어육단백질의 사후분해 1. 단백질분해의 본질과 세균기여 (Postmortem Degradation of Fish Muscle Proteins 1. Nature of proteolysis and bacterial contribution)

  • 백색 육어류의 사후 기간중에 일어나는 단백질 분해의 본질을 규명하려는 방도로써 가자미류(English sole, Paraphyrus vetulus)와 볼락류(rockfish, sebastodes caurinus)의 어육을 시료로 하여 맨 먼저 가자미 전 어체를 빙장하는 동안 취하여 어육 slurry를 만든 후 $20^{\circ}C$에서 유지시키는 동안의 단백질 분해율을 측정하였다. 근육자가 소화효소 cathepsin에 의한 단백질 분해도를 규명키 위하며 어육 slurry 일부는 0.5Mrad 감마선조사로써 무균화시키면서 세균 면식활동에서 오는 단백질 분해가능성을 제거시켰다. 어육을 기계적으로 분쇄하여 얻은 slurry를 무균화한 후 $20^{\circ}C$ 수조에서 17시간 동만 유지시켰는데도 불구하고 단백질 분해가 전혀 검측되지 안했음은 cathepsin 작용이 없었음을 뜻한다. 이와 반면에 비조사구 slurry의 경우, $20^{\circ}C$ 수조에서의 유지기간중 총함수의 증가에 따라 약간의 단백질 분해가 이뤄줬으나 slurry로 만들어지기전의 어체의 부패도가 후기에 접어 들어 총균수가 최고선에 도달하였을 때 비로소 현저한 단백질 분해를 가져 왔다. 이는 부패초기에는 부패세균에 의한 단백질 분해효소의 합성이 거의 없음을 시사한다. 이어서 볼락어육을 무균적으로 취하여 그 일부는 단백질 분해력이 강한 Pseudomonad균을 접종하고, 나머지는 각각 0, 0.5, 그리고 2.0Mrad의 감마선에 조사한 후 $0^{\circ}\~2^{\circ}C$에서의 저장기간중에 일어나는 단백질 분해과정을 규명하였다. 세균번식이 없는 무균어육의 염용선(0.5M KCl) 총질소질과 $70\%$ 에타놀 용해성 아미노태 질소량은 저장기간중 약간 감소된 반면 Pseudomonad군을 접종한 어육의 질소질의 증가는 총균수 증가에 평행하였다. 이로써 어류의 사후 기간중에 일어나는 단백질 분해는 정상적인 어류부패과정의 일부분이기는 하나 부착세균의 번식이 진행되어 총균수가 최고선에 도달할 때까지 단백질 분해는 지연되며 최소한 백색 어육에 관한한 사후기간중 cathepsin에 의한 단백질 분해에의 기여도는 거의 무시될 수 있음이 확인되었다.

  • 방어로부터 추출한 근원섬유 현탁액을 $0^{\circ}C$(빙장), $-3.5^{\circ}C$(PF저장), $-20^{\circ}C$(동결저장)에 저장하면서 Ca-과 Mg-ATPase활성을 측정하여 근육단백질의 변성을 조사하였으며 일부는 생선을 각 온도에 일정기간 저장한 후 근원섬유를 추출하여 단백질 변성정도를 비교한 결과 추출한 근원섬유를 저장하였을 때 빙장 및 PF저장한 시료의 ATPase활성은 비슷한 수준이었으나 동결 저장한 시료에서는 현저히 낮았다. 생선을 각 온도에 1주일간 저장한 후 추출한 근원섬유의 소활성은 근원섬유를 미리 추출하여 같은 기간동안 저장한 경우에 비하여 빙장과 PF저장에서는 1.2-1.8배, 동결저장에서는 2.5-3배 정도 높은 수준을 유지하고 있었다. 근원섬유를 각 온도에 저장하였을 때의 변성속도는 Ca-의 경우가 Mg-ATPase에 비 해 2-3배 빨랐으나 생선으로 저장한 경우에는 Ca-과 Mg-ATPase간에 큰 차이를 나타내지 않았다.

