산약 추출물의 항산화 및 항염증에 관한 실험적 연구 (Experimental Study on the Antioxidant and Antimicrobial Properties of Dioscoreae Rhizoma)

  • Objectives : This study evaluated the effect of Dioscoreae Rhizoma on antioxidant and antimicrobial activity in vitro. Methods : In vitro experiments were made on antioxidant by DPPH radical scavenging activity, enzyme activity of SOD, protective effect about $H_2O_2$ and DNA fragmentation in RAW264.7 cells, as well as antimicrobial by Cox-2 expression in LPS-induced RAW264.7 cells. Results : 1. Dioscoreae Rhizoma's antioxidant ability was improved by DPPH radical scavenging activity. 2. Antioxidant ability of Dioscoreae Rhizoma produced good results at more than 0.025 mg/$m{\ell}$. 3. On protective effect of Dioscoreae Rhizoma about $H_2O_2$ on RAW264.7 cells, with addition of Dioscoreae Rhizoma extract the cells' survival increased compared to adding $H_2O_2$ only. 4. About revelation-control of Cox-2 at RAW264.7 cells which induce by LPS, when Dioscoreae Rhizoma's concentration was higher from 0.0125 mg/$m{\ell}$ to 1 mg/$m{\ell}$, its control activity was stronger. Thus we found Dioscoreae Rhizoma has antimicrobial activity. 5. The power of DNA fragmentation control of Dioscoreae Rhizoma was time-independent. Conclusions : From this study we conclude that Dioscoreae Rhizoma has effects on antioxidant and antimicrobial activity. However, considerable work still needs to be done studying antioxidants by concentration, as well as a variety of experiments about antimicrobial activity.

Antioxidant activities of Erythrina stricta Roxb.using various in vitro and ex vivo models

  • Erythrina stricta, a deciduous tree widely used traditionally in indigenous system of medicine for various ailments such as rheumatism, fever, leprosy, epilepsy etc. The leaves of Erythrina stricta was extracted with ethanol (70%) and used for the evaluation of various in vitro antioxidant assays which includes H - donor activity, nitric oxide scavenging, superoxide anion scavenging, reducing ability, hydroxyl radical, hydrogen peroxide scavenging, total phenolic content, total flavonoid content, total antioxidant activity by thiocyanate and phosphomolybdenum method, metal chelating, $\beta$-carotene bleaching, total peroxy radical assays. The pro-oxidant activity was measured using bleomycin-dependent DNA damage. Ex vivo models like lipid peroxidation and erythrocyte haemolysis were also used to study the antioxidant property of the extract. The various antioxidant activities were compared with suitable standard antioxidants such as ascorbic acid, butylated hydroxyl toluene, $\alpha$-tocopherol, curcumin, quercetin and Trolox. The generation of free radicals viz. $O_2^{{\cdot}-}$, $OH^{\cdot}$, $H_2O_2$, $NO^{\cdot}$ and peroxyl radicals were effectively scavenged by the ethanolic extract of Erythrina stricta. In all the methods, the extract offered strong antioxidant activity in a concentration dependent manner. The total phenolic content, flavonoid content and total antioxidant activity in Erythrina stricta were determined as microgram (g) pyrocatechol, quercetin and $\alpha$-tocopherol equivalent/mg respectively. The extract did not exhibit any prooxidant activity when compared with ascorbic acid. The results obtained in the present study clearly indicates that Erythrina stricta scavenges free radicals and reduces lipid peroxidation, ameliorating the damage imposed by oxidative stress in different disease conditions and serve as a potential source of natural antioxidant.

  • The aim of this study is to investigate in vitro biological properties of Eucommia ulmoides Oliv. bark. Ethyl acetate(EtOAc) fraction from aqueous extract of Eucommia bark showed strong antioxidant activity of $IC_{50}$ 19.2 ${\mu}g/ml$ by 1,1-diphenyl-2-picrylhydrazyl(DPPH) free radical-scavenging assay. The Eucommia bark extract showed $\alpha$-glucosidase inhibitory activity and inhibited growth of human liver cancer cell, suggesting its potential biological value of anticancer.

Antioxidant, Anti-inflammatory and Cytotoxicity on Human Lung Epithelial A549 Cells of Jerusalem artichoke (Helianthus tuberosus L.) Tuber

  • This study investigated in vitro antioxidant, anti-inflammatory and cytotoxicity on human lung epithelial A549 cells of different solvent extracts from Jerusalem artichoke (Helianthus tuberosus L.) tuber. The EtOH extract contained amounts of phenolics (22.20 tannic acid equivalent ㎎/ɡ) and exhibited the highest antioxidant activity and anti-inflammatory activity. Several methods were employed for measure the antioxidant activity: 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity (IC50 = 206.79 ㎍/㎖), reducing power activity (21.26 ascorbic acid equivalent ㎎/ɡ) and total antioxidant activity (19.05 ascorbic acid equivalent ㎎/ɡ). Meantime, the EtOH extract inhibited the NO production completely with a concentration of 800 ㎍/㎖. Besides, the H2O extract exhibited more potent effect on human lung epithelial A549 cells. This study suggested that Jerusalem artichoke tuber had antioxidant, anti-inflammatory and cytotoxicity on human lung epithelial A549 cells.

