Effective plant regeneration from immature cotyledons of soybean [Glycine max (L.) Merr.] cultivars was achieved via somatic embryogenesis. Somatic embryogenesis was performed with the cotyledons of immature embryos 14-20 d after flowering. Immature cotyledons of cv. Whangkeum were placed abaxial or adaxial side down on modified MS medium containing 20mg/L 2,4-D. The greatest number of somatic embryos, 1.2 per cotyledon, was produced from those of 4.0-4.9 mm in length which had been placed abaxial side down. Among cvs. Pecking, Whangkeum and Baekwoon, Pecking had the highest embryo induction efficiency with 4.3 somatic embryos per cotyledon in 20mg/L 2,4-D treatment and with 1.0 embryo per cotyledon in 8mg/L NAA treatment. Germinable globular somatic embryos were induced with the highest efficiency, 27.6%, in 20mg/L 2,4-D and were proliferated efficiently on liquid medium containing 10mg/L 2,4-D. The globular somatic embryos developed into germinable mature somatic embryos on medium containing 10 $\mu$M CoCl2, 9% sucrose, and 0.5% activated charcoal. These mature somatic embryos germinated on hormone-free mediu. After transfer to the soil, regenerated plants with seeds were obtained.

To enhance in vitro plantlet regeneration efficiency of soybean through embryogenesis, the culture conditions such as material part and size of immature seed, 2,4-D, pH and solidifying agents for somatic embryogenesis were investigated. Somatic embryogenesis was induced from the immature embryo, immature cotyledon and embryonic axis explants of the immature seed on MS medium supplemented with 2.0 mg/L 2,4-D. The highest rate (up to 22.9%) of somatic embryogenesis was obtained from the immature cotyledon, following embryonic axis and the immature embryo. The rate varied with the developmental stages of seed. The maximum rate (25.4%) of embryogenesis was obtained from 3-4 mm length of the seed (after 25 days of flowering). The optimum concentration of 2,4-D for embryogenesis was 10 mg/L. The optimum pH was at 5.8 and solidifying agent for medium was better with 0.4% gelrite than with agar. For rapid multiplication of shoot tips from the germinating somatic embryos, they were cultured on MS medium containing 2 mg/L indole-3-butyyic acid (IBA) and 1 mg/L 6-benzyladenine (BA). After then somatic embryos with one and three cotyledons were transferred to the growth regulator free medium. The medium exhibited the higher rate (ca. 50%) of development than the multiplication medium.

Somatic embryos were induced from immature cotyledons and cultured on a MS medium containing 40mg/L 2,4-D. The maximum induction of embryos was obtained from immature cotyledons in a size of 3-4mm, and the highest frequency was obtained in the induction medium at pH 7.0. For embryo development, embryogenic tissues were transferred to a MSM6AC and MSM6 media. Developing embryos were placed at 27$^{\circ}C$with dim light (20$\mu$$molm^{-2}$$s^{-1}$) provided by cool fluorescent tubes (3-D wavelength light is better than standard light). Somatic embryos were clearly developed from globular stage to cotyledonary stages. The color of embryo may be a useful parameter for estimation of embryo quality. When the embryo becomes mature, embryo will be ready for desiccation in order to induce roots and shoots of embryos.
Excised cotyledon segments of ginseng zygotic embryos at various developmental stages were cultured on MS basal medium from which somatic embryos were directly induced. The frequency of somatic embryo formation on the segments declined with the advancing zygotic embryo maturity. All of the cells in the cotyledons of immature zygotic embryos were smaller and more densely cytoplasmic than those in mature embryos. Histological examinations revealed that the poly-somatic embryos formed on immature embryos were of multi-cell originand derived from the epidermal and subepidermal cell layers. However, in the cotyledon of germinating zygotic embryos, only theepidermal cells were densely cytoplasmic and singularly competent to develop into somatic embryos resulting into single embryos at a frequency of 100%.
Immature embryos of Glycine max L. was cultured on Murashige and Skoog's (MS) medium supplemented with 1 mg/L 2,4-dichlorophenoxy acetic acid (2,4-D). After 6 to 8 weeks of culture, immature embryos produced somatic embryos. Of somatic embryos, two cotyledonary embryo (14%), one cotyledonary embryo (37%), fused cotyledonary embryo (43%), and stunted globular embryos (6%) were observed. The procambial strand of cotyledons originated from circular procambial tissues of lower hypocotyl. The circular procambial tissues were independently divided into one or two procambial strand at the edge of cotyledonary-node, and then connected to each cotyledon to form somatic embryos with one or two cotyledons. When cotyledon was a fused type, the circular procambial strand in lower hypocotyl was continuously connected to the cotyledon. Also, somatic embryos with two cotyledons developed a functional shoot apex with the tunica-corpus structure. In contrast, somatic embryos with one or fused cotyledon formed an abnormal shoot apex without the tunica-corpus structure or with non-dome shape in the inter-cotyledonary area. These results indicated that the variation of cotyledon in somatic embryos is closely related to procambial differentiation and shoot apical meristem development.