Zoonotic Trematode Metacercariae in Fish from Yangon, Myanmar and Their Adults Recovered from Experimental Animals

  • A survey was performed to investigate the infection status of zoonotic trematode (ZT) metacercariae in fish from a local market in Yangon City, Myanmar. A total of 264 fish (12 species) were collected through 4 times from December 2013 to June 2015. All collected fish were transferred to our laboratory on ice and examined by the artificial digestion method. More than 7 species of ZT metacercariae, i.e., Haplorchis taichui, H. pumilio, H. yokogawai, Centrocestus spp., Stellantchasmus falcatus, Pygidiopsis cambodiensis, and Procerovum sp. were detected. Metacercariae of H. taichui were collected in 58 (42.3%) out of 137 fish (5 species), and their average density was 42.9 per fish infected. Metacercariae of H. pumilio were detected in 96 (49.0%) out of 196 fish (9 species), and their average density was 23.6 per fish infected. H. yokogawai metacercariae were found in 40 (50.0%) out of 80 fish (5 species), and Centrocestus spp. metacercariae in 91 (50.8%) out of 179 fish (8 species), and their densities were 306 and 25.8 per fish infected, respectively. Metacercariae of S. falcatus and P. cambodiensis were detected only in mullets, Chelon macrolepis. A total of 280 Procerovum sp. metacercariae were found in 6 out of 12 climbing perch, Anabas testudineus. Morphological characteristics of adult flukes recovered from experimental animals were described. It has been first confirmed that fish from Yangon, Myanmar are commonly infected with various species of ZT metacercariae.

  • Organisms living in the Arctic and Antarctic regions are exposed to strong constraints, of which temperature is a driving factor. Evolution has led to special adaptations, some with important implications at the biochemical, physiological, and molecular levels. The northern and southern polar oceans have very different characteristics. Tectonic and oceanographic events have played a key role in delimiting the two polar ecosystems and influencing evolution. Antarctica has been isolated and cold longer than the Arctic; its ice sheet developed at least 10 million years earlier. As an intermediate system, the Arctic is a connection between the more extreme, simpler Antarctic system and the very complex temperate and tropical systems. By studying the molecular bases of cold adaptation in polar fish, and taking advantage of the information available on hemoglobin structure and function, we analysed the evolutionary history of the ${\alpha}\;and\;{\beta}globins$ of Antarctic and Arctic hemoglobin using the molecular clock hypothesis as a basis for reconstructing the phylogenetic relationships among species.

Zoonotic Trematode Metacercariae in Fish from Phnom Penh and Pursat, Cambodia

  • A survey was performed to investigate the infection status of freshwater fish with zoonotic trematode metacercariae in Phnom Penh and Pursat Province, Cambodia. All collected fish with ice were transferred to our laboratory and examined using the artificial digestion method. In fish from Phnom Penh, 2 kinds of metacercariae (Opisthorchis viverrini and Haplorchis yokogawai) were detected. O. viverrini metacercariae were positive in 37 (50.0%) of 74 fish in 11 species (average no. metacercariae/fish, 18.6). H. yokogawai metacercariae were detected in 23 (57.5%) of 40 fish in 5 species (average no. metacercariae/fish, 21.0). In fish from Pursat Province, 5 kinds of metacercariae (O. viverrini, H. yokogawai, Haplorchis pumilio, Centrocestus formosanus, and Procerovum sp.) were detected; O. viverrini metacercariae (n=3) in 2 fish species (Henicorhynchus lineatus and Puntioplites falcifer), H. yokogawai metacercariae (n=51) in 1 species (P. falcifer), H. pumilio metacercariae (n=476) in 2 species (H. lineatus and Pristolepis fasciata), C. formosanus metacercariae (n=1) in 1 species (H. lineatus), and Procerovum sp. metacercariae (n=63) in 1 species (Anabas testudineus). From the above results, it has been confirmed that various freshwater fish play the role of a second intermediate host for zoonotic trematodes (O. viverrini, H. yokogawai, H. pumilio, C. formosanus, and Procerovum sp.) in Cambodia.

