A Study on Malathion-hydrolyzing Esterases of Germinating Seeds of Some Rice Plant Varieties (발아중인 벼 품종별 Malathion 가수분해효소에 관한연구)

  • Kwanok, Fujisaka #5, Paldal, and Suwon #82 as japonica type and IR-262 and CP-slo as indica type of rice seeds were selected for this experiment among varieties grown in Korea. Activities of crude enzymes extracted from germinating seeds of these varieties on malathion and p-nitrophenyl acetate as substrates, esterase zymograms with 1-naphthyl acetate as substrate, and some observations are summarized as follows: 1. Activities per unit volume of crude enzyme preparations on malathion were in the order of Kwanok>IR-262>Fujisaka #5>CP-slo>Paldal>Suwon #82. 2. Esterase zymograms on agar-gel electrophoretograms exhibited three to four bands two electrodes with little difference among varieties, nevertheless showing a wide and strongly-colored band toward cathode. Suwon #82 has a somewhat different pattern from others. 3. Enzyme activities per milligram protein with p-nitrophenyl acetate as substrate were in the order of CP-slo>IR-262>Paldal>Kwanok>Suwon #82>Fujisaka #5, indicating that activities of indica type are much stronger than those of japonica type, but not in agreement with results with malathion. 4. Malathion did not much inhibit the esterase activity at the concentration of 0.2PPM on electrophoretograms. 5. It is supposed that there is a complex esterase system hydrolyzing malathion and p-nitrophenyl acetate in germinating rice seeds.

Studies on the Varietal Differences of Tolerance to Cold Damage in Seedling Stage of the Rice Plant (수도 유묘기 내냉성의 품종간 차이에 관한 연구)

  • In Korea, two types of rice varieties are being cultivated widely, Japonica and Japonica-Indica cross. Generally speaking, the latter shows weak cold tolerance than the former and so brought about many problems in cultural practices. This study was conducted to investigate the effect . of various temperature conditions on rice seedlings growth, especially the contents of chlorophyll and the percentage of nitrogen. And the result obtained indicated the varieties of Japonica-Indica intend to weak cold tolerance than Japonica varieties.

Mapping of grain alkali digestion trait using a Cheongcheong/Nagdong doubled haploid population in rice

  • We performed a molecular marker-based analysis of quantitative trait loci for traits that determine the quality of appearance of grains using 120 doubled haploid lines developed by anther culture from the F1 cross between 'Cheongcheong' (Oryza sativa L. ssp. Indica) and 'Nagdong' (Oryza sativa L. ssp. Japonica). We therefore calculated the alkali digestion value (ADV), used to indirectly measure gelatinization temperature, to evaluate the quality of cooked rice in 2013 and 2014. The ADV score of frequency distribution was higher milled rice than brown rice. In total, nine different quantitative trait loci (QTLs) were found on 5 chromosomes in 2013 and 2014. Also, chromosome 5, 8 were detected over two years. We conclude that selected molecular markers from this QTL analysis could be exploited in future rice quality. In conclusion, we investigated ADV of brown and milled rice in CNDH population. This study found nine QTLs related to the ADV of brown and milled rice. The detected one marker can be used to select lines with desirable eating-quality traits because ADV is closely associated with the eating quality of cooked rice. Therefore, it will be useful to collect resources and distinguishable in many varieties for rice breeding program.

Effects of Seed-Soaked $GA_3$ and Inorganic Salts on Mesocotyl and Coleoptile Elongation in Rice

  • The elongation of mesocotyl and coleoptile plays important roles in the seedling emergence and stand establishment of dry direct-seeded rice. Experiments were carried out to elucidate the effects of seed-presoaking treatments of GA$_3$ and some inorganic salts on the mesocotyl, and coleoptile elongation of rice. Seed-soaked GA$_3$ promoted the elongation of mesocotyl, but little effect on the coleoptile elongation. The stimulation effects of GA$_3$ were found to be enhanced by addition of CaCl$_2$ However, the sole treatment of CaCl$_2$ showed no stimulating effect on the mesocotyl and coleoptile elongation. Mesocotyl elongation was most prominent in the combined treatments of 50ppm GA$_3$ with 100 mM CaCl$_2$. The synergistic effects of GA$_3$ and CaCl$_2$ on mesocotyl elongation varied with varietal groups. The stimulating effects of GA$_3$ were enhanced significantly by the addition of CaCl$_2$ in japonica varieties, Dongjinbyeo, Ilpumbyeo and Milyang 95, and tall indica variety, Labelle, but not in semidwarf Tongil type varieties, Tongilbyeo, Milyang 23, and Nampungbyeo, and semi-dwarf indica, Short Labelle. The promoting effects of GA$_3$ on the mesocotyl elongation were decreased in proportion to the lowered osmotic potential by PEG 6000 on the contrary to CaCl$_2$ This implies that the synergistic effects of CaCl$_2$ with GA$_3$ on mesocotyl elongation was not caused by osmotic potential lowered by CaCl$_2$ addition but by the salt itself. Salts such as Ca(NO$_3$)$_2$, MgCl$_2$ BaCl$_2$, NaCl, KCl and KNO$_3$ showed the synergistic effects with GA$_3$ on mesocotyl elongation as well. The degree of synergistic effects showed no differences among salts tested, implying that there is no specificity of ions constituting the salts.

  • Chemical compositions of Japonica and J/Indica brown rices were compared. No significant differences were noted for amino acid composition of brown rice among varieties. The first and second limiting amino acids of brown rice were lysine and isoleucine, respectively. The major fatty acids of brown rice were palmitic, oleic and linoleic acid, which comprised of 96% of total fatty acid. The contents of saturated fatty acids were lower in Japonica varieties. The average value of calcium over phosphorus was 0.052.

Studies on the Duration and Rate of Grain Filling in Rice (Oryza sativa L.) I. Varietal Difference and Effects of Nitrogen (수도의 등숙기간 및 등숙속도 연구 I. 품종간 차이 및 질소의 영향)

  • Understanding grain filling characteristics represented by grain filling duration and grain filling rate is import-ant in improving higher yielding varieties and developing better cultural methods of rice (Oryza sativa L.). Recently developed 6 Japonica and 6 Japonica/Indica varieties were grown under 3 nitrogen levels at Experimental Farm of Chungbuk National University in 1986. The range of grain filling duration of 12 varieties was 20.9-39.0 days, while grain filling rate ranged from 53.8 to 136.6 mg. panicle$^{-1}$. day$^{-1}$. Although the difference of the average grain filling duration between Japonica varieties and Japonica/Indica was less than 4 days, the average grain filling rate of Japonica/lndica varieties was greater than Japonica more than 30%. Samgangbyeo showed the shortest grain filling duration of 21.0-24.2 days and the greatest grain filling rate of 119.3-143.8 mgㆍpanicle$^{-1}$. day$^{-1}$ under 3 nitrogen levels, while Seomjinbyeo and Milyang 23 showed the quite opposite grain filling characteristics. Nitrogen levels did not show any significant effects on grain filling characteristics. Negative correlation was round between grain filling duration and grain filling rate, and significant positive correlations of grain filling rate with grains/panicle, grain weight and panicle weight indicated that grain filling rate is more important characteristics of grain filling. Pathway analysis revealed that contribution of grain filling rate to panicle weight is rather indirect through grain weight.
