Anti-hyperglycemic and Antioxidative Activities of Phenolic Acid Concentrates of Rice Bran and Hydroxycinnamic Acids in Cell Assays (미강 페놀산 농축물과 Hydroxycinnamic Acids의 세포내 항당뇨 및 항산화 활성)

  • Phenolic acid concentrates of rice bran(RB-ex) and hydroxycinnamic acids were investigated for their anti-hyperglycemic activities through glucose uptake and glucokinase activity using HepG2 cells and stimulatory effects on insulin secretion using HIT-T15 cells. RB-ex was prepared as an ethylacetate extract after alkaline hydrolysis and hydroxycinnamic acids, found as major compositions of RB-ex, such as ferulic acid(FA), sinapic acid(SA) and p-coumaric acid(p-CA) were investigated to compare with the properties of RB-ex. The properties of glucose uptake in HepG2 cells were examined in the absence of insulin and two different glucose concentrations(5.5 mM and 25 mM). RB-ex and FA showed anti-hyperglycemic activities through the increase of glucose uptake and the stimulation of glucokinase activity in HepG2 cells. RB-ex exhibited higher glucose uptakes with higher glucose concentrations, whereas FA exhibited the same increasing effects on both concentrations of glucose. RB-ex and FA exhibited doubled glucokinase activities relative to control. In the presence of insulin in the 25 mM glucose-containing medium, the levels of glucose uptake were increased in all treatments compared with control. As stimulatory effects of samples on insulin secretion were estimated, RB-ex and FA stimulated insulin secretion at a concentration of 25 ${\mu}g/m{\ell}$ and in particular, FA showed the highest amount of insulin-release in HIT-T15 cells. Antioxidative effects on HIT-T15 cells, RB-ex and hydroxycinnamic acids, excluding p-CA, showed inhibitory activities of 78% to 80% at a concentration of 100 ${\mu}g/m{\ell}$. On the basis of these results, we conclude that RB-ex and FA could help decrease blood glucose levels and prevent the cell damages via antioxidant activity.

Characterization of phenolic compounds biosynthesized in pink-colored skin of Japanese indigenous Vitis vinifera cv. Koshu grape

  • Vitis vinifera cv. Koshu is a traditional grape cultivar that has been grown for centuries in Japan. The Koshu grape has pink-colored skin and Koshu wines have slight astringency. We demonstrated for the first time the characterization of hydroxycinnamic acids, flavan-3-ols, and flavonoids in Koshu grape using high-performance liquid chromatography and liquid chromatography-mass spectrometry. The gross weight of phenolic compounds excluding anthocyanins and proanthocyanidins in Koshu grape at harvest was higher than those in Sauvignon Blanc, Chardonnay, and Merlot grapes. In addition, hydroxycinnamic acid and monomeric flavonol contents in Koshu grape were also higher than those in the other grape cultivars. Transcription analysis of cinnamic acid 4-hydroxylase, p-coumarate 3-hydroxylase, caffeate methyltransferase, and flavonol synthase genes indicated high accumulation of hydroxycinnamic acids and flavonols in Koshu grape skin compared with the other cultivars. These findings obtained by chemical and molecular approaches partially explained the phenolic characteristics and the peculiar astringency of Koshu grape.

Effect of Salinity on Lignin and Hydroxycinnamic Acid Contents in Rice

  • The lignin contents between IR-29 and Pokkali were not significantly different in the absence of NaCl, but they were slightly increased at 40 mM NaCl. Although lignin contents were not relatively significantly different between salt treated and control plants, the total yields of alkaline nitrobenzene oxidation ranged from 17.4-20.0 mg/g of cell wall residue at 40 mM NaCl were significantly different compared with control plants (11.8-12.2 mg/g). The total amounts of ester-linked hydroxycin-namic acids in IR-29 were decreased from 14.5 to 9.9mg/g, while Pokkali is almost same levels (14.9-15.0 mg/g) under treated and control with 40 mM NaCl. In contrast, the total amounts of ether-linked hydroxycinnamic acids were increased from 9.4 to 13.9 mg/g together with an opposite trend in Pokkali as a decrease 10.9 to 8.8 mg/g under treated and control with 40 mM NaCl. These results revealed that IR-29 is more sensitive in response to 40 mM NaCl in terms of hydroxycinnamic acids than Pokkali.

