Purpose: High intake of cholesterol leads to cardiovascular disruption. Estimating the actual intake of cholesterol can be beneficial for nutrition intervention. This research aimed to develop Semi-Quantitative Food Frequency Questionnaire (SQ-FFQ) to estimate cholesterol intake and analyze its reliability and validity. Methods: SQ-FFQ was developed by sorting high cholesterol food items in Indonesian food database and food items' availability. A total of 30 older adults were randomly chosen from Public Health Center in Jagir District, Surabaya, Indonesia to test its validity. Reliability test was done by measuring the same developed SQ-FFQ in one-month period, while validity test was done by comparing SQ-FFQ results with 6-days food record. Statistical analysis used for reliability test was paired t-test, the Intra-class Correlation Coefficient (ICC), and Cronbach's α to measure the internal consistency. Meanwhile, validity of developed SQ-FFQ was analyzed using paired t-test and Bland-Altman. Results: Reliability of 2 administered SQ-FFQs showed a good agreement based on paired t-test analysis (p = 0.200), ICC (0.609), and Cronbach's α (0.757). Strong agreement was found in most of food items, but agreements for egg yolk and fried duck were poor. Significant difference was found between those food items (p = 0.001 vs. p < 0.001, respectively) with mean difference were -25.3 mg and 46.2 mg. Validity of developed SQ-FFQ2 compared to 6-days food diary records also found a strong agreement based on paired t-test and the Bland-Altman analysis. Conclusion: This baseline research provides a reasonably valid and repeatable measure of cholesterol intake estimation that can be widely used in nutrition and public health study, especially in Indonesia. No study has been conducted in Indonesia on the development of tools to estimate the cholesterol intake.
This study was performed to investigate the effects of Sineui-Dae (Sasa coreana Nakai, a kind of bamboo) extracts on the improvement of serum lipid composition by using rats fed a hypercholesterol diet for 4 weeks. The experiment animals were administered with the following diets; high cholesterol diet group (HC diet) as a control and three supplemented groups with high cholesterol diets (HCW, HCM or HCH diet). Three kinds of extracts were prepared by orderly extraction with hexane, methanol, and water. We measured free cholesterol, cholesteryl ester, total cholesterol LDL- and HDL-cholesterol triglyceride and phospholipid in the serum of rats in three experimental and control groups. There were no significant differences in body weights and feed intakes between the HC control group and the extract supplemented groups. The levels of total cholesterol and LDL-cholesterol in the serum of extract supplemented groups were lower than that of the group fed with HC diet only. Serum HDL-cholesterol levet which is known as an antiatherosclerosis factor, was higher in all groups supplemented with bamboo extract by 17.2-21.9% compared to the HC control group. All groups supplemented with bamboo extracts showed the lowering effect of atherogenic index compared to the HC control group (HC group: 2.96${\pm}$0.08, HCM group: 1.48${\pm}$0.02, HCW group: 1.69${\pm}$0.04, HCH group: 1.84${\pm}$0.01). Furthermore, serum triglyceride and phospholipid decreased significantly in the HCM diet compared to the HC control diet. These results suggest that Sineui-Dae bamboo extract, especially methanol extract, has improving effects on hyperlipidemia of rats fed a high cholesterol diet.
The aim of this study is to investigate the effects of hot water soluble extract from green tea on the components of serum and the liver and the antioxidative effects in accordance with the different protein types. For this purpose, four experimental groups were set up. As for the protein source casein, isolated soy protein was supplemented to the rats, together with hot water soluble extract from green tea. Four experimental groups kept eight Sprague-Dawley rats respectively. The CP group was supplemented with casein only, the CG group was supplemented with casein and hot water soluble extract from green tea, the ISP group was supplemented with isolated soy protein only, and the ISG group was supplemented with the isolated soy protein with hot water soluble extract from green tea. After 4 weeks of feeding with experimental diet, the levels of serum and liver lipid and antioxidant enzyme activity and TBARS in the liver were measured. The results are; 1. Weight gain and FER were higher in the casein group than in the isolated soy protein group in general. In the casein group, the weight gain and FER were reduced significantly when hot water soluble extract from green tea was supplemented (P < 0.05). There was no significant difference in feed intake. 2. In general, total cholesterol and LDL-cholesterol in the serum were higher in the casein group than in the isolated soy protein group, however just the concentration of LDL-cholesterol in the casein group was significantly lower when the hot water soluble extract from green tea was supplemented (P < 0.05). 3. Triglycerides in the liver were higher in the casein group than in the isolated soy protein group general, however when hot water soluble extract from green teas was supplemented only in the isolated soy protein group, the content of triglyceride in liver was significantly lower (P < 0.05). 4. There was no significant difference in antioxidant enzyme activity in the liver in all the groups, however the content of TBARS was low only in the casein group when hot water soluble extract from green tea was supplemented (P < 0.05).

