새로운 in vitro SPF측정법 개발 (Development of a novel in vitro SPF evaluation method)

  • 본 연구에서는 SPF 측정법에 있어서 널리 사용되어온 희석법과 박막법의 문제점에 대해 검토하였으며, 이들의 문제점을 극복하기 위하여 인체를 대상으로한 in vivo평가법과 측정 및 평가 등의 실험조건에 있어서 유사성을 갖는 in vitro평가법을 개발하였다. 이 새로운 in vitro 평가결과는 in vivo 평가결과와 0.93의 높은 상관계수를 나타내었다. 한편 이 in vitro 측정법의 유용성을 더욱 확인하기 위하여 TransporeR tape에 시료를 도포한 후 SPF-290 분석기를 이용한 측정결과와 비교한 결과 SPF20 이하의 시료일 경우 0.95 이상의 높은 상관계수를 얻을 수 있었다. 또한 광량을 일정하게 유지 시키고 광세기를 변화 시켜 홍반의 정도를 조사한 결과, 광세기와 홍반의 정도에는 관련이 없음을 알 수 있었다.

생쥐난자의 시험관내 수정과 발달 (In Vitro Fertilization and Development of Mouse Eggs)

  • These experiments were carried out to obtain the information about the optimal pH osmolality affecting in vitro fertilization of the mouse eggs, to elucidate the 2-cell block to development in vitro and to find out the method of controlling the subsequent embryo development in vitro. pH and osomlality was adjusted by adding NaCl or NaHCO3 to the basic salt solution. In vitro fertilization were carried out by inroducting the cumulus masses to the suspension of epididymal spermatozoa at each pH, osmolality, and 10${\mu}$M-EDTA medium. The results obtained in these experiments were summarized as follows: 1. The fertilization rates in vitro at each medium of 235, 252, 269, 286, 306, 323, 345, 368, 393 mosmol were 15.6, 38.2, 65.7, 75.6, 80.9, 74.3, 58.1, 35.1, 24.3, 11.1%, respectively. 2. The fertilization rates in vitro at each medium of pH 6.1, 6.4, 6.7, 7.0, 7.3, 7.6, 7.9, 8.1 were 11.8, 17.9, 32.4, 61.9, 79.5, 76.7, 53.5, 13.6%, respectively. 3. In case of ICR female x ICR male embryos, the development rate of 2-cell embryos to 4-8 cell embryos was 16.2% at normal medium, but the rate was increased to 49.3% in medium containging 10 ${\mu}$M-DETA; In case of C3H female x ICR male embryos, the development rate was 41.0% at normal medium, but the rate was increased to 71.7% at 10 ${\mu}$M-EDTA-medium.

기내미세접목(器內微細接木)에 의(依)한 대추나무의 마이코플라즈마 무병주(無病株) 생산(生産) (Production of Mycoplasma-Free Jujube Trees through In vitro Micrografting)

  • 이병묘로부터 마이코플라즈마 무병주의 생산법을 확립하기 위하여 대추나무 시험관묘의 액아에서 생장점을 채취하여 기내접목을 실시하였다. 대목으로는 기내에서 발아시킨 대추나무 실생묘의 배축을 사용하였다. 접수중 10%(30점 중 3점씩 2회)가 정상적으로 생장하였고 계속적인 증식과 발근이 가능하여 유식물체로 분화되었다. 마이코플라즈마의 존재여부를 전자현미경과 형광현미경 검경방법을 이용하여 검경한 결과, 접수로 사용한 시험관묘에서는 마이코플라즈마가 관찰되었으나 접목묘에서는 관찰되지 않았다. 이로써 마이코플라즈마를 보균한 시험관묘의 생장점을 접수로 한 기내 미세접목을 통하여 건전묘 생산의 가능성을 보여주었다.

