Characterization of Ethanol Fermentation Using Alginate Immobilized Thermotolerant Yeast Cells

  • To enhance the hyperproductive and low energy-consuming ethanol fermentation rate, the thermotolerant yeast S. cerevisiae RA-74-2 cells were immobilized. An efficient immobilization condition was proved to be $1.5{\%}$ (w/v) alginate solution, neutral pH and 20 h activation of beads. The fermentation characteristics and stability at various temperatures were examined as compared with free S. cerevisiae RA-74-2 cells. The immobilized cells had excellent fermentation rate at the range of pH 3-7 at 30-$42^{\circ}C$ in 15-$20{\%}$ glucose media. When the seed volume was adjusted to 0.12 (v/v) (6ml bead/50 ml medium), $11{\%}$ (w/v) ethanol was produced during the first 34 hand $12.15{\%}$ (w/v) ethanol [$95{\%}$ (w/v) of theoretical yield] during the first 60 h in $25{\%}$ glucose medium. In repetitive fermentation using a 2 litre fermentor, 5.79-$7.27{\%}$ (w/v) ethanol [76-$95{\%}$ (w/v) of theoretical yield] was produced during the 40-55 h in $15{\%}$ glucose media. These data suggested the fact that alginate beads of thermotolerant S. cerevisiae RA-74-2 cells would contribute to economic and hyperproductive ethanol fermentation at high temperature.

Leaching and Acute Toxicity Test of Steel-making Slags for Media Contact Recycling (제강슬래그의 매체접촉형 재활용에 따른 중금속 용출특성 및 물벼룩 생태독성 평가)

  • Most of the slags generated from steel-making industry in Korea are recycled into media-contact aggregates such as fill and cover materials. For their use as media-contact aggregates, the slags must meet not only the waste quality criteria, but also the Daphnia magna acute toxicity test criteria. In this study, Korean Leaching Test ES 06150.e (Korea), Japanese Leaching Test JIS K 0058-1(Japan), Detuch Leaching Test DIN 19529 (Germany), Toxicity Characteristic Leaching Procedure (USA) were conducted for batch leaching test of slags from 6 Korean steel-making companies. In addition, Korean Standard up-flow percolation test (ES 06151.1) mimicking field conditions was conducted to assess the impact of the slag leachate on the surrounding environment indirectly. Heavy metals such as Cr6+ and Zn2+ were detected from both extractant and leachate samples, but all of them did not exceed waste quality criteria of each country. However, Daphnia magna acute toxicity tests using the leachate samples from up-flow percolation test with slag alone and slag/natural soil conditions exceeded ecotoxicity standard (TU=2) due to their high pH (11.3-12.5). After neutralizing the pH of the slag leachate to 6.5~8.5, the Daphnia magna mortality and immobilization were reduced to satisfy ecotoxicity standard. As the reducing pH of slag leachate would be extremely difficult, appropriate recycling management considering the physicochemical characteristics of he slags should be stuided further.

  • In this study plastic optical fibers(POFs) were considered as light-transmitting media and substrates for the potential use in photocatalytic environmental purification system. After the characteristics of POFs in terms of light transmittance and absorption were determined at the beginning, the detailed investigation was further performed through the photocatalytic degradation of trichloroethylene(TCE), iso-propanol and etc. with TiO$_2$-coated optical fiber reactor systems(POFR). It is concluded that the use of POfs is preferred to quartz optical fibers(QOFs) since the advantages such as ease of handling, lower cost, relatively reasonable light attenuation at the wavelength of near 400nm can be obtained. Various geometrical reactor shapes have been constructed and applied for the last one and half years. For the use of POF in water phase treatment, however, more detailed scientific and engineering aspects should be envisaged. This case requires a suitable mixture to obtain more stable and innocuous immobilization of photocatalyst on POF.

  • Abstract: In order to improve the system for biological nitrogen oxidizing process in sewage and wastewater, a bacterium having high abilities to oxidize of nitrogen was isolated from wastewater and polluted soils. The strain was identified to Bacillus sp. CH-N, based on the physiological and biochemical properties. Characteristics and oxidizing ability of both ammonia and nitrite were examined for the strain, Bacillus sp. CH-N. The strain showed the oxidizing rate about 80% to 90% on the sewage and wastewater after 48 h culture. The nitrogen oxidizing rate was increased in proportion to the initial concentration of glucose. The microorganism, Bacillus sp. CH-N cell immobilized on ceramic carrier were evaluated for the oxidation of ammonia in culture media.

