Isatis tinctoria var. yezoensis (Ohwi) Ohwi (Cruciferae) is one of major natural dyeing crops in the world and also have used as a medicinal plant in Korea. We evaluated seed purity in $F_1$-hybrid accessions using amplified fragment length polymorphism (AFLP) markers. One hundred sixty seeds from the male and female harvests were subsequently screened for seed purity with ten primers. The 13 accession-specific bands and many variable AFLP bands scored for accessions. Especially, E-AAC/M-CAA and E-AAG/M-CAT were presented clear hybrid bands for $F_1$ hybrids. $F_1$ hybrids maintained higher average level of genetic diversity compared with their correspondent parents. Self-inbred seeds from the female and male harvests were revealed 8.0% and 5.0%, respectively. The AFLP may lead to a better insight in to the hybrid seed purity test in I. tinctoria var. yezoensis.
Selection of stress-tolerant ginseng lines in fields is very difficult because it is almost impossible to control properly the environmental conditions of soil. On the contrary, it can be studied with ease to search for stress-tolerant ginseng lines through in vitro culture because of easy manipulation of stress conditions. This study was conducted for the selection of ginseng pure lines tolerant to salt stress. Murashige & Skoog(MS) media with 2.5 folds of KNO$_3$, NH$_4$NO$_3$, MgSO$_4$.7$H_2O$, KH$_2$PO$_4$, and CaC1$_2$.2$H_2O$ was established for the selection of ginseng pure lines tolerant to salt stress in vitro. Among 88 ginseng pure lines bred by Korea Ginseng and Tobacco Research Institute, Punggi Hwangsuk, 78093, 82886, 78135, 86024 and KG104 lines was tolerant to salt stress. For the stable production of quality Korean ginseng, genetic tolerance to salt stress is one of important factors since relatively high salt concentrations in the ginseng nursery soil environment of Korea. Ginseng inbred pure lines were tested for their tolerance to salt stress through in vitro culture technique.
This work was carried out to know the hereditary phenomenon of. the egg markings with 4 races preserved at Sericultural Experiment Station from 1962 to 1963. The results are as follows: 1. When crosses (Fig. 1) were made between females showing reticulate egg marks and males showing collected egg markings, the F$_1$ markings were all reticulate. From the reciprocal cross was made between the collected marking females and the reticulate marking males (Fig. 2), all the F$_1$ progeny showed the collected egg markings. 2. In the F$_2$ the expected phenotype for reticulate markings (recessive) was not expressed but the collected markings (dominance) were always appeared. 3. When each F$_2$ moth was inbred, the usual 3:1 ratio was obtained. 4. The F$_1$ egg markings as well as the spindle shape egg were not determined by its own genes but by the genes of its mother, because those were formed before fertilization. The results of such influences, when they can be identified, are called material effects. And such a phenomenon was called pseudomaternal inheritance by Tanaka. (1919)

Anthers of sweet pepper $F_1$ cultivar 'Special' were cultured on Dumas De Vaulx (C medium), supplemented with $0.1mgL^{-1}$ 2, 4-D and $0.1mg{\cdot}L^{-1}$ kinetin with 3% sucrose, and 0.32% phytagel. The calluses obtained were further sub-cultured on Murashige and Skoog (MS) medium without growth regulators for regeneration. Regenerated plantlets were grown in plastic pots under plastic house and characterized their cytological and morphological characters in spring, 2008. Twenty percent plantlets were identified as haploid plants after chromosome and ploidy analysis. Haploid plants contained 12 chromosomes, high stomatal density with small stomatal length as compared to diploid plants. Stomatal length in haploids was 23.3% smaller than diploids. Haploid plants were characterized as small leaf and petiole size, poor vigor, thin stem and short plant height, short internodes and small flower buds, fruit size and fruit weight as compared to diploid plants and most of the haploid fruits were seedless. SP55, SP62, SP68, SP72 and SP77 are found high yielding double haploids with high total soluble content (8.6, 8.7, 9.2, 9.1 and $9.8^{\circ}Brix$, respectively) and desirable fruit shape, and recommended them to exploit as inbred lines for heterosis breeding.

In crop breeding program, information about genetic dissimilarity on breeding resources is very important to corroborate genealogical relationships and to predict the most heterozygotic hybrid combinations and inbred breeding. This study aimed to evaluate the genetic variation in Kenyan sunflower breeding lines based on simple sequence repeat (SSR). A total of 83 alleles were detected at 32 SSR loci. The allele number per locus ranged from 2 to 7 with an average of 2.7 alleles per locus detected from the 24 sunflower accessions and the average value of polymorphic information contents (PIC) were 0.384. A cluster analysis based on the genetic similarity coefficients was conducted and the 24 sunflower breeding resources were classified into three groups. The principal coordinates (PCoA) revealed 34% and 13.38% respectively, and 47.38% of total variation. It was found that the genetic diversity within the Kenyan sunflower breeding resources was narrower than that in other sunflower germplasm resources, suggesting the importance and feasibility of introducing elite genotypes from different origins for selection of breeding lines with broader genetic base in Kenyan sunflower breeding program.

