Purpose : Several modulatory factors for renal peripheral benzodiazepine receptor (PBR) has been reported, but their physiological significance remains elusive. Tissue-specific, stress-induced down-regulation of renal PBR coupled with the pharmacological stimulation of these effects by angiotensin II suggested that physiological significance of renal PBR may be related to the pathophysiology of stress-induced hypertension. The boderline hypertensive rat (BHR) has been used extensively to study the interaction of environmental factors, such as stress and blood pressure. The BHR is the first-generation progeny of a cross between the spontaneously hypertensive rat and the control Wistar-Kyoto rat. The pathogenesis of stress induced hypertension in this model is not demonstrated well. Methods In this study, BHR (male, 150-200 g) and Sprague-Dawley (SD, male, 150-200 g) rats were treated by repeated immobilization to induce anxiety. We used plus-maze performance to observe the level of anxiety by measuring percent open crosses and percent time in open. Results : Percent open crosses and percent time in open in BHR were lower than in SD rats (P<0.05). Receptor densities of renal PBR in BHRs were significantly lower than those of SDs (P<0.05). We also observed that the renal PBR was upregulated in the repeatedly stressed (immobilization, 2 hours daily, for 2 weeks) rats, both in the BHR and SD. However, the density of renal PBR in the stressed BHR was still lower than that of stressed SD. Renal PBR has been suggested to be an important organs which Is responsible for the production of cholesterol-derived products during stress. Conrlusion : From these results, it can be summarized that the lowed density of renal PBR may be involved in the pathogeneis of stress-induced hypertension.
In order to investigate the Anti-stress effect of Guibiondamtang in the immobilization stressed rats, the level of serum catecholamine, the change of body weight, the humoral and cellular immune response were studied. The results were as follows; 1. The decrese of the body weight was significantly inhibited in test group for Guibiondamtang, comparting to the control group. 2. The increase of the level of serum norepinephrine was significantly inhibited in test group, comparing to the control group. 3. The increase of the level of serum epinephrine was significantly inhibited in test group, comparing to the control group. 4. In the hemagglutinaton titer, the control group was decreased on the serum antibody titer but test group was inhibitory effect on the decrease of sereum antibody titer. 5. In the plaque formation test, the control and test group were not shown significant differences. 6. In the foot pad swelling respopnse, the control group was decreased on DTH response but test group was increased comparing to the normal group. 7. There was no change on the distribution of lymphocyte subset(CD4, CD8), grnulocyte and macrophage analyzed by flow cytometry.
Introduction: Pharmacopuncture, which combines acupuncture with herbal medicine, is one of the newly developed acupuncture techniques that has recently been put into use. The possible mechanisms of scolopendra pharmacopuncture, as well as its potential effects on depressive symptoms, were investigated in this study by using a mouse model of chronic immobilization stress (CIS). Methods: C57BL/6 male mice were randomly assigned into three groups: mice not stressed with restraint and injected with distilled water, mice stressed with restraint and injected with distilled water, and mice stressed with restraint injected with scolopendra pharmacopuncture at a cervical site. Behavioral tests (an open field test, tail suspension test, and forced swimming test) were carried out after two weeks of CIS and injection treatments. The expression levels of glial fibrillary acidic protein (GFAP) in the hippocampus were determined by using western blot and immunohistochemistry analyses. Results: Mice exposed to CIS showed decreased behavioral activity, while scolopendra pharmacopuncture treatment significantly protected against the depressive-like behaviors induced by CIS. Moreover, scolopendra pharmacopuncture treatment increased GFAP protein levels in the hippocampi of the mice stressed by chronic immobilization. Conclusion: Scolopendra pharmacopuncture has an ameliorating effect on depressive behavior, which is partially mediated through protection against glial loss in the hippocampus.
