Application of Response Surface Methodology (RSM) for Optimization of Anti-Obesity Effect in Fermented Milk by Lactobacillus plantarum Q180

  • Obesity, a condition in which an abnormally large amount of fat is stored in adipose tissue, causing an increase in body weight, has become a major public health concern worldwide. The purpose of this study was to optimize the process for fermented milk for the production of a functional product with an anti-obesity effect by using Lactobacillus plantarum Q180 isolated from human feces. We used a 3-factor, 3-level central composite design (CCD) combined with the response surface methodology (RSM). Concentration of skim milk powder (%, $X_1$), incubation temperature ($^{\circ}C$, $X_2$), and incubation time (h, $X_3$) were used as the independent factors, whereas pH (pH, $Y_1$), anti-lipase activity (%, $Y_2$) and anti-adipogenetic activity (%, $Y_3$) were used as the dependent factors. The optimal conditions of fermented milk for the highest anti-lipase and anti-adipogenetic activity with pH 4.4 were the 9.5% of skim milk powder, $37^{\circ}C$ of incubation temperature, 28 h of incubation time. In the fermentation condition, the predicted values of pH, anti-lipase activity and anti-adipogenetic activity were 4.47, 55.55, and 20.48%, respectively. However, the actual values of pH, anti-lipase activity and anti-adipogenetic activity were 4.50, 52.86, and 19.25%, respectively. These results demonstrate that 9.5% of skim milk powder and incubation at $37^{\circ}C$ for 28 h were the optimum conditions for producing functional fermented milk with an anti-obesity effect.

  • The behaviors of inorganic nitrogen derived from solid animal waste in soil has been received too much concern partly because nitrate which occurred from nitrification can act as a pollutant to soil and groundwater and partly because the loss of nitrogen from surface soil by downward movement of water is disadvantageous in the view of plant nutrient. This present study was conducted to get fundamental imformations on nitrogen behavior and to provide improved basical concepts on the management of animal waste. Fresh or fermented pig manure was mixed with a sandy loam soil in the ratio of 2:1(soil:pig manure), packed into test tube and incubated at $30^+/-1^{\circ}C$ for 8 weeks under aerobic- or anaerobic condition. Sample tubes were taken at the one week interval and analyzed on pH, the amount of $CH_4$ produced under anaerobic condition and inorganic nitrogen. The pH of soil treated with fresh pig manure under anaerobic condition was lowered by 1.87 unit compared to that of under aerobic condition, but at the treatment with fermented pig manure, pH change was very little between aerobic and anaerobic condition. The coefficients of regressional equations which were obtained from pH and incubation time were -0.114 in fresh pig manure and -0.089 in fermented pig manure, and the extent of pH decrease due to incubation was greater in fresh pig manure than that of fermented pig waste. No differences in the amounts of $CH_4$ produced under anaerobic condition between fresh and fermented pig manure was observed until 3 weeks of incubation, however, after that the amount of $CH_4$ produced in fresh pig manure was abruptly increased and cumulative amont of $CH_4$ was reached 8.6 mole/g. K values on $CH_4$ production in fresh and fermented pig manure was 0.211 mole/g/day and 0.046 mole/g/day, respectively, for 5 weeks from the 3rd to the 8th week. $NH_4-N$ concentration at aerobic condition with fresh pig manure treatment was lowered by passing time of incubation, but $NO_3-N$ concentration was elevated from 11.2 ppm at initial state to 67.3 ppm after incubation and this trend on $NH_4-N$, $NO_3-N$ concentration was very similar to the treatment of fermented pig manure. While $NH_4-N$ concentration under anaerobic condition was greatly increased. $NO_3-N$ concentartion was not only very low but also no great changes, that was ranged from 4 to 8 ppm.

  • A new method to calculate the transmittable prevalence of an epidemic disease is proposed based on a back-calculation formula. We calculated the probabilities of reactivation and of parasitemia as well as transmittable prevalence (the number of persons with parasitemia in the incubation period) of malaria in South Korea using incidence of 12 years(2001-2012). For this computation, a new probability function of transmittable condition is obtained. The probability of reactivation is estimated by the least squares method for the back-calculated longterm incubation period. The probability of parasitemia is calculated by a convolution of the survival function of the short-term incubation function and the probability of reactivation. Transmittable prevalence is computed by a convolution of the infected numbers and the probabilities of transmission. Confidence intervals are calculated using the parametric bootstrap method. The method proposed is applicable to other epidemic diseases in other countries where incidence and a long incubation period are available. We found the estimated transmittable prevalence in South Korea was concentrated in the summer with 276 cases on a peak at the $31^{st}$ week and with about a 60% reduction in the peak from the naive prevalence. The statistics of transmittable prevalence can be used for malaria prevention programs and to select blood transfusion donors.

