We introduce our experiences of the design and implementation of the Page Together system that has expanded hyperlink metaphor to utilize human resources in the web. This system supports that a user connects with others in the web, communicates through video/audio channel, navigates same web pages simultaneously and cooperates some work on Internet. For these functions, it comprises Collaborative Browsing Module (CBM), Multimedia Conferencing Module(MCM) Data Conferencing Module(I)CM) and Multi Protocol Interface(MPI). We adopted three standard protocols, IEC, H.323 and T.120 for each nodule and it allows developers to use them easily. We also defined MPI to synchronize information of session among modules. Each module exchanges information each other in session creating process and session terminating process. After a session is created once, each module works independently as its won protocol. Interferences among modules are reduced as minimizing to exchange information. We also introduce a web site that provides web board service based on the Page Together system. A user may post a notice with a link to himself/herself on our web board. After then, if someone read that notice and has any question about it, he or she can try to connect to the writer as clicking the link in that notice and communicate each other. This service site shows that our system can be applied to diverse internet services such as distance teaming and distance conference.
Unlike mouse results, cloning efficiency of nuclear transfer from porcine induced pluripotent stem cells (piPSCs) is very low. The present study was performed to investigate the effect of cell cycle inhibitors on the cell cycle synchronization of piPSCs. piPSCs were generated using combination of six human transcriptional factors under stem cell culture condition. To examine the efficiency of cell cycle synchronization, piPSCs were cultured on a matrigel coated plate with stem cell media and they were treated with staurosporine (STA, 20 nM), daidzein (DAI, $100{\mu}M$), roscovitine (ROSC, $10{\mu}M$), or olomoucine (OLO, $200{\mu}M$) for 12 h. Flow Cytometry (FACs) data showed that piPSCs in control were in G1 ($37.5{\pm}0.2%$), S ($34.0{\pm}0.6%$) and G2/M ($28.5{\pm}0.4%$). The proportion of cells at G1 in DAI group was significantly higher than that in control, while STA, ROSC and OLO treatments could not block the cell cycle of piPSCs. Both of viability and apoptosis were affected by STA and ROSC treatment, but there were no significantly differences between control and DAI groups. Real-Time qPCR and FACs results revealed that DAI treatment did not affect the expression of pluripotent gene, Oct4. In case of OLO, it did not affect both of viability and apoptosis, but Oct4 expression was significantly decreased. Our results suggest that DAI could be used for synchronizing piPSCs at G1 stage and has any deleterious effect on survival and pluripotency sustaining of piPSCs.
This study was designed to examine the tissue levels of interleukin-$1{\alpha}$(IL-$1{\alpha}$), interleukin-$1{\beta}$(IL-$1{\beta}$) and tumor necrosis factor-${\alpha}$(TNF-${\alpha}$) in inflamed human dental pulps and periapical lesions, and to determine the relationship between each cytokine and pulpal and periapical pathosis. The pulps used in this experiment, were obtained in routine endodontic treatment and the periapical lesions in periapical surgery after clinical diagnoses were performed. These specimens were divided into four groups as normal pulp group(control group, n=9), acute pulpitis group(n=g), chronic pulpitis group(n= 10) and periapical lesion group(n= 18) and stored in liquid N2. For extract preparation, tissues were finely minced with a scalpel, and the fragments were incubated in $0.5m\ell$ homogenizing buffer (0.1 mol/L potassium chloride, 0.02 mol/L TRIS; pH=7.6) for two hours and grinded with glass homogenizer. Debris was removed by centrifugation and supernatants were immediately tested with enzymelinked immunosorbent assay (ELISA, R&D Co., Minneapolis, USA). Following results were obtained; 1. The concentrations of IL-$1{\alpha}$ in all experimental groups were significantly higher than in control group(p<0.05). And the concentrations of IL-$1{\alpha}$ in periapical lesion group were somewhat higher than in two pulpitis groups, but the differences among those groups were not stastically significant (p>0.05). 2. The concentrations of IL-$1{\beta}$ in all experimental groups were significantly higher than in control group (p<0.05), and all the experimental groups expressed similar concentrations. 3. The concentrations of TNF-${\alpha}$ in all experimental groups were higher than in control group but only the differences between chronic pulpitis group and control group were statistically significant(p<0.05). And the concentrations of TNF-${\alpha}$ in acute and chronic pulpit is groups were higher than in periapical lesion group but only the differences between chronic pulpitis group and periapical lesion group were statistically significant (p<0.05). 4. There was significant correlation only between IL-$1{\alpha}$ and IL-$1{\beta}$ in periapical lesion group (p<0.05).

