A study of the comparative effect of Kuibitang, Kamiondarmtang, and Kuibiondarmtang on serum levels in rats under the immobilization stress. (귀비탕(歸脾湯), 가미온담탕(加味溫膽湯) 및 귀비온담탕(歸脾溫膽湯)의 항(抗) Stress 효능(效能) 비교연구(比較硏究))

  • In order to compare and examine the effect the effect of Kuibitang, Kamiondarmtang and Kuibiondarmtang against stress, I induced immobilization stress by binding rats for 200mins under a little anesthetized with ether. I classified as normal group, control group which administrated 0.9% NaCl solution, sample A group which administrated Kuibitang, sample B group which administrated Kamiondarmtang and sample C group which administrated Kuibiondarmtang. I got some conclusions by measuring amounts of norepinephrine, epinephrine, triiodothyronine($T_3$) and thyroxine($T_4$) in 1, 3 and 5 days, respectively. 1. Norepinephrine showed high meaningful decrease after 1. 3 and 5 days in sample A group. It showed little decrease in sample B group. It showed a little meaningful decrease after 1 day and high meaningful decrease after 3 and 5 days in sample C group. 2. Epinephrine showed high meaningful decrease after 3 and 5 days in sample A group. It showed little decrease in sample B group. It showed a little meaningful decrease after 3 days and high meaningful decrese after 5 days in sample C group. 3. Triiodothyronine($T_3$) inclined to approach normal value in sample A, B, C group in comparision wit control group but didn't have high meaning. 4. Thyroxine($T_4$) inclined to approach normal va1ue in sample A, B, C group in comparision with contrl group but didn't have high meaning. As I consider the effect of experiments, I think that Kuibitang and Kuibiondarmtang have some effects against stress and it is right to apply to the treatment of stress.

The Effects of Puerariae Flos on Stress-induced Deficits of Learning and Memory in Ovariectomized Female Rats

  • Puerariae flos (PF) is a traditional oriental medicinal plant and has clinically been prescribed for a long time. The purpose of the present study was to examine the effect of PF on repeated stress-induced alterations of learning and memory on a Morris water maze (MWM) test in ovariectomized (OVX) female rats. The changes in the reactivity of the cholinergic system were assessed by measuring the immunoreactive neurons of choline acetyltransferase (ChAT) in the hippocampus after behavioral testing. The female rats were randomly divided into four groups: the nonoperated and nonstressed group (normal), the sham-operated and stressed group (control), the ovariectomized and stressed group (OS), and the ovariectomized, stressed and PF treated group (OSF). Rats were exposed to immobilization stress (IMO) for 14 d (2 h/d), and PF (400 mg/kg, p.o.) was administered 30 min before IMO stress. Results showed that treatments with PF caused significant reversals of the stress-induced deficits in learning and memory on a spatial memory task, and also increased the ChA T immunoreactivities. In conclusion, administration of PF improved spatial learning and memory in OVX rats, and PF may be useful for the treatment of postmenopausal-related dementia.

Effects of Sumsu (Bufonis venenum) Pharmacopuncture Treatment on Depression in Mice

  • Objectives: The main objective of this study was to evaluate the anti-depressant effects of pharmacopuncture using sumsu (Bufonis venenum). Methods: Animals were divided into three groups (control, sham, and experimental), with eight mice per group. The sham and the experimental groups were exposed to 2 hours of immobilization stress daily for 14 days. They were also injected with normal saline (sham) or subjected to pharmacopuncture with sumsu at the acupoints HT7, SP6, and GV20 (experimental). The depression or anxiety-like behaviors of the mice in each group were evaluated 1 day after treatment. Results: There was no difference in locomotor activity between the groups during the open-field test; i.e., all groups had normal motor function. However, the open-field and the forced-swimming tests revealed that depression and anxiety-like behaviors were decreased significantly in the group treated with sumsu pharmacopuncture. Conclusion: Sumsu pharmacopuncture attenuated depressive or anxiety-like behavior in mice stressed with chronic immobilization. These results suggest that sumsu pharmacopuncture has therapeutic potential for treating neuropsychiatric disorders such as anxiety or depression disorder.

Immobilization stress increased cytochrome P450 1A2 (CYP1A2) expression in the ovary of rat

  • Under the stressed condition, a complex feedback mechanism for stress is activated to maintain homeostasis of the body and secretes several stress hormones. But these stress hormones impair synthesis and secretion of the reproductive hormones, followed by suppression of ovarian function. Cytochrome P450 1A2 (CYP1A2) plays a major role in metabolizing exogenous substances and endogenous hormones, and its expression is recently identified at not only the liver but also several organs with respect to the pancreas, lung and ovary. Although the expression of CYP1A2 can be also affected by several factors, understanding for the changed pattern of the ovarian CYP1A2 expression upon stress induction is still limited. Therefore, CYP1A2 expression in the ovaries from immobilization stress-induced rats were assessed in the present study. The stress-induced rats in the present study exhibited the physiological changes in terms of increased stress hormone level and decreased body weight gains. Under immunohistological observation, the ovarian CYP1A2 expression in both control and the stressed ovary was localized in the antral to pre-ovulatory follicles. However, its expression level was significantly (p < 0.01) higher in the stress-induced group than control group. In addition, stress-induced group presented more abundant CYP1A2-positive follicles (%) than control group. Since expression of the ovarian CYP1A2 was highly related with follicle atresia, increased expression of CYP1A2 in the stressed ovary might be associated with changes of the ovarian follicular dynamics due to stress induction. We hope that these findings have important implications in the fields of the reproductive biology.

