The purpose of this study was to analyze whether FSH and LH hormone treatment directly or indirectly affect embryo development in embryonic development. To determine this, we compared the development of embryonic cells through the expression pattern of MMPs. As a result, 33.8% of blastocysts were formed in FSH added group, 20.8% in LH added group and 10% in FSH + LH added group. In addition, the activity of MMP-9 was highly detected in the FSH-added group, and the expression of Casp-3 was much lower than that of the other groups. These results suggest that the addition of FSH seems to increase the activity of MMP-9 in embryonic cells, and that LH, on the contrary, may activate MMP-2 activity. In addition, the expression level of MMP-2 in the FSH-added group was high in the Trophoblast cell group and in the LH-added group, the hormone ideal secretion might affect the development of the embryonic cell.
The equine chorionic gonadotropin (eCG) subunits $\alpha$ and ${\beta}$ are transcribed from different genes and associate noncovalently to form the bioactive eCG heterodimer. Dimerization is rate limiting for eCG secretion, and dissociation leads to hormone inactivation. The correct conformation of the heterodimer is alto important for efficient secretion, hormone-specific post-translational modifications, receptor binding and signal transduction. To determine whether ${\alpha}$ and ${\beta}$ subunits can be synthesized as a single polypeptide chain (tethered-eCG) and also display biological activity, the tethered-eCG molecule by fusing the carboxyl terminus of the eCG ${\beta}$-subunit to the amino terminus of the af-subunit was construe-ted and transfected into chinese hamster ovary (CHO-Kl) cells. LH- and FSH-like activities were assayed in terms of testosterone production and aromatase activity in primary cultured rat Leydig cells and granulosa cells, respectively. The tethered-eCG was efficiently secreted and showed similar LH-like activity to the dimeric eCG ${\alpha}$/${\beta}$ and native eCG. FSH-like activity of the tethered-eCG was also shown similarly in comparison with the native and wild type eCG ${\alpha}$/${\beta}$. Our data for the first time suggest that the tethered-eCG can be expressed efficiently and the produced product by the CHO-K1 cells is fully LH- and FSH-like activities in rat in vitro bioassay system. Our results also suggest that this molecular can imply particular models of FSH-like activity not LH-like activity in the eCG. Taken together, these data indicate that the constructs of tethered molecule will be useful in the study of mutants that affect subunit association and/or secretion.
eCG(말 융모성성선자극 호르몬)$\beta$와 eLH(말 황체형성호르몬)$\beta$는 하나의 유전자로 코드 되어 있으며, eCG와 eLH는 당쇄의 구조에 있어서 차이가 있는데, LH는 sulfate가 CG는 sialylate가 수식되어 있다. eCG는 다른 동물에 있어서 강력한 FSH (난포자극)와 LH의 이중활성을 나타내어 아주 특이하고, 많은 탄수화물로 수식되어 있는 당단백질 호르몬이다. eCG의 이러한 이중활성은 성선 자극호르몬의 구조, 기능 및 수용체와 이들 호르몬과의 특이결합에 대하여 분자 생물학적인 관점에서 연구하는데 아주 흥미롭다 따라서, eCG는 당쇄의 구조와 생화학적인 기능에서 아주 특이한 분자이다. 이러한 중요점을 당쇄첨가부위의 돌연변이를 통하여 분석한 결과, LH의 활성에서는 eCG$\alpha$의 56번 당쇄가 필수불가결한 역할을 하지만, eCG$\beta$의 카르복실기 말단의 O-linked 당쇄는 중요하지 않은 것으로 관찰되었다. 한편, N- 및 O-linked 당쇄 모두는 FSH활성에는 중요한 기능을 가지고 있는데, 양쪽 당쇄의 제거는 오히려 FSH 활성을 증가시켰다. 따라서, eCG의 LH와 FSH의 이중활성은 $\alpha$의 N-linked 당쇄의 제거와, $\beta$의 O-linked 당쇄를 제거함으로써 완전히 분리할 수 있으며, eCG에 있어서 당쇄는 생화학적 활성에 대하여 아주 중요하게 작용한다는 새로운 사실이 밝혀졌다. 단일체인 eCG($\beta$의 C-terminal에 u를 연결한)도 eCG$\alpha$/$\beta$ 및 천연형 eCG와 비교한 결과 효율적으로 분비되어지고 완전한 LH와 FSH 활성을 나타내었다. 이러한 결과들은 eCG 분자에 있어서 지금까지 문제시되어왔던 LH활성을 나타내지 않고, 높은 FSH 활성만을 나타내는 특이한 모델을 만들 수가 있으며, 현재 단일체인 분자에 있어서 당쇄의 기능에 대한구축은 각 단체의 결합, 분비에 영향을 미치는 당쇄 돌연변이 연구에 아주 유용할 것으로 사료된다.