Seoritae with green cotyledons refers to Korean native black beans harvested when the frost falls because the maturity is late. It is one of the beans preferred by consumers because of its softness and high sugar content. Because of late maturity, if the agricultural work is delayed by weather or agricultural schedule in green cotyledon black soybean (Seoritae), it affects seriously harvest and yield of soybeans. The aim of this study was to investigate the marginal seeding period on June 30, July 10 and July 20 in Cheongju and Jecheon area in Korea to produce stable soybean yield. The yields of green cotyledon black soybean as seeding date in Cheongju area are as follows: Seolitae (Yeoncheon), Seoltae (Goesan) and Seoritae (Gogseong), which increased by 23%, 56%, 23% and 40%, respectively, compare to July 10th to June 30th. As soybean sowing is delayed, the quality of soybean seeds has decreased due to the increase of immature seeds and fungal damaged seeds. The contents of anthocyanin in Cheongju area a functional substance of soybeans, was high on July 20 for Heukcheong, on June 30 for Seolitae (Yeoncheon), on July 10 for Seoritae (Goesan). The yields of Heukcheong and Seolitae (Yeoncheon) in Jecheon area were increased by 5% and 17%, respectively, compare to July 10th to June 30th, while Seolite (Goesan) and Seolite (Gogseong) were high in yields on June 30th. Similarly in Cheongju area, as the sowing period is delayed, the number of immature and mold damaged seeds in Jecheon area increased. The contents of anthocyanin in Jecheon area was high on July 10 for Heukcheong, on June 30 for Seolitae (Yeoncheon) and Seolitae (Goesan). From the above results, Sowing marginal date of green cotyledon black soybeans (Seolitae) in Cheongju area increased 30% in sowing on July 10 and increased 2% in sowing on July 20 compared to June 30. And Sowing marginal date of green cotyledon black soybeans (Seolitae) in Jecheon area increased 2% in July 10 compared to June 30, and the yield decreased rapidly on July 20. We have identified the seeding time limit of green cotyledon black bean in Chungbuk province. It will be possible to provide a variety of crop selection after double cropping of farmers. And by knowing the yield and seed quality of soybean according to sowing date, farmers will observe appropriate sowing period of soybeans for high quality. From the viewpoint of consumers, functional substances of Seoritae will meet the desire for health.
Embryogenic callus was induced from the cultures of immature embryos of Camellia japonica L. on Murashige & Skoog's (MS) solid medium supplemented with 1 mg/L 2,4-dichlorophenoxy acetic acid (2,4-D), and then the embryogenic callus was proliferated on same medium for 4 weeks over. The embryogenic callus was sub-cultured on MS basal medium without 2,4-D to produce coyledonary stage of somatic embryo. The frequency (%) of somatic embryogenesis was 25.1%, and the majority of somatic embryos formed had a abnormal morphology with cupshaped cotyledon (48.3%), one cotyledon (12.6%), three cotyledons (9.4%), four cotyledons (1.9%), whereas was only normal morphology with two cotyledon (27.5%). When the somatic embryos with normal or abnormal cotyledons transfer to MS basal medium or $\frac{1}{2}$ MS medium with/or without plant growth regulators ($GA_3$, IBA) for regeneration, the frequency (%) of two-cotyledon embryos regenerated into plantlets was higher 11.1% than one cotyledon (0.0~8.3 %), three cotyledons (0.0~5.8%), four cotyledons (0.0%), cup-shaped (0.3~4.2%). These results demonstrated that the anomalous cotyledons of somatic embryos could caused to decrease the rate of plant regeneration.

As the zygotic embryo of North American ginseng (Panax quinquefolius L.) matured during stratification over 203 days it grew from 0.75 to 5.2 mm. Embryo excision and culturing on media containing different concentrations of two growth regulators, gibberellic acid ($GA_3$, 1 to 10 ${\mu}M$) and benzyladenine (BA, 1 to 5 ${\mu}M$), during stratification, showed that shoot and root number and the shoot, root and cotyledon length increased with increased stratification time. Gibberellic acid was the more effective growth regulator for increasing shoot and root number and shoot, root and cotyledon lengths. Immature embryos (stratified for up to 63 days) needed growth regulators for further development. Cultures on $GA_3$ at the last culture date (stratified for 203 days) when embryos were mature, produced multiple shoots but there was no effect of $GA_3$ concentration. Benzyladenine inhibited shoot and root growth regardless of embryo stratification. Growth regulators had little effect on cotyledon length of mature embryos. Embryos cultured on $GA_3$ combined with BA were green on all culture dates whereas greening in the control and BA treatments increased with culture date. The BA treatments induced 100% swelling of the embryos on the final culture date while in the control and $GA_3$ treatments there was no swelling. There was little or no curling in the control and BA treatments and a linear decrease in curling with culture date in the $GA_3$ and $GA_3$ + BA treatments.
Immature zygotic embryos of five Korean soybean cultivars cultured on Murashige and Skoog's (MS) medium supplemented with various concentrations of 2,4-Dichlorophenoxyacetic acid (2,4-D) produced somatic embryos without forming an intervening callus. The highest frequency (up to 90%) of somatic embryo formation was obtained when zygotic embryos were cultlued on medium containing 1 to 2 mga 2, 0-D in four cultivars. However the frequency was highly variable to the cultivars. Transversely sliced primary somatic embryo halves were also capable of forming secondary embryos at frequencies of up to 70% when cultured on medium containing 0.1 to 1 mg/L 2,4-D. Somatic embryos formed on zygotic embryos cultured on medium containing 0.1 to 0.2 mg/L 2,4-D had two cotyledons more frequently than one horn-type cotyledon and those on medium containing 0.5 to 4mg/L 2,4-LD had a horm-type cotyledon at a prominently higher freequency. However somatic embryos on medium containing 10mg/L or higher concentrations of 2,4-D were usually shunted at the globular stage even after transfer to medium containing lower concentrations of 2,4-D or other growth regulators. non somatic embryos with one or two cotyledons or a hem-type cotyledon were transferred to medium containing $GA_3$, those with two cotyledons converted to plantlets at a higher frequency (25%) than the others.
Immature zygotic embryos from immature seeds of Wasabia japonica (cv Dalma) were isolated and cultured on modified MS medium supplemented with 2,4-D, IAA, and BA. Immature zygotic embryos were classified into torpedo shape and cotyledon stage.
