Microencapsulation에 의한 미세조류의 고밀도 배양 (High-Density Cultivation of Microalgae using Microencapsulation)

  • 본 연구에서는 미세조류의 고밀도 배양을 위한 경제적인 공정을 개발하는 차원에서 3종의 미세조류 즉, Chlorella vulgaris, Dunaliella salina 및 Porphyridium purpureum를 Ca-alginate 로 microencapsulation 한 후 air-bubble column형 배양조에서 고밀도 배양을 시도하였다. $2\%\;CaCl_2$ 농도로 capsule을 제조할 때 capsule의 안정성이 가장 좋았으며, capsule 막의 두께 및 $CO_2$ 공급유무에 따른 미세조류 (Chlorella vulgaris)의 생산성은 거의 차이가 없었다. 한편, 3종의 미세조류 모두 capsule로 고정화시킨 것이 bead로 고정화한 미세조류나 free cell 보다 농도가 높았으며, 특히Dunaliella salina가 microencapsulation에 의한 고밀도 배양 효과가 가장 뛰어나 free cell 보다 약 5배 정도의 생산성을 나타내었다.

  • 본 연구는 균주로 Rhodospirillum rubrum KS-301, 증식 제한 기질로 글루코오스, 고정화 담체로 Ca alginate를 사용한 고정화 생물 반응기를 조작할 때 담체에 의해 형성되는 물질전달 저항에 대한 도입 기질의 농도, 희석속도의 영향을 고찰하였다. 또한 효율인자를 평가하기 위해 속도식 변수를 구하였다. 충전층 반응기의 경우 외부 액막 물질전달 저항보다는 내부 물질전달 저항이 우세함을 알 수 있었고 총괄 효율인자는 희석속도의 증가에 따라 감소하였다. 연속 교반 탱크 반응기의 경우 외부 액막 물질전달 저항은 무시할 수 있으며 총괄 효율인자는 희석속도에 영향을 받지 않았다. 희석속도 0.2/h, 비드 반경 0.15cm, 초기 글루코오스 농도 1.0g/L의 실험조건에서 총괄 효율인자는 충전층 반응기와 연속 교반 반응기에서 각각 0.70과 0.77이었다.

Optimization of Culture Medium for Novel Cell-Associated Tannase Production from Bacillus massiliensis Using Response Surface Methodology

  • Naturally immobilized tannase (tannin acyl hydrolase, E.C. 3.1.1.20) has many advantages, as it avoids the expensive and laborious operation of isolation, purification, and immobilization, plus it is highly stable in adverse pH and temperature. However, in the case of cell-associated enzymes, since the enzyme is associated with the biomass, separation of the pure biomass is necessary. However, tannic acid, a known inducer of tannase, forms insoluble complexes with media proteins, making it difficult to separate pure biomass. Therefore, this study optimizes the production of cell-associated tannase using a "protein-tannin complex" free media. An exploratory study was first conducted in shake-flasks to select the inducer, carbon source, and nitrogen sources. As a result it was found that gallic acid induces tannase synthesis, a tryptose broth gives higher biomass, and lactose supplementation is beneficial. The medium was then optimized using response surface methodology based on the full factorial central composite design in a 3 l bioreactor. A $2^3$ factorial design augmented by 7 axial points (${\alpha}$ = 1.682) and 2 replicates at the center point was implemented in 17 experiments. A mathematical model was also developed to show the effect of each medium component and their interactions on the production of cell-associated tannase. The validity of the proposed model was verified, and the optimized medium was shown to produce maximum cell-associated tannase activity of 9.65 U/l, which is 93.8% higher than the activity in the basal medium, after 12 h at pH 5.0, $30^{\circ}C$. The optimum medium consists of 38 g/l lactose, 50 g/l tryptose, and 2.8 g/l gallic acid.

