Inhibitory Effect of Enteromorpha linza on the Melanogenesis in B16 Melanoma Cells (파래 추출물의 멜라닌 생성 억제 효과)

  • Melanin is a polymer of phenol which produces hyperpigmentation in the skin. Melanin synthesis is catalyzed by the enzyme tyrosinase. To investigate the whitening activity of the fractions from the ethanol extract of Enteromorpha linza, we studied the inhibitory effect on the tyrosinase activity and melanogenesis in the B16/F1 melanoma cells. The inhibition ratio of tyrosinase activity of ethylacetate fraction from E. linza was as strong as that of kojic acid, a positive control. Also, the melanin production was significantly inhibited by the ethylacetate fraction in a dose dependent manner. The ethylacetate fraction showed the highest activity in the fractions and as strong activity as that of arbutin, a positive control. From these results, we suggest that the E. linza extract might be able to apply to cosmetic and medical fields.

  • Mammalian tyrosinase plays an important role in the process of melanin polymer biosynthesis by catalyzing the hydroxylation of tyrosine to 3,4-dihydroxyphenylalanine(DOPA) and the oxidation of DOPA to dopaquinone. These processes are major determinant of human skin color and involved in localized hyperpigmentation. Therefore, the enzyme inhibitors have been of great concern as skin-whitening cosmetics. Methanol extracts of 174 oriental herbs were screened for the mushroom tyrosinase inhibitory activity.

Primary hypothyroidism in Jindo dog (진도견에서 발생한 후천성 갑상선 기능저하증)

  • A four-year-old female Jindo dog was admitted to the hospital because of bilaterally symmetrical alopecia and inappentence. Systemic abnomalities detected on physical examination were obesity, bradycardia, exercise intolerance, mental dullness and hypothermia. Dermatologically symmetrical alopecia on the back, trunk and dorsal tail, and ventral hyperpigmentation were observed. Also face was very tragic. The level of basal $T_4$ was 0.01ug/dl. In TSH stimulation test $fT_4$ value was 0.08ng/dl. After treatment was initiated with L-thyroxine the dog's general condition was improved at 4 weeks.

  • A female mixed miniature pinscher dog, about 5 years old, 4 kg, was referred to veterinary teaching hospital of Chungnam National University with chief complaint of excessive hemorrhagic vaginal discharge for a month, partial alopecia and hyperpigmentation. There are no specific change with CBC, blood chemistry and X-ray findings but we figured out high estrogen concentration with hormone analysis and detected a majority of the superficial epithelial cells with pyknotic nuclei by cytologic examination of vaginal smear preparation and multiple ovarian cysts on both ovaries by ultrasonography. Based on the clinical signs, hormone analysis, vaginal smear preparation and ultrasonography, the authors diagnosed this case as hyperestrogenism with ovarian cysts.

  • A 5-year-old intact male Yorkshire terrier dog was referred with a 1-week history of blindness. In both eyes, menace responses and pupillary light reflexes were absent. The indirect ophthalmoloscopy revealed multiple small gray-white and discoloration lesion in mid-nontapetum area in both eyes. Electroretinograms showed diminished amplitude of a- and b-waves. Multifocal serous retinal detachments were detected with optical coherence tomography (OCT) over the small gray-white and discoloration lesion in the fundus. After one week of treatments, the indirect ophthalmoloscopy showed that irregular patches and hyperpigmentation in the center of hyperreflectivity area. Reattachment and retinal thinning were observed by the OCT. The results indicate that OCT scanning is considered to be a useful method for retinal evaluation in dogs with chorioretinitis.