The glycoprotein hormone family represents a class of heterodimers, that includes the placental hormone equine chorionic gonadotropin (eCG) and the anterior pituitary hormones- follitropin (FSH), lutropin (LH), and thyrotropin (TSH). The 4 hormones are heterodimers, with a common $\alpha$-subunit and unique $\beta$-subunits. eCG is the most heavily glycosylated of the known pituitary and placental glycoprotein hormones. Recent observations using single chain glycoprotein hormone analogs in which, the $\beta$-and $\alpha$-subunits are linked, implied that heterodimeric-like quaternary configuration is not a prerequisite for receptor/signal transduction. To study the function and signal transduction of tethered rec-eCG, a single chain eCG molecule was constructed and rec-eCG protein was produced. Molecular mass of the single chain is about 45 kDa. All mice were ovulated by tethered rec-eCG treatment. The dual activity of tethered rec-eCG was determined in receptor cell lines of nonequid species; in fact, this dual activity was proven in species other than horse. Tethered rec-eCG in equids does not bind to FSH receptors, suggesting that eCG is primarily an LH-like hormone in the horse. Taken together, these data suggest that tethered rec-eCG has dual activity in nonequid species in vitro. However, it has only LH-like activity in equid species in vitro.
Here, we evaluated the mode of programmed cell death during porcine oocyte maturation by comparing the two major pathways associated with programmed cell death, apoptosis (type I), and autophagy (type II). We investigated the expression and localization of major genes involved in autophagy and apoptosis at mRNA and protein levels. Furthermore, the effect of hormonal stimulation on autophagy and apoptosis was analyzed. We found that the activity of autophagy-associated genes was increased in the cumulus-oocyte complexes (COCs) following follicle-stimulating hormone (FSH) treatment, while the addition of luteinizing hormone (LH) reversed this effect. The expression of proteins associated with autophagy was the highest in FSH-treated COCs. On the other hand, caspase-3 protein level was maximum in COCs cultured with LH. The treatment with rapamycin resulted in the effect similar to that observed with FSH treatment and increased autophagy activity. Thus, hormonal stimulation of pig oocytes resulted in distinct patterns of maturation. The high-quality oocytes majorly relied on the type II pathway (autophagy), while the type I pathway (apoptosis) was more prominent among poor-quality oocytes. Further investigation of this distinction may allow the development of techniques to produce high-quality oocytes in porcine in vitro fertilization.