$\small{D}$-Lactic Acid Production by Sporolactobacillus inulinus Y2-8 Immobilized in Fibrous Bed Bioreactor Using Corn Flour Hydrolyzate

  • In this study, a fibrous bed bioreactor (FBB) was used for $\small{D}$-lactic acid ($\small{D}$-LA) production by Sporolactobacillus inulinus Y2-8. Corn flour hydrolyzed with ${\alpha}$-amylase and saccharifying enzyme was used as a cost-efficient and nutrient-rich substrate for $\small{D}$-LA production. A maximal starch conversion rate of 93.78% was obtained. The optimum pH for $\small{D}$-LA production was determined to be 6.5. Ammonia water was determined to be an ideal neutralizing agent, which improved the $\small{D}$-LA production and purification processes. Batch fermentation and fed-batch fermentation, with both free cells and immobilized cells, were compared to highlight the advantages of FBB fermentation. In batch mode, the $\small{D}$-LA production rate of FBB fermentation was 1.62 g/l/h, which was 37.29% higher than that of free-cell fermentation, and the $\small{D}$-LA optical purities of the two fermentation methods were above 99.00%. In fe$\small{D}$-batch mode, the maximum $\small{D}$-LA concentration attained by FBB fermentation was 218.8 g/l, which was 37.67% higher than that of free-cell fermentation. Repeate$\small{D}$-batch fermentation was performed to determine the long-term performance of the FBB system, and the data indicated that the average $\small{D}$-LA production rate was 1.62 g/l/h and the average yield was 0.98 g/g. Thus, hydrolyzed corn flour fermented by S. inulinus Y2-8 in a FBB may be used for improving $\small{D}$-LA fermentation by using ammonia water as the neutralizing agent.

Isolation, identification and immobilized-cell characteristics of a bacterium that produces $N_2$ from $NH{_4}{^+}$ under an aerobic condition

  • To treat wastewater efficiently by a one-step process of nitrogen removal, a new strain of $N_2-producing$ bacteria from $NH{_4}{^+}$ under an aerobic condition was isolated and identified. By 16S-rDNA analysis, the isolate was identified as Enterobacter asburiae with 96% similarity. The isolate shows that the capacity of $N_2$ production under an oxic condition was approximately three times higher than that under an anoxic condition. The optimal conditions (pH, temperature and C/N ratio) of the immobilized isolate for $N_2$ production were found to be 7.0, $30^{\circ}C$ and 5, respectively. Under all the optimum reaction conditions, the removal efficiency of $COD_{Cr}$ and TN reached 56.1 and 60.9%, respectively. The removal rates of $COD_{Cr}$ and TN were highest for the first 2.5 hrs (with the removal $COD_{Cr}$ ratios of 32.1), and afterwards the rates decreased as reaction proceeded. For application of the immobilized isolate to a practical process of ammonium removal, a continuous bioreactor system exhibited a satisfactory performance at HRT of 12.1 hr, in which the effluent concentrations of $NH{_4}{^+}-N$ was measured to be 15.4 mg/L with its removal efficiency of 56.0%. The maximum removal rate of $NH{_4}{^+}-N$ reached 1.6 mg $NH{_4}{^+}-N/L/hr$ at HRT of 12.1 hr (with N loading rate of 0.08 $Kg-N/m^3-carrier/d)$. As a result, the application of the immobilized isolate appears a viable alternative to the nitrification-denitrification processes.

The cell was motile and a Gram-negative rod, and usually occurred in pairs. By 16S-rDNA analysis, the isolated strain was identified as Enterobacter asburiae with 96% similarity. The isolate showed that the capacity of $N_2$ production under an oxic condition was approximately three times higher than that under an anoxic condition. Thus, the consumption of ${NH_4}^+$ by the isolate was significantly different in the metabolism of $N_2$ production under the two different environmental conditions. The optimal conditions of the immobilized isolate for $N_2$ production were found to be pH 7.0, $30^{\circ}C$ and C/N ratio 5, respectively. Under all the optimum reaction conditions, $N_2$ production by the immobilized isolate resulted in reduction of ORP with both the consumption of DO and the drop of pH. The removal efficiencies of $COD_{Cr}$, and TN were 56.1 and 60.9%, respectively. The removal rates of $COD_{Cr}$, and TN were the highest for the first 2.5 hrs with the removal $COD_{Cr}/TN$ ratios of 32.1, and afterwards the rates decreased as reaction proceeded. For application of the immobilized isolate to a practical process of ammonium removal, a continuous operation was executed with a synthetic medium of a low C/N ratio. The continuous bioreactor system exhibited a satisfactory performance at 12.1 hrs of HRT, in which the effluent concentrations of ${NH_4}^+$-N was measured to be 15.4 mg/L with its removal efficiency of 56.0%. The maximum removal rate of ${NH_4}^+$-N reached 1.6 mg ${NH_4}^+$-N/L/hr at 12.1 hrs of HRT(with N loading rate of $0.08\;Kg-N/m^3$-carrier/d). As a result, the application of the immobilized isolate appears a viable alternative to the nitrification-denitrification processes.