Ultraviolet is the one of the main environmental factors promoting aging process via increased intracellular generation of reactive oxygen species (ROS) and decreased expression of endogenous antioxidant enzymes and molecules. Therefore, in this study, we tried to search for natural skin-protective antioxidant materials from marine origins (Porphyra Thalli, Laminariae japonicae thallus, Ostreae Concha, Sargassum Thallus, Undaria thallus, Haliotidis Concha, Codium thalli, Syngnathoides biaculeatus, Hippocampus, Stichopus Stichopus, Thalli, Hizikia fusiforme thalli) which exhibit free radical scavenging activity and protect against UVB-induced cytotoxicity and oxidative cell death. Free radical scavenging activity was shown in order of Undaria thallus. Sargassum Thallus, Laminariae japonicae thallus, Hippocampus, Haliotidis Concha, Ostreae Concha, Syngnathoides biacuJeatus. In another experiment, UVB-induced cytotoxicity and cell death were effectively suppressed by treatment of Sargassum Thallus, Haliotidis Concha, Codium thalli, or Hippocampus water extract. Furthermore, UVB-induced cell death was mediated by intracellular accumulation or ROS, which was significantly inhibited by treatment with aforementioned extracts. The protective effect of these marine natural products seemed to be mediated by increased expression of antioxidant enzymes such as catalase, superoxide dismutase, and heme oxygenase-1. These results suggest that Sargassum Thallus, Haliotidis Concha, Codium thalli, and Hippocampus may have preventive and protective potentials as new functional cosmetics against oxidative stress-mediated skin damages and aging with antioxidant properties.
The high-purity germanium gamma-ray spectrometer was used to measure the radioisotope in surface water of lakes in a Chinee petroleum industrial area. 92 samples were collected from surface water of three lakes. Activity concentrations of 232Th, 226Ra and 40K in three lakes were measured, distributed in the range of 101.8-209.4, 192.1-224.9 and 335.0-548.9 mBq/L, respectively. Results were all within the limits of WHO and China. Potential environmental and health risks were assessed by calculating some radiation hazard indicators, radium equivalent index, annual effective dose, excess lifetime cancer risk, absorbed dose rate, external hazard index, internal hazard index, annual gonadal dose equivalent, activity utilization index and representative gamma index, which ranged 0.38-0.54 Bq/L, 0.06-0.08 mSv/y, 0.23 × 10-3-0.31 × 10-3, 0.17-0.24 nGy/h, 1.01 × 10-3-1.46 × 10-3, 1.55 × 10-3-2.02 × 10-3, 1.16-1.66 μSv/y, 3.13 × 10-3-4.45 × 10-3 and 2.60 × 10-3-3.77 × 10-3. The results were all at acceptable levels, meaning no impact on human health. The relationship between the electrical conductivity of surface water and the activity concentration of 232Th, 226Ra and 40K was evaluated. The electrical conductivity value was 0.241-0.369 mS/cm, showing a significant correlation coefficient between 226Ra and 40K and electrical conductivity. Multivariate statistical methods were used to determine the relationship between the activity concentrations of 232Th, 226Ra, and 40K, radiation hazard indicators and electrical conductivity.