  • Dissolved oxygen level of cell culture media has a critical effect on cellular metabolism, which governs specific productivity of recombinant proteins and mammalian cell growth However, in the cores of cell aggregates or cell-immobilized beads, oxygen level frequently goes below a critical level. Mammalian cells have a number of genes induced in the lower level of oxygen, and the genes contain a common cis-acting element (-RCGTG-), hypoxia response element (HRE). By binding of hypoxia inducible factor-l (HIF-I) to the HRE, promoters of hypoxia inducible genes are activated, which is a survival mechanism. In this work, to develop a CHO cell capable of producing recombinant proteins in immobilization and high density cell culture efficiently, mammalian expression vectors containing human tissue-type plasminogen activator (t-PA) gene controlled by HRE were constructed and stably transfected into the CHO cells. In $Ba^{2+}$ -alginate immobilization culture, CHO/pCl/dhfr/2HRE-t-PA cells produced 2 folds higher recombinant t-PA activity than CHO/pCl/dhfrlt-PA cells without $CoCl_2$ treatment. Furthermore, in repeated fed batch culture, productivity of t-PA in immobilized CHO/pCI/dhfr/2HRE-t-PA cells was 121 ng/ml/day, total production of 0.968 mg/day at 11 days culture while CHO/pCIIdhfrlt-PA cells was 22.8 ng/ml/day. All these results indicate that HRE is very useful for the enhancement of protein productivity in mammalian cell cultures.

Microbial Removal Using Layered Double Hydroxides and Iron (Hydr)oxides Immobilized on Granular Media

  • The objective of this study was to investigate microbial removal using layered double hydroxides (LDHs) and iron (hydr)oxides (IHs) immobilized onto granular media. Column experiments were performed using calcium alginate beads (CA beads), LDHs entrapped in CA beads (LDH beads), quartz sand (QS), iron hydroxide-coated sand (IHCS) and hematite-coated sand (HCS). Microbial breakthrough curves were obtained by monitoring the effluent, with the percentage of microbial removal and collector efficiency then quantified from these curves. The results showed that the LDH beads were ineffective for the removal of the negatively-charged microbes (27.7% at 1 mM solution), even though the positively-charged LDHs were contained on the beads. The above could be related to the immobilization method, where LDH powders were immobilized inside CA beads with nano-sized pores (about 10 nm); therefore, micro-sized microbes (E. coli = 1.21 ${\mu}m$) could not diffuse through the pores to come into contact with the LDHs in the beads, but adhere only to the exterior surface of the beads via polymeric interaction. IHCS was the most effective in the microbial removal (86.0% at 1 mM solution), which could be attributed to the iron hydroxide coated onto the exterior surface of QS had a positive surface charge and, therefore, effectively attracted the negatively-charged microbes via electrostatic interactions. Meanwhile, HCS was far less effective (35.6% at 1 mM solution) than IHCS because the hematite coated onto the external surface of QS is a crystallized iron oxide with a negative surface charge. This study has helped to improve our knowledge on the potential application of functional granular media for microbial removal.

Removal of Volatile Organic Compounds using Candida tropicalis Immobilized on Polymer Gel Media in an Airlift Loop Bioreactor (Candida tropicalis 포괄고정 담체를 적용한 Airlift Loop Bioreactor에서의 복합 휘발성유기화합물 제거)

