Research Trend about the Development of White Biotech-Based Aromatic Compounds (화이트바이오텍기반 방향족화합물 개발에 관한 연구동향)

  • Due to the depleting petroleum reserve, recurring energy crisis, and global warming, it is necessary to study the development of white biotech-based aromatic chemical feedstock from renewable biomass for replacing petroleum-based one. In particular, the production of aromatic intermediates and derivatives in biosynthetic pathway of aromatic amino acids from glucose might be replaced by the production of petrochemical-based aromatic chemical feedstock including benzene-derived aromatic compounds. In this review, I briefly described the production technology for hydroquinone, catechol, adipic acid, shikimic acid, gallic acid, pyrogallol, vanillin, p-hydroxycinnamic acid, p-hydroxystyrene, p-hydroxybenzoic acid, indigo, and indole 3-acetic acid using metabolic engineering, bioconversion, and chemical process. The problems and possible solutions regarding development of production technology for competitive white biotech-based aromatic compounds were also discussed.

Improving the drought tolerance in rice (Oryza sativa L.) by exogenous application of vanillic acid and p-hydroxybenzoic acid

  • Water stress obstructs rice growth mainly by oxidative damage in biological cells to cause a reduction of leaf photosynthesis and evapotranspiration processes. In this study, exogenous application of vanillic acid (VA) and p-hydroxybenzoic acid (PHBA) to improve drought tolerance of two Oryza sativa cultivars, Q2 and Q8 was tested. The drought evaluation based on leaf phenotypes to show that both Q2 and Q8 resulted in remarkable water-stress tolerance induced by leaf spraying pretreatment of mixed solution of $50{\mu}M\;VA+50{\mu}M\;PHBA$. The mixtures of $25{\mu}M\;VA+25{\mu}M\;PHBA$ and $50{\mu}M\;VA+50{\mu}M\;PHBA$ treated on Q2 and Q8 in water deficit condition also indicated that total phenols, total flavonoids, and DPPH radical scavenging activity were significantly greater their controls. In general, the accumulation of individual phenolic acids was increased in exogenous phenolic treatments, as compared with controls. Particularly, Q2 obtained a considerable amount of endogenous PHBA after application of $50{\mu}M\;VA$, $25{\mu}M\;VA+25{\mu}M\;PHBA$, and $50{\mu}M\;VA+50{\mu}M\;PHBA$ (0.18 mg/g DW, 0.71 mg/g DW, and 1.41 mg/g DW, respectively); and a negligible content of VA (0.003 mg/g DW) appeared uniquely in the treatment of $50{\mu}M\;VA$. Similarly, Q8 also absorbed a significant quantity of PHBA in $50{\mu}M\;PHBA$, $25{\mu}M\;VA+25{\mu}M\;PHBA$, and $50{\mu}M\;VA+50{\mu}M\;PHBA$ treatments (0.15 mg/g DW, 0.15 mg/g DW, and 0.22 mg/g DW, respectively). In addition, the spraying $50{\mu}M\;PHBA$ and $25{\mu}M\;VA+25{\mu}M\;PHBA$ on Q8 leaves induced similar amount of drought tolerance of Q2 and Q8 were improved, paralleled with the increased amounts of endogenous phenolics revealed that VA and PHBA played an important role to enhance drought tolerance in rice.

The levels of drought tolerance of Q2 and Q8 were improved, paralleled with the increased amounts of endogenous phenolics revealed that VA and PHBA played an important role to enhance drought tolerance in rice.

Determination of Phenolic Compounds Responsible for Allelopathy in Upland Weeds (밭 잡초(雜草)중에 존재(存在)하는 Allelopathy 관련(關聯) Phenol 화합물(化合物)의 검색(檢索))

  • Phenolic compounds present in upland weeds (Artemisia asiatica Nakai, Capsella bursa-pastoris (L.) Medik, Portulaca orleracea L. and Trifolium repens L.) which have shown allelopathic activity were determined using paper chromatography (PC) and high performance liquid chromatography (HPLC). Effect of the determined phenolic compounds on germination and post-germination growth of test plants was also investigated. Kinds of phenolic compounds determined by PC in the four weed species were greater in the aqueous extract than in the methanol extract. Ferulic acid was found in both extracts of the weeds studied, whereas benzoic acid was that the weeds commonly contained hydroquinone, p-hydroxybenzoic, ferulic and cinnamic acids. Out of the phenolic compounds determined by PC and HPLC p-hydroxybenzoic, cinnamic and ferulic acids inhibited germination and post-germination growth of radish and sesame. Inhibition of the phenolic compounds on the radicle growth was greater than on the germination of the test plants.

