Difference of Neuronal Recovery by Incubation Condition after Transient Hypoxia (배양조건에 의한 일과성 저산소상태 후 신경세포회복의 차이)

  • Objective : The transverse hippocampal slice is one of the most commonly studied in vitro models of mammalian brain physiology. However, despite its broad usage, there has been no standardization of slice preparation techniques or recording condition. It is well known that variations in recording conditions can result in profound different effects to neuronal responses. Evoked field potentials, recorded extracellularly, were used to investigate the effects of variations in hippocampal slice preparation protocol on hypoxia responses of CA1 neurones. Material & Methods : Before hypoxic injury, hippocampal slices were incubated for 4 hours. During incubation period, the slices were placed in a incubation chamber($21^{\circ}C$) for recovery from preparation injury and then transferred to recording chamber($34^{\circ}C$) for more recovery and baseline electric recording with current stimulation(0.1Hz). Various time periods in incubation chamber and recording chamber were applied to each experimental group(group 1=60min : 180min, group 2=90min : 150min, group 3=180min : 60min, time in incubation chamber : time in recording chamber) before 10 min hypoxia produced by replacing 95% $O_2$+5% $CO_2$ mixed gas to 95% $N_2$+5% $CO_2$ gas. Calcium, Magnesium ions and several drugs effecting on glutamate receptor also were studied. Recoveries from hypoxic injury of hippocampal slices were estimated by percent recovery of population spike(PS). Statistic analysis of study were performed using paired t-test. Results : The percent recovery of PS after 10min hypoxia was considerably enhanced by increasing the period of current stimulation during incubation period before hypoxic injury. Temperature effect on the result of this experiment was also studied(group 4) but the result from this showed no statistic significance. Low magnesium ion concentration of artificial CSF(Mg-free aCSF) during incubation period enhanced the recovery of PS but low calcium (calcium-free) and high magnesium ion concentration(2mM) reduced it after hypoxic injury. L-glutamate($100{\mu}M$) and AP-5($50{\mu}M$) had no effect on the recovery of PS but CNQX($10{\mu}M$) in artificial CSF during incubation period markedly enhanced the recovery of PS. Co-treatment of AP-5($50{\mu}M$), CNQX($10{\mu}M$) and high magnesium concentration(2mM) enhanced recovery of PS in immediate following period of hypoxic injury but the effect of cotreatment after then decayed rapidly and lost statistic significance. Conclusions : Judging from above results, the condition of baseline recording is important in observing the recovery of population spike after hypoxia, and the time and the condition should be controled more strictly to obtain reliable results.

Speciation of Cd, Cu and Zn in Sewage Sludge-Treated Soils Incubated under Aerobic and Anaerobic Conditions

  • The incubation study was conducted under aerobic and anaerobic conditions to study the release of the kinetically labile forms (i. e. chelating ion or anion forms) of Cd, Cu and Zn in sludge-untreated soil ("Control"), sludge 50 and $100dry\;Mg\;ha^{-1}$ treated soils ("Soil-Sludge mixtures"), and sewage sludge ("Sludge"). The chelating ion and anion exchange membranes were embedded into the samples and incubated for 16 weeks under aerobic and anaerobic condition. The total amounts of chelating ion or anionic forms of Cd were too little to be measured during both aerobic and anaerobic incubation. On the other hand, the total amounts of chelating ion or anionic forms of Cu and Zn slightly increased throughout the incubation period under both incubation conditions. For "Control" and "Soil-Sludge mixtures" treatments, the total amounts of Cu and Zn in chelating ion and anion exchange membrane were little difference between aerobic and anaerobic condition, and the total amounts of chelating ion form of Cu and Zn were not different from the those of anionic form of Cu and Zn. However, for "Sludge" treatment, the total amounts of Cu and Zn in anion and chelating ion exchange membrane were greater under aerobic condition than under anaerobic condition, and the total amounts of chelating ion form of Cu and Zn were greater than those of anion form of Cu and Zn under both incubation conditions.