Yield and grade are the key factors that affect production value of peanut. The objective of this study was to identify QTLs for pod yield, hundred-seed weight, and total sound mature kernel (TSMK). A total of 90 recombinant inbred lines, derived from Tamrun OL07 and a breeding line Tx964117, were used as a mapping population and planted in Brownfield and Stephenville, Texas. A genetic map was developed using 1,211 SNP markers based on double digest restriction-site associated DNA sequencing (ddRAD-seq). A total of 10 QTLs were identified above the permutation threshold, three for yield, three for hundred-seed weight and four for TSMK, with LOD score values of 3.7 - 6.9 and phenotypic variance explained of 12.2% - 35.9%. Among those, there were several QTLs that were detected in more than one field experiment. The commonly detected QTLs in this study may be used as potential targets for future breeding program to incorporate yield and grade related traits through molecular breeding.

Detrimental effects of inbreeding have been studied by many researchers for a long time. However, only a few studies have shown the occurrence of inbreeding depression due to evolutionary changes as a purging process. In this study, two different populations (inbreeding and outbreeding) of Drosophila melanogaster were compared to assess inbreeding effects on artificial population bottlenecks. For inbreeding conditions, a couple of D. melanogaster (one virgin and one male) were selected from an inbred population and cultured in a vial. For outbreeding conditions, a couple of D. melanogaster were selected from different populations and cultured in a vial. There were significant differences in body lengths of adults, but not in other parameters such as the total number of adults, the rate of survival, and the rate of wing mutants. The mean body length of adults of outbreeding populations was longer than that of inbreeding populations in the first generation (G1; P = 0.004), but not in the second generation (G2; P = 0.066). Although the other three parameters (total number of adults, rate of survival, and rate of wing mutants) showed differences in their mean values between inbreeding and outbreeding populations, these differences were not statistically significant. This might be due to genetic purging. This study demonstrated one additional experimental case related to inbreeding depression in artificial bottlenecked populations. Further studies are necessary to confirm the clear interaction between inbreeding depression and genetic purging using more generations and replicates (or samples) of D. melanogaster.

Our previous study identified a resistance locus to Phytophthora sojae (isolate 2457) in an interval of 3.8-4.7 Mbp on chromosome 3 via genetic mapping using a 'Daepung'×'Daewon' recombinant inbred population. Since differential gene expression between Daepung (susceptible) and Daewon (resistant) after inoculation of P. sojae is unknown, RNA sequencing was carried out to compare transcriptomic changes between the two genotypes following inoculation with P. sojae isolate 2457. The two varieties were inoculated using hypocotyl inoculation at the VC stage and stem tissue of 1 cm above and below of the inoculated site were sampled at 0, 6, 12 hours after inoculation (hai), respectively. Differentially expressed genes (DEGs) under same cultivar in different time point and Daepung vs. Daewon in same time point were investigated. In comparison of Daepung vs. Daewon at 12 hai, a total of 3,513 DEGs were identified, including two nucleotide-binding site-leucine rich repeat (NBS-LRR) genes (Glyma.03g034800 and Glyma.03g034900) that are located in the previously reported resistance locus on chromosome 3. In addition, 14,966 DEGs were detected between 0 vs. 6 hai, containing one of candidate genes (Glyma.03g035300). This gene was upregulated by up to 4-fold in Daewon and Daepung. Additional results will be further discussed in the presentation. This study will provide valuable information for soybean crop improvement.

This present study was to identify a novel candidate gene that contribute to the elevated α-linolenic acid (ALA, ω-3) concentration in PE2166 from mutagenesis of Pungsannamul. Major loci qALA5_1 and qALA5_2 were detected on chromosome 5 of soybean through quantitative trait loci mapping analyses of recombinant inbred lines. With next generation sequencing of parental lines and Pungsannamul, and recombinant analyses, a potential gene, Glyma. 05g221500 (HD) controlling elevated ALA concentration was identified. HD is a homeodomain-like transcriptional regulator that may regulate the expression level of microsomal ω-3 fatty acid desaturase (FAD3) genes responsible for the conversion of linoleic acid into ALA in the fatty acid biosynthetic pathway. In addition, we hypothesized that combination of mutant alleles, HD and either of microsomal delta-12 fatty acid desaturase 2-1 (FAD2-1\ could reduce the ω-6/ω-3 ratio. In populations where HD, and FAD2-1A and FAD2-1B genes were segregated, combination of a hd allele from PE2166 and either of the variant FAD2-1 alleles were sufficient to reduce the ω-6/ω-3 ratio in seeds.

(-)-Epicatechin (EC), a primary form of flavan-3ol and a building block of proanthocyanidins, has health benefits as it is a potent antioxidant. So far, no quantitative trait loci (QTLs) associated with EC have yet been identified in soybean. In this study, QTLs for EC and hilum color were identified in recombinant inbred lines (RILs) derived from the varieties Jinpung and IT109098 using high-resolution single nucleotide polymorphism linkage mapping. This revealed two major QTLs for EC content, qEC06 and qEC08. qEC06 spanned the T Locus encoding flavonoid 3'-hydroxylase. qEC08, located near the I locus on Chr08, was also a major QTL for hilum color; however, allelic stacking of qEC08 and I revealed no relationship between I and EC content. RILs with IT 109098 alleles at both qEC06 and qEC08 had higher EC content than other lines. These results will enable the production of soybean varieties with high EC content via marker-assisted selection.
