Effect of Dietary Inclusion of Lactobacillus acidophilus ATCC 43121 on Cholesterol Metabolism in Rats

  • This study examined the effects of Lactobacillus acidophilus ATCC 43121 (LAB) on cholesterol metabolism in hypercholesterolemia-induced rats. Four treatment groups of rats (n=9) were fed experimental diets: normal diet, normal $diet+LAB(2{\times}10^6\;CFU/day)$, hypercholesterol diet (0.5% cholesterol, w/w), and hypercholesterol diet+LAB. Body weight, feed intake, and feed efficiency did not differ among the four groups. Supplementation with LAB reduced total serum cholesterol (25%) and VLDL+IDL+LDL cholesterol (42%) in hypercholesterol diet groups, although hepatic tissue cholesterol and lipid contents were not changed. In the normal diet group, cholesterol synthesis (HMG-CoA reductase expression), absorption (LDL receptor expression), and excretion via bile acids (cholesterol $7{\alpha}-hydroxylase$ expression) were increased by supplementation with LAB, and increased cholesterol absorption and decreased excretion were found in the hypercholesterol diet group. Total fecal acid sterols excretion was increased by supplementation with LAB. With proportional changes in both normal and hypercholesterol diet groups, primary bile acids (cholic and chenodeoxycholic acids) were reduced, and secondary bile acids (deoxycholic and lithocholic acids) were increased. Fecal neutral sterol excretion was not changed by LAB. In this experiment, the increase in insoluble bile acid (lithocholic acid) reduced blood cholesterol level in rats fed hypercholesterol diets supplemented with LAB. Thus, in the rat, L. acidophilus ATCC 43121 is more likely to affect deconjugation and dehydroxylation during cholesterol metabolism than the assimilation of cholesterol into cell membranes.

Effects of Hot Water Soluble Extract from Angelicae Gigantis Radix on the Lipid Metabolism and Antioxident Defense System in rats fed Hypercholesterol Diet (당귀 열수추출물이 고콜레스테롤 식이를 공급한 흰쥐의 지질대사와 황사화계에 미치는 영향)

  • To investigate the effects of hot water soluble extract from Angelicae Gigantis Radix on lipid metabolism and antioxidant defense system in rats fed hypercholesterol diet, Sprague-Dawley male rats were fed either hypercholesterol diet with water or hot water soluble extract from Angelicae Gigantis Radix. Hypercholesterol diet contained 18 % beeftallow and 5 % cholesterol. After 4 weeks of experimental periods serum and liver obtained. The level of serum lipid and the activities of GOT and GPT was measured. The level of lipid and TBARS and the activity of GSH-Px was measured also. The results obtained were: 1) Food intake in the experimental group supplied hot water soluble extract from Angelicae Gigantis Radix was low significantly. 2) In the experimental group supplied hot water soluble extract from Angelicae Gigantis Radix of serum total lipid and total cholesterol was significantly low. 3) Serum GOT activity was significantly low in the experimental group supplied hotwater soluble extract from Angelicae Gigantis Radix. There was no difference of serum GPT activity between control and experimental group supplied hot water soluble extract from angelical Gigantis Radix. 4) There was no difference of lipid composition, TBARS level and GSH-px activity in liver between control and experimental group. The result of this study indicates that the hot water soluble extract from Angelicae Gigantis Radix was effective in decreasing the food intake of the experimental animals in the hypercholesterol diet and also effective in decreasing the levels of serum total lipid, total cholesterol and GOT activity. However it was not effective to change the level of lipid, TBARS and GSH-Px activity of liver.

Effect of Acanthopanax Senticosus Water Extract on Lipid Metabolism in Rats Fed a Hypercholesterol Diet (가시오가피 물추출물이 고콜레스테롤식이를 공급한 흰쥐의 지질대사에 미치는 영향)

  • In order to find the effect of improving hyperlipidemia by Acanthopanax senticosus water extract, a hypercholesterol diet and Acanthopanax senticosus water extract were supplemented to week- old male Spargue Dawley rats for four weeks in different amount. Results showed that serum total cholesterol significantly decreased in the group supplemented with Acanthopanax senticosus water extract by 50mg/kg(ASW-50) and 75mg/kg(ASW-75) compared with the control group.(p<0.05). Serum triglyceride also showed a significant decrease in the group supplemented with 50mg/kg(ASW-50) and 75mg/kg(ASW-75) compared with the control group. Liver total cholesterol showed a significant decrease in the group supplemented with Acanthopanax senticosus water extract by 50mg/kg(ASW-50) and 75mg/kg(ASW-75) compared with the control group(p<0.05), but liver triglyceride did not show a significant decrease in all of the experiment groups. Total cholesterol and triglyceride in feces significantly increased in all of the groups supplemented with Acanthopanax senticosus water extract(p<0.05). Acanthopanax senticosus water extract decreased the level of serum total cholesterol and triglyceride, reduced total cholesterol in the liver, and increased the excretion of total cholesterol and triglyceride in the feces.