The aim of this study was to investigate the hypocholesterol effects of Opuntia humifusa extract in hypercholesterolemic rats. Rats (8-week-old, male) were randomly divided into four groups (n=4) as follows: N-control, normal diet; H-control, hypercholesterolemic diet; OH-1, 2% O. humifusa extract-supplemented hypercholesterolemic diet; OH-2, 4% O. humifusa extract-supplemented hypercholesterolemic diet. After 4 weeks, we observed that hypercholesterolemia induced significant increases in serum lipids (total cholesterol and LDL-cholesterol), hepatic lipids (total cholesterol and triglyceride), and hepatic function parameters (ALT, alanine aminotransferase; AST, aspartate aminotransferase) (P<0.05). Treatment with O. humifusa extract for 2 weeks normalized these indexes up to the levels of normal rats. O. humifusa extract tended to increase fecal lipid (H-control: 142.74 mg/day vs. OH-1: 214.05 mg/day; OH-2: 200.95 mg/day, P<0.05) and bile acid (H-control: 37.07 ${\mu}M/day$ vs. OH-1: 47.23 ${\mu}M/day$, OH-2: 47.93 ${\mu}M/day$, P<0.05) contents of hypercholesterolemic rats. We concluded that oral administration of O. humifusa extract effectively improved cholesterol metabolism in a hypercholesterolemic animal model induced by hypercholesterol diet.
Gender differences in the effects of gonadectomy and high cholesterol diet on body weight and body cholesterol were investigated by using Sprague Dawley rats. Body weight, plasma and liver levels of cholesterol and triglyceride and platelet aggregation were examined in ovariectomized(OVX) or orchidectomized(ODX) rats with their intacts after feeding diet with or without 0.5% cholesterol. Body weight was significantly increased(p < 0.01) in OVX rats and significantly decreased(p < 0.01) in ODX rats compared to their respective intact rats, and cholesterol diet significantly(p < 0.05) decreased body weight in gonadectomized rats. Liver lobes from rats fed cholesterol diet were opaque and larger than those from rats find control dict, resulting in a significant increase(p < 0.01) in LW/BW ratio. Plasma and liver levels of total cholesterol were significantly increased (p < 0.01) in female rats regardless ovariectomy when find 0.5% cholesterol diet, but those levels in male rats were increased only when they were orchidectomized(p < 0.0l). Plasma HDL-cholesterol was significantly decreased(p < 0.05) in both sexes when find cholesterol diet. HDL-cholesterol were higher in female than male rats regardless treatments(p < 0.05). Liver triglyceride was significantly increased(p < 0.05) in both sexes when find cholesterol dict. Plasma level of triglyceride was not different among groups except significant decrease(p < 0.05) in cholesterol find ODX rats. Maximum platelet aggregation in female rats was significantly lower(p < 0.05) than male, but ovariectomy and cholesterol diet caused an increase te the level of male rats. Microscopic examination showed cholesterol diet caused a lipid accumulation in liver. Results indicate that intact female rats have higher response to hypercholestcrolemic diet than intact male rats and orchidectomy causes male rats more responsive to hypercholesterolemic diet. However, ovariectomy causes an increase female food efficiency ratio to the level of male rats, significantly increasing body weight.

The present study was designed to investigate the effects of dietary fiber sources and levels on serum lipids in rats fed a hypercholesterol diet containing 0.5% cholesterol. Male Sprague-Dawley rats weighing 52.7 ${\pm}$ 7.27g were devided into sevel groups by completely randomized block design and fed various dietary fibers which were prepared from Rice bran, Siraegi (The dried green parts of a radish) and Bracken Fern at a level of 5% and 10% respectively. The results obtained in this study are summarized as follows: 1. Body weight gains were not significantly different among the groups and food intake was significantly higher in rats fed a diet of Siraegi 10% than those fed the control diet. 2. Fecal wet weights were significantly increased in all experimental groups. 3. Levels of serum total cholesterol in all experimental groups were significantly lower than that of the control group except the level in the R 10(Rice bran 10%) group. 4. Free Cholesterol levels in the serum were increased in all experimental groups, especially the levels in the R 10 (Rice bran 10%) and S 10 (Siraegi 10%) groups were significantly higher than that of the control group. 5. Ester Cholesterol levels in the serum were significantly lower in rats fed the experimental diets than those fed the control diet. 6. There was a tendency of higher serum HDL-Cholesterol levels in R5 (Rice bran 5%), R10 (Rice bran 10%) and B5(Bracken 5%) groups compared to the control group however no significant differences were found between the control group and all the experimental groups except in the case of the S5 (Siraegi 5%) group. 7. LDL, VLDL-Cholesterol levels in the serum sere significantly reduced in the S5 (Siraegi 5%), B5 (Bracken 5%) and B10 (Bracken 10%) groups. 8. The HDL-C./Total-C. Ratio was increased in all experimental group especially the Ratios in the S5 and B5 groups were significantly higher than that of the control group.