The effect of insemination methods on in vitro maturation outcomes

  • Objective: The aim of this study was to compare the effects of conventional insemination (in vitro fertilization [IVF]) and intracytoplasmic sperm injection (ICSI) on the fertilization, developmental competence, implantation potential, and clinical pregnancy rate of embryos derived from in vitro matured oocytes of patients with polycystic ovary syndrome (PCOS). Methods: A prospective study was carried out among 38 PCOS patients who had undergone In vitro maturation (IVM) treatment. In total, 828 immature oocytes were collected from 42 cycles and randomly assigned for insemination by IVF (416 oocytes) or ICSI (412 oocytes). After fertilization, the embryos were cultured until the blastocyst stage and single embryos were transferred after endometrial preparation and under ultrasound guidance. Results: No significant differences were found in the maturation rate (78.1% vs. 72.6% for IVF and ICSI insemination, respectively; p= 0.076), fertilization rate (59.4% vs. 66.9% for IVF and ICSI insemination, respectively; p= 0.063), or the formation of good-quality blastocysts (40.9% vs. 46.5% for IVF and ICSI insemination, respectively; p= 0.314). Implantation and clinical pregnancy also did not show significant differences. Conclusion: There was a comparable yield of in vitro matured oocytes derived from PCOS patients in terms of fertilization, blastocyst formation, implantation rate, and clinical pregnancy between IVF and ICSI insemination. These findings provide valuable insights for choosing assisted reproductive treatment in women with PCOS, as IVM offers promising outcomes and is less invasive and less costly.

우 난포란의 체외성숙에 관한 연구 III. 체외성숙 우난포란의 체외수정과 발달 (Studies on In Vitro Maturation of Bovine Follicular Oocytes III. In Vitro Fertilization and Development of In Vitro Matured Bovine Follicular Oocytes)

  • These experiments were carried out to obtain the basic information for in vitro fertilization and development of bovine follicular oocytes matured in vitro. The bovine ovaries were obtained at a slaughter house and the follicular oocytes surrounded by cumulus cells were collected by puncturing follicles with 2-6mm of diameter. Bovine oocytes were matured in vitro for 24-26 hours in a CO2 incubator with 5% CO2 in air at 39$^{\circ}C$. The medium used for maturation was TCM199 supplemented with hormones, pyruvate, FBS and antibiotics. Epididymal spermatozoa were capacitated by in vitro culture for 2-3 hours in BO solution containing bovine serum albumin(5mg/ml) and caffein(2.5mM). Insemination was made by introducing about 10-15 matured oocytes into the suspension of capacitated spermatozoa. Six hour after inseminatin the eggs were transferred to TCM 199 supplemented with FBS(10%) for in vitro development. The results obtained in these experiments were summarized as follows : 1. The maturation rate of oocytes following incubation for 24-26 hours was 78.4%(228/291). 2. Of total 250 oocytes, 172 embryos extruded 2nd polar body following in vitro culture with spermatozoa for 20 hours, and the rates of embryos developed to 2-, 4-, 8-, 16-cells and morula or early blastocyst were 64.0, 39.2, 22.0, 15.2 and 11.2%, respectively. 3. The time needed for development to 2-, 4-. 8-, 16-cell stage and morula was 42.5$\pm$5.4, 58.0$\pm$9.2, 74.4$\pm$11.5, 96.1$\pm$13.4 and 119.0$\pm$18.2 hours, respectively.

In-vitro 전분 소화 모델에서 백미와 현미 가루의 물성학적 특성 분석 (Rheological Behaviors of White and Brown Rice Flours During In-vitro Simulation of Starch Digestion)

  • The in-vitro starch digestibility of white and brown rice flours was continuously characterized from a rheological point of view. Specifically, the in-vitro viscosities of the rice digesta samples were monitored under simulated oral, gastric, and intestinal conditions. A trend of decreasing viscosities in all the digesta samples was observed during the in-vitro digestion. After cooking, the brown rice sample exhibited lower viscosity than that of the white rice flour due to the presence of more non-starch components. A similar tendency was observed during the simulated oral and gastric digestions. However, the viscosity crossover between the white and brown rice samples was observed during intestinal digestion. In addition, the amount of glucose released from the brown rice flour was significantly lower than that from the white rice flour. Thus, the slower rate of starch hydrolysis in the brown rice flour could be related to its in-vitro rheological behaviors.

The Suppression of Maturational Competence by Streptomycin during In vitro Maturation of Goat Follicular Oocytes

  • Antibiotics are common additives in culture media during in vitro embryo development, but their effects on oocyte maturation in vitro have not been tested. The effects of penicillin, streptomycin and gentamicin on the maturational competence and subsequent development potential of goat follicular oocytes were examined after parthenogenetic activation in vitro. Maturation rates at 24 h after in vitro maturation, and parthenogenetic development at 48 h after activation, were evaluated by observing the protruding first polar body and the 4 cell stage cleavage, respectively. When streptomycin was present in the maturation medium, the percentages of matured oocytes 24 h after activation were significantly (p<0.01) lower than those from the other groups (42.5-45.7% vs. 69.1-73.8%). Penicillin and gentamicin treatment did not affect the maturation rates or the percentages reaching the 4 cell stage 48 h after activation. There was no significant difference in cleavage rates among the different antibiotic treatments 48 h after activation. Therefore, streptomycin suppresses the in vitro maturation of immature goat oocytes, but does not influence their subsequent development.

