Fusarium wilt caused by Fusarium oxysporum is a devastating disease limiting production of watermelon in Korea. The best way to control diseases is to use resistant gourd rootstock on watermelon. This study was conducted to establish an efficient screening method for resistant bottle gourd to Fusarium oxysporum f. sp. lagenaria. To develop an efficient inoculation method, incubation temperature after inoculation (15, 20, 25, and 30℃), inoculum concentration (1 × 105, 5 × 105, 1 × 106, and 5 × 106 conidia·mL-1), and growth stages of seedlings (7, 10, 13, and 16 days) was investigated. Disease development of Fusarium wilt of bottle gourd was little affected by differences in incubation temperature and growth stages of seedlings. But resistant lines were more susceptible and appeared more severe symptoms at the higher inoculation level. Taken together, we suggest that an effective screening method for resistant gourd plant to Fusarium wilt is to dip the roots of 10-day old seedlings in spore suspension of 1 × 105 - 1 × 106 conidia·mL-1, for 30 min, to transplant the seedlings into a non-infected soil, and then to incubate the inoculated plants in a growth room at 25℃ for 3 weeks to develop Fusarium wilt.

The study was performed to establish an efficient screening method for resistant cucumber to Fusarium oxysporum f. sp. cucumerinum. The isolate KR5 was identified as F. oxysporum f. sp. cucumerinum based on molecular analyses of ITS and TEF genes and host-specificity test on cucurbits including melon, oriental melon, cucumber, and watermelon. Then four cucumber and two rootstock cultivars showing different resistance degrees to the Fusarium wilt pathogen KR5 were selected. And development of Fusarium wilt of the six cultivars according to several conditions, including incubation temperature after inoculation, inoculum concentration, root wounding, and growth stages of seedlings, was investigated. Disease severity of Fusarium wilt on the resistant cultivars was changed with incubation temperatures after inoculation. The resistant cultivars showed the higher resistance when inoculated plants were kept at 25 or $30^{\circ}C$ than at $20^{\circ}C$. Among four different growth stages of the seedlings, seven-day-old seedling represented the most difference of resistance and susceptibility to Fusarium wilt. From above results, we suggest that an efficient screening method for resistant cucumber to F. oxysporum f. sp. cucumerinum is to dip the non-cut roots of seven-day-old seedlings in spore suspension of $1.0{\times}10^6-1.0{\times}10^7$ conidia/ml and to transplant the seedling into a non-infected soil, and then to incubate the inoculated plants in a growth room at $25^{\circ}C$ for 3 weeks to develop Fusarium wilt.

Objective : The classic and accepted surgical method of compound comminuted depressed skull fractures (FCCD) involves total resection of all the contaminated bone and fragments at the fracture site. A second operation for cranioplasty is then performed at a later date. However, we have believed that primary repair of these bony defects can be achieved by the replacement of bone fragments at the time of the initial debridement, and this can be accomplished without danger to patient. The authors retrospectively reviewed the surgical results to assess the advantages and disadvantages, and also propose the selection criteria of replacement of fractured bone fragments as a primary procedure in FCCD. Materials and Methods : The authors analyzed the data extracted from medical records, and radiological findings in 22 of 71 patients with FCCD, who underwent immediate replacement of fractured bone fragments between April 1993 and October 1998. The mean follow-up period was 13.7 months. The selection criteria for the operation included the patients with mild to moderate severity, regardless of the degree of contamination or dural violation, which presented in hospital within 24 hours of injury. Results : The ages of the patients varied from 4 to 63 years, and there were 20 males and 2 females. Seventeen of 22 patients were fully conscious on admission and the others also had relatively good Glasgow coma scales. Sixteen fractures were located in the frontal area, 9 with involvement of the frontal sinuses, and 6 in the parietal and temporoparietal areas. Of the 22 patients, 8(36.3%) had dural lacerations with 3 of these requiring patching with pericranium, and 12(54.5%) had intracranial hematoma requiring wide craniotomy. The degree of wound contamination was also variable. Fifteen patients had relatively clean wounds, while seven(31.8%) had seriously contaminated wounds with soil, sand, hair, and wood. Only one patient(4.5%) developed infection, and the bone fragments were removed. All wounds healed primarily without pulsatile defect, the skull has remained solid, and no complications have occurred, except the infected case. Conclusion : It is proposed that bone fragments removal for FCCD, regardless of the degree of contamination or dural violation, is not necessary and that primary bone fragments replacement avoids a second operation for cranioplasty.
