To study the conditions to enhance success of embryo transfer in the dog, 20 mixed-breed bitches were used for the experiment along with 4 male dogs for mating. The bitches were paired according to synchronism of natural estrus, or the counterpart as donor or recipient was treated with gonadotropin as FSH (follicular stimulating hormone) or PMSG (pregnant mare serum gonadotropin) for induction of estrus to be synchronized with estrus of the other bitch in natural estrus. Embryo recovery was performed in two ways for comparison, either by flushing each uterine horn after ovariohysterectomy or by flushing each horn in the state of non-ovariohysterectomy. In addition, the result of pregnancy according to the embryo stage and the repeatability of the experimental animals as donor or recipient were also investigated. FSH or PMSG was administered to the bitches which had passed over 4 months from last estrus, resulting in estrus-positive in 3 dogs of 6 FSH-treated dogs (50.0%), and in 5 dogs of 9 PMSG-treated dogs (55.6%), determined by proestrus signs and vaginal smear test. Estrus-positive bitches induced with gonadotropin were used as donor or recipient resulting in one embryo-recovered bitch as donor and one offspring-delivered bitch as recipient in 5 PMSG-treated dogs, whereas no result was obtained from 3 FSH-treated dogs. The rate of embryo recovery to be compared with number of corpus luteum was 68.2% in ovariohysterectomized dogs and 55.2% in non-ovariohysterectomized dogs, respectively. The number of dogs from which embryo was collected were 4 dogs of 6 ovariohysterectomized dogs (66.7%) and 6 dogs of 7 non-ovariohysterectomized dogs (85.7%), respectively. The result of parturition was obtained from one dog of 5 estrus-induced recipients, whereas no result was obtained from 3 natural-estrus recipients. The only dog which delivered a male puppy had been transferred 3 morulae and 2 blastocysts. Of 6 repeat-used bitches in canine embryo transfer, 3 dogs showed repeatability either as donor or recipient. These results indicated that inducing estrus of a dog with gonadotropin is feasible in canine embryo transfer to be synchronized with that of a natural-estrus dog, that embryo recovery is also possible in non-hysterectomized dogs, that the estrus-induced dog is also usable as recipient to result in parturition, and that repeat-use of a bitch as donor or(and) recipient is possible in canine embryo transfer.
This study was performed to induce the estrus in 9 anestrus Shih-tzu bitches by intramuscular injection of pregnant mare serum gonadotrophin (PMSG) 50IU/kg for 10 consecutive days and by intravenous injection of human chorionic gonadotrophin (hCG) 1,000IU/Head on Day 10. The day when the first injection of PMSG was counted as Day 0 of experiment. All of the bitches were monitored by vaginal discharges, displays the perineal region, vaginal swelling and male acceptances. The 9 bitches (100%) showed vaginal discharges and vaginal swelling, and were mated. The 5 bitches out of 9 bitches were pregnant (55.6%) and 4 bitches were non-pregnant (44.4%). The 3 bitches out of 5 pregnant bitches were spontaneously delivered (33.3%) and litter size were $1.66{\pm}1.15\;(1{\sim}3\;pups)$ pups. The 2 bitches were diagnosed as early embryonic death on days 38 and 41 after first injection of PMSG. These results indicated that rates of estrus induction, pregnancy and delivery were 100%, 55.6% and 33.3%, respectively, using PMSG and hCG.
This study was focused on improvement of milk production in Mongolian dairy industry by artificial insemination (AI) technology, supported by ODA of KOICA in Republic of Korea. This program was started in January 2009 and it is in $3^{rd}$ years. This manuscript summarized the data especially on estrus synchronization and pregnancy establishment in dairy cows (Holstein) this year. A total of 81 dairy cows from 4 private farms (38 from Undarmal milk and that of 30, 8 and 5 dairy cows from Onjin (Enkhbayer), Jargalant, and BRM School farms respectively) were synchronized with 5 ml Lutalyse (i.m.) in the dump of dairy cows and then estrus was detected 2 to 3 days after $PGF_{2{\alpha}}$ injection. The synchronized dairy cows were inseminated with 0.5 ml dairy frozen semen by conventional artificial insemination (AI) techniques. Pregnancy was diagnosed about 60 days after AI by palpation method. About 96.3% (78/81) of synchronized cows were responded to single $PGF_{2{\alpha}}$ injection. Total 75 over 78 dairy cows (90.1%) inseminated were diagnosed as pregnant. The estrus induction and pregnancy rates were very effective using Lutalyse injection and conventional AI techniques in Mongolian dairy cow. The present results indicated that AI after estrus induction in Mongolian dairy cows could be applied to dairy breeding technology for improving breeding efficiency and milk production of the country.
On January 6, 2010, two months earlier than normal breeding season, a red fox vixen was implanted with synthetic GnRH analogue, Deslorelin. Blood was sampled every 2~3 days from the day of implant to identifying spermatozoa on stains of epithelial cells. Estradiol and progesterone were examined. Even though the vixen was in non-breeding season, she was mated by a male fox. Pregnancy was confirmed by canine pregnancy detection kit that detect relaxin released from placenta. Four healthy pups were born on March 9, 2010. This is the first report showing synthetic GnRH can activate ovarian function and lead to fertile estrus of red fox in non-breeding season.