This study was performed to investigate cholesterol improvement effects of adding fermented defatted soybean grit (FD) and FD added to 5 or 10% Corchorus olitorius (FDC). Cholesterol adsorption in the FD and FDC group was more than 70%. Apolipoprotein AI and CIII improved in HepG2 cells, and a greater improvement effect was shown in FDC than that in FD. We also investigated the effect of FDC on body lipid metabolism and a high-fat diet for 4 weeks. Rats were divided into control (Con), high-fat (HF), HF treated with 20% FD (HF-FD), and HF treated with 20% FDC (HF-FDC) groups. Plasma total cholesterol and low density lipoprotein-cholesterol concentrations, hepatic total cholesterol, and triglyceride contents were significantly lower in the HF-FDC group than those in the HF group. Additionally, fecal total cholesterol and triglyceride contents increased in rats treated with FDC. Hepatic 3-hydroxy-3-methylglutaryl-coenzyme A reductase activities were significantly lower in the HF-FDC group than those in the HF group.
Purpose: This study evaluates the potential protective effects of hemp (Cannabis sativa L.) seed oil supplementation in rats fed a high-cholesterol diet. Methods: Rats were fed a 1.25% cholesterol diet for 8 weeks, followed by oral administration of either of the two doses of hemp seed oil (HO) (0.5 mL/kg (HOL group) or 1 mL/kg (HOH group) body weight/day) or simvastatin at 10 mg/kg body weight/day. Oxidative stress, lipids, liver enzymes, and renal markers were measured in the serum. Western blot analysis was applied for evaluating the expressions of inflammatory makers. Results: Except for HDL-cholesterol, the altered levels of lipoproteins, aminotransferases, urea, and creatine kinases in hypercholesterolemic rats were significantly corrected by HO administration. Especially, compared to the HOH group, HOL treatment further reduced AST, ALT, creatinine, TC, and LDL-cholesterol levels. Moreover, both the atherogenic index and cardiac risk factor (CRF) in the HOL group were more restrained compared to the HOH group. Increased levels of p-AMPK coincided with the inhibition of SREBP-2 activation which subsequently suppressed the expression of HMGCR. Nuclear factor (NF)-κB activation coincided with the PI3K/Akt pathway activation and the increased phosphorylation of p38; these levels were significantly suppressed by HO treatment. In addition, HO treatment markedly reversed the changes in chemokines such as ICAM-1, VCAM-1, and MCP-1. Histological alterations induced by cholesterol overload in cardiac and hepatic tissues were ameliorated by HO supplementation. Conclusion: Taken together, our results indicate a low concentration of HO demonstrates improved dysfunctions caused by a high-cholesterol diet via inhibition of the PI3K/Akt/NF-κB signaling pathway.
Atherosclerosis is a chronic inflammatory disease of the arterial wall characterized by progressive accumulation of lipids, cells, and extracellular matrix. Matrix metalloproteinases(MMPs) and tissue inhibitor of metalloproteinases(TIMPS) contribute to vascular matrix remodeling in atherosclerosis, and some cytokines may play role in the synthesis or activation of MMPs or TIMPs. Material and Method: We produced experimental atherosclerotic plaques in 9 rabbits by atherogenic hypercholesterol diet for 12 weeks, and 10 other rabbits were used as control group with standard laboratory chow, At that time, 19 rabbits were sacrificed and aorta, coronary arteries and blood specimens were prepared. The expressions of MMP-9, TIMP-2 and interleukin(IL)-18, and the bioactivity of IL-6 were investigated with H&E stain, immunohistochemical stain, immunoblotting(Western blot analysis), and bioassay. Result: Serum cholesterol in the experimental group increased up to 1258$\pm$262 mg/dL(control group: 41$\pm$7 mg/dL). All experimental group showed well-developed atherosclerotic plaques in aorta and coronary artery. The expression of MMP-9 in aorta and coronary artery of the experimental group showed significant increase than that of the control group by immunohistochemistry. Among the experimental group, complicated lesions with intimal rupture or complete luminal occlusion, demonstrated stronger expression of MMP-9. Interestingly, there was no difference in expression of TIMP-2 between the experimental and the control group. These findings were confirmed by Western blot analysis. The bioassay revealed significant up-regulation of serum bioactivity of IL-6 in the experimental group(4819.60$\pm$2021.25 IU/$m\ell$) compared to that of IL-6 in the control group(27.20 $\pm$ 12.19 IU/$m\ell$). IL-18 was expressed in all atherosclerotic plaques, whereas little or no expression was detected in the control group. Conclusion: The increased MMP-9 expression along with the unchanged TIMP-2 expression seem to be contributory factors in extracellular matrix degradation in atherosclerosis. Focal overexpression of MMP-9 may promote plaque destabilization and cause complications of atherosclerotic plaques such as thrombosis with/without acute coronary syndrome. Elevation of IL-6 and IL-18 may be more than just markers of atherosclerosis but actual participants in lesion development. Identification of critical regulatory pathway is important to improve the understanding of the cellular and molecular basis of atherosclerosis and may open the way for novel therapeutic strategies.