Ginsenoside Rgl (Rgl), the pharmacologically active constituent of ginseng (Panax ginseng C.A. Meyer), has a variety of biological activities. The present study was undertaken to evaluate a possibility of Rgl whether it can be used in treatment or prevention of stress disorders. Animals were stressed by immobilization for 2 hours or electroshocks for 20 minutes. The normal group was not exposed to any stress. Rgl was subcutaneously injected as dosages of 5 and 10 mg/kg and red ginseng (RG) was orally administered 200 mg/kg as the positive control. Animals were given supplements for 5 days without stress, and then were given supplements for 5 days with stress. We recorded stress-related behavioral changes of experimental animals using the Etho-vision system. Weight of adrenal gland and levels of corticosterone in plasma were measured and stress related behaviors (smelling, grooming, face washing, rearing) were observed. Rgl didn't make significant behavioral changes in total open field and elevated plus maze test. Rgl did not influence on behavioral changes induced by electroshock stress. Whereas, 10 mg/kg of Rgl alleviated the increment of the freezing and face washing time and the decrement of the smelling and rearing time induced by restraint stress. The administration of Rgl 10 mg/kg has significantly increased the endurance time on rotating rod and swimming pool tests compared to the control group. These results indicate that Rgl can alleviate the damage induced by physical stress. This result suggests that Rgl may bea new candidate for treating stress related disorder.
Nonunion of the hallucal sesamoid usually occurs after stress fracture. In case of persistent disabling symptoms after non-operative treatment, surgery can be an option. We report a rare case of combined stress fractures of the medial bipartite sesamoid and the lateral sesamoid after overuse. Nonunion of the medial sesamoid developed in spite of cast immobilization, and it was eventually healed with curettage and bone grafting.
A role for brain serotonin(5-HT) in regulation of the HPA axis has been suggested but remains contoversial and poorly defined. The present experiments were designed to check kinetic parameters of 5-HT turnover in rat hypothalamus and remainder brain areas before and after stress and to test whether using various different pharmacologic approaches to stimulate or eliminate the control serotonergic system have any consistent effect on the stress-induced activation of HPA system. Steady state brain serotonin and 5-HIAA concentrations during 1 min ether stress were significantly elevated without significant rise in the levels of plasma corticosterone, which highly increased 2 minutes after stress. This suggests that the increase in serotonergic neuron activity precede that in HPA activity. Furthermore, during 1 ruin-ether stress or 30 min immobilization stress there is a marked increase in hypothalamic and remainder brain serotonin (5-HT) turnover or synthesis rates assessed by both the pargline/5-HT method and pargyline/5-HIAA method. The stress-induced corticosterone levels were increased by serotonin precursors and serotonin agonist in a dose-related fashion. The stress- induced corticosterone levels were highly elevated by L-tryptophan (100 mg/kg) and Potentiated by monoamine oxidase inhibitor, pargyline or serotonin agonist, 5-MeoDMT. The stress-induced elevation of corticosterone and 5-HT levels in rat brain were not significantly decreased by the administration of 5-HT synthesis inhibitor, PCPA and 5-HT neurotoxin, 5,7-DHT. However, the stress-induced elevation of corticosterone and 5-HT levels were decreased by the destruction of midline raphe nuclei. There was a strong positive correlation between plasma corticosterone and 5-HT concentrations changed by drugs which mainly manipulating 5-HT system in the hyhothalamus and in the remainder of the brain. In conclusion, our present data stongly suggest that 5-HT is an important key neurotransmitter involved in the stress-induced activation of the HPA system.

Ginseng is known to have antistress effects. Previously, red ginseng (RG) was shown to repress stress-induced peptidyl arginine deiminase type IV (PADI4) via estrogen receptor ${\beta}$ ($ER{\beta}$) in the brain, thus inhibiting brain cell apoptosis. Moreover, tumor necrosis factor (TNF)-${\alpha}$ plays a critical role in immobilization (IMO) stress. However, the signaling pathway of RG-mediated repressesion of inflammation is not completely understood. In this study, we determined how RG modulated gene expression in stressed brain cells. Since secretion of TNF-${\alpha}$ is modulated via TNF-${\alpha}$ converting enzyme (TACE) and nuclear factor (NF)-${\kappa}B$, we examined the inflammatory pathway in stressed brain cells. Immunohistochemistry revealed that TACE was induced by IMO stress, but RG repressed TACE induction. Moreover, PADI4 siRNA repressed TACE expression compared to the mock transfected control suggesting that PADI4 was required for TACE expression. A reporter assay also revealed that $H_2O_2$ oxidative stress induced NF-${\kappa}B$ in neuroblastoma SK-N-SH cells, however, RG pretreatment repressed NF-${\kappa}B$ induction. These findings were supported by significant induction of nitric oxide and reactive oxygen species (ROS) by oxidative stress, which could be repressed by RG administration. Taken together, RG appeared to repress stress-induced PADI4 via TACE and NF-${\kappa}B$ in brain cells thus preventing production of ROS and subsequently protecting brain cells from apoptosis.