The Hypocholesterolemic Effect of Platycodi radix Saponin in Rats Fed a Hypercholesterol Diet (길경 saponin이 고콜레스테롤식이를 공급한 흰쥐의 지질대사에 미치는 영향)

  • The objective of this study is to find the effect of improving hyperlipidemia through Platycodi radix saponin. A hypercholoesterol diet and Platycodi radix saponin methanol extract were supplemented to the 6 week old male Spargue Dawley rats for four weeks. Results showed that the serum total cholesterol, triglyceride and LDL-cholesterol level decreased in the group supplemented with Platycodi radix saponin methanol extract(p<0.05), however, the serum total lipid and HDL-cholesterol did not show any difference from the control group according to the supplement of Platycodi radix saponin methanol extract. In the group supplemented with Platycodi radix saponin methanol extract, the total cholesterol decreased by 20%, triglyceride by 36% and LDL-cholesterol by 25% respectively, and there was no difference according to the level of Platycodi radix saponin methanol extract. AI(atherogenic index) and CRF(cardiac risk factor) were low in the group supplemented with Platycodi radix saponin methanol extract. Total lipid, total cholesterol and triglyceride in liver and feces were significantly low in the group supplemented with the Platycodi radix saponin methanol extract compared with the control group(p<0.05). According to this result, it is expected that the Platycodi radix saponin methanol extract supplemented in the hypercholesterol diet can lower atherosclerosis, the cause of cardiovascular disease, by reducing not only serum cholesterol and triglyceride but also liver cholesterol and triglyceride.

Effects of the Cnidium Officinale Makino Water Extract on the Lipid Metabolism of Rats Fed a Hypercholesterol Diet (천궁 열수추출물 첨가가 고콜레스테롤 식이를 급여한 흰쥐의 지방대사에 미치는 영향)

  • The study examines the effects of the Cnidium officinale Makino water extract on hyperlipidemia. For this, three levels (low, moderate, and high) of the Cnidium officinale Makino water extract were supplemented to six-week-old male sprague Dawley rats for five weeks. According to the results, serum total lipid, triglyceride, total cholesterol, and LDL cholesterol levels showed significant decreases (p<0.05) in the group with the Cnidium officinale Makino water extract. The serum total lipid level decreased by 8~22%; the triglyceride level, by 13~25%; the total cholesterol level, by 6~23%; and the LDL cholesterol level, by 14~25%. Total cholesterol and triglyceride levels in the liver were significantly lower in the group with the Cnidium officinale Makino water extract than in the control group (p<0.05), and fecal total cholesterol and triglyceride levels were significantly higher in the group with the Cnidium officinalle Makino water extract. These results suggest that the Cnidium officinale Makino water extract in the hypercholesterol diet may reduce hyperlipidemia, a major cause of cardiovascular disease, by reducing serum cholesterol and triglyceride level in the liver through the facilitation of their excretion by feces.

Effect of Rot Water Soluble Extract from Eleutherococcus and Senticosus and Dietary Carnitine on the Lipid Metabolism and Antioxidant Defense System of Rats on Hypercholesterol Diet (가시오가피 열수추출물과 카르니틴 공급이 고콜레스테롤 식이를 공급한 흰쥐의 지질대사와 항산화계에 미치는 영향)

