QTL Mapping of Cold Tolerance at the Seedling Stage using Introgression Lines Derived from an Intersubspecific Cross in Rice

  • Low-temperature stress is an important factor controlling the growth and development of rice (Oryza sativa L.) in temperate region. In this study, a molecular linkage map consisting of 136 SSR markers was employed to identify QTL associated with cold tolerance at the seedling stage. 80 recombinant inbred lines (RILs) from an intersubspecific cross between Milyang23 (O. sativa ssp. Indica) and Hapcheonaengmi3, a japonica weedy rice and the parents were evaluated for leaf discoloration and SAPD value of seedlings. Rice plants were grown for 15 days in the low-temperature condition (13/20℃ day/night) and the control condition (25/20℃ day/night) in the growth chamber. The degree of leaf discoloration showed a highly significant correlation with the SPAD value in the low-temperature plot (r = -0.708, P < 0.0001). A total of four QTLs for SPAD were identified and the phenotypic variance explained by each QTL ranged from 5.4 to 16.0%. Two QTLs detected in the control condition were located on chromosomes 2 and 5, respectively. Two QTL on chromosomes 1 and 4 were detected at the low-temperature condition and Hapcheonaengmi3 alleles increased the SPAD values at these loci. Substitution mapping was conducted to delimit the position of qSPA-4 using introgression lines derived from the same cross. Results indicated that qSPA-4 was located in a 810-Kb region flanked by RM16333 and RM16368. The results indicated that Hapcheonaengmi3 contains QTL alleles that are likely to improve cold tolerance of Indica rice.

DNA Fingerprinting of Rice Cultivars using AFLP and RAPD Markers

  • This experiment was conducted to evaluate genetic variation in 48 rice accessions (Oryza sativa L.) using AFLP and RAPD markers. For AFLP, a total of 928 bands were generated with 11 primer combinations and 327 bands (35.2%) of them were polymorphic among 48 accessions. In RAPD analyses using 22 random primers 145 bands were produced, and 121 (83.4%) were polymorphic among 48 accessions. Each accession revealed a distinct fingerprint by two DNA marker systems. Cluster analysis using AFLP-based genetic similarity tended to classify rice cultivars into different groups corresponding to their varietal types and breeding pedigrees, but not using RAPD-based genetic similarity. The AFLP marker system was more sensitive than RAPD in fingerprinting of rice cultivars with narrow genetic diversity.

Improvement of Cultural Efficiency Using DNA Markers in Anther and Seed Culture of Rice (DNA marker를 이용한 벼의 조직배양 효율 재선)

  • The purpose of this study is to improve the culturability of a indica type rice cultivar, IR 36, using DNA marker associated with the ability of plant regeneration in anther and seed culture. The varietal difference of ability of callus formation and plant regeneration was investigated in anther and seed culture of 8 rice cultivars. Three japonica rice cultivars showed to have better culturability than those of tongil and indica type genotypes. But two indica/japonica lines, 'MGRI 079' and 'MGRI 036', which were selected to have good culturability in previous study showed the highest regenerability (20%) in anther culture of 8 rice cultivars. Thirty four $BC_2F_4$ lines were selected by marker screening using RZ400 for 100 $BC_2F_4$ lines derived from a cross $'MGRI\;079/IR\;36^{^*3}'$. The frequency of callus formation of 30 $BC_2F_4$ lines was higher than those of 'IR 36' in anther culture of the selected $BC_2F_4$ lines. The ability of plant regeneration of 15 lines was higher than that of 'IR 36' in the seed culture of 34 $BC_2F_4$ lines. A promising line, $BC_2F_4-28$, was selected to have better culturability in the anther and seed culture of the $BC_2F_4$ lines. The heading date and grain shape of the $BC_2F_4-28$ was similar to 'IR 36'. In seed culture of 50 $BC_4F_3$ lines derived from a rice cross $'MGRI\;079/IR\;36^{^*5}'$, 11 lines including $BC_4F_3-3$ showed to have higher regenerability compared with 'IR 36'. The highest frequency of plant regeneration (11%) was obtained from $BC_4F_3-46$ in seed culture of the $BC_4F_3$ lines.