체외성숙, 수정 및 배양된 한우 체외수정란의 유우이식에 의한 산자 생산 (Production of Korean Native Calf by In Vitro Maturation, Fertilization, Cultivation and Transfer of Embryos into Holstein Cows)

  • The objective of this study was to produce Korean native calves following transfer of in vitro matured, fertilized and cultured embryos into Holstein cows. The ovaries of Korean native cows or heifers were obtained from an abattoir and kept on 25 to 28$^{\circ}C$ and transported to laboratory within 2 hrs. The oocytes were matured in vitro (IVM) for 24 hrs in TCM-199 supplemented with 35$\mu\textrm{g}$/ml FSH, 10$\mu\textrm{g}$/ml LH, 1$\mu\textrm{g}$/ml estradiol-17$\beta$ and granulosa cells at 39$^{\circ}C$ under 5% CO2 in air. They were fertilized in vitro (IVF) by epididymal spermatozoa treated with heparin for 24 hrs., and then the zygotes were co-cultured in vitro (IVC) with bovine oviductal epithelial cells for 7 to 9 days. Late morulae and blastocysts produced in vitro were nonsurgically transferred to recipient cows by unilaterial. Recipients were monitored for estrus and for pegnancy by rectal plapation in 60 days after embryo transfer. One of them was pregnant to term and produced a female weighing 42.5kg at birth.

인공숙주에서 증식된 Tetrastichus sp.의 생물학적 특성 (Biological Characteristics of Tetrastichus sp. reared on Artificial Host)

  • Tetrastichus sp.는 미국흰불나방의 번데기 기생벌로서, 1개의 숙주에 여러마리의 자손을 번식하는 집단 기생벌이며, 일년에 여러 세대 발생한다. 작잠, 신생아용 분유, 난황, 로얄젤리 등을 주요성분으로 기생벌의 인공먹이를 조성하였으며, polypropylene필름으로 번데기형태를 만들어 인공숙주로 사용하였다. 인공숙주에서 발육한 기생벌은 자연숙주에서 자란 기생벌보다는 크기가 작았으나, 계대 사육으로 번식 가능한 자손을 생산하였다. 인공숙주에서 발육한 in vitro 2세대의 생물학적 특성은 in vitro 1세대보다 훨씬 개선 었다. 평균 자손 수, 산란기간, 수명 등은 in vitro 에서 45.7,7.8, 13.8일 이었고 암컷 편중의 성비 76.9%를 나타내었다. 이러한 결과는 Tetrastichus sp.가 in vitro 대량사육에 적합한 기생벌임을 보여준다.

체외성숙 소 난포란의 동결성에 관한 연구 (Study on Freezability of In Vitro-Matured Bovine Follicular Oocytes)

  • This study was investigated to test in vitro-maturation rate of bovine follicular oocytes freezability of in vitro-matured bovine follicular oocytes with different stock solution in Glycerol and Propanediol, freezability of in vitro-rnatured bovine follicular oocytes on cryoprotectants, the viability of in vitro-rnatured bovine follicular oocytes by morphologically normal and FDA staining method. 1. The maturation rates of bovine follicular oocytes classified as grade A, B and C was 88, 63 and 21%, respectively. 2. Freezability of in vitro-matured bovine follicular oocytes on stock solution, TCM-199+5% FCS and m-PBS + 5% FCS was 61%(n=105), 48%(n=62) in $_1$M Glycerol and freeability of in vitro-matured bovine follicular oocytes on stock solution, TCM-199 +5% FCS and m-PBS + 5% FCS was 68%(n=112), 42%(n=57) in 1~2 Propanediol. The results indicate that freezability of in vitro-matured bovine follicular oocytes with different stock solution is important. 3. Freezability of in vitro-matured bovine follicular oocytes on cryoprotectants was Glycerol and PROH was 56%(n=167), 57%(n=169). The results indicate that PROH was superior to Glycerol. 4. The rates of morphologically normal IVM oocytes after thawing of cryopreserved oocytes with Glycerol and PROH were 39%(n=$_1$8), 65%(n=39), respectively. The results indicate that PROH was superior to Glycerol. 5. The fluorescent light intensity after thawing of cryopreserved oocytes classified with Positive, Partial-I, Partial-II, Negative with Glycerol and PROH. The results of FDA-positive 24%, 42%, Partial-I 17%, 10%, Partial- H 20%, 12%, FDA-negative 39%, 37%, and Partial-I, II, respectively.
