Biosynthesis of Novel Poly(3-hydroxyalkanoates) Containing Alkoxy Groups by Pseudomonas oleovorans

  • Novel poly(3-hydroxyalkanoates), PHAs, having either methoxy or ethoxy groups as the terminal hydrophilic moieties, were biosynthesized by Pseudolnons oleovorans. grown either solely with 11-alkoxyundecanoic acid or 8-alkoxyoctanoic acid, or grown with a mixture of 6-alkoxyhexanoic acid and nonanoic acid. The PHA synthesized from 11-methoxyundecanoic acid consisted of 88 mol% 3-hydroxy-7-methoxyheptanoate and 12 mol% 3-hydroxy-9-methoxynonanoate. However, the PHA produced from 11-ethoxyundecanoic acid consisted of 56 mol% 3-hydroxy-5-ethoxypentanoate and 44 mol% 3-hydroxy-7-ethoxyheptanoate. The high solubility of the PHAs in methanol and ethanol indicated that the alkoxy groups in the side chains resulted in the formation of PHAs with an enhanced hydrophilicity.

A selective Assay To Measure Antioxidant Capacity in Both The Aqueous and Lipid Compartments of Plasma

  • The measurement of the total antioxidant capacity (TAC) of human plasma has been widely applied in nutritional science, for example to evaluate the antioxidant contribution of dietary components and to study, although indirectly, the bioavailability of dietary antioxidants. Several methods have been proposed for the measurement of TAC, most of them based on the ability of plasma to withstand the oxidative damage induced by aqueous radicals. Although plasma contains both hydrophilic and lipophilic antioxidants that interact through extensive cross-talk in most of the methods employed for the TAC measurement, the hydrophilic antioxidants such as ascorbic acid, uric acid, and protein thiols mainly contribute to the total antioxidant plasma capacity (almost 70%) while lipophilic antioxidants embedded in the lipoproteins (carotenoids, a-tocopherol, ubiquino1-10) participate only in a negligible amount (less than 5%). The present paper reviews the analytical methods used to assess the TAC and in particular focuses on new approaches that are capable of distinguishing the antioxidant capacity of both the aqueous and lipid compartments of plasma. The general principle of the method as well as some in vitro and ex vivo applications will be discussed within the text.

랫드 두개골 결손부에서 단삼 수용성 추출물의 골형성 효과 (Effect of water extract of Danshen on bone regeneration of rat calvarial defect model)

  • The purpose of this study was to evaluate the osteogenic capacity of water extract of danshen (Salvia miltiorrhiza Bunge). We have established in rat critical-sized calvarial defect model using the combination with collagen scaffold and danshen hydrophilic extract. All rats were extinguished at 8 weeks after bone graft surgery, and the bone regeneration ability of bone grafting sides was evaluated by plain radiography and micro-CT. These results revealed water extract of danshen had the potential to promote osteogenesis especially continuous oral administration with local treatment compared to one-shot local treatment. This compound may provide a new alternative agent for growth factors to promote bone healing and bone regeneration. In conclusion, these results suggest that danshen hydrophilic extract have the potential to promote osteogenesis in bone defects. Further studies about fusion technology with salvianolic acid B, peptides, growth factors, and scaffolds using of the combination of tissue engineering, cell engineering and mechanical engineering are needed.

Enhancement of Paracellular Transport of Heparin Disaccharide Across Caco-2 Cell Monolayers

  • The enhancement of paracellular transport of heparin disaccharide using several absorption enhancers across Caco-2 cell monolayers was tested . The cytotoxicity of these enhancers was also examined. The enhancing effects by Quillaja saponin, diponin glycyrrhizinate, $18{\beta}-glycyrrhetinic$ acid, sodium caprate and taurine were determined by changes in transepithelial electrical resistance (TEER) and the amount of heparin disaccharide transported across Caco-2 cell monolayers. Among the absorption enhancers, $18{\beta}-glycyrrhetinic$ acid arid taurine decreased TEER and increased the permeability of heparin disaccharide in a dose-dependent and time-dependent manner with little or negligible cytotoxicity. Our results indicate that these absorption enhancers can widen the tight junction, which is a dominant paracellular absorption route of hydrophilic compounds . It is highly possible that these absorption enhancers can be applied as pharmaceutical excipients to improve the transport of macromolecules and hydrophilic drugs having difficulty in permeability across the intestinal epithelium.