  • A total of 16 varieties from Korea and Indonesia were tested in Suwon, Korea (126$^{\circ}$9'E, 37$^{\circ}$16'N, 37m amsl) and Bali, Indonesia (115$^{\circ}$ 14'E, 8$^{\circ}$42' and 10m amsl). Most varieties of Indica and India/Japonica types showed adaptability to both areas. Generally Indica-type showed more number of panicles per hill than that of Indica/Japonica, while Indica/Japonica showed better performance in spike let number per panicle and 1,000 grain weight than Indica-type varieties.

Utilization of Growing Degree Days as an Index of Growth Duration of Rice Varieties (Growing Degree Days를 이용한 수도품종의 생육기간 측정방법과 이용)

  • To evaluate growing degree days(GDD) as an index of growth duration of rice plants, 30 days old seedlings of 16 japonica and 14 indica/japonica varieties were transplanted six times from May 10 at the 10-day intervals at Gyeongsan in 1982. The number of days from transplanting to heading decreased as transplanting dates delayed in all japonica varietie and 4 indica/japonica varieties but that of 10 indica/japonica varieties decreased up to June 9 or June 19 transplantings and then it levelled off or increased with further delay of transplanting. However, GDD requirement was similar among transplanting dates at appropriate base temperatures; GDD could be better than calendar day system to classify maturity of varieties especially grown in a wide range of climatic conditions. Required GDD from transplanting to heading of all indica/japonica and early japonica varieties showened a smaller coefficient of variation (CV) compared to longer season japonica varieties. Among GDD methods, an accumulation of daily Max + Min temp./2 -$l0^{\circ}C$ showed the smallest CV for the duration from transplanting to heading, but for ripening period GDD calculated with adjusted maximum temperature when it was higher than $30^{\circ}C$ showed the best results. Heading date did not affect required GDD for maturity of japonica varieties, but in indica/japonica varieties GDD decreased as heading date delayed; at late transplantings ripening period of indica/japonica varieties was less extended compared to japonica varietes due to a decrease in grain weight.

  • This study was carried out to know the physiological characteristics related to flooding tolerance of rice plants. Peroxidase specific activities and banding pattern of peroxidase isozyme of 24 days old seedlings were analyzed after 3 days of flooding treatment in the artificial flooding tank. Peroxidase activities of japonica rice varieties which were relatively susceptible to submergence were higher in comparison to those of Tongil and indica rice varieties. And a peculier band of peroxidase isozyme which was not shown in any part of rice plant if not flooded, was appeared at the around 9 of isoelectric point in the leaf blade of japonica rice varieties when flooded.

Development of a Rough Rice Handling Equipment for In-Bin Drying and Storage System (개량 곳간용 벼이송장치(移送裝置) 개발(開發))

  • The objective of this study is to develop a grain handling system for loading, unloading and transporting of rough rice stored at the in-bin drying and storage (IBDS) developed by the Korea Advanced Institute of Science and Technology(KAIST). A mechanized Fain handling system consisted of a portable auger and a gate was developed and tested. The test results can be summarized as the following: 1) The loading capacity of the handling system developed is $16.2m^3/h$ (8.3 ton/h) for the Indica type rice and $13.0m^3/h$(7.3 ton/h) for the Japonica type. It is greater than that of manual handling as much as 2.5 - 2.7 times. 2) The unloading capacity of the handling system developed is $16.0m^3/h$(8.2 ton/h) for the Indica type rice and $12.6m^3/h$(7.0 ton/h) for the Japonica type. It is greater than that of the manual as much as 4.7 - 5.5 times. 3) For 3-ton capacity of the storage, the loading and unloading can be performed for 20 and 30 minutes by one man operation of equipment. while 60 and 120 minute for the manual of 2 men, respectively. 4) The volumetric efficiency of the system developed is 0.42 - 0.54 and the power efficiency is 4.0 - 4.4. 5) The break-even quantity of the handling system developed is about 38.6 ton($68.7m^3$) of rough rice and the initial investment for the system would be returned within five years for the most owners of the KAIST IBDS system.

