Microarray and Quantitative PCR Analysis of Gene Expression Profiles in Response to Treatment with Tomato Leaf Extract in MCF-7 Breast Cancer Cells

  • We previously found cytotoxic effects of tomato leaf extract (TLE) on the MCF-7 breast cancer cell line. The aim of this study was to ascertain the molecular mechanisms associated with the usage of TLE as an anticancer agent by microarray analysis using mRNA from MCF-7 breast cancer cells after treatment with TLE for 1 hr and 48 hrs. Approximately 991 genes out of the 30,000 genes in the human genome were significantly (p<0.05) changed after the treatment. Within this gene set, 88 were significantly changed between the TLE treated cells and the untreated MCF-7 cells (control cells) with a cut-off fold change >2.00. In order to focus on genes that were involved in cancer cell growth, only twenty-nine genes were selected, either down-regulated or up-regulated after treatment with TLE. Microarray assay results were confirmed by analyzing 10 of the most up and down regulated genes related to cancer cells progression using real-time PCR. Treatment with TLE induced significant up-regulation in the expression of the CRYAB, PIM1, BTG1, CYR61, HIF1-${\alpha}$ and CEBP-${\beta}$ genes after 1 hr and 48 hrs, whereas the TXNIP and THBS1 genes were up-regulated after 1 hr of treatment but down-regulated after 48 hrs. In addition both the HMG1L1 and HIST2H3D genes were down-regulated after 1 hr and 48 hrs of treatment. These results demonstrate the potent activity of TLE as an anticancer agent.

  • With a view to developing male dress forms for educational purposes, human body measurement was made for male adults in their twenties residing in Busan, Korea. Based on the results, the following conclusion was obtained: 1. Comparison d the measurement in Busan and the data of the 5th Korean physical dimensions(Size Korea) In the comparison of the Busan male adults in their 20s with national physical dimensions(Size Korea) by way of the Mollison relational deviation line, all the 28 items revealed differences less than 0.7. As the Busan sample reflects the body type d the average Korean men in their 20s, Busan's measurement results can be safely used as the data for dress form development. 2. Results of front body type classification According to the factor analysis, seven factors of the front body type were produced to explain 77.25%. The factors were shoulder angles, waist height, hip shapes, chest shapes, proportion of body(chest, waist, hip)width to shoulder width and so on. Cluster analysis brought about three somatotype groups. First, the body type with the least differences in hip-waist width, shoulder-waist with, and hip angles (24,23% in appearance) was named Type H. Second, the body type with the most chest-waist width and hip angles (38.66% in appearance) was called Type Sam X Third, the body type with the greatest shoulder-chest width, shoulder-hip width, and shoulder angles (37.11% in appearance) was termed Type Y.

  • Ginseng is one of the most widely used medicinal plants, particularly in East Asian countries. Certain fractions or purified ingredients of ginseng have been shown to exert inhibitory effects on growth of cancer cells in culture or on tumorigenesis in experimental animals. Moreover, a recent epidemiologic study reveals that ginseng intake is associated with a reduced risk for environmentally related cancers such as esophageal, gastric, colorectal, and pulmonary tumors. Heat treatment of Panax ginseng C. A. Meyer at the temperature higher than that applied to the conventional preparation of red ginseng yielded a mixture of saponins with potent antioxidative properties. Thus, the methanol extract of heat-processed ginseng (designated as'NGMe') attenuated lipid peroxidation in rat brain homogenates induced by ferric ion or ferric ion plus ascorbic acid. Furthermore, the extract protected against strand scission in f Xl 74 supercoiled DNA Induced by UV photolysis of H2O2 and was also capable of scavenging superoxide generated in vitro by xanthine/xanthine oxidate or in differentiated human promyelocytic leukemia (HL-60) cells by the tumor promoter,12-0-tetvade- canoylphorbol-13-acetate (TPA). Since tumor promotion is closely linked to oxidative stress, we have determined possible anti-tumor promotional effects of NGMe on two-stage mouse skin tumorigenesis. Topical application of NGMe onto shaven backs of female ICR mice 10 min prior to TPA significantly ameliorated skin papillomagenesi s initiated by 7,12-dimethylbenz (a) anthracene (DMBA).'Likewise, TPA-induced epidermal ornithine decarboxylase activity and elevation of tumor necrosis factor-a were suppressed signifies%fly by NGMe pretreatment. NGMe topically applied onto surface of hamster buccal pouch 10 min before each topical application of DMBA inhibited oral carcinogenesis by 76olo in terms of multiplicity. Taken together, these results suggest that processed Panax ginseng C. A. Meyer has potential cancer chemopreventive activities.