In this study, the antioxidative effects, inhibitory effects on tyrosinase, and component of non-fermented and fermented Lavandula angustifolia extracts were investigated. The ethyl acetate fraction of fermented extract (5.95 ${\mu}g/mL$) showed the most prominent the free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}$). Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of L. angustifolia extracts on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system were investigated using the luminol-dependent chemiluminescence assay. The ethyl acetate fraction of fermented extract (1.45 ${\mu}g/mL$) showed the most prominent ROS scavenging activity. The protective effects of extract/fractions of L. angustifolia on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The L. angustifolia extracts suppressed photohemolysis in a concentration dependent manner (1 ${\sim}$ 50 ${\mu}g/mL$). The inhibitory effect of L. angustifolia extracts on tyrosinase was investigated to assess their whitening efficacy. Inhibitory effects ($IC_{50}$) on tyrosinase were determined with ethyl acetate fraction of L. angustifolia extract (144.80 ${\mu}g/mL$) and ethyl acetate fraction of fermented extract (122.40 ${\mu}g/mL$). Fractions of ethyl acetate and fermented extracts showed both 3 band in TLC and 3 peaks, 2 peaks in HPLC (340 nm), respectively. In each chromatography, fractions of ethyl acetate both from non-fermented and fermented L. angusfifolia have rosmarinic acid in common. These results indicate that the component and content of non-fermented and fermented extracts of L. angustifolia are different. Both of the extract of L. angustifolia can be used as an antioxidant.

This study was conducted to examine the effects of energy substrates in different conoentration of carbohydrates in the human oviduct and uterus on the in vitro development of mouse 2-cell embryos. Two-cell embryos were collected from ICR female mice at $46{\sim}50\;hr$ after 5 IU hCG injection and cultured in three different media [control: 0 mM, Guoup A: glucose (G) 0.5 mM + pyruvate (P) 0.32 mM + lactate (L) 10.5 mM, Group B: G 3.15 mM + P 0.1 mM + L 5.83 mM] for 72 hr. Rates of morula formation of group A (72.3%) and B (56.6%) were significantly higher higher (p<0.05) than that of control (34.9%) at 24 hr. However, blastocyst rate was significantly higher (p<0.05) in control (51.8%) than group A (39.8%) and B (28.9%) at hr. At 72 hr, no differences were found in the number of zona-intact, zona-escape and total blastocysts among groups. Mean and ICM cell numbers were significantly higher (p<0.05) in group A (78.0, 13.4) and B (64.4, 11.8) than control (53.1, 5.7), respectively, The percent of ICM were significantly higher (p<0.05) in group A (22.9%) and B (23.7%) than control (14.2%). No differences were round in the TE cell numbers ($34.1{\sim}45.1$). The ICM:TE ratio was significantly higher $34.1{\sim}45.1$ in control (1:6.0) than group A (1:3.4) and B (1:3.4). This study shows that energy substrates added to culture media especially, the oviductal level of carbohydrates increase the developmental capacity of 2-cell mouse embryos.

In this study, the antioxidative effects and inhibitory activities on tyrosinase of Lindera obtusiloba Blume (L. obtusiloba Blume) extracts were investigated. 50 % ethanol extract, ethyl acetate and aglycone fractions of L. obtusiloba Blume were used in experiments. The DPPH (1,1-diphenyl-2-picrylhydrazyl) scavenging activities of ethyl acetate fraction of L. obtusiloba Blume was higher than (+)-${\alpha}$-tocopherol, known as a typical antioxidant. Reactive oxygen species (ROS) scavenging activities of extract and fractions of L. obtusiloba Blume on ROS generated in $Fe^{3+}$-EDTA/$H_2O_2$ system were similar to L-ascorbic acid, well known as a strong antioxidant. The cellular protective effects of 50 % ethanol extract and ethyl acetate of L. obtusiloba Blume on the rose-bengal sensitized photohemolysis of human erythrocytes were increased in a concentration dependent manner ($1{\sim}25{\mu}g/mL$). Ethyl acetate fraction in $10{\mu}g/mL$ concentration showed the most protective effect among extracts (${\tau}_{50}$ = 361.0 min). The inhibitory effects on tyrosinase of ethyl acetate and aglycone fractions were higher than arbutin, known as a whitening agent. These results indicate that L. obtusiloba Blume extracts can be used as antioxidant, and could be applicable to functional cosmetic ingredient.