Response and Acetolactate Synthase Activity in Different Rice Cultivars(Oryza sativa L.) to Cinosulfuron (Cinosulfuron에 대한 벼 품종간의 생육반응과 Acetolactate Synthase 활성에 미친 영향)

  • Acetolactate synthase(ALS) activity was determined in germinating seedlings of two rice cultivars treated with cinosulfuron [3-(4,6-dimethoxy-1,3,5-triazin-2-yl)-1-[2-methoxyethoxy)-phenylsulfonyl]-urea]. IR 74(Indica type) was more tolerant than Hwajinbyeo(Japonica type) under various rates of cinosulfuron applied at the pregermination stage. In vitro response of ALS activity in the two rice cultivars was similar to $I_{50}$ values(cinosulfuron concentration required for 50% inhibition of ALS activity) of about 23ppb. In vivo, ALS activity of IR 74 increased as the seedlings grew, but that of Hwajinbyeo dropped at 5 days after 10ppm cinosulfuron treatment and shoot growth of Hwajinbyeo lagged at 4 to 5 days after herbicide treatment. ALS activity and shoot growth of Hwajinbyeo was resumed from cinosulfuron-induced inhibition at 6 days after cinosulfuron treatment. The differential response of ALS activity in two different rice cultivars against cinosulfuron may not be due to difference of ALS sensitivity, but rather due to different metabolic inactivation rates of cinosulfuron.

Genetic Analysis of Growth Response to Cold Water Irrigation in Rice

  • This study was carried out to obtain the basic information for breeding cold-tolerant rice varieties with high yield-productivity through wide crosses between indica and japonica rice. Genetic analysis was conducted using 55 F$_1$s obtained from half-diallel crosses among eleven cultivars of various origin including indica and japonica rice. Screening for cold tolerance was done with cold-water irrigation after transplanting until ripening stage. Both general combining ability (GCA) and specific combining ability (SCA) effects were highly significant in all characters associated with dry matter accumulation at 30 and 50days after cold-water irrigation (DAC). The variance of GCA was much larger than that of SCA in plant height, shoot dry weight per plant (DWP), crop growth rate (CGR) and cold-water response index (CRI) of these characters except CRI of shoot dry weight per plant. The DWP, CGR and CRI of these characters of Gaochan 102, Tong88-7 and TR22183 were markedly higher than those of the others. GCA effects of these varieties on DWP, CGR and their CRI were also higher than those of the others, indicating that they are useful as promising parents for breeding cold-tolerant varieties. Analysis of genetic parameters for 11$\times$11 half-diallel F$_1$s revealed that inter-locus gene interaction were concerned in the expression of plant height at 50 DAC, CRI of DWP at 50 DAC, and CRI of CGR, and that intra-locus gene interaction for plant height and the other characters were partial dominance and over-dominance, respectively. Narrow-sense heritability (h$^2$$_{N}$) was the highest in plant height as 0.729, and the lowest in CRI of DWP at 30 DAC as 0.048, suggesting that selection for cold tolerance will be quite effective in case that the selection criterion is the performance itself.f.

Differential Scanning Calorimetric Study of Amylose-lipid Complex and Amylose Content in Rice Starch (쌀 전분의 Amylose-lipid Complex 의 DSC 특성과 Amylose 정량)

  • Thermal properties of amylose-lysolecithin (AL) complex, amylose content and effect of lysolecithin on the gelatinization of rice starch were investigated by Differential Scanning Calorimetry (DSC). The melting temperature of AL complex was near to $108.5^{\circ}C$ and the melting enthalpy was about 1.0cal/g. The gelatinization temperature of rice starch was not affected by adding lysolecithin. However, the enthalpy of gelatinization was decreased. The amylose contents in rice varieties were calculated from melting enthalpy of AL complex. The amylose contents for Indica and Japonica types of rice were in the range of 16-19%, which were in good agreement with those determined by iodine binding method. Significant differences were not observed in the amylose contents between Indica and Japonica varieties.