표고버섯의 항산화능과 알코올분해능에 미치는 영향 (Antioxidant Activities and the Effect of Reducing Serum Alcohol Concentration of Lentinus edodes)

  • Objectives : The purpose of this study is to evaluate antioxidant activities and reducing serum alcohol concentration of extract of Lentinus edodes on the alcohol administered rats. Methods : Antioxidant effect was measured by total phenolic compound and DPPH-radical scavenging activity of extract of Lentinus edodes in vitro. Blood alcohol concentration, aldehyde concentration, malondialdehyde concentration, glutathion concentration were measured in vivo. Results : The extract of Lentinus edodes increased DPPH-radical scavenging activity dose-dependently. The water extract with boiling water showed lower antioxidant activity and phenolic content than 70% ethanol extract in vitro. Blood alcohol concentration was significantly reduced by pre-treatment of ethanol extract of Lentinus edodes. The effect was more significant than commercial product used as a positive control. Conclusions : This study suggest that Lentinus edodes can be a potential nature resource for the management of ethanol toxicity although the mechanism of action involved in the treatment remains to be explored.

In Vitro Antioxidant Activity Profiles of ${\beta}$-Glucans Isolated from Yeast Saccharomyces cerevisiae and Mutant Saccharomyces cerevisiae IS2

  • To explore the possible usefulness of ${\beta}$-glucans as natural antioxidants, the antioxidant profiles of ${\beta}$-glucan, extracted from Saccharomyces cerevisiae KCTC 7911, and water soluble and insoluble mutant ${\beta}$-glucan, isolated from yeast mutant S. cerevisiae IS2, were examined by five different in vitro evaluation methods: lipid peroxidation value (POV), nitric oxide (NO), 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, reducing power, and ${\beta}$-carotene diffusion assay. The antioxidant activities of all ${\beta}$-glucans evaluated in POV test were comparable to or better than that of the known antioxidant, vitamin C. Remarkably, the ${\beta}$-glucan and water insoluble mutant ${\beta}$-glucan possessed 2.5-fold more potent activity than vitamin C at a dosage of 2 mg. Although vitamin C showed 100-fold greater activity than all ${\beta}$-glucans in NO and DPPH tests for measuring the radical scavenging capacity, all ${\beta}$-glucans revealed higher radical scavenging activity than the known radical scavenger, N-acetyl-L-cysteine (NAC), in DPPH test. The water insoluble mutant ${\beta}$-glucan had 2.6- and 5-fold greater antioxidative activity than water soluble ${\beta}$-glucan in NO and DPPH tests, respectively, showing that all ${\beta}$-glucans were able to scavenge radicals such as NO or DPPH. While all ${\beta}$-glucans revealed lower antioxidant profiles than vitamin C in both reducing power activity and ${\beta}$-carotene agar diffusion assay, the ${\beta}$-glucan and water insoluble mutant ${\beta}$-glucan did show a marginal reducing power activity as well as a considerable ${\beta}$-carotene agar diffusion activity. These results confirmed the potential usefulness of these ${\beta}$-glucans as natural antioxidants.

홍삼복합제의 보간 및 항산화 작용에 대한 연구 (Study on Hepatoprotective and Antioxidant Activities of Korean Red Ginseng-Mixed Formula)

  • GRF (Korean Red Ginseng mixed formula) consists of six herbs such as Ginseng Radix rubra Koreana, Lycii Fructus, Artemisiae Capillaris Herba, Poria, and Glycyrrhizae Radix and Hoveniae Fructus. For the evaluation of hepatoprotective effect of GRF, the study was performed on protective effect against hepatic damage induced by galactosamine in vitro and ccl4 in vivo and also elucidate antioxidant activity. In vitro assay with 1.1 mM galactosamine, protection (%) was 44% (GR), and 58% (GRF-A) at 50 ug/ml. GRF effectively protected fatty degenertion and necrosis in murine hepatic damage induced by ccl4. For the -antioxidant study, GRF inhibited hemolysis of erythrocyte and decolored DPPH (2,2-diphenyl-l-picrylhydrazyl) free radical in a dose dependent manner more effetively than GR alone in vitro. GRF and GR significantly suppressed the time course $(1\;hr{\sim}6\;hr)-level$ of MDA (malondialdehyde) following AAPH (2,2'-azo-bis-(2-amidino -propane) dihydrochloride) treatment in vivo as compared with control data. From the results it can be concluded GR and GRF exerted the hepatoprotective effect by dint of antioxidant activity.