Various Properties and Phenolic Acid Contents of Rices and Rice Brans with Different Milling Fractions (품종 및 도정도별 백미와 미강의 특성 및 페놀산 함량)

  • Effects of rice cultivars and degree of milling (DM) on composition, pasting properties, total phenolic contents, and distribution of phenolic acids were investigated. Rice and bran fractions with 94.4, 92.0, and 90.4% milling yields from brown rice of four cultivars (Odae, Nampyung, Chucheong, and Ilmi) were used. Fat and ash contents of milled rices decreased with increasing DM, whereas protein contents were not affected. In rice bran, differences in fat and ash contents by cultivars were higher than those caused by DM. With increasing DM, gelatinization temperature of rice flour decreased, whereas peak viscosity and hold viscosity at $95^{\circ}C$ increased. While cold viscosity, final viscosity, and setback varied among cultivars, DM had little effect. Total polyphenolic contents in brown rice, milled rice, and rice bran were 93.9-88.8, 30.3-71.9, and 310.0-541.6 mg catechin eq/100g, respectively. Major phenolic compounds were identified as ferulic and p-coumaric acids. Total phenolic content of brown rice (65.9-27.9 mg%) decreased with increasing DM, whereas ratio of ferulic acid composition increased. Chucheong and Ilmi varieties showed biggest reduction of phenolic acid contents by milling. In rice bran, ferulic and p-coumaric acids were 157.8-240.2 and 31.8-90.4 mg%, respectively. Contents of sinapinic, benzoic, and m-hydroxybenzoic acids in rice bran were higher than those of brown and milled rices.

Application of Emulsion Liquid Membrane to Removal of Fermentation Inhibitors from Simulated Hemicellulosic Hydrolysates (모사 헤미셀룰로오스 가수분해액으로부터 발효 저해물질의 제거를 위해 에멀젼형 액막법의 적용)

  • Hemicellulosic hydrolysates contain not only sugars but also several kinds of ethanol fermentation inhibitory substances such as carboxylic acids, furans and phenolic compounds. In this work, emulsion liquid membrane (ELM) was chosen as a separation technology to remove the inhibitors. A basic simulated hemicellulosic hydrolysate was composed of xylose as sugar, dilute sulfuric acid solution as solvent, and acetic acid as carboxylic acid, and furfural as furan derivative or p-hydroxybenzoic acid(HBA) as phenolic compound was added to the hydrolysate when necessary. Acetic acid and HBA as weak acid could be selectively removed from the hydrolysates in all the ELM systems considered here, but furfural as aldehyde was quite hard to remove. Also, when HBA was added to the basic simulated hemicellulosic hydrolysate, both of acetic acid and HBA in the feed phase could be selectively removed up to 99% in an ELM system with tributyl phosphate as extractant.

Allelopathic Potentials of Larix leptolepis (S. et Z.) Gorden on Germination and Seeding Growth of Selected Species (植物의 發芽와 生長에 미치는 일본잎갈나무의 알레로패티 效果)

  • A number of field surveys have been performed in larch (Larix leptolepsis) forest floor in which any seeding of this species never find ther. In laboratory work, aqueous extracts from leaves, roots and stems of larch were provided and used to bioassay. Among them, germination and seedling growth of selected species were the most inhibitory in leaves extracts. Though germiation percentage of a species showed high compared with control, its dry weight after a given period grown were clearly low. The results due to be considered by allelopathic effect. And syringic acid and p-hydroxybenzoic acid were identified by HPLC from larch leaves. It is assumed that these phenolic acids affect on other species as allelochemicals of larch.