The rate of gerbera (Gerbera jamesonii Bolus.) leaf damaged by Liriomyza trifolii larvae was lower than that by adults. The adults began to be attracted to yellow sticky trap immediately after transplanting and increased gradually up to early August followed by low population density from middle to late August. In the second year, adults began to be attracted from late April after transplanting and increased abrubtly from middle May and the high density was maintained till early truly. The density decreased, however, from middle July, and then followed by the high density again early September and late October The seasonal fluctuations of larva and pupa were similar to that of adults and the peaks of pupa occurred one week later than that of larvae. Larva infected by parasitoid were observed 4 weeks after transplanting. Parasitism was 65% in average from early July to late August but decreased to 50% from early September. Number of trapped adults was the highest in the yellow sticky trap among the five different colour raps. Catches at 30~60 cm above soil surface were significantly greater than that at 90 cm.
A study on the root-knot nematodes (Meloidogyne spp.) affecting economic crops in Korea was undertaken to know the distribution of the nemic fauna. Total 66 samples were taken from soil and root of 24 host plants at different localities in 4 provinces. Four Meloidogyne species such as Meloidogyne hapla, Meloidogyne arenaria, Meloidogyne incognita and Meloidogyne javanica were identified and some morphological characteristics were described. One of these, Meloidogyne javanica was reported newly in Korea from Horticultural Experimental Station, Suweon, Gyeong Gi on potato, Geomsadong, Daegu on chinese cabbage, Sangeogdong, Daegu, Gyeong Bug on violet and Choeumri, Namhae, Gyeong Nam on pumpkin. In Jae Ju province, Meloidogyne incognita was only found except the other three species. The most common and widely distributed Meloidogyne species in Korea is Meloidogyne hapla by $50\%$ in total, next Meloidogyne incognita $33.3\%$ Meloidogyne arenaria $10.6\%$ Meloidogyne javanica $6.0\%$ in turn, The root-knot nematodes infected the most severely in Jae Ju province and Gyeong Nam, Gyeong Bug and Gyeong Gi province in turn. Twenty four plants were attacked by root-knot nematodes among them important economic crops are soybean, peanut, potato, tomato, cucumber, carrot, pumpkin, wateremelon, edible burdock, pepper, eggplant, cabbage, lettuce and tobacco in Korea.
To investigate the incidence of plant-parasitic nematodes from plastic-house fields of strawberry, 170 soil samples were collected from the main producing area of strawberry in Korea from 2014 to 2015. Based on the investigation, root-lesion nematode (RLN; Pratylenchus spp.) and root-knot nematode (RKN; Meloidogyne spp.) are the most important pests in plastic-house fields of strawberry with 57% and 36% incidences, respectively. Among the collected fields, 96 and 63 fields were infected RNL and RKN, respectively with 44 fields (38.8%) being detected at both nematode. High degree of incidence of both nematodes were observed in the regions of Jinju, Gyeonnam Province, Korea. For identification of species level, phylogenetic and distance analysis were performed using RLN and RKN isolates generated from the regions of Jinju. The results indicated that the RLN and RKN isolates were Pratylenchus vulnus and Meloidogyne hapla, respectively. This study showed that P. vulnus and M. hapla could be most dominant species in plastic-house fields of strawberry. This data will provide basic knowledge for development of effective control strategies.
Investigations were conducted to prevent entrance of Bemisia tabaci (Gennadius) with very fine mesh nets. B. tabaci vectors tomato yellow leaf curl virus in tomatoes. B. tabaci could be completely prevented to penetrate into an 83 mesh net, though few of them passed the 60 mesh net. An independent 60 mesh net cage was built within one of the three interconnected common plastic greenhouse ($21.0{\times}18.3m$) in order to find its usefulness as a crop protection tool from insect pests. TYLCV infected tomato plants were not detected, though a few eggs of B. tabaci and mines of Liriomyza trifolii (Burgess) were detected locally at the entrance of the sliding door. There were practically no significant differences between treatments of nets and the conventional plant height, leaf length and width, number of floret clusters, sugar brix, and product yield. In viewpoints of commercialization, it could be considered highly prospectful to apply to the nets to glasshouses or other greenhouses in which the growing period of crops is long, if the nets were used in the openings of sides, ceilings, and entrances of the greenhouse. However, it might be necessary to take preventive measures for occurrences of insect pests living in soil, temporarily or permanently, as well as high humidity diseases.