Equine chorionic gonadotropin (eCG), which consists of highly glycosylated α- and β-subunits, is a unique member of the gonadotropin family because it elicits response characteristics of both follicle-stimulating hormone (FSH) and luteinizing hormone (LH) in other species than the horse. This study investigated the effects of recombinant eel gonadotropin hormone (rJeGTH) produced in silkworms, with and without a carboxyl-terminal peptide from equine chorionic gonadotropin (eCG), on the induction of sexual maturation in female eels Anguilla japonica. Experiments were conducted both in vivo and in vitro. In in vitro trials, germinal vesicle breakdown (GVBD) induction did not significantly differ between rJeFSH and $rJeFSH{\cdot}eCG$ treatments and the control group. However, previous studies did find that rJeLH and $rJeLH{\cdot}eCG$ treatments induced GVBD in female eels. Our in vitro exploration of $estradiol-17{\beta}$ ($E_2$) levels in immature ovarian tissues revealed significantly higher $E_2$ levels in the group treated with $rJeFSH{\cdot}eCG$$1{\mu}g/mL$ than in the control group. In contrast, the in vivo experiments showed no effect of recombinant hormones on the sexual maturation of feminized eels. Previous studies and our own in vitro results have clearly shown that rJeGTH and $rJeGTH{\cdot}eCG$ have a positive effect on the sexual maturation of feminized eels. To develop the activity of rJeGTH in vivo, further studies should confirm circulation time and activity of these hormones in eels' bloodstream, modify the structure of the recombinant gene, and implement additional glycosylation.
eCG는 다른 포유동물에서 FSH와 LH의 활성을 나타내기 때문에 성선자극 호르몬 family에서 아주 특이적이고 많은 당쇄가 수식되어진 알파와 베타의 비공유결합으로 구성되어 있다. 유전자 재조합 $eCG{\beta}/{\alpha}$의 생물학적 기능을 규명하기 위하여 말의 LH/CGR의 포유동물발현용 벡터를 구축하였다. 재조합 $eCG{\beta}/{\alpha}$의 활성분석은 말의 LH/CGR가 일시적으로 발현되는 CHO-K1 세포와 지속적으로 발현되는 PathHunter Parental 세포를 이용하여 분석하였다. 유전자 재조합 $eCG{\beta}/{\alpha}$는 CHO-K1 부유세포의 상층으로 효율적으로 분비되었으며, 분비량은 transfection 후 1일에서 7일까지 약 200 mIU/ml이었다. Western blot 분석결과는 재조합 $eCG{\beta}/{\alpha}$의 분자량은 약 40-45 kDa으로 검출되었다. eLH/CGR가 발현되는 CHO-K1 세포에서의 cAMP분비량으로 재조합 $eCG{\beta}/{\alpha}$의 활성을 분석하였다. 그 결과 cAMP농도는 재조합 $eCG{\beta}/{\alpha}$의 농도의존적으로 증가하였다. eLH/CGR가 일시적으로 발현하는 CHO-K1 세포에서 $EC_{50}$ 값은 $8.1{\pm}6.5ng$이었다. 또한 일시적 및 지속적으로 eLH/CGR가 발현하는 PathHunter Parental 세포에서도 재조합 $eCG{\beta}/{\alpha}$의 LH 활성 분석결과 높은 활성을 나타내는 것으로 확인되었으며, 이들의 $EC_{50}$ 값은 각각 $5.0{\pm}4.7ng/ml$, $4.5{\pm}5.2ng/ml$으로 나타났다. 따라서 이러한 결과에 의하면 재조합 $eCG{\beta}/{\alpha}$는 말의 LH/CGR가 발현하는 세포에서 생물학적 활성을 나타난다는 것을 확인하였으며, PathHunter Parental 세포에서 지속적으로 발현되는 세포의 확보는 당쇄제거에 의한 재조합 eCG의 돌연변이등에 관한 기능적인 메커니즘을 밝히는데 유용할 것으로 사료된다.