This study was aimed to evaluate the anti-stress effect of danchisoyosan on the rats stressed by immobilization. The experimental animals were immobilized in the stress box($5{\times}5{\times}20cm$) for 12 hours in a day during 3 days, and administered $500mg/5m{\ell}/g$ of Danchisoyosan extract for 14 days before stress. There were measured the change of body weight and organ weight under immobilized-stress. The norepinephrine, epinephrine, dopamine, serotonine contents were measured by HPLC method in rat brain. There were measured the GOT, GPT contents in serum and tissue lipid peroxidation in the brain, liver, spleen, adrenalgland, pancreas, testes, thymus, heart. The following results were obtained: 1. The change of organ weight was significantly lower in control than normal group. Sample group inhibited decreased weight from stress comparing to control group. 2. Lipid peroxidation in the liver was significantly higher in control than normal group. Sample group shows significant decrease comparing to control group. 3. Lipid peroxidation in the kidney was significantly higher in control than normal group. Sample group shows significant decrease comparing to control group. 4. GPT contents in serum was significantly higher in control than normal group. Sample up shows significant decrease comparing to control group. 5. Dopamine contents in the brain was significantly higher in control than normal group. Sample group shows significant decrease comparing to control group. 6. Serotonine contents in the brain was significantly higher in control than normal group. Sample group shows significant decrease comparing to control group.
Stress activates hypothalamic-pituitary-adrenal (HPA) axis and subsequently increases the systemic levels of glucocorticoids. It also inhibits the release of gonadotropin-releasing hormone (GnRH) from hypothalamus. Korean ginseng (Panax ginseng CA Meyer) has been proven as an anti-stress agent. However, most of the anti-stress effects of ginseng on stresses such as immobilization, electronic foot shock, and cold swim, which subsequently cause oxidative damage in brain, were obtained by using ginseng extract or ginseng total saponin. Moreover, anti-stress and anti-oxidative effects of ginseng were demonstrated by determination of enzyme or hormone levels but not mRNA as well as transcriptome. Further studies on transcriptome, proteomics, and systems biology as well as signal transduction would be required to elucidate molecular action mechanisms of ginseng on stresses.
Objectives : This study aimed to evaluate the anti-stress effects of Guibi-tang and Yisiungyojae-dan on the contents of monoamines in the regional brain of mice immobilized stress. Methods : The experimental animals were immobilized in stress cylinder(height: 15cm, diameter: 3cm) for 15 minutes, and administered of Guibi-tang(6.0mg/10g) and Yishingyojae-dan(9.3mg/10g) water extracts for 7 days before stress. The monoamines contents were measured by HPLC method in various part(frontal cortex, hypothalamus, corpus striatum and hippocampus) of mice brain. Results: 1. In frontal cortex, the contents of norepinephrine were decreased with statistical significantly in Yishingyojae-dan administered group compared to control group. The contents of serotonin were decreased with statistical significance in all of the administered group compared to control group. 2. In hypothalamus, the contents of norepinephrine were decreased with statistical significantly in Yishingyojae-dan administered group compared to control group. The contents of serotonin were decreased with statistical significance in all of the administered group compared to control group. 3. In corpus striatum, the contents of dopamine were decreased with statistical significantly in Yishingyojae-dan administered group compared to control group. The contents of serotonin were decreased with statistical significance in all of the administered group compared to control group. 4. In hippocampus, the contents of serotonin decreased with statistical significantly in Yishingyojae-dan administered group compared to control group. Conclusions: This study shows that Guibitang and Yishingyojae-don are significantly effective on reducing and preventing stress in mice. In addition, Yishingyojae-dan is more clear effective than Guibi-tang on the monoamines change in the mice brain.