Which Couple Has More Shared Leisure Time? : The Exploration of Shared Leisure Time and Marital Relationship in Korea (어떤 부부가 함께 여가시간을 보내는가? : 부부공유여가시간의 현황과 부부관계의 질에 관한 탐색)

  • In recent years, leisure time has received increasing attention in Korean society. Despite the growing interest, however, there are very few studies that explored shared leisure time, especially among married couples. In this light, the present study examined variations in shared leisure time by demographic characteristics and associations between shared leisure time and quality of marital relationship. We used the third wave of the national data KLoWF (Korean Longitudinal Survey of Women and Families, 9,997 19-64 years old women). The study sample included 5,965 married women with no prior history of divorce. Results showed that Korean wives reported very little shared leisure time with their husbands. Given that the mean of the shared leisure time reported by employed women was significantly lower than that of the un-employed, we speculated that the time constraint may be one of the major barriers to having leisure time with one's spouse among Korean married couples. Married women in their 30's had the most frequent couple leisure time. The younger wives in their 20s had more cultural leisure time with their husbands, but sharing outdoor leisure time was not related to their age. The higher level of education for women was associated with greater shared active leisure time. However, visiting a husband's family often took place regardless of the wives' education level. After controlling for age, years of education, job status and annual family income, shared active leisure time (i.e., cultural activity and exercise) was positively related to marital satisfaction. Visiting one's own family, however, was not considered a leisure activity for Korean wives. Rather, frequent family visits were related to lower marital satisfaction. Findings shed light on how leisure life of married women with spouses are intertwined with not only their socioeconomic status and but also marital relationship in culturally unique ways.

Study on the Performances of Air Flow Fate Effect on a Structured Packed Tower at Adiabatic Condition in a Liquid Lithium Chloride Cooling System

  • The liquid desiccant air-conditioning system has been proposed as an alternative to the conventional vapor compression cooling systems to control air humidity. The complete system of liquid desiccant air-conditioning system is consisted two main components those are humidifier (regeneration) and dehumidifier. Humidifier part is connected to the load when summer season which is the air condition is hot and humid have to be turned into comfort condition on human. This paper purpose is performances study of air flow rate effect on a structured packed tower on cooling and dehumidifier system using liquid lithium chloride as the desiccant. Experimental apparatus used in this present study is consisted of three components those are load chamber, packed tower and chiller. Load chamber’s volume is $40m^3$, and packed tower dimension is cubic with length 0.4m occupied with packed column. Totally, 15 experimental has done using 5 times repeat on each variable of air velocity that varying on 2m/s, 3m/s and 4m/s with other conditions are controlled. Air inlet initial temperature and relative humidity are set respectively on $30^{\circ}C$ and 52%, desiccant flow rate is 0.63 kg/s, desiccant temperature is $10^{\circ}C$ and desiccant concentration is 0.4. The result of this study shows that averagely, the moisture removal rate and the heat transfer rate are influenced by the air velocity. Higher air velocity will increase the heat transfer and decreasing the moisture removal rate. At adiabatic condition the air velocity of 2 m/s respectively is having the higher moisture removal rate acceleration then the air velocity of 3m/s and 4 m/s until the steady state condition.

Study on Utilization of Animal By-products from Food Processing by Enzyme Treatment (효소를 이용한 동물성 식품가공부산물의 식품 소재화 탐색)

  • In order to recycle animal by-products from food processing as food seasonings, pig bone (PB), chicken bone (CB) and tuna dark flesh (TDF) were studied. PB and CB extract prepared by hot water extraction for 18 h were hydrolyzed with protease and lipase. The DHs of PB and CB extract were increased to 70% and 80%, respectively, when Flavourzyme was treated after pancreatic enzyme treatment. When TDF was hydrolyzed with Alcalase and Flavourzyme, dry matter yield and total protein yield were around 22% and 9%, respectively. Also the free ammo acid content in hydrolysate reached up to 27% of total solid. The sensory properties of three hydrolysates containing 1% NaCl were, in order of their overall acceptance, TDF, PB and CB. Therefore, tuna dark flesh turned out to be the suitable animal by-product as raw material for seasoning ingredient.

  • Purpose: The aim of this study was to evaluate the validity of swept-source optical coherence tomography (SS-OCT) for detecting sealant microleakage. Materials and Methods: A total of 31 extracted sound human molar, assigned as suitable for sealant application, were chosen and divided into two groups: (1) no microleakage group and (2) microleakage group by applying sealant using the different methods. All specimens were immersed in 1% methylene blue for 24 h and sectioned to confirm the absence or presence of sealant microleakage as a gold standard method. Subsequently, all the sectioned specimens were digitally photographed using a microscope with a magnification of ${\times}50$. Presence of microleakage was evaluated on a 2-point rating scale. The association of histological method with conventional methods (visual and tactile assessment) and SS-OCT was assessed using a chi-squared test. The intra- and inter-examiner reliability was calculated using Cohen's Kappa. Results: The SS-OCT showed a sensitivity of 0.73 and a specificity of 1.00, while visual and tactile assessment showed a sensitivity of 0.67 and a specificity of 0.86. The inter-examiner reliability of SS-OCT was 0.79, whereas that of the visual and tactile assessment was 0.53. Conclusions: SS-OCT can be used to non-invasively detect sealant microleakage and to monitor pit and fissure sealant in the clinics.