  • Uranium mill tailing ponds (UMTPs) are risk source of debris flow and a critical source of environmental U and Rn pollution. The technology of microbial induced calcium carbonate precipitation (MICP) has been extensively studied on reinforcement of UMTs, while little attention has been paid to the effects of MICP on U & Rn release, especially when incorporation of metakaolin and bacillus subtilis (MBS). In this study, the reinforcement and U & Rn immobilization role of MBS -MICP solidification in different grouting cycle for uranium mill tailings (UMTs) was comprehensively investigated. The results showed that under the action of about 166.7 g/L metakaolin and ~50% bacillus subtilis, the solidification cycle of MICP was shortened by 50%, the solidified bodies became brittle, and the axial stress increased by up to 7.9%, and U immobilization rates and Rn exhalation rates decrease by 12.6% and 0.8%, respectively. Therefore, the incorporation of MBS can enhance the triaxial compressive strength and improve the immobilization capacity of U and Rn of the UMTs bodies solidified during MICP, due to the reduction of pore volume and surface area, the formation of more crystals general gypsum and gismondine, as well as the enhancing of coprecipitation and encapsulation capacity.

Anti-stress Effect of Cholic acid Derivatives in Restraint Stress Induced Rats (구속스트레스를 가한 흰쥐에서 Cholate류의 항스트레스 작용에 관한 연구)

  • This study was done to investigate whether cholic acid derivatives have anti-stress activity and what is a cause of this anti-stress effect. Seven cholic acid derivatives (cholic acid, taurocholic acid, ursodeoxycholic acid, tauroursodeoxychoic acid, chenodeoxy cholic acid, dehydrocholic acid, hyodeoxycholic acid) were used, silymarin and valproic acid were used as positive controls. Stress was induced by restraint immobilization technique plus water immersion (24hrs) and adrenal weight, spleen weight, adrenal ascorbic acid, serum cholesterol, lactate dehydrogenase (LDH), alkaline phosphatase (ALP), adrenal cholesterol, glucose and corticosterone levels were measured as stress indicators. Most cholic acid derivatives markedly decreased the adrenal weight, and TUDCA and DHCA increased the spleen weight. The restraint stress induced increments in serum LDH, ALP and cholesterol were attenuated by most cholic acid derivatives. Cholic acid, taurocholic acid and tauroursodeoxycholic acid only increased the content of adrenal ascorbate. While valproic acid showed an inhibitory effect against stress, silymarin did not. Our findings suggest that most cholic acid derivatives have anti-stress effect and that their anti-stress effect is, in part, related to choleretic activity.

  • A yeast hydrolysate SCP-20 with anti-stress and mitigative effects for premenstrual syndromes (PMS) was composed with crude protein (57.5%) and carbohydrates (28.1%). For the investigation of anti-stress effect in the immobilization stressed rats, the weights of spleen, thyroid and kidney in the group of SCP-20 sdministration were significantly different from those in the group not given SCP-20. Total power (TP), suppression indicator of stress, was obtained from frequency of heart rate variability. TP was changed by the administration of SCP-20 suggesting that SCP-20 has the autonomous nervous control effect. In premenstrual assessment form(PAF), the administration of SCP-20 reduced the intensity of somatic symptoms, emotional symptoms, and behavioral symptoms.

Long-term prenatal stress increases susceptibility of N-methyl-D-aspartic acid-induced spasms in infant rats

  • Purpose: Infantile spasms, also known as West syndrome, is an age-specific epileptic seizure. Most patients with this condition also exhibit delayed development. This study aimed to determine the effect of long-term prenatal stress on susceptibility to infantile spasms. Methods: We subjected pregnant rats to acute or chronic immobilization stress. Resulting offspring received N-methyl-D-aspartic acid (15 mg/kg, intraperitoneally) on postnatal day 15, and their behaviors were observed 75 minutes after injection. The expression of KCC2 and GAD67 was also determined using immunohistochemistry. Results: Exposure to long-term prenatal stress increased the frequency of spasms and decreased the latency to onset of spasms compared with offspring exposed to short-term prenatal stress. Expression of KCC2 and GAD67 also decreased in the group exposed to long-term prenatal stress compared with the group exposed to short-term prenatal stress. Conclusion: Our study suggests that exposure to long-term prenatal stress results in increased susceptibility to seizures.

Anti-stress Effect by the Treatment of Vitamin C (Vitamin C 투여에 의한 항스트레스 효과)

  • The anti-stress effect by the treatment of vitamin C was investigated in this study. The treatment of ascorbic acid in the presence of $Cu^{2+}$ion induced strong time- and dose-dependent degradation of hitamine, and also the addition of histamine accelerated time-dependent decomposition of ascorbic acid in vitro. The treatment of ascorbic acid in $ODS^{od}/_{od}$rats, which cannot synthesize ascorbic acid, significantly decreased the urinary histamine. The protreatment of ascorbic acid, dexamethasone and promethazine inhibited the lethal effect induced by immobilization stress, but that of dimethylsulfoxide did not. The addition of ascorbic and to a culture of spleen cells of $ODS^{od}/_{od}$rats significantly increased the Con A-dependent T lymphocyte proliferation.