Production of Shikonin by A Hairy Root Culture of Lithospermum erythrorhizon

  • Shikonin production was examined in a bubble column bioreactor system with the hairy roots of Lithosphermum erythrorhizon. The volumetric productivity was higher than those obtained from other reactor configurations with free or immobilized cells of the same cell line. The productivities of the bubble column reactor, with and without a product absorption trap, were 7.4 and 4.5 mg of shikonin/l/d, respectively. This indicated the importance of the product removal in the design and operation of the shikonin production system with hairy root culture.

Production of Oligosaccharides from Sucrose for Animal Industry

  • The purpose of the present investigation was to develop a novel method for cell immobilization. Aureobasidium pullulans cells were mixed with an alginate solution, and the mixture was extruded to form small gel beads as hydrated- immobilized cells. The beads were then placed at $-15^{\circ}C$ for 6-24 h to induce freeze-dehydration. The freeze-dehydration resulted in shrinkage of beads due to water removal reducing bead volume by 82% and bead weight by 85%. The dehydrated beads were successfully used for the production of fructo-oligosaccharides in a model reactor system. This study showed that bioreactor performance can be improved up to 2 times by the use of the dehydrated beads.

Bioprocess Strategies and Recovery Processes in Gibberellic Acid Fermentation

  • Gibberellic acid (GA$_3$) is a commercially important plant growth hormone, which is gaining much more attention all over the world due to its effective use in agriculture and brewing industry. Industrially it is produced by submerged fermentation technique using Ascomycetous fungus Gibberella fujikuroi. Solid state and immobilized cell fermentation techniques had also been developed as an alternative to obtain higher yield of GA$_3$. This review summarizes the problems of GA$_3$ fermentation such as production of co-secondary metabolites along with GA$_3$, substrate inhibition and degradation of GA$_3$ to biologically inert compound gibberellenic acid, which limits the yield of GA$_3$ in the fermentation medium. These problems can be overcome by various bioprocessing strategies e.g. two - stage and fed batch cultivation processes. Further research on bioreactor operation strategies such as continuous and / or extractive fermentation with or without cell recycle / retention system need to be investigated for improvement in yield and productivity. Down stream processing for GA$_3$ isolation is also a challenge and procedures available for the same have been critically evaluated.

이중실관 생물 반응기에서의 구연산 생산과 Scale-up (Citric Acid Production and Scale-up in Dual Hollow Fiber Bioreactor)

  • 여러 크기의 이중실관 생물 반응기에서의 Aspergillus niger(KCTC 1232)를 이용한 구연산 생산 실험을 수행하였다. 초종 세포농도는 세포 성장구간 기준으로 300g/l에 달하였다. 공기와 산소의 공급 조건하에서의 단위 용적당의 생산성은 각각 0.63, 0.02g/l.h였고 이는 회분식 발효에 대해 10, 16배 증가한 결과이다. 공급배지의 초기 pH는 구연산의 생산에 중요한 요소이며 pH가 낮을수록 높은 구연산 생산수율을 얻을 수 있었다. Scale-up의 가능성을 알아보기 위해 반응기 unit와 배지의 공급속도를 변화시킨 결과 반응기 unit와 배지 공급속도의 증가는 기질의 높은 소비속도로 인해 생산성의 증가를 가져왔다.
