The chlorophyll-related compound pheophorbide a (Pa) was successively purified from an edible red seaweed, Grateloupia elliptica, using silica, octadecyl silica column chromatography and reversed phase-high-performance liquid chromatography, as well as the cell cycle inhibitory and apoptotic effects of Pa being investigated in U87MG glioblastoma cells. The Pa exhibited strong anticancer effects in the absence of direct photo-irradiation against various cancer cell lines, including U87MG, SK-OV-3, and HeLa cells. Among the cancer cells, the strongest anticancer activity of Pa exhibited on U87MG cells with $IC_{50}$ values of 2.8 ${\mu}g/ml$. In addition, Pa specifically had cytostatic activity on glioblastoma cells rather than human umbilical vein endothelial cells. Analysis of the cell cycle distribution showed that Pa induced G0/G1 arrest of U87 MG cells. In addition, arrested cells induced late apoptosis and DNA degradation under dark condition. These results suggest that Pa isolated from G. elliptica is a potential glioblastoma-specific anticancer agent without side effects on normal cells.
Prunus mume Sieb. et Zucc는 아시아 전역에 분포되어 있으며 전통적으로 약과 음식에 사용되어져 왔다. 이러한 매실은 각종 유기산, 무기질 및 페놀성분이 다량으로 함유되어 있는 것으로 알려져 있다. 지금까지의 매실 연구 동향은 항산화, 항암, 항균 등의 연구에만 치중되어 있고 혈관 신생에 대한 연구는 거의 이루어져 있지 않다. 혈관 신생은 전이성 암의 일반적인 특징으로 이를 통하여 부족한 산소와 영양분을 타 조직에서 가지고 온다. 이 연구에서는 매실 부탄올 분획물의 총 폴리페놀 및 플라보노이드 함량과 VEGF로 유도된 인체 혈관 내피 세포의 증식과 이동성, 침윤 및 모세혈관 형성 억제능을 평가함으로서 혈관신생 억제 활성에 대하여 조사하였다. 매실 부탄올 분획물은 높은 항산화능을 가지고 있는 폴리페놀(12.81 mg GAE/g)과 플라보노이드(28.4 mg QE/g)를 함유하고 있다. 매실 부탄올 분획물은 25-200 ㎍/ml의 농도에서 인체 혈관 내피 세포의 증식을 억제하지 않았으며 정상 세포에 독성을 나타내지 않았다. 또한 매실 부탄올 분획물은 인체 혈관 내피 세포에서 VEGF에 의한 이동성, 침윤 및 모세혈관 형성을 억제하였다. 이 결과는 매실 부탄올 분획물이 VEGF에 의해 유도된 인체 혈관 내피 세포의 혈관 신생활성을 억제하는 것을 시사하였다. 따라서 본 연구는 매실 부탄올 분획물이 혈관 신생 억제를 위한 잠재적인 치료제로서의 가능성이 있음을 확인하였다.
본 연구는 복분자의 수용성 추출물(RCE) 이 배양된 제대정맥내피세포에서 내피세포 NO 합성 효소(eNOS)의 발현과 활성에 미치는 효과를 연구하고, 이러한 RCE의 효과가 어떤 신호전달 과정을 거치는지를 밝히기 위한 것이다. 연구에 따르면 RCE가 제대정맥내피세포에서 NO의 생성을 증가시키는데 이는 iNOS 보다는 eNOS의 활성화에 의한 것임을 이들의 특이 억제제를 사용한 실험으로 확인할 수 있었다. 나아가 eNOS에 의한 NO 생성 증가는 이 효소의 활성 증가뿐만 아니라 mRNA 수준에서의 발현증가에도 기인함을 확인할 수 있었다. PKC-특이억제제인 RO-317549는 RCE에 의한 NO 생성의 증가에 별다른 영향을 주지 않았으나, 에스트로젠 수용체-특이 억제제인 Tamoxifen, ERK-특이 억제제인 PD98059와 PI3K/Akt-특이 억제제인 LY-294002는 제대정맥내피세포에서 RCE에 의해 증가된 NO 생성을 억제하였으며, 이는 두 저해제가 eNOS의 활성형인 pSer1177의 양을 감소시키며, 특히 PD98059는 비활성형인 pThr495의 양도 증가시키기 때문임을 알 수 있었다. 이상의 결과로써, 복분자 수추출물은 사람의 제대정맥내피세포에서 NO 생성을 증가시키며, 이는 eNOS의 발현을 증가시킬 뿐만 아니라, ERK와 PI3K/Akt의 신호전달과정을 거쳐서 eNOS의 활성도 증가시키기 때문임을 확인할 수 있었다.