  • 본 연구는 직접메탄을 연료전지(Direct Methanol Fuel Cell)에 적용가능한 양이온교환막 개발에 관한 것으로 poly(vinyl alcohol) (PVA)에 가교제로 poly(acrylic acid-co-maleic acid) (PAM)와 hydroquinonesulfonic acid (HQSA)를 이용하여 가교제의 함량을 변화시키면서 막을 제조하였다. 제조한 막은 가교제의 함량 변화에 따라 메탄을 투과도, 이온전도도를 측정하였으며 기본적인 이온교환막의 특성인 함수율, 이온교환용량 그리고 고정이온농도 등을 측정하였다. PAM 함량이 증가함에 따라 메탄을 투과도와 이온전도도 및 함수율이 조금 증가하는 추세를 보이다 9 wt%부터 감소하는 경향을 보였는데 이는 PAM의 친수성기보다는 가교효과의 영향이라 사료되며 HQSA 함량을 변화시켰을 때는 이온전도도, 함수율 그리고 이온교환용량이 전반적으로 증가하였는데 그 증가폭은 미비하였다.

  • 잔류성 유기오염물질인 과불화합물 중 perfluorooctanesufonic acid (PFOS)와 perfluorooctanoic acid (PFOA)의 잔류분석은 환경부 "잔류성 유기오염물질 공정시험기준"에 따라 hydrophilic-lipophilic balance (HLB) solid phase extraction (SPE) 전처리 후 LC-$MS^n$를 이용해 정량 분석하고 있다. 본 연구에서는 환경부 설정 공정시험법에 따라 농업용수 전처리 후, 시험법에서 제거하지 못한 미지의 불순물을 Envi-Carb$^{TM}$을 통해 회수율에 영향을 주지 않고 손쉽게 제거하였다. 또한, 과불화합물 분석에 사용되는 LC-$MS^n$ 중 quadrupole-time-of-flight mass spectrometry (qTOFMS)에서 측정된 PFCs의 정량한계를 평가한 결과, 시험에 사용된 장비간 정량한계 편차가 크게 관찰되었으나, 시험대상 장비 모두 농업용수 중 ng/L 수준으로 잔류하는 과불화합물의 정밀 잔류분석에 사용가능 한 것으로 확인되었다.

물-알코올 혼합물의 분리를 위한 알긴산막에 관한 연구 (A Study of Alginic acid Membrane for the Separation of Water-Alcohol Mixtures)

  • 음이온성 고분자 전해질의 하나인 알긴산을 이용 하여 알긴산막의 분리성능을 개선함과 동시에 막의 얀정성을 높이고, 막과 분리 대상물과의 화학적 친 화성을 제어하기 위하여 막의 가능기에 다양한 금속 이온으로 가교시킨 금속복합체 막을 제조하고, 그막 들이 투과증발 특성에 미치는 영향을 조사하여 다음 과 같은 결론을 얻였다. 1) 금속복합체 막들 중 IA족 금속이온으로 치환 한 경우에 저농도 에탄올 수용액에서 크게 팽윤되거 나 쉽게 부스러지는 경향이 있어 투과증발막으로 샤 용할 수 없었다. 2) 알긴산막에 비해 2가 금속으로 가교한 금속복 합체 막의 경우 막의 안정성 및 분라성능이 크게 향 상되는 것을 알 수 있었다. 3) 2가 금속 복합체 막들의 경우 분리계수는 Na a alg 막에 비해 약간 증가하는 경향을 나타낸 반면, 투과속도는 비슷한 경향을 나타내였다. 4) Ca-alg 금속복합체 막의 경우에 다른 금속복 합체 막들에 비하여 분리계수가 크게 증가하여 가장 우수한 성능을 나타내였다. 5) Ca-alg막의 경우 90wt.% 에탄올 수용액에서 분리계수는 6,420이었으며 투과속도는 $0.269kg/m^2.hr$로써 우수한 생능을 나타내었다.

Fabrication of nanoaggregates of triple hydrophilic block copolymers by binding of ionic surfactants

  • Nanoaggregates of triple hydrophilic block copolymers comprised of poly(ethylene oxide), poly(sodium 2-acrylamido)-2-methylpropanesulfonate), and poly(methacrylic acid) (PEO-PAMPS-PMAA) and the cationic surfactant, dodecyltrimethylammonium chloride (DTAC) have been fabricated. The formation of $^{\circ}^{\circ}$the nanoaggregates is based on electrostatic interaction of sulfonate and carboxylate groups of PAMPS and PMAA blocks with the cationic surfactant, which results in insolubilization of these blocks. The formation of micelle is observed by dynamic light scattering measurements. Binding of DTAC to the anionic blocks of PEO-PAMPS-PMAA is confirmed by electrophoresis measurements.

  • In summary, we have demonstrated soft lithographic application of HA (or polysaccharides in general) by means of ${\mu}CP$ and molding and constructed well-defined patterns of proteins and cells of various substrates including glass, silicon dioxides, poly(HEMA), polystyrene culture dishes, and biodegradable PLGA. These results suggest that HA could be used as a general platform on hydrophilic substrates.