Neonatal Adrenoleukodystrophy Presenting with Neonatal Seizure (경련을 동반한 신생아 부신백질이영양증)

  • Disorders resulting from defects in peroxisomal biogenesis include Zellweger syndrome, neonatal adrenoleukodystrophy, and infantile Refsum disease. The three diseases are now considered as a continuum of clinical features. Neonatal adrenoleukodystrophy is intermediate between Zellweger syndrome and infantile Refsum disease in severity, and is characterized by profound hypotonia, intractable seizures and premature death. We report a cases of neonatal adrenoleukodystrophy presenting with neonatal seizure and hypotonia. At the age of 43 months, she had clinical evidence of adrenal insufficiency with skin hyperpigmentation and electrolyte imbalance. She was diagnosed having neonatal adrenoleukodystrophy based on abnormally high levels of plasma very long-chain fatty acids, pipecolic acid and phytanic acid.

A Case of Using Hominis Placenta and Soyeom Mixed Pharmacopuncture to Remove Melanocytic Nevus

  • Objective: It has been attempted to find out that Homine Placents and Soyeom mixed Pharmacopuncture (HSP) procedure is effective for the removal of melanocytic nevus of considerable size which cannot be applied to general acupuncture. Methods: Hominis placenta (0.5ml) and Soyeom (0.5ml) mixed pharmacopuncture (1.0ml) was subcutaneously injected in several places of melanocytic nevus. The treatment was performed about 20 times, once a week. Layer analysis of pigmentation was carried out using ECOSKIN. Results: One year after the HSP procedure, the pigmentation of the melanocytic nevus was slightly reduced and completely removed after 2 years of procedure. Conclusion: The HSP could be effective for removing hyperpigmentation in the melanocytic nevus.

Inhibitory Effects of Aqueous Extracts from Nardostachys chinensis on ${\alpha}$-Melanocyte Stimulating Hormone-induced Melanogenesis in B16F10 Cells

  • For the purpose of the development of skin-whitening or therapeutic agents against hyperpigmentation, aqueous extract from Nardostachys chinensis (AENC) was evaluated for melanogenesis inhibitory activity in B16F10 melanoma cell. The treatment with AENC at the 0.2, 0.5 and 1.0 mg/ml level significantly inhibits the biosynthesis of melanin compared with untreated control. The tyrosinase activity also significantly decreased in AENC-treated cells at the 0.2 and 0.5 mg/ml level and inhibitory effects were more efficient than commercial arbutin at 0.1 mg/ml. The Western analyses confirmed the significantly decreased expression of tyrosinase and tyrosinase-related protein-2 by AENC treatment. These results indicate that AENC may contribute to the inhibition of melanin biosynthesis through regulating the expression as well as activity of tyrosinase and AENC may be useful as a new candidate in the design of new skinwhitening or therapeutic agents.

  • The authors observed 2 cases of fibrous dysplasia on the cranial and maxillofacial bones in 31, 28 aged sisters, who had come to the Infirmary of Dental College, Yonsei University. The serial roentgenograms and clinical findings had been taken and the results established as polyostotic fibrous dysplasia according to the findings in their images. The authors have obtained the results as follows: 1. Bony expansion of the mandible occured at 18 years of age and the facial assymetry appeared due to development of the lesions. 2. The traumatic history were not noted but weak tendency of familial history noted 3. Endocrine disturbances, hyperpigmentation on the skin and premature puberty in the infancy were not noted. 4. We have concluded these diseases as polyostotic fibrous dysplasia on the cranial and maxillofacial bones with weak familial tendency according to the findings

The Study on Depigmentation of Kamikwibi-Tang (가미귀비탕의 미백효과에 관한 연구)

  • Objective: This study was performed to investigate the depigmentation effects of the extracts of Kamikwibi-Tang. Methods Inhibition of tyrosinase activity, melanin production & cell viability in cultured B16 melanoma cells, UV screen and cytoprotective effects on PC12 cells injured by hydrogen peroxide were measured. Results: The extracts of Kamikwibi-Tang did not have any inhibitory effects on tyrosinase activity and did not show any inhibitory effects on melanin production in melanoma cells and also did not have any inhibitory effects on UV screen. But the extracts showed high cytoprotective effects on PC12 cells injured by hydrogen peroxide. Conclusion : These results suggest that Kamikwibi-Tang indrectly inhibits melanin biosynthesis which is involved in hyperpigmentation and could be used as a whitening agent for the skin.
