Effect of Food and Salinity on Larval Growth and Survival of the River Puffer, Takifugu obscurus (황복, Takifugu obscurus의 초기 발달 동안 성장 및 생존에 있어 먹이와 염분의 효과)

  • In the present study, we investigated the effects of food and salinity on growth and survival of river puffer, Takifugu obscurus offsprings in indoor land-based tank during the early development. In the food experiment, the river puffer larvae (TL 8.7$\pm$0.1 mm, BW 20.0$\pm$3.2 mg) were fed with tubificid, Limnodrilus gotoi, water flea, Daphnia carinata, mysid, Neomysis awatschensis, Manila clam, Ruditapes philippinarum and artificial food together Artemia nauplii for 30 days, and the growth and the survival of larvae were investigated. In the salinity experiment, the river puffer larvae and juvenile at three stages (Stage I: TL 5.8$\pm$0.4 mm; Stage II: TL 12.0$\pm$0.2 mm, Stage III: TL 44.5$\pm$0.7 mm) were supplied with Artemia nauplii, water flea and/or artificial food and were reared in 0, 10, 20 and 30 psu at respective stages. The results shows that the growth rates of larvae feeding with artificial food and tubificid together Artemia nauplii were higher than those of larvae in other groups, but the survival rates of larvae feeding with water flea and mysid together Artemia nauplii were higher than those of larvae in other groups. In salinity, the results shows that the growth and survival rates of offsprings were significantly higher in 10∼20 psu than those in 0 and 30 psu at Stage I and n. At Stage III, the growth and the survival rates were the highest in 20 psu, but the lowest in 0 psu. However, it should be noted that the rates in 0 and 30 psu were significantly enhanced at Stage III in comparison with those at Stage I and II. Therefore, it is concluded that river puffer, T. obscurus is euryhaline marine species, which can normally grow and live at salinities from 0 to 30 psu, and that a combinative supplement with Artemia nauplii and water flea, D. carinata may confer an advantage on growth and survival of the river puffer offsprings in indoor land-based tank.

Dynamics of Air Temperature, Velocity and Ammonia Emissions in Enclosed and Conventional Pig Housing Systems

  • This study aimed to compare the dynamics of air temperature and velocity under two different ventilation and housing systems during summer and winter in Korea. The $NH_3$ concentration of both housing systems was also investigated in relation to the pig's growth. The ventilation systems used were; negative pressure type for the enclosed pig house (EPH) and natural airflow for the conventional pig house (CPH). Against a highly fluctuating outdoor temperature, the EPH was able to maintain a stable temperature at 24.8 to $29.1^{\circ}C$ during summer and 17.9 to $23.1^{\circ}C$ during winter whilst the CPH had a wider temperature variance during summer at 24.7 to $32.3^{\circ}C$. However, the temperature fluctuation of the CPH during winter was almost the same with that of EPH at 14.5 to $18.2^{\circ}C$. The NH3 levels in the CPH ranged from 9.31 to 16.9 mg/L during summer and 5.1 to 19.7 mg/L during winter whilst that of the EPH pig house was 7.9 to 16.1 mg/L and 3.7 to 9.6 mg/L during summer and winter, respectively. These values were less than the critical ammonia level for pigs with the EPH maintaining a lower level than the CPH in both winter and summer. The air velocity at pig nose level in the EPH during summer was 0.23 m/s, enough to provide comfort because of the unique design of the inlet feature. However, no air movement was observed in almost all the lower portions of the CPH during winter because of the absence of an inlet feature. There was a significant improvement in weight gain and feed intake of pigs reared in the EPH compared to the CPH (p<0.05). These findings proved that despite the difference in the housing systems, a stable indoor temperature was necessary to minimize the impact of an avoidable and highly fluctuating outdoor temperature. The EPH consistently maintained an effective indoor airspeed irrespective of season; however the CPH had defective and stagnant air at pig nose level during winter. Characteristics of airflow direction and pattern were consistent relative to housing system during both summer and winter but not of airspeed. The ideal air velocity measurement favored the EPH and therefore can be appropriate for the Korean environment. Further emphasis on its cost effectiveness will be the subject of future investigations.

Quantification of Reproductive Effort and Microscopic Observation on the Larval Development of Manila clam Ruditapes philippinarum (Adams and Reeve, 1850) (바지락 (Ruditapes philippinarum)의 번식량 측정 및 유생발달에 관한 연구)

  • Larval development of the Manila clam Ruditapes philippinarum reared in an indoor tank system was examined in this study using light microscope and scanning electron microscope. To induce spawning and subsequent larval development, clams were collected from the intertidal zone at Gim-nyeong harbor in Jeju Island in August 2011. After 2 days of rearing in the tank, all Manila clams spawned in the midnight. Non-feeding trochophore larvae appeared 7hrsafter fertilization and the first D-shape larvae could be observed at 19 hrs. Twenty one days after fertilization the pediveliger larvae crawling on the bottom of the tank with well-developed foot were observed. Histology indicated that all the clams used in this study were in the ripe stage prior to spawning and the gonad-somatic index (GSI), a ratio of the egg mass to the tissue weight, of the ripe female measured by ELISA was 28.6%. The GSI of female clam declined to 17.3% after the massive spawning in the tank, suggesting that Manila clam discharged 40% of the total eggs during the first spawning event. In conclusion, spawning and subsequent larval development of Manila clam was successfully carried out in this study using an indoor tank system, and the information obtained in the present study could be useful in future Manila clam hatchery development.