Larval Gnathostomes and Zoonotic Trematode Metacercariae in Fish from a Local Market in Yangon City, Myanmar

  • A survey was performed to investigate the infection status of zoonotic helminth larvae in fish from a local market of North Dagon District in Yangon City, Myanmar. A total of 486 fish in 13 species were collected 8 times from December 2015 to December 2019. All fish were transported under ice to a laboratory in Korea and examined for helminth larvae using artificial digestion method. Larval gnathostomes and metacercariae of more than 8 zoonotic trematode species, i.e., Opisthorchis viverrini, Haplorchis taichui, H. pumilio, H. yokogawai, Centrocestus spp., Stellantchasmus falcatus, Pygidiopsis cambodiensis, and Procerovum sp., were detected. Larval gnathostomes were found in 58 (16.0%) out of 362 fish of 6 species, with mean intensity of 2.8 per fish infected. Metacercariae of O. viverrini were detected in 10 (2.9%) out of 349 fish of 5 species, with mean intensity of 16.9 per fish infected. Metacercarial prevalences of 4 intestinal flukes, H. taichui, H. pumilio, H. yokogawai, and Centrocestus spp., were 16.8%, 26.0%, 12.5%, and 15.0% in the positive fish species, respectively, and mean metacercarial intensity was 63.3, 26.8, 86.2, and 8.7 per fish infected. Metacercariae of S. falcatus and P. cambodiensis were detected only from the mullet, Chelon macrolepis. Metacercariae of Procerovum sp. were found in Channa striata and Anabas testudineus. Collectively, it was confirmed that the fish were infected with gnathostome larvae and metacercariae of O. viverrini and intestinal flukes in Yangon City, Myanmar.

기존 어도의 개선에 관한 조사 연구 - 우리나라 동해안 하류 하천 내 어도를 중심으로 - (A Study on Improvement of Existing Fishways - Focusing on Fishways Located at Downstream of Eastern Coast Rivers in Korea -)

  • This study aims at examining the structural and managemental problems of the existing fishways which affect the migrations of fishes, and at suggesting alternatives to improve the fishways' efficiency. The study focuses on the fishways constructed in the diversion weirs in the lower reaches of 41 rivers(2 national rivers and 39 rural rivers) in the eastern coast of Korea. Most of water-using facilities in Korean rivers are diversion weirs, and fishways are established as subsidiary facilities of most of the weirs. Among the 58 or so fishways examined in this study, only a few are doing their proper function, and most of them need improvement. This study aims at strengthening those fishways function and increasing their economic efficiency and migration efficiency. Based on the results of this study, the followings are suggested. In case the fishway sticks out downstream from the apron, a subsidiary fishway diverging from the main is suggested to attract fish. The entrance to the subsidiary should head below the raised part of the apron and a pool should be constructed in the connecting part with the main. To attract fish and let the upstream migrators rest for a while, an entrance pool should be constructed in the fishway. Most of the examined fishways have low side walls, which allow the upstreamers to jump out of the fishway. Those side walls are suggested to be raised above the jumping height of target fishes. The exit parts of most fishways are too shallow, or have no grass to protect the fish from the birds. Protective facilities are suggested to keep the birds from entering the fishway. By the time Ice-harbor type and Vertical-slot type fishways are scientifically proved to be defective, the fishway should not diverge from the standard design.

Cloning, expression, and activity of type IV antifreeze protein from cultured subtropical olive flounder (Paralichthys olivaceus)