A Study of matrix metalloproteinase-9 inhibitor in root bark of ulmus davidiana planchon (유근피내의 Matrix Metalloproteinase-9 활성 억제제에 관한 연구)

  • Several solvents were used to fractionate an extract obtained from the chapped root bark of Ulmus davidiana Planchon. The each fractionary part was condensed under reduced pressure and then examined to investigate the inhibitory effect on MMPs by modified gelatin zymography, where EA fraction showed the inhibition effect on the activity of MMPs. A compound showing inhibition effect on the MMPs was isolated and purified from EA fraction. Under IR, $^1H$- and $^{13}C$- NMR analyses it is very close to a catethin. This substance showed 48% inhibition effect on measurement of MMP-9 activity at 5 mM and 43% at 10 mM. To verify the effect of this substance on cells, human hepatoma, SK-Hep-1 cells as a cancer model, and Chang liver cells as a normal model were selected. MTT assay was performed to examine the cell viability by treatment of $1{\mu}L/mL$ of the purified substance on cells. The purified substance showed negligible toxicity on human liver cell line.

Comparative Analysis of Open- Spike between Excellent and Non-excellent Players in Volleyball (배구 우수선수와 비우수선수간의 오픈 스파이크 동작의 비교 분석)

  • This study aims at finding the structure of spike technique by analysing comparatively the spike action by excellent and by non-excellent players throughout the section from a flying jump to the time of landing for the correct analysis of spike action and tries to help athletes and coaches to execute a scientific training. For the objected person of this study, six of H College athletes three of excellent athletes and three of non-excellent athletes, presently registered as athlete with the Korea Volleyball federation) were chosen, and the factors of analysis were analysed upon performance time of action by section, human body centered displacement, change of articulation angle, speed change of articulation of the upper limbs, uniformity of the articulation of the upper limbs upon impact, etc. The conclusion of this study is as follow: 1. In the time required for taking action, it shows to take $1.067{\pm}0.057$ seconds for the group of excellent athletes and $1.034{\pm}0.033$ seconds for the group of non-excellent athletes. Although there was not big difference between two groups in the performance time of action, it showed that the group of excellent athletes takes longer compared to the group of non-excellent athletes. And it was found by the result of this study that the group of excellent athletes stays longer in the duration of flight. 2. In the displacements of horizontal movement and vertical movement, it was found that the group of excellent athletes have moved more than the group of non-excellent athletes in the horizontal movement of the center of human body 3. In the angles of wrist and knee, it was found that the excellent athletes have shown little than the non-excellent athletes in the entire sections, but that in the angle of elbow, the non-excellent athletes have shown bigger than the excellent athletes.. 4. In the speed of the articulation of the upper limbs upon impact, it was found that the group of excellent athletes have shown bigger than the group of non-excellent athletes, and that in the maximum value of the articulation of the upper limbs, the maximum value for the hand was indicated upon impact and that forearm and upper arm have shown the maximum value just before the impact. 5. In the uniformity of articulation of the upper limbs at the time of impact, the group of excellent athletes showed bigger than the group of non-excellent athletes in all the articulations.

Research on Professional Groups through Learning of Professional Game Players (전문가 집단 양성을 위한 프로게이머 발달 및 학습 모형 연구)

  • The current interests in e-sports is being extended to the fields of education these days. Professional game players, so called as 'Pro-Gamers', therefore, should be recognized as human resource for education, and the theoretical foundation for them needs to be established. This study examines informal learning styles, motivation, and interactions among professional game players in South Korea. The aim of this grounded theory study is to discover the trajectory of professional game players' experiences and explain what properties and interactions they are facing depending on the stage of the trajectory. This study conceptualizes educational meaning within and across the society of StarCraft Pro-Gamers, providing suggestions for the management of human resource using models constructed. Data was analyzed by interviewing 1 consultant, 2 directors and 9 Pro-Gamers. By analyzing the data, this study explored what learning strategies Pro-Gamers construct and apply in their trajectory as Pro-Gamers. It includes how they organize learning, how they formulate their motivation and goals, how they cooperate and compete, what curricula they adapt, how they become one of the ace players overcoming their slump, and how informal education works in practice in the interaction among members of a StarCraft Pro-Gamer team. Finally, in this paper the stage theory was presented. It is argued that when the stage of the players shifts (Stage Shifting). It also brings changes to proficiency properties, emotional properties, interactional properties and educational properties related to each stage. Stages are categorized by five levels: Enjoying, Struggling, Achieving, Slumping, and Recovering. Although each stage has its own properties, the stages are grouped by two main properties, one of which is a Communicative Stage and the other is a Practicing Stage.
