Effect of the Various Heavy Metals on the Growth and Phosphorus (P) Removal Capacity of the Phosphorus Accumulating Microorganism (Pseudomonas sp.) (다양한 중금속이 인 축적 미생물 (Pseudomonas sp.)의 생장과 인 제거에 대한 효과)

  • The removal of phosphorus (P) in the wastewater is essential for the prevention of eutrophication in the river and stream. This study was initiated to evaluate the effect of the various heavy metals on the growth and P removal capacity of Pseudomonas sp., which was well known as phosphorus accumulating microorganism(PAO's) in the EBPR(Enhanced Biological Phosphorus Removal) process. The five heavy metals used in the study were Cu, As, Zn, Ni, and Cd. The growth rate of Pseudomonas sp. was the greatest at $25^{\circ}C$, but the removal efficiency of P was the highest at $30^{\circ}C$. The $IC_{50}$ (median Inhibition Concentration) values of Pseudomonas sp. for the Cu, As, Zn, Ni, and Cd were 2.35, 11.04, 1.80, 4.92, and 0.24 mg/L, respectively. Therefore, it appears that the sensitivity of the heavy metals to Pseudomonas sp. was in the following order: Cd> Zn> Cu> Ni> AS. Also, the P removal efficiencies by Pseudomonas sp. were correspondingly decreased as the concentrations of heavy metals were increased.

  • Docosahexaenoic acid (DHA, 22:6n-3) and ecosapentaenoic acid (20:5n-3) have attracted increasing attention since the first epidemiological report on the importance of n-3 essential fatty acids. It is thought that DHA has important functions in brain and retinal tissues. Thraustochytrids, a group of marine protists, are capable of heterotrophic growth, and are potential omega-3 producers for industrial use, especially the members of the Schizochytrium and Thraustochytrium genera. The aims of this work were to isolate, identify and screen thraustochytrids from 17 different locations. Twenty-three isolates were screened for biomass, total fatty acid (TFA) and DHA content. Analysis of the fatty acid methyl esters revealed four distinct clusters biomass ranged from $8.68-9.36gL^{-1}$, and lipid and DHA contents ranged from $3.11-4.10gL^{-1}$ and $1.05-1.93gL^{-1}$ biomass, respectively. B-12 isolates were screened for biomass ($9.36gL^{-1}$), TFA ($4.10gL^{-1}$) and DHA (47.01%, w/w) content. C-6 isolates were also screened for biomass ($8.92gL^{-1}$), TFA ($3.30gL^{-1}$) and DHA (49.41%, w/w) content. The 18S rRNA gene sequencing results identified Schizochytrium mangrovei as B-12 and Crypthecodium cohnii as C-6.

  • Ammonia is a useful substance which is widely used in various industries. It is generally released by the decomposition of agricultural wastes and known to have toxic effects on human beings. Due to the common usage, it is possible to cause water pollution through either direct or indirect leakage. Such cases, it is preferable to use the adsorption capacity of zeolite to rapidly remove ammonium ions, but it is not sufficiently removed by the adsorption only. In this paper, the removal efficiency of ammonium ion through both the adsorption capacities of commercial synthetic zeolites and the biological mechanism of microorganisms were compared. In addition, microorganisms were immobilized on the zeolite in order to enhance the removal efficiency by applying a chemo-biological process. As a result, the standard commercial zeolite showed 67~81% of the removal efficiency in 2~4 hours at a 100 ppm concentration of ammonium, whereas the selected microorganism Klebsiella pneumoniae subsp. Pneumoniae showed up to 97% within 8 hours. When the microorganism was immobilized on the zeolite, the highest removal efficiency of approximately 98.5% were observed within 8 hours.

  • On the basis of fabrication, the utilization of nano material in numerous industrial and technological system, obtained the utmost significance in current decade. Therefore, the current investigation presents a theoretical disposition regarding the flow of electric conducting Williamson nanoliquid over a stretchable surface in the presence of the motile microorganism. The impact of thermal radiation and magnetic parameter are incorporated in the energy equation. The concentration field is modified by adding the influence of chemical reaction. Moreover, the splendid features of nanofluid are displayed by utilizing the thermophoresis and Brownian motion aspects. Compatible similarity transformation is imposed on the equations governing the problem to derive the dimensionless ordinary differential equations. The Homotopy analysis method has been implemented to find the analytic solution of the obtained differential equations. The implications of specific parameters on profiles of velocity, temperature, concentration and motile microorganism density are investigated graphically. Moreover, coefficient of skin friction, Nusselt number, Sherwood number and density of motile number are clarified in tabular forms. It is revealed that thermal radiation, thermophoresis and Brownian motion parameters are very effective for improvement of heat transfer. The reported investigation can be used in improving the heat transfer appliances and systems of solar energy.