혈관 신생은 암의 성장 및 전이뿐만 아니라 염증, 관절염, 건성, 동맥경화 등의 병적인 진행에 주요한 역할을 하며, 혈관신생 억제를 통한 암의 치료를 시도하는 연구들이 활발하게 진행되고 있다. 혈관 신생 시 내피세포의 증식, 이동을 유도하는 활성화 과정이 필수적으로 일어나는 것으로 알려져 있다 본 연구에서는 in vitro에서 내피세포를 배양하여, 각종 growth factor가 풍부한 배지에서 활성화 시켰을 때, 그렇지 않는 세포들과의 유전자 발현 형태를 비교 조사하였다. HUVEC을 70∼80% cofluency로 배양시킨 후에 endothelial cell growth supplement (ECCS), 20% fetal bovine serum, heparin이 첨가된 Ml99 배지에서 13 시간 활성화시킨 세포(AHUVEC)와 대조군 세포(RHUVEC)로부터 분리한 total RNA로부터 CDNA를 제작하였고, 이것을 18,864 개의 유전자가 올려져있는 인간 올리고 칩과 hybridization 반응을 시켰다. 반응된 유전자를 이용하여 random clustering분석을 실시한 결과, 활성화 시켰던 HUVEC과 그렇지 않은 HUVEC으로 dendrogram 상에서 두개의 subgroup으로 나뉘어 지는 것을 확인할 수 있었다. 최소 2배 이상 발현 변화가 있는 유전자 122종이 활성화 시켰던 HUVEC으로부터 추출되었다. 이중에서 기능이 알려진 32 개의 유전자는 활성화시킨 HUVEC에서 발현이 증가하였고, 38 개의 유전자 발현은 감소하였다. 흥미롭게도 세포 증식과 이동, 염증, 면역반응에 관련한 유전자의 발현이 증가된 반면에 세포 흡착과 혈관 조직과 기능에 관련한 유전자의 발현이 감소된 것이 관찰되었다. 예상외로 규명이 잘된 혈관신생 인자와 관련한 유전자들의 발현에는 크기 차이를 보이지 않았으나, Eph-B4의 발현은 약 4 배 감소된 것으로 관찰되었다 또한, 2배 이상 발현에 차이를 보이고 기능이 알려져 있지 않은 유전자 52종이 발견되었다. 따라서, 이러한 연구 결과로부터 새로운 혈관 표적 물질 개발에 대한 기회가 제공될 수 있을 것이라 사료된다.
Aqueous extraction of Citrus unshiu peels (AECUP) is mainly comprised with pro-angiogenichesperidin and narirutin. In this study, we report approaches to increasing the yields of extracted hesperidin and narirutinfrom Citrus unshiu peels using proper solvents. Significantly improved yields of both compounds were obtained using methanol and dimethyl sulfoxide (DMSO) compared to acetonitrile, ethyl acetate, ethanol, and isopropyl alcohol. Especially, effect of DMSO was by far the better of the two solvents in extraction of hesperidin. In addition, the DMSO extracted hesperidin significantly induced the pro-angiogenic effects of human umbilical vein endothelial cells (HUVECs) and markedly up-regulated phosphorylation of the ERK1/2 signaling pathway. These results demonstrate that pro-angiogenic inducer; hesperidin and narirutin can be simply, easily, and effectively extracted from Citrus unshiu peels.
This study was conducted to investigate antioxidant activity and anti-immunological inflammatory effect of Allium victorialis subsp. platyphyllum extracts (AVPEs). Antioxidant activities of AVPEs were determined by free radical scavenging assay and reducing power test. Leaf-part extract had comparatively better antioxidant activity than other-part extracts. Antioxidant activity of extracts had protective effect for human umbilical vein endothelial cells (HUVECs) against superoxide anions secreted from activated neutrophils. Also, we observed AVPEs had inhibitory effects on the adherence of monocytic THP-1 to HUVEC monolayer to the basal level. Inhibitory effect on cell adhesion was caused by suppression of tumor necrosis factor-${\alpha}\;(TNF-{\alpha})-upregulated$ expression of vascular cellular adhesion molecule-1 (VCAM-1) and E-selectin in HUVECs. From these results, we expect to support the evidence of anti-immunological inflammatory effects of Allium victorialis subsp. platyphyllum (AVP) as a Korean traditional pharmaceutical.
Interleukin-2 plays a significant role in T cell proliferation. Here, we report the role of IL-2 in angiogenesis. IL-2 increased the ROS level and phosphorylation of Akt in human umbilical vein endothelial cells (HUVECs). IL-2 increased angiogenesis in an animal model and tube formation in HUVECs. The effect of IL-2 on angiogenesis and tube formation was mediated by ROS and Akt. This is the first report that IL-2 promotes angiogenesis.
Vascular inflammatory process has been suggested to play a key role in initiation and progression of atherosclerosis, a major complication of diabetes mellitus. Thus, in this study, we attempted to determine whether three structurally related polyphenols found in the Chinese herb Huang Qui, namely baicalin, baicalein, and wogonin, can suppress vascular inflammatory processes induced by high glucose (HG) in human umbilical vein endothelial cells (HUVECs) and mice. Data showed that HG induced markedly increased vascular permeability, monocyte adhesion, expressions of cell adhesion molecules (CAMs), formation of reactive oxygen species (ROS) and activation of nuclear factor (NF)-κB. Remarkably, all of the above mentioned vascular inflammatory effects of HG were attenuated by pretreatment with baicalin, baicalein, and wogonin. Vascular inflammatory responses induced by HG are critical events underlying development of various diabetic complications, therefore, our results suggest that baicalin, baicalein, and wogonin may have significant therapeutic benefits against diabetic complications and atherosclerosis. Objectives : The purpose of this study is to evaluate the wound healing potential of Glycyrrhiza uralensis extract. Methods : Free radical scavenging activity tests for DPPH, peroxynitrite (ONOO) and hydroxyl radical (${\cdot}OH$) and total phenolic contents of Glycyrrhiza uralensis extract were conducted. Tube formation assay was performed using human umbilical vein endothelial cells (HUVECs). Results : The results showed that Glycyrrhiza uralensis extract exerted inhibitory effects on ONOO and ${\cdot}OH$. Tube formation in HUVEC was increase in a dose dependent manner. Conclusions : These results show the potential to promote the wound healing process by Glycyrrhiza uralensis extract.