Oyster (Crassostrea gigas) is a marine bivalve mollusk widely distributed in coastal areas, and have been long widely used in industrial resources. Several studies demonstrated that fermented oyster (FO) extract attribute to bone health, but whether administration of FO play as an endocrine disruptor has not been studied. Therefore, in the present study, we investigated the effect of FO on the endocrine system in vitro and in vivo. As the results of the competitive estrogen receptor (ER) and androgen receptor (AR) binding affinities, FO was not combined with ER-α, ER-β, and AR. However, 17β-estradiol and testosterone, used as positive control, were interacted with ER and AR, respectively. Meanwhile, oral administration of 100 mg/kg and 200 mg/kg of FO doesn't have any harmful effect on the body weight, androgen-dependent sex accessory organs, estrogen-dependent-sex accessory organs, kidney, and liver in immature rats. In addition, FO supplementation has no effect on the serum levels of luteinizing hormone (LH), follicle stimulating hormone (FSH), testosterone, and 17β-estradiol. However, the relative weight of androgen- and estrogen-dependent organs were significantly increased by subcutaneously injection of 4.0 mg/kg of testosterone propionate (TP) and by orally administration of 1.0 ㎍ of 17α-ethynyl estradiol (EE) in immature male and female rats, respectively. Furthermore, TP and EE administration markedly decreased the serum LH and FSH levels, which are similar those of mature Sprague-Dawley (SD) rat. Furthermore, the testosterone and 17β-estradiol levels were significantly enhanced in TP and EE-treated immature rats. Taken together, our findings showed that FO does not interact with ER and AR, suggesting consequentially FO does not play as a ligand for ER and AR. Furthermore, oral administration of FO did not act as an endocrine disruptor including androgenic activity, estrogenic activity, and abnormal levels of sex hormone, indicating FO may ensure the safety on endocrine system to develop dietary supplement for bone health.
배아줄기세포(embryonic stem cell, ESC)는 미분화상태로 지속적인 계대가 가능하며, 정상 핵형과 전 분화능(pluripotency)을 가져 생체내-외에서 분화 유도시 삼배엽성의 모든 세포로 분화 가능하다. ESC를 feeder 세포 없이 부유배양하면 배아체(embryoid body, EB)를 형성하고, 초기 배아 발생과 유사한 분화 양상을 갖는다. ESC의 분화 유도가 초기배아 발생처럼 생식호르몬(GTH: FSH, LH; steroids)의 영향을 받는지는 불명하다. 본 연구는 ESC가 분화과정중 생식호르몬처리에 의해 그들 수용체가 발현되는가를 알아보고자 하였다. 순계혈통 생쥐인 C57BL/6J에서 과배란 유도후 포배를 수획하고, 유사분열적으로 불활성화된 feeder 세포와 공배양하여, 계대배양 하는 중 배아줄기세포주(JHYl)를 확립하였다. JHY1의 alkaline phosphatase 활성과 SSEA-1, 3, 4 발현을 통해 ESC임을 확인하였다. Feeder 세포 없이 ESC를 계대배양 후 호르몬처리(FSH LH E$_2$, P$_4$, T)하에서 5일 동안 부유배양하여 배아체를 형성시키고, 이후 7일 동안 부착배양하여 분화를 유도하였다. GTH와 스테로이드의 수용체 발현 실험에서 ESC에 E$_2$ 처리에 의한 LHR의 발현 증가를 제외한 나머지 호르몬 처리군에서 ESC보다 낮은 생식호르몬의 수용체 발현이 관찰되었다. 생식호르몬을 농도별 수용체 발현 정도는 증감되지 않았다. 미분화 ESC 표지유전자인 Oct-4는 호르몬 처리군에서도 발현되었다. 각 배엽의 표지유전자들(영양세포, handl; 외배엽성, keratin와fgf-5; 중배엽성, enolase와 $\alpha$ -globin; 내배엽성, gata-4와 $\alpha$ -fetoprotein) 등의 발현 양상을 조사한 결과 호르몬 처리후 내배엽성 표지유전자외에는 발현 증가가 관찰되지 않았다. 즉 생식호르몬에 의해 gata-4, $\alpha$-fetoprotein의 발현이 증가되는 것으로 보아 내배엽성 계열로의 분화 유도가 이루어진 것으로 사료된다.
