Perspectives of AIDS Vaccine Development: T Cell-based Vaccine

  • Estimated number of adults and children newly infected with HIV-1 during 2001 alone is 5 million in total. An effective vaccine, in addition to education & public health approaches, has been believed to be the best option to stop the HIV-1 transmission, especially for developing countries. Among AIDS vaccine candidates, DNA vaccine is relatively safe and, in a certain extent, mimics some attributes of live attenuated vaccine, with regard to in vivo gene expression & the type of immunity induced. We recently demonstrated that DNA vaccines expressing SIVmac239 structural and regulatory genes, augmented with coadministration of IL-12 mutant induced the strongest T cell responses, resulting in low to undetectable setpoint viral loads, stable $CD4^+$ T cell counts, and no evidence of clinical diseases or mortality by day 420 after challenge. This finding is the second demonstration, following the protective result of live attenuated SIV vaccine in SIVmac-rhesus monkey model, which was known to have safety problem. So, our DNA vaccines could give a significant impact on HIV-1 epidemic by slowing or stopping the spread of HIV-1, leading to eventual eradication of HIV-1 and AIDS in the population.

  • Fiber reinforcement can significantly improve the properties of concrete. Particulary, toughness or energy-absorbing ability of fiber reinforced concrete is frequently higher than that of unreinforced concrete. Toughness is a measure of energy absorption capacity and used to characterized fiber reinforced concrete's ability to resist fracture when subjected to static, dynamic and impact loads. However, the current standard methods of characterizing the toughness of fiber reinforced concrete have proven to be some inadequate and problems and have caused a great deal of dissent and confusion. This study research some of the inadequate and problems with these toughness measurement methods and proposes the evaluation method for Fiber Reinforced Concrete toughness.

  • A SMART anchor is a kind of friction mount anchor, the load is diffused and applied to the various parts of the distributed bond length, having less impact on the grout strength, and being able to secure necessary anchoring force in relatively soft grounds. Smart anchor can have strong loads in soft and weak grounds as in rock beds.

  • This paper examined the probabilistic load analysis for the tailplane during pitching maneuvering in the conceptual aircraft design phase. The flight load analysis based on the probabilistic distribution of design variables are compared with the results of the deterministic analysis. Two forms of variable distribution are used in this paper. One is standard normal distribution, the other distribution is calculated from the results of short-period longitudinal equation of aircraft motion. The influence of the distribution parameter on the probabilistic load analysis was investigated and the significant design variables that have an impact on the mean and variance of probabilistic load were identified. The comparison indicates that probabilistic load analysis provides more reliable probabilistic load distribution for the structural design than the traditional deterministic analysis.

  • Dynamic load models which show the practical behavior of truss bridge subjected to moving train load are presented. Three basically approaches are available for evaluating structural response to dynamic effects : moving force, moving mass, and influence moving force and mass. Simple warren truss bridge model is selected in this research, and idealized lumped mass system, modelled as a planar structure. In the process of dynamic analysis, the uncoupled equation of motion is derived from simultaneous equation of the motion of truss bridge and moving train load. The solution of the uncoupled equations of motion is solved by Newmark-$\beta$ method. The results show that dynamic response of moving mass and static analysis considering the impact factor specified in the present railway bridge code was nearly the same. Generally, the dynamic response of moving force is somewhat greater than that of moving mass. The dynamic load models which are presented by this study are obtained relatively adequate load model when apply to a truss bridge.

  • A huge offshore structures such as immersed tunnel, ice-resisting wall are continuously subjected to large force from water pressure, wave action and impact loads. Composite steel-concrete structure of sandwich system has profitable advantages for a huge offshore structures. This composite structures should exhibit a high degree of strength and ductility, because of concrete confining effect and the property of steel plate. Therefore, it endures large deformation and absorbs a great deal of energy until failure. In this study, nonlinear analysis for composite steel-concrete structure of sandwich system was carried out, and certify the effects of various parameters, elastic·plastic behavior characteristic, load-carrying and failure mechanism.

Identification of multiple sources in a plate structure using pre-filtering process for reduction of interference wave

  • This paper presents novel research into the source localization of multiple impacts. Source localization technology for single impact loads in a plate structure has been used for health monitoring. Most of research on source localization has been focused only on the localization of single impacts. Overlapping of dispersive waves induced by multiple impacts and reflection of those waves from the edge of the plate make it difficult to localize the sources of multiple impacts using traditional source localization technology. The method solving the overlapping problem and the reflection problem is presented in the paper. The suggested method is based on pre-signal processing technology using band pass filter and optimal filter. Results from numerical simulation and from experimentation are presented, and these verify the capability of the proposed method.

  • Resonance of railroad bridge can be broken out when natural frequency of the bridge coincides with exciting frequency of moving forces. In order to avoid aforementioned unpleasant response of the structure, exact determination of dynamic structural properties is important to understand dynamic behavior of the structure under moving train loads. In the present paper, a 25 meters long full scale IPC girder and 15m Precom girder models were fabricated as a test specimen and modal testing was carried out to evaluate modal parameters including natural frequencies and modal damping ratios. In the modal testing, a digitally controlled vibration exciter as well as an impact hammer is applied to obtain frequency response functions more exactly and the modal parameters are evaluated varying with structural status.

  • In recent railroad markets, the use of steel sleepers is gradually increased due to various advantages in resistance for impact as well as economical efficiency for production, construction. maintenance and recycle. The typical steel sleepers which are successfully used in railroad markets are n-type of Corus Inc. in England and Y-type of ThyssenKrupp Gft Gleistechnik in Germany. Both types have merits and demerits in safety and economical efficiency. In 1990, n-type steel sleeper was developed in Korea, but was failed in putting into practical use, due to the subsidence into the ballast by Jive loads and welding crack, etc.. In this paper, in order to develop optimal shape of n-type steel sleepers for domestic rail mads, parametric studies for cross section, thickness, are performed.

  • Delaying and objection for the construction of storage spent-fuel disposal has prompted to consider expanding on-site storage of spent reactor fuel since it can eliminate the need for costly and difficult shipping and control of the spent fuel completely under the direction of the owner-utility. The dry storage unit developed in Canada can accommodate Korea heavy water reactor fuel elements and become a candidate for the Korean market. In this paper, finite element analyses were carried out in order to investigate the structural behavior of the nuclear spent fuel dry storage system, which is subjected to impact loads such as collision of a truck load and dropping of flask under the irregular operation.
