects stanches the epistaxis collectively.

Pharmacological Effects of Asaronaldehyde Isolated from Acorus gramineus Rhizome

  • Antibacterial and antiplatelet activities of Acorus gramineus rhizome-derived asaronaldehyde and asaron were analyzed using platelet aggregometer and six human intestinal bacteria. Active constituent of A. gramineus rhizome was isolated and characterized as asaronaldehyde by spectral analyses. At 2 and 1 mg/disk, asaronaldehyde exhibited strong inhibition of Clostridium perfringens and C. difficile without adverse effects on growth of beneficial bacteria such as Bifidobacterium bifidum, Lactobacillus acidophilus, and L. casei. Asaron also revealed moderate growth inhibition against C. perfringens and C. difficile at 2 mg/disk, no growth-inhibiting activity was observed on B. bifidum, L. acidophilus, L. casei, and E. coli. At 50% inhibitory concentration ($IC_{50}$) value, asaronaldehyde was effective in inhibiting platelet aggregation induced by collagen ($IC_{50}$, $27.6\;{\mu}M$) and arachidonic acid ($IC_{50}$, $53.7\;{\mu}M$). These results suggest asaronaldehyde may be useful as lead compound for inhibiting platelet aggregation induced by collagen and arachidonic acid.

In vitro Inhibitory Activities of Essential Oils from Oenanthe javanica DC against Candida and Streptococcus species

  • The composition of essential oil from O. javanica was analyzed by gas chromatography-mass spectrometry. Using the broth dilution method and disk diffusion test, anti-microbial activities of the oil fraction and its main components were evaluated against various antibiotic-susceptible and resistant strains of pathogenic microorganisms. As a result of GC-MS analysis, 57 compounds, including ${\alpha}-terpinolene$ (28.1%), dl-limonene (16.0%), ${\gamma}-terpinene$ (10.3%), ${\beta}-pinene$ (9.7%) and ${\alpha}-pinene$ (6.0%) were identified in the essential oil fraction. The essential oil fraction of O. javanica and its main components exhibited significant inhibitory activities, particularly against Candida albicans (antibiotic-susceptible strains) and Streptococcus pneumoniae (antibiotic- susceptible and resistant strains). The main components of the O. javanica oil fraction displayed different patterns of activity against the three tested Candida species as exemplified by the differential minimum inhibiting concentration (MIC) values. The disk diffusion test showed that the activities were dose dependent.

In Vitro Cancer Chemopreventive Activities of Polysaccharides from Soybeans Fermented with Phellinus igniarius or Agrocybe cylindracea

  • Chemopreventive activities of polysaccharides from soybeans fermented with either Phellinus igniarius or Agrocybe cylindracea were investigated by measuring the induction of quinone reductase (QR), glutathione S-transferase (GST) activities, and glutathione (GSH) levels in the cell culture along with inhibition of polyamine biosynthesis. The polysaccharides from soybeans fermented with P. igniarius strongly (p<0.005) induced QR activity at all concentrations tested. The extract not only induced GST activity in a dose-dependent manner in the concentration range of 0.1-1.0 mg, but significantly induced GSH revels in cultured Hepa 1c1c7 cells with a maximal 1.4-fold increase at 0.1 mg. The polysaccharides from soybeans fermented with A. cylindracea were effective in inhibiting polyamine metabolism. These results suggest that polysaccharides from soybeans fermented with P. igniarius or A. cylindracea have cancer chemopreventive activities in in vitro models and, therefore, could be considered as potential agents for cancer chemoprevention.

전호(Anthriscus sylvestris) 뿌리 정유의 항균 및 항산화 작용 (Antibacterial and Antioxidant Activities of the Essential Oil from the Roots of Anthriscus sylvestris)

  • To develop a new effective and safe natural antibiotics and antioxidant the essential oil was extracted from the roots of Anthriscus sylvestris by steam distillation. Its composition was analyzed by GC-MS. The activities of the essential oil fraction and its main components were evaluated against antibiotic-susceptible and -resistant strains of some food-born bacteria. In addition the synergism was examined with this oil combined with antibiotic by checkerboard titer test. The antioxidant activities were determined by in 1,1-diphenyl-2-picryl-hydrazil (DPPH) free radical scavenging activity test and reducing power assay. The essential oil fraction of A. sylvestris revealed significant inhibiting activities against antibiotic-susceptible and -resistant species of Vibrio and Shigella with MICs ranged from 1.00~4.00 mg/ml. It showed synergistic or additive effects when it was combined with amphicillin or trimethoprim/sulfamethoxazole (1 : 9). Additionally, the essential oil fraction of A. sylvestris exhibited significant DPPH free radical scavenging activity and the reducing power.

비타민나무(Seabuckthorn, Hippophae rhamnoides L.) 부위별 추출물의 생리활성 비교 (Comparison of Biological Activities of Extracts from Different Parts of Seabuckthorn (Hippophae rhamnoides L.))

