  • We studied the inhibitory effect of the aqueous extract of Salvia plebeia (SPAE) on immediate hypersensitivity reactions. SPAE inhibited immediate hypersensitivity reaction induced by compound 48/80 in mice. When SPAE was employed in an immediate hypersensitivity reaction test, the plasma histamine levels were reduced in a dose-dependent manner. SPAE also inhibited the histamine release from rat peritoneal mast cells (RPMC) by compound 48/80. The level of cAMP in RPMC, when SPAE was added, significantly increased compared with that of a normal control. These results indicate that SPAE may be beneficial in the treatment of immediate hypersensitivity reactions.

  • We investigated the effect of aqueous extract of Terminalia chebula (Combretaceae)(TCAE) on the immediate hypersensitivity reaction in vivo and in vivo. TCAE (0.01 to 1 g/kg) dose-dependently inhibited compound 48/80 induced systemic anaphylaxis in mice. When TCAE was pretreated at concentrations ranging from 0.01 to 1 g/kg, the plasma histamine levels were reduced in a dose-dependent manner. TCAE (0.1 and 1 g/kg) significantly inhibited local immunoglobulin E (IgE)-mediated passive cutaneous anaphylactic reaction. TCAE (0.001 to 1 mg/ml) also dose-dependently inhibited the histamine release from rat peritoneal mast cells (RPMC) activated by compound 48/80 or anti-dinitrophenyl (DNP) IgE. TCAE (0.01 to 1 mg/ml) had a significant inhibitory effect on anti-DNP IgE-induced tumor necrosis $factor-{\alpha}$ production from RPMC. These results indicate that TCAE inhibits immediate hypersensitivity reaction in vivo and in vitro.

  • We studied the effect of aqueous extract of Magnolia officinalis bark (Magnoliaceae) (MOAE) on the immediate hypersensitivity reaction. MOAE (0.01 to 1g/kg) dose-dependently inhibited compound 48/80 induced systemic anaphylaxis in rats. MOAE (0.1 and 1g/kg) also significantly inhibited local immunoglobulin E (lgE)-mediated passive cutaneous anaphylactic reaction. When MOAE was pretreated at concentrations ranging from 0.01 to 1g/kg, the levels of plasma histamine were reduced in a dose-dependent manner. MOAE (0.001 to 1 mg/ml) dose-dependently inhibited the histamine release from rat peritoneal mast cells (RPMC) activated by compound 48/80 or anti-dinitrophenyl (DNP) Igl. The level of cyclic AMP (CAMP) in RPMC, when MOAE was added, significantly increased compared with that of the normal control. Moreover, MOAE (0.01 to 1 mg/ml) had a significant inhibitory effect on anti-DNP Igl-induced tumor necrosis factor-$\alpha$ production from RPMC. These results indicate that MOAE inhibits immediate hypersensitivity reaction in vivo and in vitro.

  • Spirodela polyrhiza(L.) Schleid (Lemnaceae) have been used as a traditional drug in treating urticaria and itching. However, the exact role of Spirodela polyrhiza in allergic reaction has not been clarified yet. Type 1 hypersensitivity (immediate hypersensitivity), popularly known as allergy, is a major clinical problem in humans. It has been found that the histamine release from mast cells is an essential step in the pathological process of immediate hypersensitivity. In the present study, the effect of aqueous extract of Spirodela polyrhiza (AESP) on immediate hypersensitivity was investigated. AESP inhibited the antigen-induced passive cutaneous anaphylaxis (PCA). AESP in vitro exhibited a dose-dependent inhibition of degranulation in RPMC stimulated by compound 48/80. AESP also suppressed the morphological changes and the increase of intracellular free calcium level induced by compound 48/80. These results suggest that inhibitory effect of AESP on immediate hypersensitivity may be mediated through the decrease of intracellular free calcium levels, and AESP importantly contributes to the treatment of anaphylaxis and may be useful for other allergic disease.

  • Object : We investigated the effect of Gamiokyaek-tang(GOYT) on immediate type hypersensitivity. Methods : We investigated anti-dinitrophenyl(DNP) IgE-mediated passive cutaneous anaphylaxis(PCA) and acetic acid-induced vascular permeability in rodents. Also we measured MTT assay, ${\beta}-hexosminidase$ activity and IL-4 from RBL-2H3 and nitric oxide from Raw264.7. Results : GOYT inhibited passive cutaneous anaphylaxis and acetic acid-induced vascular permeability by oral administration. All the concentrations of GOYT from 0.1 to 5mg/ml didn't have an effect on cell viability and cytotoxicity. In RBL-2H3, ${\beta}-hexosminidase$ release and IL-4 production were significantly reduced by 1, 2 and 5mg/ml of GOYT. In Raw264.7, nitric oxide level was decreased by 5mg/ml of GOYT. Conclusion : These results indicate that GOYT have inhibition effects on immediate type hypersensitivity.

