Radiation Grafting of Hydrophilic Monomers onto Polyester

  • Radiation grafting of acrylic acid and 4-vinylpyridine at room temperature has been studied by an impregnation method to improve the hygroscopic properties, the antistatic behavior and the dyeability of polyester fabric. Polyester fabric was impregnated with acrylic acid or aqueous emulsion of acrylic acid-4-vinylpyridine by immersion at 25$^{\circ}$or 7$0^{\circ}C$. The impregnated fabric was irradiated under nitrogen gas with ${\gamma}$-rays from Co-60. When acrylic acid grafted polyester fabric was treated with sodium carbonate, calcium acetate and potassium persulfate, tne rate of water absorption was increased and most parts of polyacrylic acid formed were extracted off from the fabric with 0.1% solution of sodium hydroxide at 10$0^{\circ}C$. In the case of the impregnation of a mixture of acrylic acid and 4-vinylpyridine the petcent of grafting has been shown to be proportional to the ratio of 4-VP/AA and radiation dost. Estimating by contact angle measurements of water on the various polymer surfaces, the antistatic behavior was decreased with the increase of grafting percent. The investigation of electron micrograph disclosed the existence of certain type of discontinuities in the acrylic acid grafted polyester fiber which was treated with various salts.

  • As an effective separation method for biomolecules, aqueous two-phase systems based on ionic liquids were suggested. Hydrophobic ionic liquids are more expensive and viscous in spite of their usage in the ionic liquid/water biphasic extraction compared with hydrophilic ionic liquids. In case of aqueous two-phase systems using hydrophilic ionic liquids, they can be diluted in aqueous phase. Experimental results show that aqueous two phase systems can be formed by adding appropriate amount of ionic liquids to aqueous salts solutions. The viscosity of ionic liquid aqueous phase is proportional to the cation chain length in ionic liquids. It is founded that the ionic liquid based aqueous two phase systems are effective for the separation of biomolecules such as acrylic acid.

Molecular cloning of a rhoptry protein (ROP6) secreted from Toxoplasma gondii

  • Monoclonal antibody (mAb) Tg786 against Toxoplasma gondii has been found to detect a 42-kDa rhoptry protein (ROP6) which showed protease activity and host cell binding characteristics after secretion. Using the mAb, a colony containing a 3'-UTR was probed in a T. gondii cDNA expression library. A full length cDNA sequence of the rhoptry protein was completed after 5'-RACE, which consisted of 1,908 bp with a 1,443 bp ORF. The deduced amino acid sequence of ROP6 consisted of a polypeptide of 480 amino acids without significant homology to any other known proteins. This sequence contains an amino terminal stop transfer sequence downstream of a short neutral sequence, hydrophilic middle sequence, and hydrophobic carboxy terminus. It is suggested that the ROP6 is inserted into the rhoptry membrane with both N- and C-termini.

Screening, Characterization, and Cloning of a Solvent-Tolerant Protease from Serratia marcescens MH6

  • solvent-tolerant bacterium strain, MH6, was isolated by hydrophilic organic solvent DMSO enrichment in the medium and identified as Serratia marcescens. The extracellular protease with novel organic-solvent-stable properties from strain MH6 was purified and characterized. The molecular mass of the purified protease was estimated to be 52 kDa on SDS-PAGE. The open reading frame (ORF) of the MH6 protease encoded 504 amino acids with 471 amino acid residues in the mature protease. Based on the inhibitory effects of EDTA and 1,10-phenathroline, the MH6 protease was characterized as a metalloproteinase. The enzyme activity was increased in the presence of $Ni^{2+}$, $Mg^{2+}$, and $Ca^{2+}$. The protease could also be activated by the nonionic surfactants Tween 80 (1.0%) and Triton X-100 (1.0%). The protease showed remarkable solvent stability in the presence of 50% (v/v) solutions of long-chain alkanes and long-chain alcohols. It was also fairly stable in the presence of 25% solutions of hydrophilic organic solvents. Owing to its high stability in solvents and surfactants, the MH6 protease is an ideal candidate for applications in organic catalysis and other related fields.

  • Preliminary study was perfomed to develop a biocompatible filter material using radiation energy. Electrosppined PLLA nano sheets were surface-modificated with hydrophilic groups(acrylic group) by using radiation. The physico-chemical and morphological characteristics of modified PLLA sheets were measured by ATR, SEM, contact angle, and hydrophilic (acryl group) introduction rate (TBO). As a result, there was no morphological(fiber structure) structure change due to radiation, and it was confirmed that an acrylic group was successfully introduced onto PLLA fiber sheet by radiation.