The present study was conducted to explore the possibilities of estrus induction and superovulation in a native Indian breed of goats called 'Black Bengal'. Forty-two adult non-pregnant females were divided in two groups, of which 18 goats were subjected to a superovulatory treatment comprising of equine chorionic gonadotropin (eCG), Prostaglandin (PGF2$\alpha$) and human chorionic gonadotropin (hCG) to induce superovulation. The remaining 24 goats received no treatment and served as controls for the parameter under study as well as recipients for embryo transfer studies. The average duration of estrus was found to be significantly increased in treated goats (34.2${\pm}$3.4 h) compared to controls 3.0${\pm}$2.4 h). The average duration between PGF administration and occurrence of estrus was 2.0${\pm}$5.2 h. After mid ventral laparotomy, superovulatory responses indicated a significant increase in the number of follicles, which was 8.27${\pm}$0.37 in the treatment group compared to 4.16${\pm}$0.17 in the control group. The number of corpora lutea was also significantly increased in treated animals compared to control (2.90${\pm}$0.86 vs. 0.74${\pm}$0.04) respectively per ovary per goat.
Buffalo heifers weighing from 400 - 500 kg and having a history of reproductive problems like anestrus, irregular estrus or failure to conceive after repeated inseminations were administered subcutaneously with estradiol-$17{\beta}$ and progesterone in two dosage rate 0.1 mg and 0.25 mg, respectively, per kg body weight per day for 7 days in experiment-I and 0.1 mg and 0.125 mg, respectively, per kg body weight per day for 7 days in experiment II. In experiment-I, 9 out of 10 buffaloes responded positively to the hormonal treatment. Milk secretion started between 14-20 days after the start of the treatment. The total milk yield in the successfully induced animals varied from 471.98-625.40 kg. The average daily milk yield varied from 2.08-2.76 kg and peak yield from 3.6-5.3 kg. The time taken to reach peak yield varied from 12-14 weeks. In experiment - II, the established lactation response was absent, although milk secretion process was initiated, the yield could not reach more than 50 - 100 gm at each milking. In experiment - I, the first estrus occurred between days 87 - 231 following the hormonal treatment. Four animals in which lactation was established successfully got pregnant after one or two services. In experiment - II the first estrus occurred between 85 - 173 days following the treatment and only one animal got pregnant.
To establish a protocol of estrus induction and synchronization in European mouflon, we performed artificial insemination using frozen-thawed semen and exogenous hormones. CIDR was inserted into vaginas of four mouflons for 16 days. A day before removal of CIDR, PG 600 was injected intramuscularly. $PGF_2{\alpha}$ was injected when removing CIDR. Artificial insemination was cervically conducted with injecting LHRH 48 hours after CIDR withdrawal. Even though no pregnancy was confirmed, estrous signs were notified like open cervix, congestion of vaginal wall and discharge of cervical mucus. Further research in the wild sheep would be needed for development of artificial breeding methods and advancing sustainability of domestic zoos.
The objectives of this study were to test the efficacy of induction of estrus and determine the timing of ovulation in relation to preovulatory LH and estrogen surges in cycling Murrah buffaloes subjected to Heatsynch protocol (GnRH-$PGF_2{\alpha}$-Estradiol benzoate). In experiment 1, the buffaloes (n = 10) were treated with Heatsynch protocol and observed for estrus and ovulation. In experiment 2 and 3, 30 cycling Murrah buffaloes were used to investigate the efficacy of Heatsynch protocol in terms of conception rates in summer (experiment 2) and winter (experiment 3) seasons. Fixed time A.I. was performed in all the buffaloes at 48 and 60 h post-estradiol benzoate (EB) injection. All buffaloes responded to the Heatsynch protocol with expression of estrus for which ovulations were induced in 8 buffaloes (80%). Mean time interval from the EB injection to ovulation was 50.0${\pm}$2.0 h (range 44.0 to 60.0 h). The interval from the end of LH surge to ovulation was 18.5${\pm}$2.47 h (range 8 to 26 h). The interval from end of estrogen surge to ovulation was 26.75 ${\pm}$2.07 h (range 22 to 36 h). Mean LH peak after EB injection occurred at 20.81${\pm}$1.61 h (range 14 to 28 h) and mean estrogen peak after EB injection occurred at 9.62${\pm}$1.03 h (range 7 to 16 h). Hence, the mean estrogen peak preceded the mean LH peak by 11 h. It was observed that the percentage of conceptions to total number of estruses for control buffaloes was 18 and 30 in summer and winter, respectively, whereas it increased to 26 and 40 in Heatsynch treated buffaloes in respective seasons. The results suggest the possibility of using Heatsynch treatment followed by fixed time A.I. in buffaloes for fertility improvement, especially since the incidence of silent heat in buffaloes is very high.
Morphometric observation on the ovarian follicles of ICR strain-female mice has been conducted to investigate the effect of induced inflammation on the follicular populations. Inflammation was induced by administering 25${\mu}\ell$ of turpentine oil intraperitoneally 2 times at the interval of 3 days. Mice showing 5-day estrous cycle about 20 days after induction of inflammation are placed in experimental group. Normal follicles of 100~399$\mu\textrm{m}$ in diameter increased at estrus but those of over 400$\mu\textrm{m}$ decreased. super-ovulation increased the number of normal follicles of over 400$\mu\textrm{m}$. The number of normal follicles of over 400$\mu\textrm{m}$ 48 hours after superovulation was not significantly different between experimental and control groups. Present results indicated that the number of preovulatory follicles 48 hours after PMSG injection did not decrease in the mice which showed regular estrous cycle even after the induction of inflammation.
Successful induction of lactation in 5 nulliparous repeat breeding cows was achieved by administering diethyl stillboestrol (0.1 mg/kg B.W.) and hydroxyprogesterone caproate (0.25 mg/kg B.W.) for 3 days followed by reserpine (4 mg/day) for 4 days, and dexamethasone (16 mg/day) for one day. The treatment caused cystic ovarian condition in all the animals. Intravaginally progesteronesoaked sponges suppressed but did not eliminate the behavioural estrus. Administration of LHRH ($300{\mu}g$) regressed the cysts in all animals in the first instance but the cysts reappeared in 2 animals. One animal became pregnant and delivered a normal calf.