  • Incubation condition affecting the chlamydospore formation and isolation from mycelia and conidia of Cylindrocarpon destructanse (isolate ACY-9701), isolated from the root rot lesion of the American ginseng (Panax quinquefolium) was investigated. Chlamydospores were formed from mycilia but not from conidia on the Czapek-Dox agar without carbon or nitrogen source after 20 days incubation at 2$0^{\circ}C$. In the medium added with nitrogen and carbon sources, immatured chlamy-dospore-like cells were formed from microconidia and mycelia as well. Immatured chlamydospore-like cells were formed from mycelia as well as microconidia In corn, kidney bean, and pea root extracts after 20 days incubation at 20"C, while typical chlamydospores were formed from both of them in the root extract of Panax quinquefolium. The 3.6 log chlamydospore/mm" was converted from microconidia in the medium, which was equal to 2.5% conidia formed. Under the light condition (251.1 pmol/m" sec, 12 hrs dark and light cycle), 4.2 log/mm" of chlamydospores were converted from interracially or terminal cells of macroconidia, which was 4.0% of macroconidia produced on Potato dextrose agar (PDA). When mycelia and microconidia were stored at -7$0^{\circ}C$ for 32 days and incubated on PDA after thawing at room temperature to isolate chlamydospores from them, microconidia and mycelia were still alive. Meanwhile, microconidial lysis was found after heating them at 32$^{\circ}C$ for 7 days, but the chlamydospores converted from macroconidia were not lysed up to 13 days at 32"C. to 13 days at 32"C.ot;C.

Effect of Phase Separation on Anaerobic Degradation of Phenol (페놀의 혐기성분해에 대한 상분리의 영향)

  • With the rapid industrialization, an ever-increasing quantity and kind of new organic compounds pose environmental problems due to their toxicity and physiological effect. However, research on the biodegradation of these compounds under anaerobic condition is very limited inspite of its efficiency and economical advantage. In this research, the pH effect on the ring cleavage of phenol under anaerobic condition was investigated, and the theory of phase separation was applied to the degradation of phenol for investigating the role of acidogenic bacteria. Results, obtained from biochemical methane potential(BMP) assay for 15.5 days of incubation, showed that acidic condition was more desirable for phenol degradation than alkaline condition. By both unacclimated methanogenic granular sludge and two mixed cultures, phenol was completely removed within six weeks of incubation with a gas conversion rate of over 86% of theoretical one. However, phenol was not degraded by unacclimated acidogenic culture, and thus it is considered as a syntrophic substrate. In case of phase separated biochemical methane potential(PSBMP) assay, in which acidogenic and methanogenic culture were seeded separately and consecutively, those that had been subjected to normal acidogens for 3 to 4 weeks showed higher gas production than those seeded with sterile or frozen culture.

Effects of Strains of Saccharomyces cerevisiae and Incubation Conditions on the In vitro Degradability of Yeast and Roughage

  • The in vitro degradability of yeast and the effect of yeast on the in vitro degradability of forage may differ in terms of the specific yeast strains or their incubation conditions. Thus in experiment 1, two strains of sake yeast (strainK7 and strainK9) and one strain of bakers' yeast (KY5649) were incubated in an aerobic condition. In experiment 2, aerobically or anaero bically incubated K7 was used for investigating the in vitro degradability of yeast, the effect of yeast on the in vitro degradability of forage, and the degradability of yeast by pepsin and pronase treatment. The in vitrodegradability of bakers' yeast was significantly (p<0.05) higher than those of sake yeasts. The in vitro degradability of anaerobically incubated yeast was significantly (p<0.01) higher than that of aerobically incubated yeast. The degradability of bakers' yeast by pepsin treatment was significantly (p<0.01) higher than that of the sake yeasts. The degradability of bakers' yeast by pronase treatment was slightly higher than that of the two sake yeasts, while the degradability of anaerobically incubated yeast by both enzymes, respectively, was significantly (p<0.01) higher than that of aerobically incubated yeast. The degradability of forages was increased significantly (p<0.05) by the addition of yeasts. The degradability of roughage by sake yeast tended to be higher than that by the bakers' yeast. The degradability of roughage was significantly (p<0.05) higher by anaerobically incubated yeast than by aerobically incubated yeast. Given the above results, it seems that in vitro degradability of yeast and the magnitude of the increment of roughage degradation differ among the yeast strains and their incubation conditions.

