In batch bioreactor fermentations for cyclosporin A (CyA) production, good potential for bioprocess improvement was demonstrated in the immobilized cell system, providing appreciably better utilization of the catalytic activity of the biomass than the freely suspended cells, especially during the exponential phase. When concentrated nutrient medium was added pulsely during the exponential phase of cell growth(at hour 139 of fermentation), reactivation and regermination in both immobilized and suspended cell cultures were observed to contribute to the longevity of CyA production, maintaining maximum CyA titre until 250 hours of fermentation. Contrarily, simple batch fermentations without any supplement of medium in both systems showed repid decrease in CyA concentrations during the late stationary phase. Notably, the CyA yield coefficient $(Y_p/x)$ for the immobilized cell system was maintained quite high even after the pulse addition of the concentrated full medium, reaching almost 80% of the level attained during the exponential phase. This is in sharp contrast when compared with the corresponding value of 58% in the case of parallel-suspended cells. This pattern of CyA production resulted in considerably enhanced CyA production in the immobilized cell system, reaching almost 2 time higher maximum CyA production in comparison with the free cell system.
A marine bacterium, Pseudomonas aeruginosa BYK-2 KCTC 18012P was immobilized in modified polyvinyl alcohol for the continuous production of rhamnolipids. The stability of rhamnolipids production, the mechanical strength of beads and the scanning electron microscope of immobilized cell were determined in a repeated batch culture. The rhamnolipids production was maintained $80{\sim}90%$ stability of initial production, and the mechanical strength also was stable during the repeated batch culture more than 14 cycles. In the case of SEM studies, the internal distribution pattern of the cell entrapped in modified PVA beads was observed. On the basis of optimal conditions, the continuous culture was investigated in 1.8L air lift bioreactor. The result suggested 0.1g/h rhamnolipids was obtained from 1%(v/v) fish oil continuously in conditions of 1.2L working volume, 0.5vvm and 20ml/h flow rate.
Plant cell cultures of Lithospermum erythrorhizon were performed in stirred tank and packed-bed reactors with in situ extraction by n-hexadecane. The specific shikonin production and volumetric shikonin productivity of stirred tank reactor reached 1.5 mg shikoninlg cell and 400$\mu g$ shikonin/(L.day), respectively. In packed-bed reactor with calcium alginate-immobilized cells specific shikonin production and volumetric productivity reached 2.0 mg shikoninlg cell and 2857$\mu g$ shikonin/(L.day), which were 1.3 and 7.1 times higher than those of stirred tank reactor, respectively. The higher shikonin production and productivity of packed-bed reactor seemed to be due to high cell loading capacity of calcium alginate immobilized cells in packed-bed reactor and improved cell-cell contact.
A denitrification bacterium was isolated from riverbed soil and identified as Ochrobactrum sp., whose specific enzymes for denitrification metabolism were biochemically assayed or confirmed with specific coding genes. The denitrification activity of strain G3-1 was proportional to glucose/nitrate balance, which was consistent with the theoretical balance (0.5). The modified graphite felt cathode with neutral red, which functions as a solid electron mediator, enhanced the electron transfer from electrode to bacterial cell. The porous carbon anode was coated with a ceramic membrane and cellulose acetate film in order to permit the penetration of water molecules from the catholyte to the outside through anode, which functions as an air anode. A non-compartmented electrochemical bioreactor (NCEB) comprised of a solid electron mediator and an air anode was employed for cultivation of G3-1 cells. The intact G3-1 cells were immobilized in the solid electron mediator, by which denitrification activity was greatly increased at the lower glucose/nitrate balance than the theoretical balance (0.5). Metabolic stability of the intact G3-1 cells immobilized in the solid electron mediator was extended to 20 days, even at a glucose/nitrate balance of 0.1.
Removal of nitrogen compound from waste water is essential and often accomplished by biological process. To prevent washout and to develop an efficient bioreactor, immobilization of sutibal microorganisms could be sensible approach. Strains and permeabilized cell encapsulated in cellulose nitrate microcapsules and immobilized on polystyrene films were prepared by the method described in the previous study. In the wastewater treatment system, nitrification of ammonia component is generally known as rate controlling step. To enhance the rate of nitrification, firstly nitrifying strains Nitrosomonas europaea(IFO14298), are permeabilized chemically, and immobilized on polystyrene films and secondly oxidation rates of strain system and permeabilized strain system are compared in the same condition. with 30 minute permeabilized cells, it took about 25 hours to oxidize 70% of ammonia in the solution, while it took about 40 hours to treat same amount of ammonia with untreated cells. All the immobilization procedures did not harm to the enzyme activity and no mass transfer resistance through the capsule well was shown. In the durability test of immobilized system, the system showed considerable activity for the repeated operation for 90 days. With these results, the system developed in this study showed the possibility to be used in the actual waste water treatment system.