In vitro 상에서 유자(Citrus junos SIEB ex TANAKA) 용매 추출물의 암세포 억제효과 및 항산화성 (In Vitro Effect of Yuza (Citrus junos SIEB ex TANAKA) Extracts on Proliferation of Human Prostate Cancer Cells and Antioxidant Activity)

  • 유자는 성숙할수록 과피와 과육의 총페놀 함량은 많아지고 자유기 소거능이 높아지는 경향을 보였다. 유자로부터 추출한 18가지 용매 추출물의 항산화 활성과 전립선 암세포 DU 145, LN-CaP에 대한 세포독성 실험을 실시한 결과 과피와 과육 모두 미성숙한 유자 보다 성숙한 후 수확한 유자가 그 활성이 높게 나타냈다. 용매별로 살펴보면 과성숙 유자 과피, 과육의 클로로포름, 메탄올 추출물은 전립선 암세포 DU 145에 대하여 60% 이상의 세포사멸 효과를 나타났으며 메탄올 추출물의 효과도 농도에 따라 상당히 높은 것으로 나타났다. 또한 세포 조직을 현미경으로 살펴본 결과 추출물의 처리 농도가 증가할수록 세포 사멸이 쉽게 이루어지는 것을 확인 할 수 있었다. 이런 세포 사멸의 경향은 추출물의 자유기 소거 환성이 높을수록 높게 나타났다.

Antioxidant Activity of Vitex rotundifolia Seeds and Phytochemical Analysis Using HPLC-PDA

  • This study assessed in vitro antioxidant activity (ABTS+ and DPPH) of Vitex rotundifolia seeds collected from two different regions in Korea (Jungjang City and Sindu City). Three extraction methods using ethanol, methanol, and water were prepared separately and subjected to quantification by reverse-phase high-performance liquid chromatography-photodiode array (HPLC-PDA) analysis as well as antioxidant testing. Among them, the water-based extract exhibited superior activity in the ABTS+ compared with the ethanol- and methanol-based extracts, while the DPPH assay analysis, revealed that the methanol-based extract had very low antioxidant activity. The concentrations of vanillic acid (1), luteolin (2), vitexicarpin (3), and artemetin (4) were quantified using HPLC-PDA analysis. Vanillic acid (1) was identified as the main antioxidant in V. rotundifolia seeds. Combining the antioxidant activity and quantitative analysis results, the water-based extract was considered to have the highest antioxidant activity. Furthermore, vanillic acid (1) was detected in the leaves and stems of V. rotundifolia plants from different regions, indicating that this species has the potential for use in future antioxidant-applications.

Antioxidant and Free Radical Scavenging Potential of Justicia gendarussa Burm. Leaves in vitro.

  • Antioxidant activity of 70% aqueous ethanolic extract of leaves of Justicia gendarussa (EJ) was evaluated. EJ was prepared by cold maceration method. The antioxidant potency of EJ was investigated employing various established in vitro systems, such as DPPH radical scavenging, nitric oxide (NO) scavenging, ${\beta}-carotene$ linoleic acid module system (${\beta}$ CLAMS), hydroxyl (OH) radical scavenging, anti lipid peroxidation. $IC_{50}$ values were determined in each experiment. Also, ferric ion reduction capacity of extracts in presence and absence of chelating agent (EDTA) and total antioxidant capacity were determined. Preliminary phytochemical investigation was carried out to know the nature of constituents present in the leaves and correlate it with antioxidant activity. Further total phenolic content was determined in EJ. $IC_{50}$ values of EJ were 123.09 ${\pm}$ 3.01, 643.0 ${\pm}$ 61.10, 132.3 ${\pm}$ 6.03, 68.5 ${\pm}$ 11.5 and 68.13 ${\pm}$ 1.38 ${\mu}g/mL$ in DPPH radical scavenging, NO scavenging, ${\beta}$ CLAMS, OH radical scavenging and anti lipid peroxidation activity respectively. In total antioxidant capacity assay, ascorbic acid equivalent value was found to be 205.56 ${\pm}$ 4.69 ${\mu}g/mg$ of extract. Total phenolic content was found to be 43.76 ${\pm}$ 4.27 ${\mu}g$ equivalent of gallic acid per mg of extract. Phytochemical investigation reveals the presence of flavonoids. The results indicate that EJ possess antioxidant activity and flavonoids are responsible for this activity.