The aim of this study was to evaluate various aspects of Vitex negundo L. leaf extract, such as the antioxidative activity, tyrosinase inhibitory effects, and inhibitory activities on ${\alpha}$-MSH induced melanogenesis, and active component analysis. The DPPH (1, 1-diphenyl-2-picrylhydrazyl) scavenging activities ($FSC_{50}$) of the ethyl acetate fraction and aglycone fraction of V. negundo L. leaf extract were $14.51{\mu}g/ml$ and $13.96{\mu}g/ml$, respectively. A luminol-dependent chemiluminescence assay revealed that the reactive oxygen species (ROS) scavenging activity ($OSC_{50}$) of the aglycone fraction of V. negundo L. leaf extract on ROS generated in an $Fe^{3+}$-$EDTA/H_2O_2$ system was the most prominent at $0.22{\mu}g/ml$. The protective effects of the extracts fractions of V. negundo L. leaf against the rose-bengal sensitized photohemolysis of human erythrocytes were increased in a concentration dependent manner ($1{\sim}50{\mu}g/ml$). In particular, there were greater protective effects of the aglycone fraction on the cellular membrane than that of the fat-soluble antioxidant (+)-${\alpha}$-tocopherol. The inhibitory effects ($IC_{50}$) on mushroom tyrosinase were the highest for the ethyl acetate fraction ($IC_{50}$ = $48.58{\mu}g/ml$). The inhibitory effect on ${\alpha}$-MSH induced melanogenesis in B16 melanoma cells was 41.80% at $50{\mu}g/ml$ of ethyl acetate fraction. Active component analyses by TLC, HPLC and LC/ESI-MS revealed luteolin and isoorientin. These results indicate that V. negundo L. leaf extract can be used as an antioxidant for ROS scavenging. Particularly, the luteolin and isoorientin of the ethyl acetate fraction may be applicable to new whitening cosmetics because of its inhibitory effect on mushroom tyrosinase and ${\alpha}$-MSH induced melanogenesis in B16 melanoma cells.

In this study, the antioxidative effects and inhibitory activities of unripened fruit extract of Rubus coreanus Miquel (R. coreanus Miquel) on tyrosinase were investigated and the potential applicability as a cosmeceutical ingredients was evaluated. All experiments were performed with 50% ethanol extract, ethyl acetate fraction and aglycone fraction of unripened fruit extract of R. coreanus Miquel. The DPPH (1,1-diphenyl-2-picrylhydrazyl) scavenging activites ($FSC_{50}$) of 50% ethanol extract (6.56 ${\mu}g/mL$) and ethyl acetate fraction (6.14 ${\mu}g/mL$) of unripened fruit extract of R. coreanus Miquel were higher than (+)-${\alpha}$-tocopherol (8.98 ${\mu}g/mL$), which is known as a typical hydrophobic antioxidant. Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of 50% ethanol extract (0.83 ${\mu}g/mL$), ethyl acetate fraction (0.84 ${\mu}g/mL$) and aglycone fraction (1.13 ${\mu}g/mL$) of R. coreanus Miquel on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system were higher than L-ascorbic acid (1.5 ${\mu}g/mL$), which is known as s typical hydrophilic antioxidant. The cellular protective effect of extract and fraction of unripened fruit extract of R. coreanus Miquel on the rose bengal sensitized photohemolysis of human erythrocytes was increased in a concentration dependent manner (1 ~ 50 ${\mu}g/mL$). And 50% ethanol extract in 50 ${\mu}g/mL$ showed the most protective effect among extracts (${\tau}_{50}$ = 296.3 min). The inhibitory effects on tyrosinase of ethyl acetate and agylcone fractions were higher than arbutin. These results indicate that unripened fruit extracts of R. coreanus Miquel can be applied to antioxidant scavenging ROS including radical as an alternative whitening agent to replace arbutin.