An Experiment on the Rearing of Eels with Pellet Feed (Pellet을 이용한 수조내 뱀장어 사육실험)

  • The eels Anguilla japonica were reared in an indoor aquarium of $1m{\times}1m{\times}0.67m$ from June 7 to August 8, 1980. The eels used for the experiment were the smallest ones which were left over from 1974 to 1976 groups, of which the others grew out years earlier. The eels were fed pelletized feed, with the results of 1.05 to 1.678 in feed coefficient, and 0.96 to $1.46\%$ in daily growth rate. The results are well comparable to those of paste feed, which has been being used in the eel farming industry up to date and will be as well used in the future, if any innovative way will not be developed. If pelletized feed can be used without any other hazards, the cost of feed will be remarkably reduced by eliminating costly $\alpha-starch$ which is absolutely necessary for the paste feed preparation, and also by preventing scattering of feed materials which is always associated with paste-feed during feeding time.

Efficacy of Wood Ash as an Antiviral Agent against Cytoplasmic Polyhedrosis Virus of Tasar Silkworm, Antheraea mylitta D.

  • The efficacy of wood ash from Terminalia arjuna (arjun) and T. tomentosa (asan) has been tested against virosis of tasar silkworm, Antheraea. mylitta D. The Polyhedral Occlusion Bodies (POBs) of Cytoplasmic Polyhedrosis Virus of A. mylitta (AmCPV) were exposed to the aqueous solution (0.5 to 4%) of wood ash for 5 to 30 minutes. The treated suspension of POBs was orally inoculated once to tasar silkworm larvae after 24 hours of $1^{st}$ moult, and larvae reared in indoor on arjun leaves till spinning. The application of aqueous solution of wood ash has established its potential as antiviral agent against cytoplasmic polyhedrosis virus. Two percent aqueous solution of wood ash from arjun and asan dissolved the Polyhedral Occlusion Bodies (POBs) of cytoplasmic polyhedrosis virus of tasar silkworm and inactivated the virions within a short period of 20 to 30 minutes. In vivo efficacy of aqueous solution of wood ash resulted in reduction of larval mortality due to virosis. The mortality was reduced to $2.56{\pm}0.21\;and\;3.03{\pm}0.32%$ when treatment of 2.0% solution of wood ash of arjun and asan respectively were applied for 20 minutes, compared to inoculated control $(92.18{\pm}7.52%)$. No mortality was recorded when treatment of 2.5% solution of wood ash of arjun and asan were applied for 10 minutes or more.

Early Gonadal Differentiation of the Protogynous Red Spotted Grouper, Epinephelus akaara

  • Red spotted grouper, Epinephelus akaara is a popular aquaculture species in many Asian countries. This species is a protogynous hermaphrodite that first differentiates into female and changes to male later. Due to this reproductive characteristic, stable supply of male and female gametes is a key to the success of seed production in this species. Thus, understanding early gonadal differentiation is required to develop effective sex control techniques. Red spotted grouper were reared in indoor tanks and sampled every 5 days from 40 days post-hatch (DPH) to 130 DPH. Changes of gonadal tissues were examined and analyzed by means of histology. A pair of gonadal primordium has already existed underneath the kidney in the posterior part of the body cavity at 38 DPH when this study began. Gonadal primordia of 38, 40 DPH consisted of germ cells surrounded by a few somatic cells. The blood vessel was observed in the gonadal primordium at 45 DPH. The number of somatic cells and size of gonadal primordium increased age-dependently up to 60 DPH. Formation of ovarian cavity was obvious by two protuberant aggregations of somatic cells at 65 DPH. Completed ovarian cavity and oogonia were first observed in the gonad of one fish sample at 105 DPH. Based on these histological observations, it can be suggested that induction of primary male differentiation could be more successfully applied at around 60 DPH in this species.

  • Strongylocentrotus pulcherrimus is one of the very important fishery stock in this country since its high demand for raw gonad. Although the demand has been continuously increasing, the natural stock of this species seems to be decreasing. Since fresh macroalgae are not always available, the substitution of terrestrial vegetables for algae as a principal food was examined for the intensive indoor culture of the sea urchin. The results from the studies on selective and non-selective feeding habits and nutritional efficiencies using 13 food stuffs (6 macroalgae, 5 vegetables and 2 animal products) indicate that algae can be substituted with vegetables in sea urchin culture. Although the growth of body weight was the highest when sea urchins were fed Undaria Pinnatifida, the gonad index of the sea urchins fed on spinach, radish leaf and lettuce was higher than that of this brown alga. Considering the different food efficiencies of body and gonad growth, spinach and radish leaf will be adequate during gonad growth season, while lettuce and Chinese cabbage will be suitable for test growth season. In particular, radish leaf, which is usually not eaten by humans, could be highly beneficial and very economical in the sea urchin culture.