Sclerotium rot of cucumber (Cucumis sativus L.) occurred at the experimental field of Gyeongsangnam-do Agricultural Research and Extension Services in July 2012. The typical symptoms included wilt, rot, and water-soaking on stems and fruits and severely infected plants eventually died. White mycelial mats spread over lesions, and then sclerotia were formed on fruit and near soil line. The sclerotia were globoid in shape, white to brown in color and 1-3 mm in size and the hyphal width was 4-8 ${\mu}m$. The optimum temperature for mycelial growth and sclerotia formation on PDA was $30^{\circ}C$. The typical clamp connections were observed in the hyphae of the fungus grown on PDA. For further identification, the complete internal transcribed spacer (ITS) rDNA region was amplified and sequenced. On the basis of mycological characteristics, ITS rDNA region comparison, and pathogenicity to host plants, this fungus was identified as Sclerotium rolfsii Saccardo. This is the first report of sclerotium rot on cucumber caused by S. rolfsii in Korea.
Fifty three diseased root samples of Chinese cabbage and 22 soil samples were taken from six alpine areas in Kangwon province to examine the organisms causing root rot of Chinese cabbage. Among thirteen microorganisms detected from diseased roots samples, Pythium ultimum, Pythium echinocarpum and Rhizoctonia solani were detected frequently and were pathogenic to Chinese cabbage. Five to 100% of young Chinese cabbage plants grown in the sample soils taken from alpine cabbage fields were infected either alone or together with P. ultimum, P. echinocarpum and R. solani, depending upon the or gin of sample soils. When relative density of the three organsims in the sample soils was estimated based on the infection frequency on the plants grown in those sample soils, P. ultimum was prevalent only in Chahang-i-li area. P. echinocarpum was prevalent in Yongsan-il-li, Hoengwe-li, and Munmaek areas. R. solani was solely found in Maebongsan area where both P. ultimum and P. echinocarpum were not detected. In the three areas where both P. ultimum and P. echinocarpum were present, density of P. echinocarpum was generally greater than that of P. ultimum. Seedlings of thirty major Chinese cabbage varieties were inoculated independently with the three organisms in the greenhouse. Six varieties were resistant to P. ultimum. Three and two varieties were moderately resistant to P. ultimum and P. echinocarpum, respectively. All varieties were highly susceptible to R. solani.
This study was investigate the occurrence of bottom rot caused by Rhizoctonia solani at the crisphead lettuce fields in Uiryeong-gun, Gyeongsangnam-do from November to December in 2003. Incidence of bottom rot on crisphead lettuce was up to 5.3% at the six plastic houses. A total of 30 isolates of R. solani were obtained from diseased leaves of plants and were tested by artificial inoculation to the host. Among them, PY-1 isolate was selected showing highly virulent on the whole plant and was identified as R. solani AG1 (IB) based on the anastomosis test, morphological and cultural characteristics. Symptoms of bottom rot by PY-1 isolate produced small dark brown, depressed and elliptical spots on the lower part of leaves in the early stage as same as at the fields, were enlarged onto the upper part of leaves later, and the infected plant wilted and ultimately died in the end. For the pathogenicity test, triturated mycelia-inoculum (A$_{550}$=1.0) of PY-1 isolate was selected the most effective inoculum showing disease incidence of 51.1% for the mycelial inoculation at pot assay. Otherwise, WSRP media-inoculum (wheat brane : sawdust : rice brane : PDB media=30 g : 10 g : 10 g : 100 ml, w/w/w/v) of PY-1 isolate was effectual inoculum showing disease incidence of 61.6% for soil inoculation at the plastic house. Also, in selection of density and amount of inoculum, most suitable density of triturated mycelia-inoculum and amount of WSRP media- inoculum were determined as $A_{550}$=1.0 and 40 ml, respectively. This is the first report on the pathogenicity test using by WSRP media-inoculum of R. solani PY-1 isolate for the bottom rot of crisphead lettuce.