Chitosan-alginate capsules were formed by electrostatic interactions and exhibited an appropriate mechanical strength, permeability, and stability for the culture of hepatocytes. Pig hepatocytes were isolated and hepatocyte spheroids formed and immobilized in chitosan-alginate capsules. An encapsulation procedure of 3 min and spheroid formation period of 24 h were the optimum conditions for the best liver functions. Pig hepatocytes with a cell density of $6.0{\tomes}10^6$ cells/ml in the capsules were found to be most suitable for application in a bioartificial liver support system. The encapsulated pig hepatocyte spheroids exhibited stable ammonia removal and urea secretion rates in a bioreactor for 2 weeks. Accordingly, chitosan-alginate encapsulated hepatocyte spheroids in a packed-bed bioreactor would appear to have potential as a bioartificial liver.
Anabaena variabilis ATCC 29413 grew in chromium (Cr) containing Chu-10 (basal) and nitrate-supplemented media, and the growth of the organism in $100{\mu}M$ chromium was found to be 50% of that in control medium. The growth in nitrate $({NO_3}^-)$ supplemented cultures was better as compared to cultures grown in basal medium. Free cells from basal and nitrate-supplemented media removed 5.2 and 7.4 nmol of chromium $mg^{-1}$protein in 8 h, respectively, from the medium containing $30{\mu}M$ chromium. The efficiency of chromium removal increased 7-fold in imidazole buffer (0.2 M, pH 7.0). A cell density equivalent to $100{\mu}g$ protein $ml^{-1}$ was found to be optimum for maximum Cr removal. Entrapment of cells in calcium-alginate beads did not affect the rate of Cr uptake by the cells. The efficiency of the laboratory-scale continuous flow bioreactor $(12.5{\times}2cm)$ loaded with alginate-immobilized cells (10 mg protein) and fed with $30{\mu}M$ chromium solution was compared at different flow rates. The efficiency of the bioreactor varied with flow rates. In terms of percent removal of Cr from influent, a flow rate of 0.1 ml $min^{-1}$ was found to be optimum for 6 h (54% Cr removal efficiency). Maximum amount of Cr (883 nmol) was removed by the cells in 3 h at a flow rate of 0.5 ml $min^{-1}$. The potential use of A. variabilis in removing Cr from industrial effluents is discussed.
Optimum conditions for production of casein phosphopeptides (CPP) from sodium casenate by immobilized cell culture of Streptococcus faecalis var. liquefaciens were investigated. Immobilized cells were made by mixing 60% sodium alginate solution with an equal volume of culture broth at the end of exponential phase and subsequently dropping the mixture into $CaCl_{2}$ solution. Optimum conditions for CPP production by the immobilized cells were the same as those ($50^{\circ}C$, pH 7.0, and 10% substrate concentration) by the crude enzyme solution from the supernatant of culture broth. Optimum loading volume of the immobilized cells into a batch reactor was 30% (w/v). Using a continuous reactor loaded by the immobilized cells under the identified optimal conditions, we were able to produce CPP continuously up to 30 days with a maximum CPP conversion efficiency of 20%.
A one-stage, continuous-flow bioreactor with both immobilized and suspended cells was used to investigate the roles of lipid supplements in ethanol production by Saccharomyces cerevisiae. The reactor performance and the level of alcohol dehydrogenase(ADH) activities of the suspended cells, grown under various conditions, were measured. When ergosterol and/or oleic acid were added with surfactants to the yeast culture grown under non-aerated conditions, remarkable increases in ethanol production and cell growth was achieved, but specific ADH activities were not affected. Especially, no difference of specific ADH activities of the suspended cells grown under aerated and non-aerated condition was observed. The addition of the surfactant as a supplement also resulted in significant increases in ethanol production, cell growth, and specific ADH activity. When ergosterol and oleic acid were added to the yeast culture exposed to higher ethanol concentration($>40\;g/{\ell}$) level, ethanol production, cell growth, and specific ADH activity were increased, but the addition of surfactant was as effective as at lower ethanol concentration level. The results indicated that lipid supplements were more effective at higher ethanol concentration level than at lower ethanol concentration level during ethanol production. ADH isozyme patterns of the yeast cultures grown under various conditions on starch gel electrophoresis showed only one major band, probably ADH I. The migrating distance of the major isozyme, however, varied slightly according to the culture conditions of the cells. No apparent correlation was found between specific ADH activity and amount of ethanol produced. Cell mass was more important factor for ethanol production than specific ADH activity of the cells.
The porcine epidemic diarrhea virus(PEDV) production yield in spinner flask cultures using Vero cells immobilized on microcarriers was improved by the selective adsorption of ammonium ions in a Carberry type bioreactor which was equipped with Phillipsite-Gismondine synthetic zeolite. Though the apparent cell growth seemed to be lower than that of control due to the aggregation of microcarriers between impeller shaft and the adsorbent, zeolite was found to not to be toxic to Vero cell, considering estimated glucose and lactate changes. Zeolite was observed to remove ammonium ions effectively in both steps of cell growth and virus production. In virus production, the virus titer with zeolite was two times higher than that without zeolite. Consequently, zeolite was found to be an ideal adsorbent for higher production of virus vaccine with the effective removal of ammonium ions.