Characterization of beer-spoilage microorganism and its rapid detection by specific PCR primer (맥주오염미생물의 동정과 specific PCR primer의한 신속한 검출 방법)

  • Several contaminated bacteria such as Lactobacillus brevis and Pediococcus damnosus in beer production cause beer spoilage by producing off flavours and turbidity. Detection of these organisms is complicated by the strict anaerobic conditions and lengthy incubation times required for their cultivation, consequently there is a need for more rapid detection methods. Recently, two contaminated strains were isolated from vessel of beer production and identified as Lactobacillus species by API kit identificaton as well as 16S-23S ITS sequencing analyses. Two isolated strains were named as Lactobacillus sp. HLA1 and Lactobacillus HLB2, respectively. A polymerase chain reaction (PCR) method was developed for the rapid and specific detection of Lactobacillus sp.. Two sets of primer pairs (HLA1-F/HLA1-R and HLB2-F/HLB2-R) were designed for the amplification of a 1576 base pair (bp) fragment of the HLA1 16S-23S rRNA gene and 1888 bp fragement of the HLB2 16S-23S rRNA. Amplified PCR products were highly specific to detect corresponding bacteria when other contaminated strains were used as PCR templates. However, detection of both strains were limited when $100{\mu}{\ell}$ of cultured samples were mixed with $100m{\ell}$ of beer sample in arbitrary manner. The sensitivity of the assay still needs to be improved for direct detection of the small amounts of bacteria present in beer.

Stabilization of Industrial Wastes Landfill using Lab-lysimeter (모형매립조를 이용한 산업폐기물 매립지의 안정화 조사 기초 연구)

  • An experimental research was conducted to establish primary data for the stabilization assessment of industrial wastes landfill with analysis of waste components and investigation of leachate and gas generation, using three sets of lysimeter as experimental apparatus. Comparing results of lysimeter from data of landfill, it is suggested that lysimeter of this study can be used to accomplish the stabilization assessment of the real landfill site. Moisture content was lower as landfill period was older and combustible component was the highest in lysimeter C. The C/N ratio of waste was 7.4~14.4 and, with the elemental analysis, the theoretical gas generation rate based on the modified Buswell equation was 0.47~0.49 $m^3/kg-dry$ waste in lysimeter C. Considering the C/N ratio of leachate, it is concluded that the addition of carbon source is needed to biodegrade leachate hereafter. Gas generation rate($m^3/kg-dry$ waste) from lysimeter A, B and C was 0.0009, 0.014 and 0.0067, respectively, and different from each other according to the landfill period of wastes. The results in this study show that the biodegradation of microorganism for stabilization of landfill was inhibited and more activated in acidogenic step than in methanogenic of anaerobic degradation.

Field Applicability Evaluation Using Effective Microorganism Brewing Cycle for Contaminated Soil in Water Retention Basin (복합발효미생물을 이용한 하천유수지 오염토의 현장적용성 평가)

  • In this study, by using a Effective Microorganisms Brewing Cycle, it confirmed the purification effect of pollutants that are adsorbed on the basins stench removal and retarding soil. On the basis of on-site application test, a soil decontamination system will be suggested. Using a Effective Microorganisms Brewing Cycle, the odor concentration is reduced 2.5 times than that of natural purification treatment method. It was measured and found that the quality of the pore water discharged from the soil is improved. In addition, it was found that a composite of copper and lead with the fermentation microorganisms adsorbed on soil particles from the surface of the stirred experiments lagoon mixed soil is reduced to 65% and 66%, respectively, The TPH organic component was confirmed that the reduction effect of 85%. Restoration of reservoir contaminated soils using the effective microorganism brewing cycle needs to be more developed and implemented as a long-term purification system. This study may be a good reference of developing more complete microorganism brewing system which will efficiently reduce the odor and soil contamination based on optimal stirring and mixing ratio of the compound solutions and contaminated soils in reservoir.

  • Aspergillus oryzae is a filamentous fungus classified in the group Aspergillaceae Ascomycetes. It is an important microorganism for industrial production of enzymes and fermented food productions. The genetic transformation system in A. oryzae was used to protoplast mediated transformation with PEG/$CaCl_2$. When the protoplast was used, the regeneration efficiency was decreased and then transformation frequence was also effected. In this study, fungal transformation was carried out by bypassing the protoplast isolation step, changing enzymes, such as hemicellulase and celluclast, and decreasing the culturing time for the increment of the transformation efficiency. 83 transformants/10ug of DNA with hemicellulase were obtained, compared with less than 10 transformants with novozyme234 and celluclast.

  • This paper discussed about the effect of Zoogloea ramigera on the initial microorganism attachment and the biofilm growth. The additions of 5, 10 and 15%(w/w) of Zoogoea ramigera were facilitated for the initial attachment on the surface of the acryl disk. At biofilm growth, the more Zoogoea ramikera added to the activated sludge, the more biofilm dry weight was obtained. In order to get the stable blofllm and to mlnlmlze the start-up periods, Initial biofilm formation using activated sludge with floe forming microorganisms like Zoogloea ramigera was recommended rather than that without floe forming microorganisms.

  • 본 연구는 하 ${\cdot}$ 폐수 중의 유기물뿐만 아니라 질소, 인을 생물학적으로 제거하는 실험을 수행하였다. 공정 내 미생물의 유실을 방지하고 미생물이 고농도 상태로 유지 가능한 부착성장의 한 공법인 RBC에 끈상 나선형 미생물 접촉재를 설치한 반웅기를 이용하였다. 원수는 Glucose 1,800 mg/L, $NH_{4}Cl$ 500 mg/L, $KH_{2}PO_{4}$ 5mg/L를 혼합한 인공폐수를 제조하여 공정에 주입하였고, 그 결과 각각 미생물이 폐수에 적응하는 단계인 Period 1에서는 각 수질 분석 항목의 농도가 점차적으로 감소하는 경향을 보였으며, 정상상태라고 판단한 Period 2에서는 최종적으로 유입수에 대한 유출수의 제거율은 각각 $TCOD_{Cr}$ 94%, BOD 87%, T-N 85%, T-P 89%의 결과를 나타내었다.