  • 국내에서 재배된 비타민나무(Hippophae rhamnoides L.)의 생리활성을 부위별(줄기, 잎, 뿌리, 열매), 용매별(물, 에탄올)로 시험한 결과는 다음과 같다. $\alpha$-Glucosidase 활성 억제 실험에서, 비타민나무의 저해활성은 줄기의 에탄올 추출물이 93%로 가장 높았고, 다음이 잎의 에탄올 추출물이 88.7% 순으로서 대조구 acarbose의 20.2%보다 훨씬 높았다. Pancreatin 기원의 $\alpha$-amylase 억제 활성 측정 실험에서는 잎의 물 추출물이 54.7%로 다른 추출물에 비해 가장 높았다. HT-29 cell과 DU-145 cell의 암세포에 대한 항암효과 시험 결과 뿌리의 물 추출물에서 각각 47.1%, 32.3%의 항암효과가 나타났다. 또한 항균활성 실험 결과 잎의 에탄올 추출물에서 위해균주인 Clostridium butyricum, Proteus mirabilis, Shigella flexneri에 대한 저해활성이 나타났다. 향후 연구에서는 생리활성 효과가 나타난 물질에 대한 분리, 정제 등의 연구가 계속 수행되어져야할 것으로 생각된다.

Immunohistochemical Changes of Apoptotic Control Genes by Chronic Inhibition of Nitric Oxide in Rats

  • Sprague-Dawley (SD) rats were orally administered with NG-nitro-L-arginine methyl ester (L-NAME), which inhibits or blocks the production of nitric oxide from L-arginine in vascular endothelial cells and vessel tissue. We examined the effects of nitric oxide on some physiological changes such as blood pressure and heart rate, and confirms the apoptosis induced by the suppressed nitric oxide activity in the kidney. This study was performed to investigate correlation between the activities of nitric oxide and apoptosis by immunohistochemical changes of apoptotic control proteins with regulated chronic inhibition of nitric oxide. In the kidney from L-NAME-treated group, immunohistochemical reaction to the antigens of apoptosis inhibiting proteins such as bcl-2 and bcl-xL, exhibited a time-dependent reduction. The expression of apoptosis-inhibiting proteins such as bax and p53 increased expression in proportion to the duration of treatment. The most sensitive apoptosis regulating proteins to L-NAME were p53 in stimulation and bcl-2 in inhibition, respectively.

Comparisons between White Ginseng Radix and Rootlet for Antidiabetic Activity and Mechanism in KKAy Mice

  • The mechanisms responsible for the antidiabetic activity of both the white ginseng radix (Ginseng Radix Alba, GRA) and the rootlet (Cinseng Radix Palva, GRP) were investigated. After a four week oral administration, the fasting blood glucose levels in the GRA- and GRP-treated groups were lower when compared to the control group. To elucidate the hypoglycemic mechanism(s) of the ginseng radices, glucose absorption from the small intestine, hepatic hexokinase and glucose-6-phosphatase activities, in addition to PPAR-${\gamma}$ expression in adipose tissue were examined. The results strongly suggest that GRA can improve hyperglycemia in KKAy mice, possibly by blocking intestinal glucose absorption and inhibiting hepatic glucose-6-phosphatase, and GRP through the upregulation of adipocytic PPAR-$\gamma$ protein expression as well as inhibiting intestinal glucose absorption.

Cytotoxic, Antioxidative, and ACE Inhibiting Activities of Dolsan Leaf Mustard Juice (DLMJ) Treated with Lactic Acid Bacteria

  • This study was performed to know whether there is any change of physiological activity in DLMJ which is inoculated by lactic acid bacteria. Lactic acid bacteria were isolated from Dolsan leaf mustard Kimchi (DLMK) at $20^{\circ}C$. In the optimum ripening period, the population of Leuconostoc and Lactobacilli in the DLMK were found to be high. The Leuconostoc, Lactobacilli and Lactococci strains were identified as Leuconostoc mesenteroides, Leuconostoc gelidum, Weissella confusa, Lactobacillus plantarum, Lactobacillus raffinolactis, Lactococcus lactis and Weissella confusa using the Biolog system. The most predominant strain which was isolated from DLMK was Weissella confusa. As the results of the phylogenetic analysis using 16s rDNA sequence, the Weissella confusa turned out to be Weissella kimchii, with 99.0% similarity. To investigated the change of physiological activity in DLMJ by lactic acid bacteria, 7 predominant strains inoculated to DLMJ (Dolsan Leaf Mustard Juice). The cytotoxicity was found to be under $19.55\%$ all cases. Also, the antioxidative activity of the DLMJ treated with lactic acid bacteria was very low, which might have been due to the reduced antioxidative phytochemicals during the preparation of the sterile sample. The ACE inhibiting activity of DLMJ by inoculation with Weissella kimchii was shown to be the highest ($94.0\%$). This could be that the degradation of sulfur containing materials in DLMJ by Weissella kimchii gave rise to ACE inhibiting activity.