  • Antifreeze proteins (AFPs) lower the freezing point but not the melting point of aqueous solutions by inhibiting the growth of ice crystals via an adsorption-inhibition mechanism. However, the function of type IV AFP (AFP IV) is questionable, as its antifreeze activity is on the verge of detectable limits, its physiological concentration in adult fish blood is too low to function as a biological antifreeze, and its homologues are present even in fish from tropic oceans as well as freshwater. Therefore, we speculated that AFP IV may have gained antifreeze activity not by selective pressure but by chance. To test this hypothesis, we cloned, expressed, and assayed AFP IV from cultured subtropical olive flounder (Paralichthys olivaceus), which do not require antifreeze protein for survival. Among the identified expressed sequence tags of the flounder liver sample, a 5'-deleted complementary DNA (cDNA) sequence similar to the afp4 gene of the longhorn sculpin was identified, and its full-length cDNA and genome structure were examined. The deduced amino acid sequence of flounder AFP IV shared 55, 53, 52, and 49 % identity with those of Pleuragramma antarcticum, Myoxocephalus octodecemspinosus, Myoxocephalus scorpius, and Notothenia coriiceps, respectively. Furthermore, the genomic structure of this gene was conserved with those of other known AFP IVs. Notably, the recombinant AFP IV showed a weak but distinct thermal hysteresis of $0.07{\pm}0.01^{\circ}C$ at the concentration of 0.5 mg/mL, and ice crystals in an AFP IV solution grew star-shaped, which are very similar to those obtained from other polar AFP IVs. Taken together, our results do not support the hypothesis of evolution of AFP IV by selective pressure, suggesting that the antifreeze activity of AFP IV may have been gained by chance.

빙어의 난발생과정과 자어의 형태발달 (Eggs Development and Larval Development of the Ice Fish, Hypomesus transpacificus nipponensis McAllister)

  • 1990년 3월, 경상북도 영덕군 강구면에 위치한 오십천과 경상남도 밀양시 밀양강에서 채집하여 인공수정한 난을 실험실로 운반하여 난발생과정과 부화자어의 성장에 따른 형태변화를 관찰한 결과는 다음과 같다. 1. 빙어의 수정난은 구형의 침성부착난으로 무색 투명하며, 난경은 $0.85\~1.05mm$ (평균 0.97mm, n=50)이다. 2. 부화에 소요된 시간은 평균 $16.5^{\circ}C$에서 약 170시간만에 부화한다. 3. 부화직후의 자어는 전장 $3.85\~4.25mm$ (평균 4.05 mm, n=10)로, 아직 입과 항문이 열려 있지 않고, 눈에 색소포가 착색되어 있으며, 근절수는 $52\~54$개이다. 4. 부화 $4\~5$일째의 자어는 전장이 $5.20\~5.65mm$ (평균 5.37mm, n=10)로 난황은 거의 흡수되고, 눈의 색소는 더욱 짙어진다. 5. 부화 20일째의 후기자어는 전장이 $8.20\~8.55\;mm$(평균 8.38mm, n=10)로 척색말단이 $45^{\circ}$ 각도로 위로 굽어진다. 6. 부화 25일째의 후기자어는 전장이 $9.15\~9.75mm$ (평균 9.63mm, n=10)로 등지느러미와 뒷지느러미에 각각 융기하기 시작하여 원기가 나타난다.

Trap을 이용한 공주보 아이스하버식 어도의 효과분석 (Efficiency Analysis of the Ice Harbor Type Fishway Installed at the Gongju Weir on the Geum River using Traps)

  • 댐과 보의 건설로 인한 하천의 단절 및 어류의 이동 문제를 해결하기 위해 다수의 어도를 건설하고 있다. 하지만 이에 대한 어도 효율성 평가나 적합성에 대한 연구는 상당히 제한적으로 이루어지고 있다. 따라서 본 연구는 2012년 6월부터 10월까지 매달 1회씩 trap을 이용하여 공주보에 설치된 아이스하버식 어도에서 시기별, 시간대별, 어도 위치별 어도 이용 차이를 분석하였다. 연구 결과, 6~8월보다 9~10월에 어도를 이용하는 종 및 개체가 감소하였다. 그리고 어도 좌안 측면이 통계적으로 유의하지 않지만 많은 종 및 개체가 출현하였고, 또한 다른 위치보다 다양한 크기의 개체들이 출현하였다. 상류수위와 채집된 소상 어류 개체수는 양의 상관성을 나타내고 있었다. 그러나 일평균 상류수위가 증가할수록 전장 70 mm 이하 소형개체들의 출현율이 감소하는 경향이 나타났다. 어도 내 시간대별 이동은 종별 생태적인 특성에 따라 차이를 보였다. 전체적으로 크기가 작은 개체부터 큰 개체까지 다양한 크기의 어류가 어도를 이용했다. 이러한 결과는 국내에 건설된 아이스하버식 어도관리 및 평가에 도움이 될 것으로 판단된다.