Effect of Phenolic Compounds Identified from Crop Residues (Wheat.Rye) on the Germination and Growth of Various Weeds (맥류작물(麥類作物)(밀.호밀) 잔여물(殘餘物)로부터 동정(同定)된 Phenolic Compounds 가 잡초(雜草)의 발아(發芽) 및 생육(生育)에 미치는 영향(影響))

  • The phenolic compounds identified from rye and wheat residues seemed to be p-coumaric, p-hydroxybenzoic, vanillic, ferulic, salicylic and syringic acids. Total phenol content of rye and wheat straw determined at heading stage was 0.1803% and 0.1702%, respectively. Total phenol content in straw was higher than that of root at all growth stages. The germination and growth of plants, such as Oryza sativa, Echinochloa creugalli, Cyperus serotinus, Portulaca oleacea, Amaranthus retroflexus, Digitaria saguinalis, and C. album were inhibited by treatments of authentic phenolic compounds as the concentrations increased. However, at the early stage, the germination and shoot growth of P. distinctus were markedly stimulated by them, and then the further growth of shoot and root was markedly inhibited by the prolonged treatment of phenolic compounds. The aqueous extracts from rye and wheat straw completely inhibited the germination of A. retroflexus and C. album at a high concentrations. The content of starch and protein in bulbs of P. distinctus was lower in the p-hydroxybenzoic acid treated plot, at $10^{-2}M$, than the untreated control.

Ginsenoside, Phenolic Acid Composition and Physiological Significances of Fermented Ginseng Leaf (발효처리가 인삼잎의 진세노사이드 및 페놀산 조성 변화와 생리활성에 미치는 영향)

  • This study was carried out to investigate the compositional changes of ginsenosides and phenolic acids of ginseng leaf by fermentation in order to promote the utilization of ginseng leaf. The chief ginsenosides in non-fermented ginseng leaf (NFGL) were ginsenoside-Rg1 (26.0 mg/g), -Re (47.3 mg/g) and -Rd (23.9 mg/g). By fermentation, ginsenoside-Rg1, -Rb1, -Rb2, -Rb3, -Rc and -Re were decreased tremendously and new ginsenoside-Rh2, -Rh1, -Rg2 and -Rg3 appeared. Especially, ginsenoside-Rg3 (3.7 mg/g) on FGL was increased 15-fold compared to that of NFGL (0.2 mg/g). Total phenolic compound content of NFGL and FGL measured by colorimetric analysis was 350.4 and 312.5 mg%, respectively. There were 8 free and 6 ester forms of phenolic acids in NFGL. Among them, content of ferulic acid was the highest, comprised of 12.6 and 50.7 mg%, respectively. In FGL, total content of protocatechuic acid, p-hydroxybenzoic acid, and vanillic acid were increased by 28, 5 and 7.8 fold and ferulic acid was decreased greatly. Tyrosinase inhibitory activity of FGL was stronger than NFGL, while electron donating abilities of FGL were similar to NFGL.

Effects of Major Phenolic Acids Identified from Barley Residues on the Germination of Paddy Weeds (보리 잔여물(殘餘物)속에 함유(含有)된 주요(主要) Phenolic Acids가 논 잡초(雜草) 발아(發芽)에 미치는 영향(影響))

  • Effects of major phenolic acids identified from barley residues (straw, root) on the germination of rice and 3 paddy weeds such as Echinochloa crusgalli, Cyperus serotinus, and Potamogeton distintus were evaluated to obtain the basic informations on the development of naturally occurring herbicides. The predominant phenolic compounds extracted from barley residues in both straw and root were identified as p-coumaric, p-hydroxybenzoic, ferulic, vanillic, and salicylic acids by means of paper chromatography. Total phenol content of barley straw and root at the harvesting stage was 0.169% and 0.127% per dry weight, respectively. During the decomposing process, total phenol content slightly increased and then decreased. The germination of test plants was inhibited by treatments of 4 major authentic phenolic acids identified, most significantly on rice, and less on E. crusgalli, and C. serotinus. P. distintus, however, was markedly stimulated by them as the concentration increased, and then sprouted buds of pondweeds were changed to dark brownish color, resulting in the death as the treatment prolonged. The greater inhibitory effect appeared on shoot growth rather than germination. The aqueous extracts of barley residues showed the similar inhibitory effect on the germination and shoot growth of rice and three paddy weeds as the treatments of 4 authentic phenolic acids.