Observations on Some of the Mycelial Growth and Pigmentation Characteristics of Cordyceps militaris Isolates

  • Characteristic growth patterns of Cordyceps militaris isolates on various media, under varying light conditions and at varying incubation periods were examined. Light was found to be the most critical single factor in determining the density, texture, and pigmentation of the mycelial culture of the fungus. However, under the light condition, the degree of pigmentation and mycelial density were found to be affected by the incubation period and type of medium. Irrespective of the variations in medium type or incubation period, there was no pigmentation of the mycelium under dark condition. Radial growth of the mycelium was faster under dark incubation rather than under light incubation. Abundant mycelial density and darkest pigmentation of C. militaris isolates were produced in nutritionally rich media like SDAY, SMAY and CZYA, suggesting that these media may fulfill all the requirements for vegetative growth of the fungus. Growth characteristics of C. militaris isolates could be easily observed by the simple agar culture method, which would be useful to characterize the phenotypic characteristics of large number of pure cultures of the fungus under given conditions of growth factors such as medium, light and temperature.

The Effect of Incubation Behaviour on the Daily Body Mass Regulation of Streaked Shearwaters (Calonectris leucomelas) (슴새(Calonectris leucomelas)의 포란행동이 일별 몸무게 변화에 미치는 영향)

  • During incubation, pelagic seabirds undergo large variations in their body mass as a result of concomitant fasting. In Procellariifomes undergoing important changes in body mass through the incubation, body mass regulation could be an important factor in behavioural decisions for changing activities during the incubation. The aim of this study was to investigate the effect of individuals' quality and incubation behaviour on their body mass regulation in Streaked Shearwaters Calonectris leucomelas breeding at Sasudo Island, in Jeju, South Korea during incubation period, June to August in 2013. In our results, we found that there was no relationship between the daily body mass loss and body size or condition of Streaked Shearwaters. However, the daily body mass loss increased significantly as the incubation bout increased. Moreover, the daily incubation bout was positively related to their body size. Consequently, although breeders' qualities were not related to their daily body mass loss during the incubation, the incubation behaviour affecting their daily body mass loss was regulated by their body size. Therefore, the body size of Streaked Shearwater breeders is one of important factors in their decisions for the incubation behaviour.

Anaerobic Biodegradation of PCP in Japanese Paddy Soils

  • Seven soil samples were collected from paddy fields located nearby Nagoya city in Japan. All the soils were subjected to flooded condition and incubated with PCP at $30^{\circ}C$ for two months, and their anaerobic PCP degradation have been monitored by checking the PCP concentration of the soils at regular intervals. The degradation of PCP did not occur in the soils autoclaved two times before pre-incubation. On the other hand, all the soils showed significant PCP degradation in non-sterilized condition after 30 days of incubation, except far one soil sample (Yatomi), in which PCP was rarely degraded until 30 days of incubation. This result showed PCP disappearance in the pad(rf soils was mainly caused by microbiological activity, and depended upon the physicochemical characteristics of the soils.

Effective Screening Method for Viviparous Germination of Rice

  • The viviparity of 28 rice varieties was tested at 25 days after heading(DAH), 35DAH, and 45DAH in the laboratory and field condition for 12 days. The incubation temperature was 20/l$0^{\circ}C$ (day/night), 25/15$^{\circ}$C$ and 30/20$^{\circ}$C$ in the laboratory test, and under field water conditions in the field test. The biggest varietal difference of viviparity was found in the laboratory test when examined at 45DAH with the 6-day incubation under 25/15$^{\circ}$C$ . At this conditions the mean viviparous ratio was 32.1 % with the range of 53.9 and the variance of 259.5. In the field test, the significant varietal difference in the viviparity was also found in the lodging treatment at 45 DAH for 6 days. Correlation coefficient analysis between the field and laboratory tests was highly significant from 4 days after incubation at 45 DAH and after 6-day incubation at 35 DAH, and correlation coefficient was higher as incubation days in the laboratory and submerged days under field water became longer. Considering the correlation between the field and laboratory tests, varietal difference of viviparity and convenience of testing, the laboratory test at 45 DAH for 6-day incubation under 25/15$^{\circ}$C$ was the most efficient evaluation method for the viviparity of rice cultivar.