  • This study investigated the synergy effects of hot water soluble extract from Eleutherococcus senticosus and dietarty carnitine on the lipid metabolism and antioxidant defense system of rats on hypercholesterol diets. Sprague-Dawley male rats were fed either a hypercholesterol diet with water or a hot water soluble extract from Eleutherococcus senticosus and dietarty carnitine. The experimental groups consisted of the control group (CO), the group supplied with hot water soluble extract from Eleutherococcus senticosus(ES), and the group supplied with hot water soluble extract from Eleutherococcus senticosus and dietarty carnitine(ESC). Eleutherococcus senticosus was extracted, made into 0.5% solution, and fed to the ES and ESC groups. A 3% carnitine diet was supplied to the ESC group. Hypercholesterol diets contained 18% beef tallow and 5% cholesterol. After 4 weeks of administering these diets, serum and liver were obtained and the level of serum lipid and the activities of GOT and GPT were measured. In addition, the level of liver lipid and TBARS and the activity of GSH-Px were measured. The results were as follows: 1) Weight gain and FER in the group supplied with hot water soluble extract from Eleutherococcus senticosus and dietarty carnitine(ESC) was low significantly(P<0.05). 2) In the groups supplied with hot water soluble extract from Eleutherococcus senticosus(ES) and the group supplied with hot water soluble extract from Eleutherocorcus senticosus and dietarty carnitine(ESC), total serum cholesterol levels were both significantly low (P<0.05,P.0.01). 3) Serum GOT activity was significantly low(P<0.05) in the group supplied with hot water soluble extract from Eleutherococcus senticosus(ES) and in the group supplied with hot water soluble extract from Eleutherococcus senticosus and dietary carnitine(ESC). There was no difference between serum GPT activity in the control group and the groups supplied with hot water soluble extract from Eleutherococcus senticosus or dietary carnitine. 4) Liver triglyceride was low (P.0.05) in the group supplied with hot water soluble extract from Eleutherococcus senticosus and dietarty carnitine(ESC). No difference was observed in other lipid composition, TBARS level, and GSH-px activity in liver between the control group and the experimental groups. In summary, the effects of hot water soluble extract from Eleutherococcus senticosus and dietarty carnitine on the lipid metabolism and antioxidant defense system (such as weight gain, FER, total serum cholesterol and liver triglyceride) were low when Eleutherococcus senticosus and dietarty carnitine were supplied simultaneously.

Treatment Effect of Mulberry Root-Bark (Mori radicis Cortex) Extract on Hypercholesterolemia in Rabbits

  • This study reports that the effect of Mori radicis cortex, Mulberry root-bark, on the lipid peroxidation products formation in the liver, blood serum and atherosclerosis of hypercholesterolemia rabbits fed Mulberry root-bark for eight weeks. The rabbits were permitted a fixed quantity to extract of Mulberry root-bark mixed hypercholesterol fodder. The lipid peroxidation products, malondialdehyde (MDA) was significantly diminished. Total cholesterol in serum worthily decreased. Also, triglyceride and high density lipoproteins (HDLs) increased after feeding Mori radicis cortex. Besides, thicky atheromatous plaque on the wall of aorta of rabbits fed on hypercholesterol diet were markedly attenuated by Mulberry root-bark extract. These results support and substantiate the extract of Mulberry root-bark as protection against lipid peroxidation in the liver, blood serum and atherosclerosis.

  • We treated a 45 year-old male patient. He had Hypertension, Diabetes, Hyperlipidemia and Obesity. The main symptoms were Low-back pain, General body weakness, Abdominal discomfort. We diagnosed him as metabolic syndrome and prescribe Yagwanchunghyul-tang three times a day. We observed clinical symptoms Blood pressure, Capillary blood glucose, BMI, Body weight. Cholesterol, Triglyceride. After the treatment, The main symptom, blood pressure, capillary blood glucose(PP2hr), Hypercholesterol, Hypertriglyceride were improved. and BMI levels fell. This case shows that Yagwanchunghyul-tang has an effect on Metabolic syndrome like a Hypertension, Diabetes, Hyperlipidermia, Hypertriglyceride.

Cholesterol Improvement Synergistic Effects of Fermented Soybean Grits Caused by Added with Mung Bean in vitro (녹두 첨가로 인한 탈지대두 Grits(Defatted Soybean Grits) 발효물의 in vitro 상에서의 콜레스테롤 개선능 상승효과)

  • This study was performed to investigate cholesterol improvement of fermented defatted soybean grits (FD) and FD added with 2.5, 5, 10% mung bean (FDM). The FD and FDM were prepared by the solid state fermentation using Bacillus subtilis NUC1 at $40^{\circ}C$ for 24 hr. More than 70% cholesterol adsorption of FD and FDM groups was shown. Particularly, FDM added with 2.5% mung bean (2.5% FDM) showed highest cholesterol adsorption by 90% among FD and FDM groups. 2.5% FDM showed 42% inhibition effect on HMG-CoA reductase, and significantly decreased the intracellular cholesterol contents in HepG2 cells. Apolipoprotein AI, CIII improvement effects of FD and FDM group in HepG2 cells showed most effects in the 2.5% FDM. The results suggest that FDM added with 2.5% mung bean may be beneficial to the prevention of hypercholesterol.