  • Plant phenolics은 식물 생체에 널리 분포되어 있으므로 매일 일정량이 식품으로 섭취되어지고 있으며, 이 물질들은 식품 외에도 일부 향신료 및 의약용으로 이용되어지고 있다. 본 논문에서는 plant phenolics이 free radical scavenger로서의 주요 작용을 포함한 항산화 메카니즘과 기타 항산화 특성, 그리고 plant phenolic계 free radical scavenger 물질인 flavonoids와 hydroxycinnamic acids 및 hydroxycinnamic acid derivatives의 항산화 활성과 구조 간의 관계에 초점을 두어 고찰하였다. 또한 주요 식물별 plant phenolics 물질의 특성과 항산화 활성에 대하여서도 검토하였다.

Scavenging Effects of Hydroxycinnamic Acids on Paraquat Induced Pulmonary Toxicity (III) (Paraquat 유도 페독성에 대한 Hydroxycinnamic Acid계 화합물의 독성 경감 효과 (III))

  • The scavenging effects of two hyaroxycinnamic acids such as caffeic acid and chlorogenic acid on paraquat induced pulmonary toxicity were investigated. The results are summerized as follows: 1. In the 5-lipoxygenase assay, caffeic acid and chlorogenic acid inhibited the enzyme activities whose inhibition concentration (IC$\_$50/) were 4.1 and 9.6 ${\mu}$M respectively. 2. To evaluate the antiinflammatory effects on mediator related to the mechanism of inflammation, ADP-induced platelet aggregation assay and histamine degranulation assay were used. Caffeic acid and chlorogenic acid inhibited on ADP-induced platelet aggregation and histamine release at a concentration dependent manners. 3. Arachidonic acid-induced ear edema were inhibited by administration of caffeic acid and chlorogenic acid. 4. Cytologicad analysis of branchoalveolar lavage fluid (BALF) which was the useful tool for detection of an inflammatory response in the lungs of animals intoxicated with chemicals were used. Alveolar macrophages and neutrophils in BALF, as well as the protein content and the LDH activity in BALF supernatant increased by intoxication of paraquat, but decreased by administration of caffeic acid and chlorogenic acid. Therefore, two hydroxyeinnamic acids tested were the useful candidates for scavenger and antiinflammatory agents on paraquat induced pulmonary toxicity.

Stabilization of Ascorbic Acid by Hydroxycinnamic Acid Incorporation and BGsome Encapsulation (Hydroxycinnamic Acid 첨가와 BGsome 포집에 의한 Ascorbic Acid의 안정화)

  • Ferulic and caffeic acids are hydroxycinnamic acid derivatives, which are potent plant antioxidants. Their free radical scavenging abilities in aqueous solution exposed to DPPH radical, and chemical stabilities against oxidative stress like high temperature and metal ion, were evaluated. To improve the stability of ascorbic acid solution, ferulic acid or caffeic acid was incorporated into ascorbic acid solution. Stability improvement of ascorbic acid was verified through $SC_{50}$ value change according to storage time. Ascorbic acid in combination with ferulic acid or caffeic acid was encapsulated with high efficiency inside BGsome. In this form, its stability was remarkably enhanced compared to that in free aqueous solution.

  • The effective components of Korean ginseng showing the lipid-peroxide depressing activity were isolated. From the ether-soluble acidic fraction of fresh ginseng three phenolic acids were obtained. Salicylic acid and vanilic acid exhibited the potent antioxidant activity, where p-hydroxycinnamic acid did not.