When Chong Ju and Chung Ju soils possessing different physicochemical properties were treated with 500 ppm of TOK and incubated in flooded anaerobic condition for 2, 4, and 6 months, respectively, they produced 4-Chloro-4'-amino diphenyl ether, 2,4-Dichloro-4'-amino diphenyl ether(amin-TOK), N-[4'-(4-Chlorophenoxy)] phenyl acetamide, and N-[4'-(4-Chlorophenoxy)] phenyl formamide as the metabolities. This result indicates that TOK undergose the reduction of its $NO_2\;to\;NH_2$ group, dechlorination, acetylation, and formylation under this condition. The cleavage of ether linkage does not occur. In addition, TOK degrades more readily in Chung Ju soil which is characterized by pH 6.43 and higher contents of $Ca^{++}$ and C.E.C. than in Chong Ju soil which is lower in pH, $Ca^{++}$, and C.E.C. 2. In the aerobic incubation of TOK of 25ppm in Chung Ju soil suspension for 21 days, the ratio of the resulting metabolites, TOK : amino-TOK : 4-Chloro-4'-amino diphenyl ether was 100 : 130 : 76. Meanwhile, in the 42 day incubation, the ratio was 100 : 19 : 5, which indicates that TOK in aerobic condition dose not necessrily degrade as a function of the incubation period. 3. The citrate buffer extract of Chung Ju soil has the capability of degrading TOK, which was verified to be due to the action of the microorganisms involved. 4. Twelye strains of soil bacteria were isolated from the TOK-treated soils. In the incubation of TOK in pure cultures of the respective isolates, the strain T-1-1 isolated from Chong Ju soil had almost no degradability whereas the strain T-2-3 was the most potent. The degradation of TOK by the isolates constituted mostly the reduction of the nitro group to amino group. 5. In a test for the degradability of TOK by some selected microorganisms, Pseudomonas species were more potent than fungi. Yet, Isolate B which had been isolated from Chung Ju soil suspension was the most potent.

  • 표면상태가 $LaNi_{4.7}Al_{0.3}$의 활성화에 미치는 영향을 조사하기 위해서 여러가지 산화 상태에서의 incubation time을 관찰하였다. Incubation time은 입자의 크기, 파쇄후 경과시간, 합금방법에 따라 다르게 나타났다. 재료를 파쇄한후 실험한 날짜에 따라 incubation time은 초기에는 증가하다가 최대점을 지난후 감소하고 다시 최소점을 보인후 증가하는 현상을 보였다. 이것은 Ni cluster와 산화층의 복합적인 작용인 것으로 생각된다. Ni cluster의 양과 산화층의 두께는 공기에 노출되는 시간이 증가함에 따라 증가하였다.

  • P-type (100) Si wafer patterned with 1000$\AA$ SiO$_2$island was used as substrate and the Si films were deposited under low pressure using Si$_2$H$_{6}$-H$_2$gas mixture where the total gas flow rate and deposition pressure were 16.6sccm and 3.5mtorr, respectively. In this condition, we selectively obtained high-quality epitaxial Si layer of the 350~1050$\AA$ thickness. In order to extend the incubation period, we kept high pressure H$_2$ environment without Si$_2$H$_{6}$ gas for few minutes after first incubation period and then we conformed the existence of second incubation period.iod.

Effect of Defaunation on In Vitro Fermentation Characteristics and Methane Emission When Incubated with Forages

  • An in vitro study was conducted to determine the effects of defaunation (removal of protozoa) and forage sources (rice straw, ryegrass and tall fescue) on ruminal fermentation characteristics, methane ($CH_4$) production and degradation by rumen microbes. Sodium lauryl sulfate, as a defaunation reagent, was added into the mixed culture solution to remove ruminal protozoa at a concentration of 0.375 mg/ml. Pure cellulose (0.64 g, Sigma, C8002) and three forage sources were incubated in the bottle of culture solution of mixed rumen microbes (faunation) or defaunation for up to 24 h. The concentration of ammonia-N was high under condition of defaunation compared to that from faunation in all incubations (p<0.001). Total VFA concentration was increased at 3, 6 and 12 h (p<0.05~p<0.01) but was decreased at 24 h incubation (p<0.001) under condition of defaunation. Defaunation decreased acetate (p<0.001) and butyrate (p<0.001) proportions at 6, 12 and 24 h incubation times, but increased propionate (p<0.001) proportion at all incubation times for forages. Effective degradability of dry matter was decreased by defaunation (p<0.001). Defaunation not only decreased total gas (p<0.001) and $CO_2$ (p<0.01~0.001) production at 12 and 24 h incubations, but reduced $CH_4$ production (p<0.001) at all incubation times for all forages. The $CH_4$ production, regardless of defaunation, in order of forage sources were rice straw > tall fescue > ryegrass > cellulose (p<0.001) up to 24 h incubation.
