Botrytis cinerea 균주들이 생산하는 Polygalacturonase, Laccase, ${\beta}$-glucosidase의 균주 간 화성 및 병원성과의 상관관계 (Relationships Between Pathogenicty and Activities of Polygalacturonase, Laccase, and ${\beta}$-Glucosidase Produced by Botrytis cinerea)

  • 기주, 분리 연도 및 장소가 다른 Botryis cinerea 20개 균주를 이용하여 오이세포벽을 탄소원으로 첨가한 배지에서 배양한 뒤 병원성과 관련이 있는 것으로 알려진 polygalacturomase, Iaccase, $\beta$-glucosidase의 활성을 측정하고 병원성과의 상관관계를 조사하였다. 조사된 효소들의 활성과 병원성과의 상관관계는 상관계수가 모두 0.5 이하로 직접적인 연관성을 인정할 수 없었다. 그러나 Bovendamm 배지에서의 균사생장 정도의 병원성은 r=0.522(p=0.05)로 정의 상관관계를 보였다. 또한 병원성이 강한 3균주와 약한 3균주를 선발하여 각 기주의 세포벽을 첨가하여 배양 후 poly-galacturonase 활성정도를 비교하였으나 뚜렷한 경향을 찾을 수 없었다. Exo-PG 항체를 이용한 면역혈청 blotting 결과 병원성이 다른 거의 모든 균주가 66kDa 크기의 1 band를 보이는 유사한 효소 pattern을 보였다. 이상의 결과로 볼 때 측정된 이들 효소는 병원성과 관련된 일차적 요인이 아닐 것으로 생각한다.

임파구 CD38의 효소학적 연구 (Enzymatic study on lymphocyte CD38)

  • Murine CD38 is a 42 kDa type II glycoprotein expressed on cell surface of both B and T lymphocytes. CD38 is a multifunctional enzyme that catalyzes the formation and hydrolysis of cyclic adenosine diphosphoribose (cADPR): ADP-ribosyl cyclase activity of CD38 catalyzes the formation of cADPR from NAD and cADPR hydrolase activity of CD38 catalyzes the hydrolysis of cADPR to ADP-ribose (ADPR). And also, CD38 has the catalytic activity of NAD glycohydrolase (NADase) which catalyzes the hydrolysis of catalyzes the formation and hydrolysis of cyclic adenosine diphosphoribose (cADPR): ADP-ribosyl cyclase activity of CD38 catalyzes the formation of cADPR from NAD to ADPR. In this study, we attempted to purify CD38 from mouse lymphocytes by using the immobilized anti-CD38 monoclonal antibody. The single step immuno-affinity column chromatography resulted in homogeneous purification, showing a single protein of 42 kDa on a SDS polyacrylamide gel. We have investigated the effects of various inhibitors on the enzyme activities of the purified CD38. Cibacron blue (0.5 mM) inhibited all three enzyme activities of CD38, NADase, ADP-ribosyl cyclase and cADPR hydrolase activities. ADPR (2 mM) showed inhibitory effect on both cADPR hydrolase activity and NADase, but not on ADP-ribosyl cyclase activity. However, ATP (2 mM) inhibited only cADPR hydrolase activity. $Zn^{2+}$ (1 mM) showed similar inhibitory effect as that of ADPR, but activated cyclase activity These results suggest that CD38 has three different catalytic activity domains which might be differentially regulated by their specific inhibitors.