  • Objectives : Hyaluronic acid(HA) is a mucopolysaccharide, occuring naturally in living organisms. It is one of the most hydrophilic molecules, so it has been known as being related to skin hydration and anti-aging. The purpose of this study is to examine the effects of Angelica acutiloba ethanol extract on hyaluronic acid synthesis. Methods : To determine cytotoxicity and hyaluronic acid synthase 2 gene expression, hyaluronic acid production in HaCaT cells, MTT assay and RT-PCR ELISA was used. Results : There was no cytotoxicity in $50{\mu}g/ml$ concentration Angelica acutiloba extract in MTT assay. Hyaluronic acid synthase 2(HAS2) gene expression was increased by all treated concentration Angelica acutiloba extract. Hyaluronic acid production was higher in $50{\mu}g/ml$ & $100{\mu}g/ml$ concentration Angelica acutiloba extract than control group. Conclusions : Hyaluronic acid production was increased by Angelica Acutiloba extracts. Therefore, We suggest that Angelica acutiloba can make a contribution to the moisturing effect on human skin. Adhesion improvement between metals and fluoropolymers by ion assisted reaction (이온보조반응에 의한 금속과 불소계 고분자의 접착력 증진)

  • Polyvinylidenefluoride and Polytetrafluoroethylene have been irradiated by 1 keV Ar+ ion beam in an $O_2$ environment. Hydrophilic functional groups (such as -(C-O)-,-(C=O)-,-(C=O)-O- and so on) were formed on fluoropolymers. Contact angles of water to PVDF were reduced from $75^{\circ}$ to $31^{\circ}$. Re-increase of contact angle was originated from carbonization phase in case of high dose irradiation above $1{\times}10^{16} Ar^+cm^2$. Contact angles to PTFE decreased at low dose irradiation and were exaggerated to about $140^{\circ}$ due to cone type surface at high dose irradiation. Hydrophilic functional groups have played an important role on adhesion between metal and fluoropolymers by acid-base interaction and chemical bond formation. Adhesion of Pt/PVDF was enhanced by acid-base interaction because Pt is inert metal. Chemical bond formation between Cu and PTFE could enlarge the adhesion strength of Cu/PTFE.

  • Metal distribution in the reaction products and system of reactions between organo chloro acid or ester and zinc, silicon, magnesium, and tin under acetonitrile, dioxane, and toluene solvent were determined by means of radioactive tracer prepared by means of a (n, ${\gamma}$) reaction. It was found that the solubility of the organo halogen metal complex was markedly increased in a hydrophilic solvent and was decreased in a nonpolar solvent which resulted in an increased metal distribution in the recovered metal or water washing of the recovered metal mixture. This was also true in the case of the reaction conducted in the presence of a carbonyl compound. The relative increase of the solubility of the metal complex in a hydrophilic solvent was in order of zinc, silicon, tin and magnesium, and in a nonpolar solvent, it was in order of silicon, tin, magnesium, and zinc. There was no formation of organo metallics throughout the reaction sequence. The result was discussed and the observed solvent influence on the present reaction path was criticized.

  • O/W emulsions were prepared by adding water to the solution containing amphiphilic resin and the mixed emulsifier of cetyl alcohol polyoxyethylene(20) sorbitan monooleate. Phase behavoir of these emulsions was studied at various HLB(Hydrophilic Lipophilic Balance) values and temperatures. The polar oil emulsion containing the amphiphilic resin showed improved phase stability at various temperatures. Model compounds which contain one of the functional groups in the amphiphilic resin were used in the polar oil phase in order to study the effect of interaction between the functional group and the emulsifier on the phase stability of emulsion. These model compound emulsions showed the phase stability order of poly(acrylic acid)

Efficiency of Antioxidants on Types of Substrate Systems (기질 system의 유형에 따른 항산화제의 효과에 관한 연구)

  • The purpose of this study was to investigate any differences in the efficiency of various antioxidants for the three types of substrates such as corn oil in water (O/W) emulsion, water in com oil (W/O) emulsion, and bulky corn oil. ${\alpha}$-Tocopherol (${\alpha}$-Toc) at 0.01 or 0.02%, ascorbic acid (AsA), ascorbyl palmitate (AP), and BHT at 0.02% were added separately to the prepared O/W emulsion, W/O emulsion, and bulk oil, and their antioxidative effects were compared. The mixture of ${\alpha}$-Toc ind AsA or AP at the level of 0.02% also was tested to observe any synergistic effect. Oxidation was made by storing at 42${\pm}$1$^{\circ}C$ for 25 days and the oxidative stability was determined by peroxide value and conjugated dienoic acid with time fluctuation of storage. The results were as follows: 1. In case of O/W emulsion, the order of antioxidative effect was AP> ${\alpha}$-Toc+AP>${\alpha}$-Toc+AsA>AsA>BHT. 2. In case of W/O emulsion, the order of antioxidative effect was AsA>AP>${\alpha}$-Toc+AsA>BHT. ${\alpha}$-Toc+AP mixture showed the prooxidant effect rather than synergistic effect. 3. In case of bulk oil, the order of antioxidative effect was AsA>AP>${\alpha}$-Toc+AsA>${\alpha}$-Toc+AP\ulcornerBHT. Therefore, AsA, a hydrophilic antioxidant, was more effective in W/O emulsion system than in O/W emulsion system, while the opposite trend was found in AP, a lipophilic antioxidant. AsA, a hydrophilic antioxidant, was more efficient in bulk oil of anhydrous substrate. ${\alpha}$-Toc showed prooxidant effects in all substrates.