  • The Aspergilli can be isolated from many of damaged domestic fowl's lung which were collected at fowl slaughter house from July to August 1973, during two months at Dapsimri, Dongdaemoon-ku, Seoul city. The survey of incubation condition for carbon source and nitrogen source were summerized as follows: 1. One of the Aspergillius fumigatus group were isolated from damaged domestic fowl's lung. 2. In the preparation of isolation media, adding the 4 gs of silkworm pupa and Meju to Czapek's agar medium is more preferable than Austick's media (2% malt extract agar) 3. In survey of incubation condition, the growth rate and sporulation shown that higher differences according to varieties and amounts of C-source. And in the case of 20% C-source concentration shown that low growth rate than lower concentration of C-source. 4. In N-source, the growth rate indicated that some difference according to varieties of N-source, difference of growth rate demonstrated almost not shown at various amounts differences of mycelium and sporulation was only detected reversely.

Estimation of N Mineralization Potential and N Mineralization Rate of Organic Amendments in Upland Soil

  • Management of renewable organic resources is important in attaining the sustainability of agricultural production. However, nutrient management with organic resources is more complex than fertilization with chemical fertilizer because the composition of the organic input or the environmental condition will influence organic matter decomposition and nutrient release. One of the most effective methods for estimating nutrient release from organic amendment is the use of N mineralization models. The present study aimed at parameterizing N mineralization models for a number of organic amendments being used as a nutrient source for crop production. Laboratory incubation experiment was conducted in aerobic condition. N mineralization was investigated for nineteen organic amendments in sandy soil and clay soil at $20^{\circ}C$, $25^{\circ}C$, and $30^{\circ}C$. N mineralization was facilitated at higher temperature condition. Negative correlation was observed between mineralized N and C:N ratio of organic amendments. N mineralization process was slower in clay soil than in sandy soil and this was mainly due to the delayed nitrification. The single and the double exponential models were used to estimate N mineralization of the organic amendments. N mineralization potential $N_p$ and mineralization rate k were estimated in different temperature and soil conditions. Estimated $N_p$ ranged from 28.8 to 228.1 and k from 0.0066 to 0.6932. The double exponential model showed better prediction of N mineralization compared with the single exponential model, particularly for organic amendments with high C:N ratio. It is expected that the model parameters estimated based on the incubation experiment could be used to design nutrient management planning in environment-friendly agriculture.

Studies on the Intestinal Microflora of Chicken Under Tropical Condition

  • Three media, i. e., MOD-SD, M98-5 and M98-5 supplemented with chicken fecal extract were tested as isolation media for anaerobic bacteria present in the duodenum, jeju-ileum and cecum of chicken. The results showed that the mean colony counts of medium M98-5 were similar with those of MOD-SD medium in all intestinal samples at the incubation periods of 2, 6 and 10 days. Supplementation with chicken fecal extract of M98-5 medium significantly increased (p < 0.05) the colony counts of bacteria from the duodenum, jeju-ileum and cecum. The colony counts at 6-day incubation were similar with those at 10-day incubation, but were much higher than the counts at 2-day incubation. The major types of bacteria found in the duodenum and jeju-ileum of chicken were tentatively identified as Lactobacillus, Streptococcus and E. coli. In the cecum, ten tentatively identified groups of bacteria, namely, Streptococcus, Staphylococcus, Lactobacillus, E. coli, anaerobic coccus, Eubacterium, Propionibacterium, Clostridium, Fusobacterium and Bacteroides were isolated. Anaerobes were found to comprise nearly the entire microbial population of the cecum. Predominating in all sections of the intestine were homofermentative lactobacilli. The main Lactotacillus species in chicken intestine were L. acidophilus, L. fermentum and L. brevis.

Selective Si Epitaxial Growth by Control of Hydrogen Atmosphere During Heating-up (승온중 수소 분위기 제어에 의한 선택적 Si 에피텍시 성장)

  • we proposed the use of $Si_2H_ 6/H_2$ chemistry for selective silicon epitaxy growth by low-pressure chemical vapor deposition(LPCVD) in the temperature range $600~710^{\circ}C$ under an ultraclean environment. As a result of ultraclean processing, an incubation period of Si deposition only on $SiO_2$ was found, and low temperature epitaxy selective deposition on Si was achieved without addition of HCI. Total gas flow rate and deposition pressure were 16.6sccm and 3.5mtorr, respectively. In this condition, we selectively obtained high-quality epitaxial Si layers of the 350~1050$\AA$ thickness. In older to extend the selectivity, we kept high pressure $H_2$ environment without $Si_2H_6$ gas for few minutes after first incubation period and then we conformed the existence of second incubation period.