고로쇠와 우산고로쇠 나무의 부위별 항암 및 면역조절능 비교 (Comparison of Anticancer and Immuno-Modulatory Activities in the different parts of the Acer mono Max. and Acer okamotoanum)

  • 인간 정상 폐세포인 HEL299을 이용한 세포 독성을 살펴본 결과, 각 추출물은 1.0 $mg/m{\ell}$의 농도에서 최고 19.5%의 세포독성을 나타내었다. 또한 항암활성 효과를 살펴보기 위하여 2가지의 암세포를 이용하였는데, AGS, A549, MCF-7, Hep3B에서 수피 추출물의 경우 1.0 $mg/m{\ell}$의 농도에서 55% 이상의 높은 억제활성을 나타내었고, 또한 암세포의 생육활성에 대한 정상 세포의 세포독성 비로 나타낸 선택적 사멸도는 고농도에서는 모두 3이상으로 나타났다. B cell과 T cell을 이용한 면역세포의 생육 촉진 실험에서는, BT cell 은 배양 5일째 수피 추출물이 각각 $7.5{\times}10^4cells/m{\ell}$, $8.2{\times}10^4cells/m{\ell}$로 가장 높은 세포농도를 나타냈다. 면역세포를 이용한 cytokine의 분비량 측정실험에서는 수피 추출물이 배양 시간에 따른 cytokine의 분비가 가장 높게 나타나는 것을 확인할 수 있었다. 각 세포의 $TNF-{\alpha}$의 분비량은 6일째 최고의 분비량을 나타내었고, 수피 추출물의 경우 대조군에 비해 7-8 배가량 정도 더 높은 분비량을 나타낸 것을 확인하였다. IL-6의 경우 6일째 최고의 분비량을 나타내었고, 수피 추출물에서 가장 높은 분비량을 나타내었다. 또한 우산고로쇠 수피 추출물을 첨가한 배지에 의한 NK cell의 면역활성을 측정하였으며, 모든 추출물에서 배양 시간에 따라 유의적으로 증가하는 것을 확인하였다. 그 중 우산고로쇠 수피 추출물에서 B cell의 경우 $18.0{\times}10^4cells/m{\ell}$, T cell의 경우 $17.2{\times}10^4cells/m{\ell}$으로 가장 높은 생육 활성을 나타내었다. 본 실험 결과를 통하여 고로쇠와 우산고로쇠의 기능성 소재로서의 가능성을 확인하였으며, 유용성분 분리와 생리활성에 대한 연구가 보다 다각적으로 가져야 할 것으로 생각된다.

한외거르기(Ultrafiltration)에 의하여 분리된 백수오 고분자 분획물의 면역증진 활성 (Immuno-stimulatory Activities of a High Molecular Weight Fraction from Cynanchum wilfordii Radix Obtained by Ultrafiltration)

  • 백수오의 국내 재배활성화와 함께 농가소득을 높이기 위한 기능성 식품소재로서의 우수성을 산업적으로 응용하고자 백수오 추출물의 면역증진 활성을 평가하였다. 낮은 온도에서 액체상태의 추출물을 효과적으로 농축하고 정제하기 위하여 비열처리 공정인 한외거르기를 통해 분자량(MWCO)이 30 kDa인 여과막을 이용하여 고분자 분획물(CWUF>30) 및 저분자 분획물(CWUF<30)을 얻었으며 이를 큰포식세포 활성화를 통한 면역 증진 효과를 알아보기 위해 RAW 264.7 세포를 이용하여 면역활성의 지표가 되는 산화질소(II) 생성량 및 사이토카인인 TNF-${\alpha}$, 인터루킨-6의 생성량을 측정한 결과, 정상대조군(CON)에 비해 CWUF>30을 처리하였을 때 농도 의존적으로 유의하게 증가하였다. 세포실험을 통해 면역증강활성이 높은 CWUF>30은 정상동물모델에서 면역기관인 비장 및 흉선의 무게를 증가시켰고 비장세포 유래 림프구 증식도 유의하게 증가시켰다. 이는 CWUF>30의 투여가 비장세포를 증식시키는 mitogen 활성이 있음을 보여주며 외부의 항원에 노출 시 항원에 대한 면역반응을 유도하는 면역세포의 수를 증가시켜 항원에 대한 효과적인 방어에 도움을 줄 것으로 보여진다. 또한 CWUF>30을 투여한 마우스 비장세포(splenocyte)의 Yac-1 세포 살해 정도를 측정한 결과, 200 mg/kg BW의 농도로 CWUF>30을 투여한 마우스의 비장세포는 정상대조군(CON)에 비하여 effector 세포와 target 세포의 비율이 10:1에서 유의적으로 증가하였으며 이는 양성대조군(PC)과 유사한 높은 활성을 보이는 것을 확인 할 수 있었다. 이와 같은 결과를 통해 백수오 고분자 분획물(CWUF>30)이 강력한 면역활성 증진효능을 갖고 있으며 각종 바이러스 등 외부 항원들에 대응하여 초기 면역세포를 자극하고 면역매개물질을 생성함으로써 인체의 비특이적 면역반응을 증가시키는데 중요한 역할을 할 수 있을 것으로 기대된다.

