한국산 땅콩에 감염되어 있는 바이러스를 동정하기 위하여 땅콩 재배지 에서 모자이크, 괴저를 동반한 얼룩무늬, 황화, 줄무늬, 엽맥녹대, 위축 등의 바이러스 감염 증상을 나타내는 땅콩잎 및 땅콩을 채집하였다. 이들 시료로부터 기주범위, 면역전자현미경(ISEM), 감염 세포내의 바이러스의 존재양식, direct immune staining assay(DISA), RT-PCR 등에 의하여 Bean common mosaic virus(BCMV-PSt)와 Peanut mottle virus(PeMoV)를 분리하였다. 이들 시료를 DN법에 의거하여 전자현미경으로 관찰한 바 길이가 약 780 nm의 사상형 입자와 세포질 봉입체를 관찰하였다. 초박절편의 시료 관찰에서도 길이 약 700 nm의 사상 입자가 엽육세포 등의 세포질과 액포에 산재 혹은 병행 배열로 존재해 있는 영상 및 세포질 봉입체가 반드시 확인되었다. 또한, 이들 시료를 BCMV-PSt와 PeMoV의 항혈청으로 ISEM을 실시한 결과, 두 항체에 모두 decoration 되었음을 확인하였다. 두 바이러스가 종자전염이 되는 지를 확인하기 위하여 땅콩을 직접 BCMV-PSt와 PeMoV의 항혈청으로 DISA를 실시하였다. 그 결과, BCMV-PSt 및 PeMoV 항혈청에 발색이 되었으며, 이들을 발아시켜 유식물체를 획득하였다. 이들 유식물체에서도 바이러스 감염 증상인 얼룩무늬 증상이 관찰되었고, 이들을 BCMV-PSt와 PeMoV의 항혈청으로 ISEM방법으로 검경한 결과, 두 항체에 모두 decoration 되었다. 또한, 자연감염 식물체 및 DISA에 의해 바이러스 감염이 확인된 종자를 발아시킨 2년생 식물체로부터 생물검정 법 및 ISEM에 의해 두 종의 바이러스가 검출되었다. Rl-PCR을 실시한 결과, 약 1.2Kb크기의 BCMV-PSt coat protein유전자가 증폭 되었다. 이 연구결과, 한국산 땅콩에 BCMV-PSt가 가장 많이 감염되어 있음을 확인하였다.
Cronobacter spp.는 Salmonella spp.와 함께 조제분유에서 발견되는 미생물 중 위험도가 가장 높은 category A에 속하는 균으로 알려져 있다. 1958년 영국에서 처음으로 유아의 뇌수 막염의 원인균으로 보고되었으며, 감염 후 신생아에게서 괴사성 장염, 패혈증 등을 일으켜 후유증으로 시력과 청력 상실 및 신경마비를 일으키기도 한다. 이러한 위험성 때문에 Cronobacter spp.의 신속 검출 및 진단은 식중독 예방에 있어서 중요하다. 따라서 2002년 미국 식품의약품안전국에서는 분유에서의 Cronobacter spp.를 EE broth, VRBG 배지를 이용해 균을 분리 하고, TSA 배지에 순수분리 후 노란색을 발하는 colony를 생화학적 실험을 통해 검출하는 방법을 제시했다. 또한, 우리나라 식품공전에서도 배지를 이용하여 Cronobacter spp.를 검출 하는 방법에 대해 등재하였다. 하지만 배지배양법을 기반으로 하는 Cronobacter spp.의 검출방법은 검출에 소요되는 시간과 노동력 등이 비효율적이라는 단점이 있다. 따라서 많은 시간이 소요되는 배지배양법을 보완하고자 PCR, real-time PCR 방법 및 최근에는 PCR기법과 ELISA, CE-LIF 등을 결합하여 검출하는 방법이 개발되어 Cronobacter spp.의 검출에 사용되기도 하였다. Cronobacter spp.의 검출에 사용되고 있는 분자생물학적 기반의 PCR 및 real-time PCR 기법은 민감도와 특이도가 좋으며, 배지배양법에 의한 검출방법에 비해 검출 시간을 획기적으로 줄일 수 있다는 장점이 있다. 하지만 까다로운 실험과정과 조작에 있어서 필요한 전문적인 기술이 필요함은 한계점으로 사료된다. 면역학적 방법에 의한 Cronobacter spp. 검출에 관한 연구를 통해, 분석에 소요되는 상당시간을 단축할 수 있었으며, 민감하고 특이적인 검출이 가능함을 보여주었다. 특히 일부 연구보고에서 면역학적 검출방법은 고가의 장비가 필요 없으며, 간단한 지침에 따라 모니터링 업무의 수행이 가능하다고 하였다. 면역학적 검출방법에는 ELISA 방법 외에도 immunomagnetic bead, liposome, immunochromatographic strip 등이 개발되고 있다. 우리나라 식품의약품안전처에서는 영 유아 대상 식품의 안전관리를 강화하고자 Cronobacter spp.에 대해서는 '불검출'로 기준을 설정 운영하고 있으며, 지속적으로 이에 관한 규정을 강화하고 있는 실정이다. 따라서, 식품산업체 및 식품 의 제조, 가공, 유통 현장에서 쉽게 모니터링이 가능하며, 신속, 민감하고 특이적인 검출방법의 개발은 지속되어야 한다.