Investigation of Habitat and Development of Indoor-rearing Condition of Peacock Butterfly, Inachis io(Linnaeus) (공작나비(Peacock butterfly), Inachis io (Linnaeus)의 서식지 조사 및 실내사육 조건 구명)

  • This study was conducted for investigation of habitat and development of indoor-rearing conditions of peacock butterfly, Inachis io (Linnaeus). Three different region, Mt. Kwangduk-san, Mt. Hae-san and Mt. Taebaek-san in Kangwon-do, was selected for investigation of domestic distribution and ecological environment of peacock butterfly. In result, there are many butterflies in the only two region, Mt. Kwangduk-san and Mt. Hae-san. On 16th April, 4 overwintered butterflies were observed in Mt. Hae-san. At the end of June and early in July, 51 individual next generation's butterflies were observed in Mt. Kwangduk-san and Mt. Hae-san. For development of indoor-rearing conditions, collected each 15 male and female peacock butterflies was reared in a room condition. Female butterflies laid eggs in the egg cluster on the underside of host-plant's leaves, Urtica angustifolia Fisch., in the ovipositioning room. We took 11 egg cluster with average $404.6{\pm}23.6$ eggs. Hatchablity of eggs was 92.3%, and the eggs hatched within $5.1{\pm}0.9$ days from the day of oviposition under high temperature, long day condition($25^{\circ}C$, 14L:10D). The larval period was $18.3{\pm}1.0$ days under high temperature, long day condition($25^{\circ}C$, 14L:10D), showing 81.0% pupal ratio. The head width of each developmental larval stage were $0.37{\pm}0.01mm$(1st instar), $0.66{\pm}0.02mm$(2nd instar), $1.07{\pm}0.05mm$(3rd instar), $1.81{\pm}0.07$(4th instar), $2.76{\pm}0.08$(5th instar). The pupal period was $8.4{\pm}0.6$ days, and the emergence rate was 87.5%.

Ecological Characteristics of Cotesia glomerata L. (Hymenoptera: Braconidae) and Its Parasitism Rates for Diamondback Moth (Plutella xylostella L.) in a Kimchi Cabbage Field in The Korean Highland Area (배추나비고치벌(Cotesia glomerata L.)의 생태적 특성 및 고랭지 배추밭에서 배추좀나방(Plutella xylostella L.)에 대한 기생률)

  • Cotesia glomerata L., an internal parasitoid wasp, attacks the larvae of both the cabbage white butterfly (Artogeia rapae L.) and the diamondback moth (Plutella xylostella L.). It can be utilized as a natural biological enemy to control these two insect pests in the summer cabbage fields of the Korean highland areas. The developmental response and sex ratio of C. glomerata to various temperatures and its longevity were examined in the laboratory. The egg-to-larva and pupa stages of C. glomerata were 12.1 ± 2.1 and 6.4 ± 1.8 days, respectively, at 20℃, The developmental threshold for egg-to-larva and pupa stages were 7.7 and 8.5℃, respectively. The sex ratios of C. glomerata when reared under various temperatures were 61.0 ± 4.5% at 15℃, 44.2 ± 1.0% at 20℃, and 39.0 ± 2.3% at 25℃, and the incidence of females increased as the temperature decreased. The longevity of C. glomerata when fed a 10% sugar solution was 20.4 ± 0.2 days, while in adults without any feed, the longevity was 3.6 ± 0.1 days. Indoor reared C. glomerata adults were released into cabbage fields from 2007 to 2018, in early August of each year, and the outdoor parasitism rates were surveyed. The parasitism rates were found to increase gradually as the year passed (Y = 0.2696X + 2.8633, R2 = 0.3994). The highest parasitism rate was observed in 2013 at 7.6%, and the lowest was in 2018 at 6.5 %. These results could be used as basic information for biological control of kimchi cabbage pests at highland fields.

  • Artificial fertilization of flounder, Limanda herzensteini caught at Pohang brook was performed in April 13, 1995 and hatched larvae were reared for 25 days to describe the development of eggs and the morphogenesis of larvae. The fertilized egg was buoyant, colorless and spherical in shape, measuring 0.86~0.96 mm in diameter (mean: 0.90 mm), The egg has no oil globule, and its perivitelline space was narrow. Hatching in the indoor tank with $16.0^{\circ}C$ in mean water temperature started from the 64 hours after fertilization. The newly hatched larva, measuring about 2.80 mm in total length, had 10~11+25~27=35~38 myomeres. The anus is open at about the anterior point of the body. The 3 to 4 days old larva was 3.90 mm in total length. In this larva the eyes were black and the mouth was open. The yolk was consumed in 8 to 9 days. In 20 to 22 days after hatching, the larvae grew 6.1 mm in total length. The eyes were located symmetry on both sides of head.