콩으로부터 추출한 Pinitol의 면역억제 활성 비교 (Comparison of Immuno-Suppressive Activities of Pinitol Isolated from Soybean)

  • The experiment was conducted to validate anti-inflammatory effects of pinitol from bean. It was evaluated for some molecule targets by wound healing assay and RT-PCR. The results of wound healing assay was shown dose-dependent inhibition of cell migration in cancer cells and inhibited RNA expression of ICAM-1, CD 44, MMP-17, MMP-14 and ARF2. Immune suppression activity in a mouse provoked by DNFB observed that inflammatory reaction with pinitol were reduced ear swelling and inflammatory cells infiltration in mouse atopic models. The result confirmed that pinitol have the effect of dose-dependent immune suppression activity.

Distribution and Antimicrobial Susceptibility of Clostridium Species in Soil Contaminated with Domestic Livestock Feces of Korea

  • Soil samples from five different areas in Korea were collected during 2001/02 and examined for presence of the genus Clostridium. Direct immuno-fluorescent assay (IFA) examination showed that Clostridium septicum, Cl. novyi and Cl. chauvoei were detected in the soil of specific areas in Korea. Sixteen species of Clostridium were isolated and cultivated from the soil samples. Cl. peifringens was detected in all sampling locations, while the other species were not. The in vitro activity of 14 antibiotic agents was determined against 421 clostridia isolated from the soil contaminated with animal feces in Korea. Trovafloxacin was effective against all isolates of the genus Clostridium except one isolate of Cl. subterminale, two of Cl. tetani, and three of Cl novyi with $MIC_{50}$ $8- 16\mu$g $ml^{-1}$. Thirteen species of Clostridium were resistant to vancomycin except for Cl. perfringens, Cl. sporogenes, and Cl. subterminale. Imipenem and trovafloxacin showed high antimicrobial activities (>95%) against all strains in the clostridia investigated. Therefore, antibiotic agents such as imipenem and trovafloxacin are the most suitable agents for polymicrobial infection as broad-spectrum monotherapy.

Immunomodulating and Anticoagulant Activity of Glycosaminoglycans Derived from Porcine Testis

  • Glycosaminoglycans (GAGs) were isolated from the porcine testis, and their immuno-modulating and anticoagulant activity was investigated. From anion exchange chromatography (Dowex Macropolous Resin) used for further isolation of porcine testis GAGs (PT-GAGs), two fractions (PT-GAG-1.5 and PT-GAG-16) eluted by different salt concentration were obtained. In immunomodulating activity test, PT-GAG-1.5, but not PT-GAG-16, significantly enhanced the growth of murine peritoneal macrophages. In addition, treatment with PT-GAG-1.5 induced the production of cytokines, interleukin-1$\beta$ (IL-1$\beta$), interferon-${\gamma}$ (IFN-${\gamma}$) and tumor necrosis factor-$\alpha$ (TNF-$\alpha$), from murine microphages. Unexpectedly, both of PT-GAGs had no effect on the growth of murine splenocytes. The anticoagulant activity of PT-GAG-1.5 and PT-GAG-16 was examined by activated partial thromboplastin time (aPTT) assay and thrombin time (TT) assay. Both of PT-kGAGs significantly increased the clotting times of aPTT and TT in a dose-dependent manner. The anticoagulant activity of PT-GAG-16 was found to be higher than that of PT-GAG-1.5. These results suggest that PT-GAGs possess biological activities such as immunomodulating activity and anticoagulant activity.