목적 : 본 연구는 염증유발 내독소인 LPS 자극을 통한 면역 스트레스에 대한 약침의 치료효과와 기전을 알아보기 위해 설계되었다. LPS를 통한 염증반응은 다양한 기전을 통해 이루어지는 데 특히, 산화질소(NO)를 생성하는 효소인 iNOS의 활성은 시상하부에서 NO의 생성을 촉진함으로써 염증반응을 발현시키는 중요한 역할을 하는데 약침이 LPS로 인해 활성화된 iNOS의 발현에 어떠한 영향을 미치는지 알아보고자 하였다. 실험방법 : 흰쥐에 LPS를 투여하고 2시간 경과한 후 약침 (소염방, 대한약침학회)을 경혈 (합곡, 족삼리)에 좌,우 각각 0.2cc 씩 피하 투여하고 RT-PCR 방법을 통하여 면역활성에 중요한 역할을 하는 시상하부에서의 iNOS의 mRNA 발현을 알아 보았다. 실험결과 : LPS 투여로 인하여 증가하였던 iNOS mRNA가 약침치료를 통해 현저하게 감소하였다. 그러나 비경혈점 (미추부위 임의혈)에 약침을 투여한 군 경혈점에 생리식염수를 투여한 군에서는 iNOS mRNA 가 감소하지 않았다. 이는 약침이 LPS를 투여한 흰쥐의 시상하부에서 iNOS의 mRNA 발현에 조절작용을 가지고 있는 것을 보여주는 결과로 약침이 면역활성에 중요한 역할을 하는 NO의 생성을 억제함으로써 면역 스트레스에 치료효과를 나타내는 것으로 보여진다. 결론 : 본 연구는 약침치료가 면역반응의 중추기관인 시상하부에서 면역반응의 중요한 조절인자인 NO의 생성을 조절하는 기전을 통해 그 치료효과를 나타내는 것을 밝혔고 더 나아가 약침에 대한 현대과학적 연구에 중요한 발판을 마련했다는 점에서 그 의의를 가지고 있다고 사료된다.
Hox 유전자는 호메오도메인을 포함한 전사인자로써, 발생 과정 중 전후축을 따라 몸의 형태 형성을 조절하는 역할을 한다. 레티노산(RA)은 발생 과정에서 필수적인 형태형성인자이며 세포의 특성을 결정하는데 중요한 조절자이다. 특히, RA는 생쥐나 인간으로부터 만들어진 배아암종(EC)세포에서 Hox 유전자의 발현을 조절한다고 밝혀져 있다. 또한 RA에 의한 세포 분화와 유전자 조절 과정에 히스톤 변이가 중요한 역할을 하는 것으로 보고되어 있다. 히스톤 변이가 RA에 의해 유도되는 Hox 유전자의 발현에 특이적인 역할을 할 것으로 유추되기 때문에, 이 연구의 목적은 F9 생쥐배아 기형암종세포에서 RA에 의해 유도되는 Hoxc 유전자의 순차적인 발현이 히스톤 변이에 의해 일어나는 것인지를 조사하는 것이다. Hox 유전자의 발현 양상과 히스톤 변이는 semi-quantitative RT-PCR, RNA-sequencing과 chromatin immuno-precipitation (ChIP)-PCR 기법을 이용하여 관찰하였다. RA 처리 후(0일(D0), 1일(D1), 3일(D3)), Hoxc4 유전자의 발현(D1)은 Hoxc5부터 –c10 유전자(D3)보다 먼저 시작되었다. Hox가 발현하지 않는 D0 샘플은 전사 억제 마커인 H3K27me3이 모든 Hoxc 좌위에 강하게 표지 되어 있었으나 D1과 D3 샘플에서는 모든 좌위의 H3K27me3 표지가 확연히 줄어들어 있었다. 전사 발현 마커인 H3K4me3가 Hoxc 유전자의 순차적인 발현과 더 연관성이 있는 것으로 보이는데 D1에서 Hoxc4 발현과 함께 H3K4me3이 표지 되어 있었고, D3에서는 Hoxc 유전자 발현과 함께 모든 좌위에서 H3K4me3 마커가 존재했기 때문이다. 모든 결과를 종합해 보았을 때 F9 세포에서 RA에 의해 유도된 Hoxc 유전자의 순차적인 발현은 Hoxc 좌위에서 H3K27me3가 사라지고, H3K4me3가 표지 되는 히스톤 메틸화의 변이에 의해 결정되는 것으로 사료된다.