Scavenging Effects of Hydroxycinnamic acids on Paraquat Induced Hepatotoxicity (II) (Paraquat 유도 간독성에 대한 Hydroxycinnamic acid계 화합물의 독성 경감 효과 (II))

  • Antioxidative and scavenging effects were investigated by using two hyaroxycinnamic acids (caffeetannins). such as caffeic acid and chlorogenic acid, on oxidative stress and hepatotoxicity that induced by paraquat. The results are summerized as follows: 1. To assess radical scavenging ability, reduction concentration (IC$\sub$50/) of 1.1 diphenyl-2-dipicrylhydrazine (DPPH) were measured. IC$\sub$50/ values of caffeic acid and chlorogenic acid were 29.7 ${\pm}$0.6 ${\mu}$M and 26.0${\pm}$0.5 ${\mu}$M respectively. Their radical scavenging activities showed concentration-dependent manner. 2. In H$_2$O$_2$-induced hemolysis assay to rat blood, caffeic acid and chlorogenic acid led to different effects, whose hemolysis inhibition ratios at 100 ${\mu}$M were 45.2${\pm}$7.1% and 11.6${\pm}$3.1% respectively 3. In hypoxanthine-xanthine oxidase system producing superoxide anion, caffeic acid and chlorogenic acid showed different inhibitory activities of xanthine oxidase showing 36.8${\pm}$4.3% and 5.4${\pm}$2.3% respectively. 4. To microsomal NADPH dependent cytochrome p-450 reductase in rat liver, paraquat consumed NADPH at a dose-dependent manner from 0 to 1 ${\mu}$M paraquat concentration. Caffeic acid and chlorogenic acid blocked NADPH consumption rates at concentration-dependent manner and inhibition ratios at 100 ${\mu}$M were 67.6% and 59.2% respectively. 5. Administration (30mg/kg, iv) of paraquat to rats caused the marked elevation of glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), lactate dehydrogenase (LDH), alkaline phosphatase (ALP) and lipid peroxides (LPO) in the serum and lipid peroxides in the microsome as compared to the control group. Serum GOT, GPT, LDH, ALP and LPO and liver microsomal LPO were reduced significantly by caffeic acid (50mg/kg), chlorogenic acid (25mg/kg) and silymarin (150 mg/kg) as compared to the paraquat group. From these results, caffeic acid and chlorogenic acid exerted their antioxidative agents by removing reactive oxygen substance (ROS) and scavenging effects by inhibiting ROS generating enzyme. As a general, two hydroxyeinnamic acids showed the useful compounds for scavenger and reducer on the paraquat induced hepatotoxicity.

  • Inhibitory effects of 16 cinnamic acid analogs on formyl-Met-Leu-Phe(fMLP)-induced chemotaxis of rat polymorphonuclear leukocytes were determined by using a microchemotaxis appa ratus. 3,4-Dlhydrocinnamic acid called as caffeic acid exhibited the highest inhibitory effect on the chemotaxis among cinnamic acid analogs tested in this study. Hydroxycinnamic acids exhibited stronger inhibitory effects on the chemotaxis than cinnnamic acid. Hydroxycinnamic acids with one hydroxy group at ortho, meta or para position exhibited similar inhibitory effects on the chemotaxis with corresponding methoxy cinnamic acids, but 3,4-dihydroxycinnamic acid did stronger inhibitory effects than 3,4-dimethoxycinnamic acid. 3,4-Dimethoxycinnamic acid exhibited weaker inhibitory effects on the chemotaxis than 1,2-dimethoxy-4-propenylbenzene and 3,4-dimethoxy cinnamonitrile with -CH=CHCN or -CH=$CHCH_3$, group instead of -CH=CHCOOH group. 4-Hydroxy cinnamic acid and 3,4-dihydroxycinnamic acid exhibited stronger exhibitory effects on the chemotaxis than 3-(4-hydroxyphenyl) propionic acid and 3,4-dihydroxyhydrocinnamic acid with -$CH_2CH_2$COOH group instead of -CH=CHCOOH group.

  • 식물 특히, 우리가 섭취하고 있는 식품에 널리 존재하고 있는 phenolic compounds는 simple phenols과 phenolic acids, hydroxycinnamic acid derivatives 그리고 flavonoids 등의 세가지 종류들로 나누고 있다. 이들 물질은 일정수준의 항산화성, 항돌연변이원성, 항암성을 지니고 있다. 본 총설에서는 식물성 phenolic compounds의 특성, 산화작용과 항산화작용, phenolic compounds의 항산화 메카니즘, 그리고 식물성 phenolic compounds들의 항산화능 등에 대하여 차례로 살핀다. 특히, 우리가 일상적으로 섭취하고 있는 식품에서의 이들 물질과 그들의 항산화성에 대한 최근 연구결과들을 중심으로 고찰한다.