Four-Week Repeated Oral Toxicity Study of AIP1, a Water-soluble Carbohydrate Fraction from Artemisia iwayomogi in Mice

  • Artemisia iwayomogi, a member of the Compositae, is a perennial herb easily found in Korea and used as a traditional medicine to treat liver disease. AIP1, a water-soluble carbohydrate fraction from Artemisia iwayomogi, showed anti-tumor and immuno-modulating activities in animal studies. A subacute toxicological evaluation of AIP1 was performed for 4 weeks in ICR mice. After administration of AIP1 (0, 20, 100, 500 mg/kg/day), the clinical signs, mortalities, body weight changes, hematology, blood clinical biochemistry, urinalysis, organ histopathology, organ weights and gross finding were examined. The results showed that there were no significant differences in body weight changes, food intakes, water consumptions, or organ weights among different dose groups. Also we observed no death and abnormal clinical signs during the experimental period. Between the groups orally treated with AIP1 and the control group, there was no statistical significance in hematological test or serum biochemical values. Histopathological examination showed no abnormal changes in AIP1 groups. These results suggest that no observed adverse effect level (NOAEL) of the oral administration of AIP1 for 4 weeks was considered to be more than 500 mg/kg/day in mice under the condition investigated in current study.

Modulation of Immunosuppression by Oligonucleotide-Based Molecules and Small Molecules Targeting Myeloid-Derived Suppressor Cells

  • Myeloid-derived suppressor cells (MDSCs) are immature myeloid cells that exert suppressive function on the immune response. MDSCs expand in tumor-bearing hosts or in the tumor microenvironment and suppress T cell responses via various mechanisms, whereas a reduction in their activities has been observed in autoimmune diseases or infections. It has been reported that the symptoms of various diseases, including malignant tumors, can be alleviated by targeting MDSCs. Moreover, MDSCs can contribute to patient resistance to therapy using immune checkpoint inhibitors. In line with these therapeutic approaches, diverse oligonucleotide-based molecules and small molecules have been evaluated for their therapeutic efficacy in several disease models via the modulation of MDSC activity. In the current review, MDSC-targeting oligonucleotides and small molecules are briefly summarized, and we highlight the immunomodulatory effects on MDSCs in a variety of disease models and the application of MDSC-targeting molecules for immuno-oncologic therapy.

Purification and Characterization of Complement-activating Acidic Polysaccharides from the Fruits of Capsicum annuum

  • Hot water-soluble crude polysaccharide (HCAP-0) that was obtained from the fruits of Capsicum annuum showed potent anti-complementary activity. The activity was unchanged by pronase digestion, but decreased by periodate oxidation. The HCAP-0 was fractionated by DEAE ion-exchange chromatography to give two major fractions, HCAP-II and III. These two fractions were finally purified by gel filtration to give HCAP-IIa, HCAP-IIIa1, and IIIa2 fractions that had high anti-complementary activities. The HCAP-IIIa1 and IIIa2 consisted of homogeneous polysaccharides. The anti-complementary activities were unaffected by treatment with polymyxin B, indicating that the modes of complement activation were not due to preexisting lipopolysaccharide. The molecular weight and sugar content of HCAP-IIIa2 had potent anti-complementary activity. The highest yields were 55 kDa and 75.9%, and the molar ratio of galactose (Ara:Gal, 1.0:4.6) was higher than other sugars. The crossed immuno-electrophoresis showed that both classical and alternative pathways were activated by HCAP-IIIa2.