The experiment was conducted to validate anti-inflammatory effects of pinitol from bean. It was evaluated for some molecule targets by wound healing assay and RT-PCR. The results of wound healing assay was shown dose-dependent inhibition of cell migration in cancer cells and inhibited RNA expression of ICAM-1, CD 44, MMP-17, MMP-14 and ARF2. Immune suppression activity in a mouse provoked by DNFB observed that inflammatory reaction with pinitol were reduced ear swelling and inflammatory cells infiltration in mouse atopic models. The result confirmed that pinitol have the effect of dose-dependent immune suppression activity.

The present study identified the favorable conditions for isolation, enrichment and in vitro culture of highly purified, undifferentiated pig spermatogonial stem cell (SSC) lines that proliferate for long periods of time in culture. The colonies displayed morphology similar to miceSSC and were positive for markers of SSC (PGP9.5), proliferating germ cell (PigVASA), pre-meiotic germ cell (DAZL) and pluripotency (OCT4, SSEA-1, NANOG, and SOX2) based on immuno-cytochemistry and RT-PCR. The purity of these colonies was confirmed by negative expression of markers for sertoli cell (GATA4 and SOX9), peritubular myoid cell (${\alpha}$-SMA), differentiating spermatogonial and germ cells (c-KIT). The colonies could be maintained with undifferentiated morphology for more than two months and passaged more than 8 times with doubling time between 6-7 days. Taken together, we conclude that pigSSC could be successfully isolated and cultured in vitro and they possess characteristics similar to miceSSC.
Apple (Malus domestica) is one of the most economically important fruits in Korea. But virus infection has decreased sustainable production of apple and caused the serious problems such as yield loss and poor fruit quality. Virus or viroid infection including Apple chlorotic leaf spot virus (ACLSV), Apple stem pitting virus (ASPV), Apple stem grooving virus (ASGV), Apple mosaic virus (ApMV) and Apple scar skin viroid (ASSVd) has been also reported in Korea. In many cases, apple is infected with virus and viroid with no specific symptoms, the damage caused by the virus are unaware significantly. In our research, we tried to eliminate viruses in the rootstock for the disease-free seedlings of the apple dwarfing rootstock M.9 and M.26. The method of virus elimination was meristem culture, heat($37^{\circ}C$, 6weeks) treatment and chemistry($Ribavirin^{(R)}$) treatment. The analytical methods commonly used for the detection of virus is Enzyme-linked Immuno-Sorbent Assay(ELlSA) and Reverse Transcription-polymerase Chain Reaction(RT-PCR). RT-PCR method was more 30% sensitive than ELISA method. Efficiency of method eliminate virus appeared meristem method > heat treatment > chemistry treatment. The higher acquisition rate of disease-free seedlings is 30~40% on meristem treatment. In meristem treatment, the apple dwarfing rootstock M.9 gained infection ratio of ACLSV, ASPV and ASGV were 45%, 60% and 50% respectively. In the apple dwarfing rootstock M.26, infection ratio of ACLSV, ASPV and ASGV were 40%, 55%, 55%, respectively. Based on our results, it was found that most effective method of disease-free seedlings apple dwarfing rootstocks was by meristem treatment than heat method and chemistry treatment.