  • The flowers of Lonicera japonica Thunb. (Caprifoliaceae) have been used as a traditional effective drug in treating bacillary dysentery, purulence. However, the exact role of Lonicera japonica in allergic reaction has not been clarified yet. Immediate hypersensitivity, popularly known as allergy, is a major clinical problem in humans. It has been found that the histamine release from mast cells is an essential step in the pathological process of immediate hypersensitivity. In this study, the effect of aqueous extract of Lonicera japonica (AELJ) on allergic reaction was investigated. • Objectives: A previous study showed that bee venom (BV) could cause anaphylaxis or other hypersensitivity reactions. Although hypersensitivity reactions due to sweet bee venom (SBV) have been reported, SBV has been reported to be associated with significantly reduced sensitization compared to BV. Although no systemic immediate hypersensitive response accompanied by abnormal vital signs has been reported with respect to SBV, we report a systemic immediate hypersensitive response that we experienced while trying to use SBV clinically. Methods: The patient had undergone BV treatment several times at other Oriental medicine clinics and had experienced no adverse reactions. She came to acupuncture & moxibustion department at Semyung university hospital of Oriental medicine (Je-cheon, Korea) complaining of facial hypoesthesia and was treated using SBV injections, her first SBV treatment. SBV, 0.05 cc, was injected at each of 8 acupoints, for a total of 0.40 cc: Jichang (ST4), Daeyeong (ST5), Hyeopgeo (ST6), Hagwan (ST7), Yepung (TE17), Imun (TE21), Cheonghoe (GB2), and Gwallyeo (SI18). Results: The patient showed systemic immediate hypersensitive reactions. The main symptoms were abdominal pain, nausea and perspiration, but common symptoms associated with hypersensitivity, such as edema, were mild. Abdominal pain was the most long-lasting symptom and was accompanied by nausea. Her body temperature decreased due to sweating. Her diastolic blood pressure could not be measured on three occasions. She remained alert, though the symptoms persisted. The following treatments were conducted in sequence; intramuscular epinephrine, 1 mg/mL, injection, intramuscular dexamethasone, 5 mg/mL, injection, intramuscular buscopan, 20 mg/mL, injection, oxygen ($O_2$) inhalation therapy, 1 L/minutes, via a nasal prong, and intravascular injection of normal saline, 1 L. After 12 hours of treatment, the symptoms had completely disappeared. Conclusion: This case shows that the use of SBV does not completely eliminate the possibility of hypersensitivity and that patients who received BV treatment before may also be sensitized to SBV. Thus, a skin test should be given prior to using SBV.

  • Cloves are the dried flower buds of Syzygium aromaticum (L.) Mere et Perry (Myrtaceae). They have been successfully used for the management of various allergic disorders by oral administration in Korea. In this study, the author investigated the effect of an aqueous extract of Syzygium aromaticum on immediate-type hypersensitivity by anal administration. Anal administration of Syzygium aromaticum showed a marked inhibition rate in systemic hypersensitivity with a dose of 1 mg/kg 1 hr before intraperitoneal injection of compound 48/80. Anal administration of Syzygium aromaticum significantly reduced plasma histamine contents induced by compound 48/80. Anal administration of Syzygium aromaticum (1 mg/kg) also inhibited to $61.4\%$ (P<0.01) local a1lergic reaction activated by anti-dinitrophenyl (DNP) IgE. In addition, Syzygium aromaticum dose-dependently inhibited the histamine release from the peritoneal mast cells by compound 48/80 or anti-DNP IgE. When Syzygium aromaticum was added, the level of cAMP in peritoneal mast cells transiently and significantly increased about 47-fold at 10 second compared with that of basal cells. These results provide evidence that anal therapy of Syzygium aromaticum may be beneficial in the treatment of systemic and local immediate-type hypersensitivity by inhibition of histamine release from mast cells in vivo and in vitro.

  • Anal therapy is another way of taking medicine It is a traditional pathway but not available in common situation. Nevertheless. It has many benefit and usefulness, it has not treated so much. The dried immature fruit of Poncirus trifoliata L(Ji-Sil) is widely used to treat urticaria, itch and indigestion in traditional Korean Medicine. So this study was carried out to examine the effect of an aqueous extract from immature fruit of Poncirus trifoliate L. on immediate-type allergic reaction by anal administration. Anal administration of Poncirus trifoliata L. showed a marked inhibition rate in systemic immediate-type allergic reaction with a dose of 0.1 - 1 g/kg 1 hr before intraperitoneal injection of compound 48/80. Anal administration of Poncirus tnjoliata L. (0.1 -1 g/kg) significant1y reduced plasma histamine contents induced by compound 48/80. Anal administration of Poncirus trifoliata L. (10 g/kg) also inhibited to $44.6\%$ (P<0.01) local allergic reaction activated by anti-dinitrophenyl (DNP) IgE. In addition, Syzygium aromaticum dose-dependently inhibited the histamine release from the peritoneal mast cells by compound 48/80 or anti-DNP IgE. These results provide evidence that anal therapy of Poncirus trifoliata L. may be beneficial in the treatment of systemic and local immediate-type hypersensitivity by inhibition of histamine release from mast cells in vivo and in vitro.