  • This research was performed to improve removal efficiency of toluene and methyl ethyl ketone (MEK) using Candida tropicalis, one of the yeast species. An airlift loop bioreactor (ALB) was employed to enhance the capability of mass transfer for toluene and MEK from the gas phase to the liquid, microbial phase. Polymer gel media made from PAC, alginate and PEG was applied for the effective immobilization of the yeast strain on the polymer gel media. The experimental results indicated that the mass transfer coefficient of toluene without polymer gel media was 1.29 $min^{-1}$ at a gas retention time of 15 sec, whereas the KLa value for toluene was increased to 4.07 $min^{-1}$ by adding the media, confirming the enhanced mass transfer of volatile organic compounds between the gas and liquid phases. The removal efficiency of toluene and MEK by using yeast-immobilized polymer gel media in the ALB was greater than 80% at different pollutant loading rates (5, 10, 19 and 37 g/$m^3$/hr for toluene, 4.5, 8.9, 17.8 and 35.1 g/$m^3$/hr for MEK). In addition, an elimination capacity test conducted by changing inlet loading rates stepwise demonstrated that maximum elimination capacities for toluene and MEK were 70.4 and 56.4 g/$m^3$/hr, respectively.

  • The purpose of this study is to improve the system for biological nitrogen oxidizing process in sewage and wastewater. A bacterium having high abilities to oxidize of nitrogen was one of the possessed on Lab. The strain was identified to Bacillus sp. A8-8, based on the physiological and biochemical properties. And the strain has ability degradation crude oil. In comparison with oxidizing rates with changing initial pH and temperature, the strain Bacillus sp. A8-8 was nitrogen oxidizing ability and growth rate on the various of pH, temperature. oxidizing rates of the strain in sewage and wastewater were about 48% and 62%, respectively. The nitrogen oxidizing rate was increased in proportion to the initial concentration of glucose. The microorganism, Bacillus sp. A8-8, immobilized in ceramic carrier were evaluated for the oxidation of ammonia in culture media.

Optimization of Culture Medium for Novel Cell-Associated Tannase Production from Bacillus massiliensis Using Response Surface Methodology

  • Naturally immobilized tannase (tannin acyl hydrolase, E.C. 3.1.1.20) has many advantages, as it avoids the expensive and laborious operation of isolation, purification, and immobilization, plus it is highly stable in adverse pH and temperature. However, in the case of cell-associated enzymes, since the enzyme is associated with the biomass, separation of the pure biomass is necessary. However, tannic acid, a known inducer of tannase, forms insoluble complexes with media proteins, making it difficult to separate pure biomass. Therefore, this study optimizes the production of cell-associated tannase using a "protein-tannin complex" free media. An exploratory study was first conducted in shake-flasks to select the inducer, carbon source, and nitrogen sources. As a result it was found that gallic acid induces tannase synthesis, a tryptose broth gives higher biomass, and lactose supplementation is beneficial. The medium was then optimized using response surface methodology based on the full factorial central composite design in a 3 l bioreactor. A $2^3$ factorial design augmented by 7 axial points (${\alpha}$ = 1.682) and 2 replicates at the center point was implemented in 17 experiments. A mathematical model was also developed to show the effect of each medium component and their interactions on the production of cell-associated tannase. The validity of the proposed model was verified, and the optimized medium was shown to produce maximum cell-associated tannase activity of 9.65 U/l, which is 93.8% higher than the activity in the basal medium, after 12 h at pH 5.0, $30^{\circ}C$. The optimum medium consists of 38 g/l lactose, 50 g/l tryptose, and 2.8 g/l gallic acid.

  • This research is to investigate the reaction kinetics by air-lift bioreactor using calcium hydroxide, the neutralization agent and immobilization media, for removing ethylene glycol remained after chemical pretreatment. It was found that the optimum hydraulic retention time was obtained as 24.2hours at the optimum F/M ratio of 1.32kg-$TCOD_{Mn}$/day.kg-MLVSS, and then, infiuent $TCOD_{Mn}$ and MLVSS concentration were 3,290mg/l and 2,472mg/l, respectively. During the steady state, the kinetics constants such as maximum specific substrate removal rate, half saturation velocity coefficient, yield coefficient and endogenous respiration coefficient were estimated in the base of $TCOD_{Mn}$ as substrate concentration. And they were 1.47day$^{-1}$, 3.95mg/l, 0.391 and 0.092day$^{-1}$, respectively. And also, the oxgen use coefficients for cell synthesis, a', and energy of maintenance, b', were obtained as 0.4kg-O$_{2}$/kg-$TCOD_{Mn}$ and 0.056day$^{-1}$, at the steady state by the experimental result of oxygen uptake rate.