Human embryonic stem (ES) cells are derived from the inner cell mass of the preimplantation embryo. Human ES cells have the capacity to differentiate into various types of cells in the body. Human ES cells are indefinite source of cells for cell therapy in various degenerative disorders including neuronal disorders. Directed differentiation of human ES cells is a prerequisite for their clinical application. The objective of this study is to develop the culture condition for the derivation of neural precursor cells from human ES cells. Neural precursor cells were derived from human ES cells in a stepwise culture condition. Neural precursor cells in the form of neural rosette structures developed into neurospheres when cultured in suspension. Suspension culture of neurospheres has been maintained over 4 months. Expressions of nestin, soxl, sox2, pax3 and pax6 transcripts were upregulated during differentiation into neural precursor cells by RT-PCR analysis. In contrast, expression of oct4 was dramatically downregulated in neural precursor cells. Immunocytochemical analyses of neural precursor cells demonstrated expression of nestin and SOX1. When induced to differentiate on an adhesive substrate, neuro-spheres were able to differentiate into three lineages of neural systems, including neurons, astrocytes and oligo-dendrocytes. Transcripts of sox1 and pax6 were downregulated during differentiation of neural precursor cells into neurons. In contrast, expression of map2ab was elevated in the differentiated cells, relative to those in neural precursor cells. Neurons derived from neural precursor cells expressed NCAM, Tuj1, MAP2ab, NeuN and NF200 in immunocytochemical analyses. Presence of astrocytes was confirmed by expression of GFAP immuno-cytochemically. Oligodendrocytes were also observed by positive immuno-reactivities against oligodendrocyte marker O1. Results of this study demonstrate that a stepwise culture condition is developed for the derivation of neural precursor cells from human ES cells.
Anabolic steroids are frequently used to increase the growth rate of meat-producing animals. Exposure to an anabolic-androgenic steroid, nandrolone decanoate (ND), is associated with expressional reduction of testicular steroidogenic enzymes. However, the effect of withdrawal of ND exposure on the expression of these testicular molecules has not been thoroughly explored. The current research investigated expression changes of testicular steroidogenic enzymes in rats at several recovery periods (2, 6, and 12 weeks) after the stop of ND treatment with different doses (2 and 10 mg/kg body weight) for 12 weeks. Body and testis weights were recorded, and transcript levels of molecules were determined by quantitative real-time polymerase chain reaction (PCR). The immunohistochemistry was used to examine the changes of immuno-intensities of molecules. At 6 and 12 weeks of the recovery period, the 10 mg/kg ND-treated rats were lighter than other experimental groups. The interstitial compartment vanished by ND treatment filled up as the recovery period became longer. The expression of steroidogenic acute regulatory protein was returned to the control level at 12 weeks of the recovery period. Expression levels of cytochrome P450 side-chain cleavage and 17a-hydroxylase were increased in 2 mg/kg ND-treated group at 6 weeks of the recovery period, and transcript levels of these molecules in 2 and 10 mg/kg ND-treated groups at 12 weeks of the recovery period were significantly lower than the control. Expression levels of 3β-hydroxysteroid dehydrogenase (HSD) type I and 17β-HSD type 3 in 2 mg/kg ND-treated group were comparable with those of control at 12 weeks of the recovery period, but not in 10 mg/kg ND-treated group. Expression of cytochrome P450 aromatase (Cyp19) was reverted to the control level at 2 weeks of the recovery period. Except for Cyp19, there was a visible increase of immuno-staining intensity of other testicular steroidogenic enzymes in the Leydig cells as the recovery period progressed. This research has demonstrated that the cease of ND administration could restore the expression of testicular steroidogenic enzymes close to the normal level. Nevertheless, a relatively long recovery period, compared to the ND-exposure period would be required to retrieve normal expression levels of testicular steroidogenic enzymes.
Clostridioides (Clostridium) difficile is a Gram (+), anaerobic, spore forming, rod shaped bacterium that can produce toxin. The objective of this study is to reveal the presence of C. difficile in meat products, to analyze the ribotype diversity by PCR and to evaluate the antibiotic susceptibility of isolated strains. The organism was isolated in 22 out of 319 (6.9%) examined meat product samples and 9 out of 22 (40.9%) isolates were identified as RT027 and all isolates had the ability of toxin production. In terms of antibiotic susceptibility, all isolates were susceptive to amoxicillin-clavulanic acid, tetracycline and vancomycin and 21 (95.4%) isolates to metronidazole. On the other hand, imipenem and cefotaxim resistance was observed in all. In conclusion, the results of this comprehensive study conducted in Turkey deduced the presence of C. difficile in different meat products. Therefore, these products can be evaluated as a potential contamination source of C. difficile from animals to humans especially for elders, youngsters, long terms wide spectrum antibiotic used and immuno-suppressed individuals.

제 5 장 계약 해지 및 이용 제한

② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
